FA/Mel@ZnO nanoparticles as drug self-delivery systems for RPE protection against oxidative stress

  • Drug self-delivery systems can easily realize combination drug therapy and avoid carrier-induced toxicity and immunogenicity because they do not need non-therapeutic carrier materials. So, designing appropriate drug self-delivery systems for specific diseases can settle most of the problems existing in traditional drug delivery systems. Retinal pigment epithelium is very important for the homeostasis of retina. However, it is vulnerable to oxidative damage and difficult to repair. Worse still, the antioxidants can hardly reach the retina by non-invasive administration routes due to the ocular barriers. Herein, the targeted group (folic acid) and antioxidant (melatonin) have been grafted on the surface of ZnO quantum dots to fabricate a new kind of drug self-delivery systems as a protectant via eyedrops. In this study, the negative nanoparticles with size ranging in 4~6 nm were successfully synthesized. They could easily and precisely deliver drugs to retinal pigment epithelium via eyedrops. And they realized acid degradation to controlled release of melatonin and zinc in retinal pigment epithelium cells. Consequently, the structure of retinal pigment epithelium cells were stabilized according to the expression of ZO-1 and β-catenin. Moreover, the antioxidant capacity of retinal pigment epithelium were enhanced both in health mice and photic injury mice. Therefore, such new drug self-delivery systems have great potential both in prevention and treatment of oxidative damage induced retinal diseases.

Effect of Controlling Exhaust Valve Timing on Engine Efficiency in LIVC and EIVC States in a 2-Cylinder Small Turbo Gasoline Engine (2기통 소형 터보가솔린엔진에서 배기 밸브 타이밍 제어에 따른 LIVC, EIVC 상태에서의 엔진 효율 영향)

  • This study examines whether engine fuel efficiency is improved by optimization of the exhaust valve timing in a state where the intake valve timing has been optimized in a small turbo gasoline engine that has intake cams and exhaust cams with fixed valve opening periods. When the exhaust valve is opened late, the expansion stroke is longer, and the efficiency can be improved. A 2-cylinder turbo gasoline engine with 0.8 liters of displacement and an MPI (Multi Point Injection) fuel system was used. The engine was operated at 1,500 and 3,000 rpm, and the load conditions included a partial load of 50 N·m and a high load of 70 N·m. Data was recorded as the exhaust valve timing was controlled, and this was used to calculate the efficiency of combustion using a heat release, the fuel conversion efficiency, and the pumping loss. Results and the hydrocarbon concentrations in the exhaust gas were compared for each condition. Experiment results confirmed that additional fuel efficiency improvements are possible through exhaust valve timing control at 1,500 rpm and 50 N·m. However, in other operating conditions, fuel efficiency improvements could not be obtained through exhaust valve timing control because cases where the pumping loss and fuel/air mixture slip increased when the exhaust valve timing changed and the fuel efficiency declined.

Microencapsulated basil oil (Ocimum basilicum Linn.) enhances growth performance, intestinal morphology, and antioxidant capacity of broiler chickens in the tropics

  • Objective: Microencapsulation is a technique to improve stability, bioavailability, and controlled release of active ingredients at a target site. This experiment aimed to investigate the effects of microencapsulated basil oil (MBO) on growth performance, apparent ileal digestibility (AID), jejunal histomorphology, bacterial population as well as antioxidant capacity of broiler chickens in a tropical climate. Methods: A total of 288 one-day-old female broilers (Ross 308) were randomly allocated into 4 groups (6 replicates of 12 birds), based on a completely randomized design. Dietary treatments were as follows: i) basal diet (NC), ii) basal diet with avilamycin at 10 ppm (PC), iii) basal diet with free basil oil (FBO) at 500 ppm, and iv) basal diet with MBO at 500 ppm, respectively. Results: Dietary supplementation of MBO improved average daily gain, and feed conversion ratio of broilers throughout the 42-d trial period (p<0.05), whereas MBO did not affect average daily feed intake compared with NC group. The broilers fed MBO diet exhibited a greater AID of crude protein and gross energy compared with those in other groups (p<0.05). Lactobacillus spp. and Escherichia coli populations were not affected by feeding dietary treatments. Both FBO and MBO had positive effects on jejunal villus height (VH), villus height to crypt depth ratio (VH:CD) and villus surface area of broilers compared to NC and PC groups (p<0.05). Superoxide dismutase level in the duodenal mucosa of MBO group was significantly increased (p<0.01), whereas malondialdehyde level was significantly decreased (p<0.01). Conclusion: Microencapsulation could be considered as a promising driver of the basil oil efficiency, consequently MBO at 500 ppm could be potentially used as a feed additive for improvement of intestinal integrity and nutrient utilization, leading to better performance of broiler chickens.

