The fungal isolate Isaria javanica pf185 has potential as a mycopesticide because it demonstrates insecticidal activity against the green peach aphid and antifungal activity against Colletotrichum gloeosporioides. For commercialization of this isolate, determination of the optimal and least expensive culture conditions is required; however, these data are not currently available. This study describes the conditions for optimal development of conidia and production of metabolites for the biocontrol of the fungal pathogen. The optimal culture conditions were examined using cultures on solid agar and liquid media. High growth temperature enhanced spore formation but reduced antifungal activity in both solid and liquid media. The highest spore yield was obtained in a medium containing glucose as a carbon source and yeast extract as a nitrogen source. Soybean powder and wheat bran were effective nitrogen sources that promoted spore production and antifungal activity of the isolate. These results revealed the basic, cost-effective growth media for commercial production of a biopesticide with insecticidal and antifungal properties for use in integrated pest management.
BACKGROUND: Entomopathogenic fungi have been studied to develop for biological control agents as an alternative to chemical control agents in insect pest management. This investigated to determine the optimal culture conditions in ceramic balls for maximal sporulation of entomopathogenic fungi Beauveria bassiana M130 by use rice bran extract.METHODS AND RESULTS: METHODS AND RESULTS: A culture of entomopathogenic fungi for 12day on rice bran extract(1:8, w/v) incubated in ceramic matrix at 28℃. Natural zeolite ceramic ball was high production of 4.2×108 conidial/mL. The culture condition optimized initial pH, temperature, rice bran extract concentration, adhesives substance and concentration of NaCl, respectively. The high production of spore optimal conditions were temperature 28℃, initial pH 3, rice bran extract 3 mL, starch 33 g, 5 % NaCl and sopre suspension 7 mL, respectively.CONCLUSION: This study was carried out for the mass production of entomopathogenic fungi conidia recover rate 65% in matrix of natural zeolite ceramic ball, and to develop ingredient-used formulation of Beauveria bassiana M130 conidia for biological control agents.

Several fungal isolates were isolated from diseased monochoria(Monochoria vaginalis, weed of paddy field), which has an resistance to sulfonyl urea(S.U.) herbicide, and were evaluated in the laboratory and greenhouse as potential mycoherbicide. Eight fungi, Alternaria sp., Colletotrichum sp., Curvularia sp., Paenicillium sp and etc. were observed in the isolates. Pathogenicity testing were done on the monochorias in the greenhouse. Monochorias were inoculated with suspensions containing conidia of each isolate at the rates of 1.0 ${\times}$ 10$\^$5/ conidia/ml and 0.1% Tween 80 with hand-gun sprayer. Curvularia sp. and an unidentified fungal isolate caused 90-95% mortality on the monochorias 15∼20 days after inoculation. However the other isolates induced slight symptom of disease on the monochorias. In the early stage of disease development sun-burn appearance was shown at the infected site and the last infected leaves and stems were withered to death. Subsequently the pathogenicity on the rice was evaluated with above two effective isolates. From the test an unidentified isolate showed pathogenicity on the rice but Curvularia sp., named as DBB2003, didn't. Now the mass production and formulation using Curvularia sp. DBB2003 are in progress and the field test will be followed. Combination product with Curvularia sp. DBB2003 and chemical herbicide will be more effect to control the monochoria resisted on S.U. herbicide and need to be further tested.
To investigate Phomopsis species causing fruit decays of Japanese apricot, apple and kiwifruit, we collected diseased fruits from the fruit markets in 1995 and 1996 respectively. Phomopsis mali Roberts was identified based on cultural characteristics, morphological aspects and pathogenicity. There were no remarkable differences with respect to $\alpha$ and $\beta$ conidia, growth rates and colony characters among the isolates from Japanese apricot, apple and kiwifruit. The pathogens grew more than 70 mm on potato dextrose agar in 5 days at $25^{\circ}C$. The agar was slightly discolored by the production of a reddish purple pigment under the light at $25^{\circ}C$ and 3$0^{\circ}C$ respectively. Only $\alpha$ spores of the different isolates of P. mali were formed at 15$^{\circ}C$ and $\beta$ spores were mainly produced at 3$0^{\circ}C$, but and $\alpha$ and $\beta$ spores were produced in approximately equal numbers at 2$0^{\circ}C$ and $25^{\circ}C$. Pycnidia were a few under the dark condition but were abundant at wide range of 15~3$0^{\circ}C$ under near ultra violet illumination. Conidia were two types : $\alpha$ spores were unicellar, fusoid, hyaline and biguttulate, whereas $\beta$ sores were unicellar, acicular to filiform, straight or hooked and hyaline. An ascigerous stage was not formed in cultures or in nature. Isolates of Phomopsis mali from japanese apricot, apple and kiwifruit could infect fruits of apple, pear, apricot, Japanese apricot and kiwifruit. There were some differences in pathogenicity depending on stocks of fruit crops tested.

