Cloning and Expression of Phytochelatin Synthase 1 Gene from Rhizophora stylosa Exposed to Cadmium and Copper (카드뮴과 구리에 노출된 Rhizophora stylosa 의 phytochelatin synthase 1 유전자 클로닝 및 발현)

  • The mangrove ecosystems have the capacity to act as a sink of heavy metals entering aquatic ecosystems. Despite their potential exposure to metal contaminated sediments, mangroves appear to be highly tolerant to heavy metals. In this study, we cloned metal tolerance gene from mangrove plant. Using CTAB method, RNA were isolated from leaves and root tissue of Rhizophora stylosa habitated at Weno island in Micronesia Chuuk lagoon using CTAB method and phytochelatin synthase 1 (PCS1) gene was cloned using gene specific primers. Expression of PCS1 gene was increased 1.91 fold and 2.72 fold in mangrove propagules exposed to 100 ppb Cd and 10 ppb Cu, respectively. These results indicate that expression of PCS1 gene are promising tools for health assessment of mangrove ecosystem.

Study on the Effect of Corrosion Gas and Acidic Falling Materials for Conservational Wax on the Copper based Sculpture (동(銅)이 주종을 이루는 조형물 보존용 왁스의 산성강하물 및 부식 가스에서의 영향성 연구)

  • The influence of noxious corrosive gas, which is the product of fossil fuel, and the acidic descent material, which becomes the reason for such reaction, on wax which protects the cultural assets, and the habitat factor were researched. It could be viewed that corrosion occurred rapidly due to the acidic descent material, which have the long staying on the material, rather than harmful corrosive gas. Also, it could be figured out that such corrosion began from whitening phenomenon of coated wax. As the acidity increased, more whitening phenomenon occurred, and eventually, it could be found out that whitening effect began due to the acidic descent material, corrosive gas and various 0.1 M acid solution, which lead to the exposure of internal metal, and corrosion occurred. The wax with the greatest resistance to the acidity due to its least changing aspect seemed to be Renaissance wax, except sulfuric acid.

Monitoring of Heavy Metal Contaminations using Feathers of Feral Pigeons Columba livia in Seoul (서울지역에 서식하는 집비둘기 Columba livia의 깃털을 이용한 중금속오염 모니터링)

  • To monitor the heavy metal exposure conditions of Feral Pigeons Columba livia, we collected them at commercial area, industrial area, park area and resident area in Seoul. In feathers, iron concentrations were significantly different among habitats(ANOVA, p<0.05), and were the highest at park area. Manganese concentrations were relatively higher at industrial and park area than other areas(ANOVA, p<0.05). In case of copper levels were the significantly highest at park area(ANOVA, p<0.05). Zinc, lead and cadmium had no difference among habitats. Correlation between livers and feathers concentrations were highly significant to copper(r²=0.983, p<0.01) and cadmium(r²=0.632, p<0.01), but iron, zinc, manganese and lead were not significantly different.

The Evaluation of the Preservative Treated Plywood Produced by Factory Processing (야외사용을 목적으로 공장라인에서 생산한 방부합판의 성능평가)

  • To make up original defects of the wooden materials for decks, and to supply the wooden material for outdoor, we fabricated preservative treated plywood(PTP). Copper azole (CUAZ-1) preservative was treated with a normal full-cell process. Bond Strength of PTP was not affected after the preservative treatment. The anti-fungal efficiency and dimensional stability were obtained from PTP. A little discoloration of the surface was detected, but the dimensional change or peel bonded area off were not observed after accelerated weathering test. Although some strength of PTP was reduced after 17 months of field exposure, the PTP should be applicable for outdoor applications.

Risk Assessment of Pesticide for Earthworms (농약의 지렁이에 대한 위해성 평가)

  • To assess the risk of pesticides on earthworm, the acute toxicities of 10 pesticides were investigated and their toxicity exposure ratios(TERs) were calculated. As the TERs of paraquat dichloride and pendimethalin were more than 100, their risks were rated negligible. Risk of benfuracarb, cadusafos, chlorpyrifos-methyl, endosulfan, isazofos and parathion which have TERs of $10\sim100$ were rated low. However, risk of imidacloprid and phorate which have TER of less than 10 were estimated highly to need a reproduction study. Earthworms were exposed to twenty two pesticides including dazomet 98% GR having PECs of more than $5mg{\cdot}kg^{-1}$ in artificial soil at standard and double dose for 14 days. All the earthworms exposed to dazomet 98% GR and metam-sodium 25% SL were died to show their high risk, while no serious adverse effects were observed in the soil treated with 15 pesticides, calcite 95% WP, calcium polysulfide 36% CF, chlorothalonil 75% WP, daminozide 85% WP, dichlonil 6.7% GR, etridiazole 25% EC, fosetyl-Al 80% WP, glyphosate 41 % SL, hymexazol 30% SL, iprodione 50% WP, machine oil 95% EC, mancozeb 75% WP, propineb 70% WP, terbuthylazine 80% WP and triazophos 40% EC. In case of thiophanate-methyl 70% WP, copper hydroxide 77% WP, dimethoate 46% EC, tolclofos-methyl 50% WP and propamocarb hydrochloride 67% SL, any effect did not show clearly, suggesting an additional subchronic toxicity study. The risk of thiophanate-methyl 70% WP to earthworm was estimated high, considering its subchronic effect, while effects of copper hydroxide 77% WP, dimethoate 46% EC, tolclofos-methyl 50% WP and propamocarb hydrochloride 67% SL to earthworms were negligible, considering no adverse effects in subchronic tests.

