Influence of Site-Directed Mutagenesis on Protein Assembly and Solubility of Tadpole H-chain Ferritin

  • In order to understand the influence of ferroxidase center on the protein assembly and solubility of tadpole ferrin, three mutant plasmids, pTH58K, pTH61G, and pTHKG were constructed with the aid of site-directed mutagenesis and mutant proteins were produced in Eshcerichia coli. Mutant ferritin H-subunits produced by the cells carrying plasmids pTH58K and pTHKG were active soluble proteins, whereas the mutant obtained from the plasmid pTH61G was soluble only under osmotic stress in the presence obtained from the plasmid pTH61G was soluble only under osmotic stress in the presence of sorbitol and betaine. Especially, the cells carrying pTH61G together with the plasmid pGroESL harboring the molecular chaperone genes produced soluble ferritin. The mutant ferritin H-subunits were all assembled into ferritin-like holoproteins. These mutant ferritns were capable of forming stable iron cores, which means the mutants are able to accumulate iron with such modified ferroxidase sites. Further functional analysis was also made on the individual amino acid residues of ferroxidase center.

Development of Cermet Cutting Tool by Powder Injection Molding

  • Chip breaker of cutting tool is an important feature to enhance cutting performance. Powder injection molding process was used to produce a triangular-shape cermet grooving insert which has three chip breakers. Attrition milled cermet powders were mixed with wax-based binder system in continuous twin screw extruder. Three-plate injection mold with slide cores was used to produce injection-molded parts. After molding, solvent and thermal debinding was carried out. Sintering was conducted in a batch furnace with a graphite heater. The sintered parts satisfy the requirements of dimensional tolerances and material properties.

  • TiC-and Ti(C,N)-based cermets are excellent in semi-and final finishing of work piece during cutting operations. Typical microstructure of the cermets is a core/rim structure. The undissolved Ti(C,N) cores contribute to their high hardness while the rim phases, (Ti,M1,M2)(C,N)-type solid solutions, play great roles in enhancing the toughness. In this paper, various ultrafine pre-mixed MeC-Ni powders were synthesized and the powders were sintered or hot pressed after mixing in order to control the size and volume fractions of core and rim phases in the system. This paper will present the factors determining the microstructure along with mechanical properties.

Historical Record of Alexandrium spp. (Dinophyceae) in Southern Coastal Area of Korea

  • To investigate the historical record of Alexandrium spp. in southern coastal areas of Korea, two sediment cores were collected from Gamak Bay and Yeoja Bay. Germination experiments revealed that the ellipsoidal Alexandrium cysts isolated from Gamak Bay and Yeoja Bay are morphologically identical to a toxic dinoflagellate A. tamarense. The ellipsoidal Alexandrium cysts in Yeoja Bay appeared from 30 to 32 cm depth upwards (ca. 1980s), and their concentration increased around 10 to 12 cm depth (mid-1990s). Similarly, cyst concentration in Gamak Bay also increased from 40 to 44 cm depth (ca. 1990s). These results coincide with the reports of Paralytic Shellfish Poisoning caused by A. tamarense in 1980s and 1990s along the southeast coast of Korea.

Monte-Carlo Simulation to the Color Distribution within Galactic Globular Clusters

  • According to the CCD photometric studies, the color distributions of globular clusters with collapsed cores, which are characterized by a power law cusp in thier surface brighness pronto, become bluer toward their centers, but this is not the case in the flat core clusters which are fit by the King model. To test the statistical implication of the color distribution within globular clusters, we built the sample dusters which follows the surface brightness pofile of the King model and power law cusp profile with the Sandage's standao luminosity function for M3 and the Salpter's initial mass functions. On the results from simulations based on the uniform random number generation the color gadients within globualr dusters mar be not likely to come from the statistical random distributions of stars but from the dynamical process on the cluster evolution.

  • In this paper, Take two perment magnets and three iron bars and wind copper wire around one or the bars. This paper demonstrate that the method or calculation yields the right results with MAXWELL-2D program package for computing magnetic field, also indicate the value to which the current in the coil has to be limited to prevent saturation of the iron or the cores. The Finally procedures for design of LDM are discussed.

  • This paper proposes a new algorithm of underground cable fault location based on the analysis of distributed parameter circuit. The proposed method firstly makes voltage and current equations for each of cores and sheathes respectively, and then establishes an equation of the fault distance according to the analysis of the fault conditions. Finally the solution of this equation is calculated by Newton-Raphson iteration method. The effectiveness of this proposed algorithm has been proven through PSCAD/EMTDC simulations.

  • In the field of composite structures, the use of carbon tube for the confinement of concrete has been arisen since 1990's. However, experimental and analytical studies were limited to those of reinforced concrete and concrete filled steel tube. The carbon tube provides excellent confinement capabilities for concrete cores, enhancing compressive strength and ductility of concrete significantly. The carbon tube has high tensile strength, light weight, corrosion immunity and high fatigue strength properties. Since carbon fiber is an anisotropic material, carbon tube could be optimized by adjusting the fiber orientation, thickness and the number of different layers. In this study, both experimental and analytical studies of axial and lateral behavior of full-scale CFCT (Concrete Filled Carbon Tube) columns subjected to monotonic axial load were carried out using Drucker-Prager theory. And, based on comparison results between experiment results and analytical results, k factor estimation was proposed for effective analysis.

Benchmarks of subcriticality in accelerator-driven system at Kyoto University Critical Assembly

  • Basic research on the accelerator-driven system is conducted by combining $^{235}U$-fueled and $^{232}Th$-loaded cores in the Kyoto University Critical Assembly with the pulsed neutron generator (14 MeV neutrons) and the proton beam accelerator (100 MeV protons with a heavy metal target). The results of experimental subcriticality are presented with a wide range of subcriticality level between near critical and 10,000 pcm, as obtained by the pulsed neutron source method, the Feynman-${\alpha}$ method, and the neutron source multiplication method.

Ground Stability Assessement for the Mining Induced Subsidence Area (지하공동에 의한 지표침하지역의 지반안정성 평가)

  • Surface subsidence is one of the problems caused by mined out caverns. Depending on the geologic conditions and mining methods, subsidence can occur in various forms. This report describes the ground stability assessment for the mining induced subsidence area where unfilled caverns still exist abandoned. Geologic features which could affect the stability of the ground were investigated and all the possible geophysical methods were employed to obtain data that could explain the state of the ground in question. Basic rock tests were conducted from the drill cores and rock mass classification was performed by core logging and borehole camera investigation. Numerical analyses were carried out to predict the ground stability using data obtained by various investigations. The result could have been more reliable if in-situ stress were measure and reflected in the numerical analysis.
