In Vitro Screening of Tannic Acid-based Eco-friendly Farming Material (notice no. 2-4-064) against Plant Pathogenic Bacteria (탄닌산을 함유한 친환경농자재(공시번호 2-4-064)의 식물병원세균 기내 억제효과)

  • To date, chemical managements of plant bacterial diseases are complicated by limitations on use of antibiotics in agriculture, antibiotic resistance development, and limited efficacy of alternative control agents. In this study, thus, we performed screening of eco-friendly farming material (notice no. 2-4-064) containing tannic acid as new antibacterial-activity against 7 plant bacterial pathogens: Pectobacterium carotovorum subsp. carotovorum (Pcc), Ralstonia solanacearum (Rs), Acidovorax avenae subsp. citrulli (Aac), Xanthomonas cirti pv. citri (Xcc), Erwinia pyrifoliae (Ep), Clavibacter michiganensis subsp. michiganensis (Cmm), and Streptomyces scabies (Sc), Initial screening of antibacterial effects of eco-friendly farming material was performed using the paper disk diffusion method and co-cultivation in broth culture. Tannic acid based eco-friendly farming material showed inhibitory effect against Pcc, Rs, Aac, Xcc, Cmm, and Ss, whereas it did not show inhibition zone against Ep. However, when it tested by co-cultivation in broth culture, it showed strong inhibition effect against all pathogens that declined growth curve compared to bacterial pathogen only. Interestingly, when we observed morphological changes on those pathogens by SEM, cell morphologies of 7 pathogens were slightly changed that seem to be malfunction in their cell envelope.

Microstructure and Strength of Class F Fly Ash based Geopolymer Containing Sodium Sulfate as an Additive (황산나트륨 첨가제에 따른 플라이애시 기반 지오폴리머의 미세구조 및 강도 특성)

  • This paper presents an investigation of the mechanical and microstructural properties of Class F fly ash based geopolymer containing sodium sulfate as an additive. Sodium sulfate was used as an chemical additive at the dosage levels of 0, 2, 4, and 6wt% of fly ash. Sodium hydroxide and sodium silicate solutions were used to activate fly ash. The compressive strengths of geopolymer pastes were measured at the age of 28 days. The microstructures of the geopolymer pastes were examined using XRD, MIP and SEM tests. The additions of 2wt% and 4wt% sodium sulfate produced geopolymers with high strength, while increasing the dosage of levels to 6% resulted in almost no changes in strength, comparing with the control geopolymer. The optimum increase in strength was obtained with the addition of 4wt% sodium sulfate. As the amount of sodium sulfate is increased, no additional crystalline phase was detected and no change of amorphous phase indicated despite the change in the strength development. The increase in the strength was due to the change of pore size distribution in samples. As addition of sodium sulfate altered the morphologies of reactive productions and Si/Al ratios of the reaction products, the strengths were thus affected. It was found that the strengths of geopolymer were larger for lower Si/Al ratios of reaction products formed in samples. The optimal amount of sodium sulfate in the fly ash based geopolymer helps to improve mechanical properties of the geopolymer, on the other hand, the high percentage of sodium sulfate could exist as an impurity in the geopolymer and hinder the geopolymer reaction.

Effects of the immobilization of heparin and rhPDGF-BB to titanium surfaces for the enhancement of osteoblastic functions and anti-inflammation

  • PURPOSE. This study was to investigate the effects of recombinant human platelet-derived growth factor (rhPDGF-BB) and heparin to titanium surfaces for enhancement of osteoblastic functions and inhibition of inflammation activity. MATERIALS AND METHODS. The anodized titanium discs, not coated with any material, were used as a control group. In heparinized-Ti group, dopamine was anchored to the surface of Ti substrates, and coated with heparin. In PDGF-Ti group, rhPDGF-BB was immobilized onto heparinized Ti surface. The surface morphologies were investigated by the scanning electron microscope in each group. The release kinetics of rhPDGF-BB were analyzed, and cytotoxicity tests for each group were conducted. The biocompatibilities were characterized by measuring cell proliferation, alkaline phosphatase activity, and calcium deposition using MG-63 cells. Statistical comparisons were carried out by one-way ANOVA tests. Differences were considered statistically significant at $^*$P<.05 and $^{**}$P<.001. RESULTS. The combination of rhPDGF-BB and heparin stimulated alkaline phosphatase activity and OCN mRNA expression in osteoblastic cells ($^*$P<.05 and $^{**}$P<.001). MG-63 cells grown on PDGF-Ti had significantly higher amounts of calcium deposition than those grown on anodized Ti ($^{**}$ P<.001). Heparinized Ti was more anti-inflammatory compared to anodized Ti, when exposed to lipopolysaccharide using the transcript levels of TNF-${\alpha}$ and IL-6 of proinflammatory cytokine ($^*$P<.05 and $^{**}$P<.001). CONCLUSION. The result of this study demonstrated that the incorporation of rhPDGF-BB and heparin onto Ti surface enhanced osteoblastic functions and inhibited inflammation.