Effects of a Combination of Scapular Stabilization and Thoracic Extension Exercises on Respiration, Pain, Craniovertebral Angle and Cervical Range of Motion in Elementary School Teachers with a Forward Head Posture: A Randomized Controlled Trial

  • PURPOSE: This study examined the effects of an exercise program for the thoracic spine and scapula rather than the neck, which is the primary site of pain. METHODS: Thirty-two elementary school teachers with a forward head posture (FHP) were assigned randomly to either the experimental group (n = 16) or the control group (n = 16). The experimental group performed scapular stabilization exercise (SSE) and thoracic extension exercise (TEE), and the control group performed cervical self-myofascial release exercise and stretching exercise. The pulmonary functions, pain, craniovertebral angle (CVA), and cervical range of motion (CROM) were measured before the intervention and six weeks after. RESULTS: The within-group comparisons showed that the VAS and CROM (except for extension) in both groups were significantly different before and after the intervention (p < .05). The changes in the maximum inspiratory pressure (MIP), maximum expiratory pressure (MEP), forced vital capacity (FVC), forced expiratory volume at 1 sec (FEV1), and CVA were significant only in the experimental group (p < .05). The between-group comparisons showed a significant difference in the FVC, FEV1, VAS, CVA, and left lateral flexion (p < .05). CONCLUSION: The combination of SSE and TEE in the experimental group was more effective in improving the FHP and breathing ability. Moreover, the experimental group and control combination appeared to be effective in reducing pain and improving the CROM. The combination of SSE and TEE, which are exercises that do not target the cervical spine directly, was effective in improving the posture, respiration, neck pain, and CROM in elementary school teachers with FHP.

Extracts of abalone intestine regulates fat metabolism in 3T3-L1 adipocytes and high fat diet-induced zebrafish larvae

  • Obesity is a disease involving mechanisms of fat accumulation, low-grade inflammatory cytokine release, and mitochondrial dysfunction. The aim of the current study was to investigate the effects of abalone intestine extract on fat metabolism in 3T3-L1 adipocytes and high fat diet-induced zebrafish larvae. The total phenol content was highest in subcritical water extract at 210℃ (SW210) among hot water, ethanol, and subcritical water extracts of abalone intestine. In addition, SW210 of male abalone intestine (MASW210) most effectively controlled the lipid accumulation and expression of adipogenic or lipogenic regulators (PPAR-γ, C/EBPα, SREBP-1c, and FAS) in 3T3-L1 adipocytes. Likewise, in zebrafish larvae fed high fat, MASW210 significantly suppressed body weight, glucose levels, and lipid accumulation. The mRNA expression related to adipogenesis (PPAR-γ and C/EBPα), lipogenesis (SREBP-1c and FAS), inflammatory cytokines (TNF-α and IL-6), energy m/;.etabolism (AMPK, lepr, SIRT1, and adiponectin), and mitochondrial biogenesis (PGC-1α and CPT-1) were significantly regulated by treatment with MASW210. These results suggest that abalone intestine extract such as MASW210, are useful biomaterials for improving obesity and metabolic diseases.

