Change in the Binding Cooperativity of Ethidium with Calf Thymus DNA, Induced by Spermine Binding (Spermine에 依한 Ethidium의 Calf Thymus DNA와의 結合 Cooperativity 變化)

  • At the spermine concentration to cover the number of the binding site of spermine 0.016 per nucleotide, the Hill coefficient of the ethidium binding to the calf thymus DNA was 1.7, while the value was 0.38 in the absence of the spermine. On the basis of the data, together with other present data on the viscometric titration of the DNA with spermine and anomalous absorbance-temperature profile at 260nm and viscosity-temperature profile, it can be speculated that allosteric propagation of the conformational transition induced by the binding of the spermine may be involved in the monomolecular collapse of the DNA to a condensed structure.

  • An anomalous absorbance-temperature profile of calf thymus DNA, having a trough preceding the initiation of the melting, occurs at the spermine concentration, where the DNA collapses into a compact structure. The cooperativity, enthalpy, and the midpoint of the phase transition to the trough region are more sensitive to ethidium bromide than those of the Tm region. As the concentration of ethidium bromide added is increased, the peak size of the trough is decreased, while the Tm remains essentially constant.

Surface Tentiometric Studies on the Interaction of Anionic Polyelectrolytes with Cationic Surfactants

  • The interaction of cationic surfactants, n-alkyltrimethylammonium bromide ($C_nTAB$; n = 12, 14, 16) with anionic polyelectrolyte, poly(styrenesulfonate) (PSS) has been studied by surface tension measurement. In the absence of added salt, the cationic surfactants bind to PSS quantitatively up to ca. 60% coverage of anionic sites of the polyanion and the complexes were surface inactive. Further binding of the surfactant cations on PSS caused a sharp conformational transition of the surfactant/ PSS complexes to surface active complexes and accompanied precipitation. The binding showed a biphasic behavior in the presence of NaCl and cooperativity of the binding became less as the concentration of NaCl increased. Binding of the cationic surfactants on poly(vinylsulfonate) also showed the biphasic behavior and the cooperativity of the binding was much less even in the absence of NaCl. The binding of surfactant to PSS provided hydrophobic environment to solubilized pyrene and reduced the viscosity of the solution greatly even at surfactant concentrations well below cmc. This study indicated that the surfactant bound to PSS up to $60{\%}$ coverage of PSS sites are present as surfactant aggregates which are wrapped up with PSS chains, and hydrophobic interaction is an important factor in the binding of the surfactants to PSS.

Kinetic Characterization of an Iron-sulfur Containing Enzyme, L-serine Dehydratase from Mycobacterium tuberculosis H37Rv (Mycobacterium tuberculosis H37Rv로부터 유래된 철-황 함유 효소인 L-세린 탈수화효소의 동력학적 특성)

  • L-Serine dehydratase (LSD) is an iron-sulfur containing enzyme that catalyzes the conversion of L-serine to pyruvate and ammonia. Among the bacterial amino acid dehydratases, it appears that only the L-serine specific enzymes utilize an iron-sulfur cluster at their catalytic site. Moreover, bacterial LSDs are classified into four types based on structural characteristics and domain arrangement. To date, only the LSD enzymes from a few bacterial strains have been studied, but more detailed investigations are required to understand the catalytic mechanism of various bacterial LSDs. In this study, LSD type II from Mycobacterium tuberculosis (MtLSD) H37Rv was expressed and purified to elucidate the biochemical and catalytic properties using the enzyme kinetic method. The L-serine saturation curve of MtLSD exhibited a typically sigmoid character, indicating an allosteric cooperativity. The values of $K_m$ and $k_{cat}$ were estimated to be $59.35{\pm}1.23mM$ and $18.12{\pm}0.20s^{-1}$, respectively. Moreover, the plot of initial velocity versus D-serine concentration at fixed L-serine concentrations showed a non-linear hyperbola decay shape and exhibited a competitive inhibition for D-serine with an apparent $K_i$ value of $30.46{\pm}5.93mM$ and with no change in the $k_{cat}$ value. These results provide insightful biochemical information regarding the catalytic properties and the substrate specificity of MtLSD.

  • The synthesis and characterization of third- and fifth-generation poly(propylene imine) dendrimers terminated with imidazole moieties is reported. Functionalization was achieved using simple peptide coupling reagents. These materials were characte rized by MALDI-MS, NMR, and titration. The use of these endgroup-functionalized dendrimers as catalysts for the hydrolysis of 2,4-dinitrophenyl acetate is described. Molecular simulations provide a basis for interpreting the catalytic data.

  • The reaction of [(2-N,N-dimethylaminomethyl) pheny] methylvinychlorosiane with t-BuLi in hexane solvent gave dimers, five isomeric 1,3-disilacyslobutanes which were weparated and charaterized. In trapping experiments with various trapping agents, no corresponding silene-trapping aduct was observed. We suggest that more important species for the formation of five isomeric dimers might be the zwitterionic species generated by virtue of intramolecular donor atom rather than the silene.