Effect of Developmental Disorders of Maxillary First Molars on Orofacial Morphology (악안면 형태에 대한 상악 제1대구치 발육장애의 영향)

  • This study was aimed to evaluate orofacial morphologies on the cases of developmental disorders of maxillary first molars. Panoramic radiographs, lateral cephalographs, and clinical photos of 2983 children who attended the Pediatric Dental Clinic of Pusan National University Dental Hospital from 2006 to August 2017 were assessed retrospectively. 34 patients were selected whose maxillary first molars were missed or developmentally delayed unilaterally or bilaterally. Demirjian's method was used for estimating dental age, then which was compared to chronologic age of children. Parameters expressing skeletal and dentoalveolar disharmony were checked and compared with control. Additionally, occlusion relationship was evaluated. Maxillary dental age was significantly delayed compared to chronologic age. Several parameters which show skeletal open-bite tendency and skeletal class III malocclusion with maxillary retrusion were statistically significant. Anterior crossbite and edge-bite were expected in most of these cases, but compensation by occlusion and soft tissue was also verified which might mask skeletal class III tendency. Congenital missed or developmentally delayed maxillary first molars might be related with declined growth of maxilla. If developmental disorders of maxillary first molars were verified during clinical examination, careful monitoring of orofacial growth was necessary during puberty and timed orthopedic and orthodontic intervention were considered.

Induction of G1 Phase Cell Cycle Arrest and Apoptotic Cell Death by 5-Fluorouracil in Ewing′s Sarcoma CHP-100 Cells (CHP-100 Ewing′s 육종세포에서 5-fluorouracil에 의한 G1 arrest 유도 및 apoptosis 유발에 관한 연구)

  • 5-fluorouracil (5-FU), a pyrimidine analog, is a widely used anticancer drug, which works through irreversible inhibition of thymidylate synthase. In the present study, it was investigated the anti-proliferative effects and molecular mechanisms of 5-FU using Ewing's Sarcoma CHP-100 Cells. The present data indicated that treatment of 5-FU to CHP-100 cells induced a G1 phase arrest of the cell cycle in a time-dependent manner. 5-FU-induced G1 arrest was correlated with the accumulation of the hypophosphorylated form of the retinoblastoma protein (pRB) and association of pRB with the transcription factors E2F-1 and E2F-4. Although 5-FU treatment did affect the levels of cyclin-dependent kinases, the levels of cyclin A and B were markedly down-regulated as compared with the untreated control group. In addition, 5-FU-induced G1 arrest of CHP-100 cells was also associated with the induction of apoptosis, as determined by apoptotic cell morphologies, degradation of poly(ADP-ribose) polymerase and Annexin V staining. Furthermore, 5-FU induced the loss of mitochondrial membrane potential with up-regulated pro-apoptotic Bax expression, down-regulated anti-apoptotic Bcl-2 expression and cytochrome c release from mitochondria to cytosol. Collectively, the data suggest that 5-FU is effective in inducing cell growth reduction and apoptosis, in part, by reducing phosphorylation of pRB and activating mitochondrial dysfunction in CHP-100 cells.

