  • The Gold-spotted pond frog, Rana plancyi chosenica, designated as a vulnerable species by IUCN Red list. This species is a typical example facing local population threats and extinction due to human activities in South Korea. A strategic conservation plan for this endangered species is urgently needed. In order to provide information for future conservation planning, accurate information on the genetic diversity and taxonomic status is needed for the establishment of conservation units for this species. In this study, we used a molecular genetic approach using the mitochondrial cytochrome b gene and control region sequences to find the genetic diversity of gold-spotted pond frogs within South Korea. We sequenced the mitochondrial DNA cytochrome b gene and control region of 77 individuals from 11 populations in South Korea, and one from Chongqing, China. A total of 15 cytochrome b gene haplotypes and 34 control region haplotypes were identified from Korean gold-spotted pond frogs. Mean sequence diversity among Korean gold-spotted pond frogs was 0.31% (0.0-0.8%) and 0.51% (0.0-1.0%), respectively. Most Korean populations had at least one unique haplotype for each locus. The Taean, Ansan and Cheongwon populations had no haplotypes shared with other populations. There was a sequence divergence between Korean and Chinese gold-spotted pond frogs (1.3% for cyt b; 2.9% for control region). Analysis of genetic distances and phylogenetic trees based on both cytochrome b and control region sequences indicate that the Korean gold-spotted pond frog are genetically differentiated from those in China.

  • "Buliseonrando," meaning "Buddhist virtues and the orchid are one and the same," was painted by Chusa Kim Jeong-hee. Four appreciation sentences are written in various fonts around the orchid drawn in the center of the painting, along with a total of 15 seals stamped. Hyperspectral image analysis(HSI), microscopy, and X-ray fluorescence (XRF) were conducted with a focus on the seals and the parts of the painting that have been applied with a conservation treatment. As a result of the analyses, the seals were classified into two types-seals with or without barium content. Stamp shade was identified only in five of themstamps, which allows the assumption that the composition and material characteristics of the stamp inks varied depending on the period. In particular, hyperspectral image analysis confirms traces of conservation treatment on the seals and the lost parts identified in addition to the 15 seals, which also demonstrates the utility of hyperspectral image analysis.

  • Microorganisms were isolated from Dongchundang(wooden cultural heritage) with PDA medium culture. Nineteen species shows the cellulolytic activity. Methylobacterium sp. was the most active in cellulose degradation. The growth curve and pH were measured during incubation of the microorganism for 72 hours. The pH was increased with the increasing of microbial growth. The degree of cellulose degradation was determined with the amount of reducing sugar by use of dinitrosalicylic acid (DNS) method. The amount of reducing sugar was decreased after 45 hours. As a results, It should suggested that wood component were deteriorated by Methylobacterium sp..

  • In order to clarify the effect of consolidant, the artificial porous material with low intensity was manufactured using granite powder and Portland cement. We have prepared four kinds of alkoxysilane system consolidants, a acrylic resin and a epoxy resin and investigated about characteristics before and after consolidation. As a result of the research, Silres BS OH 100 was effective for density and surface hardness. SS-101 with hydrophobicity and Site SX-RO with hydrophilicity had the good durability over salts weathering. On the other hand, Syton HT-50 and Paraloid B72 were easily destructed by salt weathering because they were concentrated on surface area by the low penetration depth. Araldite 2020 was the most effective consolidant for improvement of physical properties.

  • In this paper, we present a nondestructive analysis using X-ray and microscopic investigation to detect the structure, manufacturing technique and preservation status of the Goryeo lacquered box Inlaid with Mother-of-pearl Chrysanthemum and Scroll Design (Goryeo Lacquered Box). We confirm that the Goryeo Lacquered Box consists of the soft wood as the basic material. The soft wood was coated with textile and then lacquered. The box structure of the Goryeo Lacquered Box was formed of wooden boards with wood plants added to the side, after processing into a trefoil-shaped. The wooden sides of the Goryeo Lacquered Box were cut at regular intervals for easier processing into a curved shape. Moter-of-pearl, tortoiseshell, and metal wire were used to decorate the surface. mother-of pearl was the cutting processing, and tortoiseshell was used for back coloring. The metal line was constructed using one line and twist line.

  • The Conservation Science Team of National Museum of Korea has established the data base of lead isotope ratio as the scientific research of bronze patina, which was acquired from conservation process of metal objects, and based on this result, it intends to conduct the research related to the production places of raw material for the bronze objects. As the equipment for analysis of lead isotope ratio of the bronze patina, the thermal ionization mass spectrometer (TIMS) was used. As part of the research, in 2007, the 11 samples of the bronze Patina from the objects, believed to belong to of Nangnang(Lelang), the 6 items of bronze patina from the flower-shaped bronze dishes, belong to Unified Silla period and 4 items from the Goryeo bronze bell were acquired and analyzed the lead isotope ratio. Based on the data of the lead isotope ratio from the galena of Korea, China and Japan, the provenance raw materials were estimated through the comparison of their lead isotope ratios. In the future, the Conservation Science Team of the National Museum of Korea will collect as much bronze patina as possible from the process of conservation treatment of metal objects, and analyze them in order to conduct the study of provenance with the type, period and the region of the objects.

  • The Stone Seated Buddha Triad and Stone Standing Buddha in Bijung-ri are state-designated heritage (treasure) statues having the Buddha style of the Goryeo dynasty from the 6th century. Conservation scientific investigations were conducted to understand the preservation status of these stone Buddha statues and to establish a conservation plan. The Stone Seated Buddha Triad and Stone Standing Buddha are composed of fine-medium grained biotite granite, which is considered to be of the same origin owing to their low magnetic susceptibility distribution of less than 0.2 (×10-3 SI unit) and similar mineral characteristics. The Stone Seated Buddha Triad has highly homogenous mineral composition and particle size, whole-rock magnetic susceptibility, and geochemical characteristics very similar to those of the nearby outcrop. It was confirmed that a combination of physical, chemical, and biological factors affects the Stone Buddha statues. In particular, both the Stone Seated Buddha Triad and Stone Standing Buddha tend to be chipped off from the front and cracked and scaled from the back. The Stone Standing Buddha located outdoors experiences granularity decomposition and black algae formation, which accelerate the weathering under unfavorable conservation environments. The result of non-destructive physical property diagnosis using ultrasonic velocity showed that both the Stone Seated Buddha Triad and Stone Standing Buddha have been completely weathered (CW), indicating very poor physical properties.

  • While excavating relics in Uksu-dong of Daegu and Oksan-dong of Gyeongsan, 38 units of the earthen kilns used during the Three Kingdom era were discovered. Nine pieces of charcoal were collected for examination from 8 kilns out of 38. The result identified 4 pinus densiflora, 3 Quercus sp. and 1 Platycarya strobilacea Sieb. The remaining 1 tree could not be identified because most of the remaining wood was mixed and tangled with clay.