Effect of Application Rate of a Controlled Release Fertilizer on the Changes in Medium EC and Growth of Subirrigated Vinca and Salvia (저면관수 재배에서 완효성 비료의 양이 배지의 EC 및 일일초와 살비아의 생장에 미치는 영향)

  • Plug seedlings of vinca (Catharanthus roseus L. 'Pacifica Punch') and salvia (Salvia splendens F. Sellow ex Roem & Schult 'Maestro') were transplanted into square plastic pots (145 mL volume) filled with a soilless growing medium. To determine the effect of application rate on the growing medium EC and growth of plants, 0, 0.5, 1.0, 1.5, 2.0 and 4.0 g per pot of a controlled release fertilizer (14-14-14 Osmocote, 14N-6.2P-11.6K) were mixed with the growing medium. Plants were subirrigated daily with tap water. In both vinca and salvia, growing medium EC increased as application rate was elevated. Growing medium EC was relatively constant over a whole crop period when the application rate was less than 1.5 g per pot, while it decreased throughout the experiment at higher application rates such as 2.0 to 4.0g per pot in both species. The greatest leaf area, plant height, and shoot dry weight of vinca were obtained when plants were fertilized with 2.0 to 4.0 g per pot of the fertilizer, resulting in a growing medium EC of $1.0{\sim}1.7dS{\cdot}m^{-1}$ throughout the experiment. Leaf area, shoot dry weight, and chlorophyll content of salvia increased with elevated application rates. Leaf area, shoot dry weight, and chlorophyll content of salvia were the greatest when plants were fertilized with 4.0 g per pot, resulting in growing medium EC of $1.0{\sim}4.0dS{\cdot}m^{-1}$ throughout the experiment. Plant height of salvia was the greatest when plants were fertilized with 2.0 to 4.0g per pot. Concentrations of nitrogen (N), phosphorus (P), potassium (K), magnesium (Mg), and sulfur (S) in the shoots of vinca increased, while concentration of calcium (Ca) decreased with elevated application rates. Concentrations of boron (B) and manganese (Mn) in the shoots of vinca increased as the application rate decreased.

Quantification of Temperature Effects on Flowering Date Determination in Niitaka Pear (신고 배의 개화기 결정에 미치는 온도영향의 정량화)

  • Most deciduous trees in temperate zone are dormant during the winter to overcome cold and dry environment. Dormancy of deciduous fruit trees is usually separated into a period of rest by physiological conditions and a period of quiescence by unfavorable environmental conditions. Inconsistent and fewer budburst in pear orchards has been reported recently in South Korea and Japan and the insufficient chilling due to warmer winters is suspected to play a role. An accurate prediction of the flowering time under the climate change scenarios may be critical to the planning of adaptation strategy for the pear industry in the future. However, existing methods for the prediction of budburst depend on the spring temperature, neglecting potential effects of warmer winters on the rest release and subsequent budburst. We adapted a dormancy clock model which uses daily temperature data to calculate the thermal time for simulating winter phenology of deciduous trees and tested the feasibility of this model in predicting budburst and flowering of Niitaka pear, one of the favorite cultivars in Korea. In order to derive the model parameter values suitable for Niitaka, the mean time for the rest release was estimated by observing budburst of field collected twigs in a controlled environment. The thermal time (in chill-days) was calculated and accumulated by a predefined temperature range from fall harvest until the chilling requirement (maximum accumulated chill-days in a negative number) is met. The chilling requirement is then offset by anti-chill days (in positive numbers) until the accumulated chill-days become null, which is assumed to be the budburst date. Calculations were repeated with arbitrary threshold temperatures from $4^{\circ}C$ to $10^{\circ}C$ (at an interval of 0.1), and a set of threshold temperature and chilling requirement was selected when the estimated budburst date coincides with the field observation. A heating requirement (in accumulation of anti-chill days since budburst) for flowering was also determined from an experiment based on historical observations. The dormancy clock model optimized with the selected parameter values was used to predict flowering of Niitaka pear grown in Suwon for the recent 9 years. The predicted dates for full bloom were within the range of the observed dates with 1.9 days of root mean square error.