  • We examined the state of Korean pomfret(Pampus echinogaster) in Korean waters and considered the management strategy of the stock based on the theory yield Per recruit. It is not facile to discriminate Korean pomfret distributed in Korean waters from silver pomfret (p. argenteus) due to the similarities in their external morphologies. For this rea- son, Korean pomfret has been treated in silver pomfret in fisheries statistics of Korea. In this study, we asserted Korean pomfret from pomfrets caught commercially by the morphology, from which we recognized that Korean pomfret took $60\~70\%$ in catch(in weight) and that the smaller the body length, the higher the proportion of Korean pomfret. Parameters estimated for Korean pomfret were as follows: natural mortality(M) = 0.6, fishing mortality(F) =0.924(mean value for $1986\~1988$), age at recruit to fishery($t_r$) =0.19 yrs, age at first capture($t_c$) : 0.49 yrs, and the rate of recruit of age-0 fish to fishery(Q) = 0.29. The results obtained from the theory of yield per recruit indicated that the present state of stock was below the optimum level of exploitation and that the control of fishing intensity rarely had an effect on the increasing of yield. Accordingly, we conclude that proper management can be made by increasing the current age of 0.49 yrs at first capture to 1.5 yrs.

  • Fabrication of monolayer is important method for enhancing physical and chemical characteristics such as light shielding and antireflection while maintaining thin film properties. In previous studies, monolayers were fabricated by various methods on small substrates, but processes were complicated and difficult to form monolayers with large area. We used rod coating equipment with a small amount of coating liquid to form a HCP (hexagonal closed packing) coating of PMMA beads on PET(poly(ethylene terephthalate)) substrate with $20cm{\times}20cm$ size. We observed that changes in morphologies of monolayers by using the solvents with different boiling points and vapor pressures, by adapting surfactants on particles and by applying plasma treatment on substrates. The coverage was increased by 20% by optimizing the coating conditions including meniscus of beads, control of the attraction - repulsion forces and surface energy. This result can potentially be applied to optical films and sensors because it is possible to make a uniform and large-scale monolayer in a simple and rapid manner when it is compared to the methods in previous studies.

Importance of Strain Improvement and Control of Fungal cells Morphology for Enhanced Production of Protein-bound Polysaccharides(β-D-glucan) in Suspended Cultures of Phellinus linteus Mycelia (Phellinus linteus의 균사체 액상배양에서 단백다당체(β-D-glucan)의 생산성 향상을 위한 균주 개량과 배양형태 조절의 중요성)

  • Strain improvement and morphology investigation in bioreactor cultures were undertaken in suspended cultures of Phellinus linteus mycelia for mass production of protein-bound polysaccharides(soluble ${\beta}$-D-glucan), a powerful immuno-stimulating agent. Phellineus sp. screened for this research was identified as Phellinus linteues through ITS rDNA sequencing method and blast search, demonstrating 99.7% similarity to other Phellinus linteus strains. Intensive strain improvement program was carried out by obtaining large amounts of protoplasts for the isolation of single cell colonies. Rapid and large screening of high-yielding producers was possible because large numbers of protoplasts ($1{\times}10^5{\sim}10^6\;protoplasts/ml$) formed using the banding filtration method with the cell wall-disrupting enzymes could be regenerated in relatively high regeneration frequency($10^{-2}{\sim}10^{-3}$) in the newly developed regeneration medium. It was demonstrated that the strains showing high performances in the protoplast regeneration and solid growth medium were able to produce 5.8~6.4%(w/w) of ${\beta}$-D-glucan and 13~15 g/L of biomass in stable manners in suspended shake-flask cultures of P. linteus mycelia. In addition, cell mass increase was observed to be the most important in order to enhance ${\beta}$-D-glucan productivity during the course of strain improvement program, since the amount of ${\beta}$-D-glucan extracted from the cell wall of P. linteus mycelia was almost constant on the unit biomass basis. Therefore we fully investigated the fungal cell morphology, generally known as one of the key factors affecting cell growth extent in the bioreactor cultures of mycelial fungal cells. It was found that, in order to obtain as high cell mass as possible in the final production bioreactor cultures, the producing cells should be proliferated in condensed filamentous forms in the growth cultures, and optimum amounts of these filamentous cells should be transferred as active inoculums to the production bioreactor. In this case, ideal morphologies consisting of compacted pellets less than 0.5mm in diameter were successfully induced in the production cultures, resulting in shorter period of lag phase, 1.5 fold higher specific cell growth rate and 3.3 fold increase in the final biomass production as compared to the parallel bioreactor cultures of different morphological forms. It was concluded that not only the high-yielding but also the good morphological characteristics led to the significantly higher biomass production and ${\beta}$-D-glucan productivity in the final production cultures.