Effect of Copper on Lethal Dose and Pupation of Aedes albopictus (Diptera: Culicidae) (흰줄숲모기, Aedes albopictus (Diptera: Culicidae)의 치사농도 및 용화에 대한 구리의 영향)

  • The mosquito, Aedes albopictus is important vector of humen phatogen such as dengue fever. And Aedes albopictus has widly distribution in oriental regions. This study was conducted to investigate the biological effects of cooper on the development of the Asian tiger mosquitoes, Aedes albopictus (Skuse). Aedes albopictus was affected with pupation ratios, mortalities, and wing lengths of the 3th and 4th instars by exposure to different concentration $CuCl_2$ (0.0, 2.5, 25.0 and 50.0 ppm) for 24 h and 48 h depending on dose and exposure period. The lowest $LC_{50}$ (median lethal concentration) value of $CuCl_2$ was 18.1 ppm at the 3rd instars for 48 h exposure. The pupation ratio of the larvae exposed at 50.0 ppm for 48 h was 14% which was much lower than those exposed to other treatment groups Changes of adult wing length showed significant decreased by Cu treatment. In conclusion, the most prominent effects of high concentration and longer exposure period were to reduce in the survival and pupation rates of larvae and wing length of Ae. albopictus.

Dose and Image Quality Analysis According to The Type of Composite Additional Filter (복합 부가필터 종류에 따른 선량 및 화질 분석)

  • In this study, four types of composite added filtration (aluminum, nickel, copper, and zinc) were combined for each thickness to evaluate dose reduction and optimal images due to X-ray attenuation. To evaluate dose and image quality. X-ray generators, Dose Area Product(DAP) and ICY programs with RQR9 standard quality were used. In the image quality evaluation element (PSNR, RMSE, SSIM), only images with PSNR value of 30 dB or more were analyzed. As a result, the best combination in dose evaluation was 3 mmAl + 0.6 mmNi (0.16µGy㎡), and the best filter in image quality evaluation was 0.9 mmAl (PSNR 34.24dB, RMSE 79.52, SSIM 0.24). In this study, the dose aspect and the image quality aspect are mentioned, So it is considered that further studies on patient's exposure dose and optimal image will be needed in the future.

  • The fact that hazardous metals do not bio-degrade or bio-deteriorate translates to long-lasting environmental effects. In the context of evidently rapid global industrialization, this ought to warrant serious caution, particularly in developing countries. In the Republic of Fiji, a developing country in the South Pacific, several different environmental studies over the past 20 years have shown levels of lead, copper, zinc and iron in sediments of the Suva Harbor to be 6.2, 3.9, 3.3 and 2.1 times more than the accepted background reference levels, respectively. High levels of mercury have also been reported in lagoon shellfish. These data inevitably warrant thorough assessment of the waste practices of industries located upstream from the estuaries, but in addition, an exposure and health impact assessment has never been conducted. Relevant government departments are duty-bound, at least to the general public that reside in and consume seafood from the vicinities of the Suva Harbor, to investigate possible human effects of the elevated hazardous metal concentrations found consistently in 20 years of surface sediment analysis. Furthermore, pollution of the intermediate food web with hazardous metals should be investigated, regardless of whether human effects are eventually confirmed present or not.

A Study on the Heavy Metal Content of Permanent Wave Products (퍼머넌트 웨이브제의 중금속 함량에 관한 연구)

  • This study is to measure the heavy metal content of permanent wave products which on marketing correctly as estimating the extent of exposure by a hair permanent wave scientifically. We would like to prevent an affair from arising health obstruction as to the heavy metal who is using those and also show the basic data for proposing the new standard. The results were as follows.: in case of the average heavy metal content for a wave type thioglycol acid ingredient includes 1.61ppm(Pb), 0.03ppm(Cd), 0.05ppm(Ni), 0.27ppm(Mn), 0.82ppm(Cu) and those were recognized the significant gap between products all the heavy metals. In case of a cysteine acid ingredient includes 0.86ppm(Pb), 0.01ppm(Cd), 0.05ppm(Ni), 0.20ppm(Mn) and 0.66ppm(Cu) and those were recognized the significant gap between products except a nickel. Straight type of permanent wave reductant includes 2.11ppm(Pb), 0.01ppm(Cd), 0.27ppm(Ni), 0.66ppm(Mn), 2.53ppm(Cu) and those were recognized the significant gap between products all the heavy metals. Permanent wave reducing agent includes 1.43ppm(Pb), 0.01ppm(Cd), 0.09ppm(Ni), 0.66ppm(Mn), 0.75ppm(Cu) and those were approved the significant gap between products except a cadmium. Exposure level of the heavy metal contents per onetime permanent waving were 242.3ppm(Pb), 2.5ppm(Cd), 17.7ppm(Ni), 89.0ppm(Mn), 174.7ppm(Cu).

Wood Decay Properties of Difference MCQ Retention Level (MCQ 보유량에 따른 목재의 부후 특성)

  • In this study, MCQ (Micronized copper Quat) which was copper-based wood preservative, was investigated on decay properties by difference of the its infiltration volume. After pressure-treatment with different MCQ concentration, test specimens (Japanese Red Pine) were exposed by a brown-rot fungus (Fomitopsis palustris). At the end of the 12 weeks exposure to the fungus, untreated specimen was showed the mass loss of more than 35%, and the value of preservative effectiveness of MCQ by indoor decay was $3.99{\pm}0.42kg/m^3$. Therefore, MCQ should be pressure-injected at least $3.99kg/m^3$. The three dimensional of the specimens were observed by using the light microscope and scanning electron microscope, Untreated and treatment specimens with low concentration generally had underwent serious decays and had a lot of fungal hyphae. Howere, the formation of bore hole by hyphae were not observed. Mass loss and decay properties of specimens were affected by amount of MCQ injection. Therefore, it is necessary to the review of the adequacy of the MCQ injection amount in domestic environments.