  • Electrochemical reductions of $trans-[Co(en)_2X_2](ClO_4)_n$ (where X is cyanide, nitrite, ammonia, and isothiocyanate) were investigated by cyclic voltammetry and polarography at mercury and glassy carbon electrode. $trans-[Co(en)_2(CN)_2]ClO_4$ was reduced to Co(II) complex followed by adsorption to the mercury electrode. Cyanide ion was not released from the reduced Co(II) complex but the cyanide and (en) were released after the reduction to metallic cobalt. The other complexes except $trans-[Co(en)_2(CN)_2]ClO_4$ were reduced to cobalt(II) complexes followed by release of monodendate ligand, and (en) was released at the reduction step to metallic cobalt. $trans-[Co(en)_2(NO_2)_2]ClO_4$ was reduced to cobalt(Ⅱ) complex, and $NO_2^-$ ion was released followed by electroreduction through ECE mechanism at pH 2. On glassy carbon electrode, all complexes of Co(III) were reduced to Co(II) complexes with irreversible one-electron diffusion controlled reaction in which (en) was not released at this step. Increasing absorption wave number of complexes caused to negative shift of peak potential.

Breakthrough Cancer Pain (돌발성 암성 통증)

  • Breakthrough cancer pain is a transient exacerbation of pain that occurs despite relatively well controlled background pain with around-the-clock analgesia. It is highly prevalent in patients with cancer pain, with an overall prevalence of 70~90%. Breakthrough cancer pain has several negative effects on quality of life, including a decrease in functional status and social relationship, and higher incidence of anxiety/depression. It also places a detrimental burden on their families, society, and the healthcare system. According to the pathogenic mechanism, breakthrough cancer pain is classified into two categories: idiopathic (or spontaneous) pain and incident pain. Episodes of breakthrough cancer pain have typical characteristics, including rapid onset (5~10 min), severe intensity, and short duration (30~60 min). However, there are some variations in timing and severity of pain among patients and episodes. Therefore, a thorough assessment of pain episodes is needed and management plan must be individualized to provide optimal treatment. Several immediate-release formulations such as oxycodone, morphine, and hydromorphone are widely used despite relatively slow onset of action. Recent studies have shown that transmucosal fentanyl preparations were effective for faster control of breakthrough pain. We hope to improve management of breakthrough cancer pain with more efficient analgesics in line with currently available evidence.

A Nucleotide Exchange Factor, BAP, dissociated Protein-Molecular Chaperone Complex in vitro (In vitro에서 핵산치환인자 BAP이 단백질-분자 샤페론 복합체 해리에 미치는 영향)

  • Molecular chaperones and folding enzymes in the endoplasmic reticulum (ER) associate with the newly synthesized proteins to prevent their aggregation and help them fold and assemble correctly. Chaperone function of BiP, which is a Hsp70 homologue in ER, is controlled by the N-terminal ATPase domain. The ATPase activity of the ATPase domain is affected by regulatory factors. BAP was identified as a nucleotide exchange factor of BiP (Grp78), which exchanges ADP with ATP in the ATPase domain of BiP This study presents whether BAP can influence folding of a protein, immunoglobulin heavy chain that is bound to BiP tightly. We first examined which nucleotide of ADP and ATP affects on BAP binding to BiP The data showed that endogenous BAP of HEK293 cells prefers ADP for binding to BiP in vitro, suggesting that BAP first releases ADP from the ATPase domain in order to exchange with ATP. Immunoglobulin heavy chain, an unfolded protein substrate, was released from BiP in the presence of BAP but not in the presence of ERdj3, which is another regulatory factor for BiP accelerating the rate of ATP hydrolysis of BiP The ADP-releasing function of BAP was, therefore, believed to be responsible for immunoglobulin heavy chain release from BiP. Grp170, another Hsp70 homologue in ER, did not co-precipited with BAP from $[^{35}S]$-metabolic labeled HEK293 lysate containing both overexpressed Grp170 and BAP. These data suggested that BAP has no specificity to Grp170 although the ATPase domains of Grp170 and BiP are homologous each other.