Cooperativity of ${\alpha}$- and ${\beta}$-Subunits of Group II Chaperonin from the Hyperthermophilic Archaeum Aeropyrum pernix K1

  • ${\alpha}$ and ${\beta}$-subunits (ApCpnA and ApCpnB) are group II chaperonins from the hyperthermophilic archaeum Aeropyrum pernix K1, specialized in preventing the aggregation and inactivation of substrate proteins under conditions of transient heat stress. In the present study, the cooperativity of ${\alpha}$- and ${\beta}$-subunits from the A. pernix K1 was investigated. The ApCpnA and ApCpnB chaperonin genes were overexpressed in E. coli Rosetta and Codonplus (DE3), respectively. Each of the recombinant ${\alpha}$- and ${\beta}$-subunits was purified to 92% and 94% by using anionexchange chromatography. The cooperative activity between purified ${\alpha}$- and ${\beta}$-subunits was examined using citrate synthase (CS), alcohol dehydrogenase (ADH), and malate dehydrogenase (MDH) as substrate proteins. The addition of both ${\alpha}$- and ${\beta}$-subunits could effectively protect CS and ADH from thermal aggregation and inactivation at $43^{\circ}C$ and $50^{\circ}C$, respectively, and MDH from thermal inactivation at $80^{\circ}C$C and $85^{\circ}C$. Moreover, in the presence of ATP, the protective effects of ${\alpha}$- and ${\beta}$-subunits on CS from thermal aggregation and inactivation, and ADH from thermal aggregation, were more enhanced, whereas cooperation between chaperonins and ATP in protection activity on ADH and MDH (at $85^{\circ}C$) from thermal inactivation was not observed. Specifically, the presence of both ${\alpha}$- and ${\beta}$- subunits could effectively protect MDH from thermal inactivation at $80^{\circ}C$ in an ATP-dependent manner.

Purification and Some Properties of Arginine Deiminase in Euglena gracilis Z (Euglena gracilis Z로부터 Arginine Deiminase의 정제 및 그의 특성)

  • In Euglena gracilis arginine deiminase was located in the mitochondrial matrix. The highly purified enzyme required $Co^{2+}$ for the enzyme reaction with the $K_m$ value of 0.23 nM, and its optimum pH was 9.7 to 10.3. The molecular weight of the native enzyme protein was 87,000 by gel filtration, and SDS-acrylamide gel electrophoresis showed that the enzyme consisted of two identical subunits with a molecular weight of 48,000. Euglena arginine deiminase was inhibited by sulfhydryl inhibitors, indicating that a sulfhydryl group is involved in the active center of the enzyme. It exhibited negative cooperativity in binding with arginine. $L-{\alpha}-amino-{\beta}-guanidino-propionate$, D-arginine, and L-homoarginine strongly inhibited the enzyme while ${\beta}-guanidinopro-pionate$, ${\gamma}-guanidinobutyrate$, and guanidinosuccinate did not. Considerable inhibition was also observed with citrulline and ornithine. We discuss the effects of the unique properties of the Euglena arginine deiminase on the regulation of arginine metabolism in this protozoon.

A Case Study on the Students' Attitude toward User-Created Contents in Discretionary Classes of Elementary School (UCC 활용교육에 관한 학습자 태도 연구 - 초등학교 재량교육 사례를 중심으로 -)

  • User-created content(UCC) is on the rapid increase both in terms of quantity and the quality of sharing and enjoying each others. However, not much research has been done on school's adoption of this innovative contents for educational purposes. The aim of this case study is to identify the determinants of students' attitude toward using UCC in elementary school classes. This study employs the perceived educational value, ease of use, social influence, fun, computer self-efficacy, cooperativity and concentration as the antecedents of attitude, and collected 246 survey responses from 5th and 6th grade students. The results indicate that the perceived educational value, ease of use and socal influence have significant impact on the attitude toward using UCC. It was also found that the cooperativity, fun and ease of use determine the level of perceived educational value, and that the computer self-efficacy, fun and concentration influence the perceived ease of use. Some useful suggestions, concerning the utilizing UCC for educational purpose in elementary school classes, are also presented.

Characteristics of a Novel Acinetobacter sp. and Its Kinetics in Hexavalent Chromium Bioreduction

  • Cr-B2, a Gram-negative hexavalent chromium [Cr(VI)] reducing bacteria, was isolated from the aerator water of an activated sludge process in the wastewater treatment facility of a dye and pigment based chemical industry. Cr-B2 exhibited a resistance for 1,100 mg/l Cr(VI) and, similarly, resistance against other heavy metal ions such as $Ni^{2+}$ (800 mg/l), $Cu^{2+}$ (600 mg/l), $Pb^{2+}$ (1,100 mg/l), $Cd^{2+}$ (350 mg/l), $ZN^{2+}$ (700 mg/l), and $Fe^{3+}$ (1,000 mg/l), and against selected antibiotics. Cr-B2 was observed to efficiently reduce 200 mg/l Cr(VI) completely in both nutrient and LB media, and could convert Cr(VI) to Cr(III) aerobically. Cr(VI) reduction kinetics followed allosteric enzyme kinetics. The $K_m$ values were found to be 43.11 mg/l for nutrient media and 38.05 mg/l for LB media. $V_{max}$ values of 13.17 mg/l/h and 12.53 mg/l/h were obtained for nutrient media and LB media, respectively, and the cooperativity coefficients (n) were found to be 8.47 and 3.49, respectively, indicating positive cooperativity in both cases. SEM analysis showed the formation of wrinkles and depressions in the cells when exposed to 800 mg/l Cr(VI) concentration. The organism was seen to exhibit pleomorphic behavior. Cr-B2 was identified on the basis of morphological, biochemical, and partial 16S rRNA gene sequencing chracterizations and found to be Acinetobacter sp.