The value of silkworms as functional health food materials has increased, as has the interest in its disease control for stable production, and in the economic value of entomopathogenic microorganisms. In this study, we isolated and identified disease-causing fungi from white muscardine silkworms, and confirmed whether this strain could produce white muscardine silkworms. For the analysis of the cause of white muscardine disease in the infected silkworms, the fungi and prokaryotes causing the disease were identified, isolated, and identified using metagenome analysis. Metagenomic analysis detected a large amount of the fungus Metarhizium rileyi in silkworms, and a large amount of the bacterium Enterococcus mundtii, which was presumed to be the causative agent of the disease. For accurate identification of the fungi, these were purified by culture medium, and sequencing and phylogenetic tree analyses were performed using an internal transcribed spacer. As a result, M. rileyi, Cladosporium cladosporioides, and C. tenuissimum were identified. In general, M. rileyi is known to form green conidia, but in this study, white-yellow conidia were formed, indicating that the exact causative agent of the fungal disease cannot be estimated by diagnosing the symptoms. Thus, a diagnostic method is necessary for the continuously collection of required pathogens, and identifying their morphological and genetic characteristics.

Air-borne plant pathogenic fungus Fusarium graminearum and seed-borne plant pathogenic bacterium Burkholderia glumae are cause similar disease symptoms in rice heads. Here we showed that two pathogens frequently co-isolated in rice heads and F. graminearum is resistant to toxoflavin produced by B. glumae while other fungal genera are sensitive to the toxin. We have tried to clarify the resistant mechanism of F. graminearum against toxoflavin and the ecological reason of co-existence of the two pathogens in rice. We found that F. graminearum carries resistance to toxoflavin as accumulating lipid in fungal cells. Co-cultivation of two pathogens resulted in increased conidia and enhanced chemical attraction and attachment of the bacterial cells to the fungal conidia. Bacteria physically attached to fungal conidia, which protected bacterium cells from UV light and allowed disease dispersal. Chemotaxis analysis showed that bacterial cells moved toward the fungal exudation compared to a control. Even enhanced the production of phytotoxic trichothecene by the fungal under presence of toxoflavin and disease severity on rice heads was significantly increased by co-inoculation rather than single inoculation. This study suggested that the undisclosed potentiality of air-born infection of bacteria using the fungal spores for survival and dispersal.
The research was carried out for the purpose of finding effects of gerbal saponins on aflatoxin synthesis by Aspergillus parasitics NRRL 2999. A. parasiticus with $10^6$ conidia were grown at $30^{\circ}C$ for 9 days on the enriched medium that is optimum for the frowth and aflatoxins production by the mold. The inhibitory effect on the growth and aflatoxins produced by the mold occurred in the presence of 0.36% of crude red-ginseng saponin showing both the growth and aflatoxins production come to 62.3% (growth), 38.7% (aflatoxin $B_1$) and 22.9% (aflatoxin $G_1$) of the control. Thd next effective saponin to inhibit the growth and aflatoxins production was from burdock seeds. However, saponin extracted from honeysuckle flowers had no inhibitory effect. The mold caused no changes in the pH of the medium when it contained red-ginseng saponin. Red-ginseng saponin was more effective than the white-ginseng in inhibiting both the growth and aflatoxin production.
Rusty root of ginseng has been known as one of the limiting factors in ginseng production in Korea. An attempt was, therefore, made to elucidate biological and chemical natures of the rusty root, and the redox Potential of the ginseng cultivated soils were measured and compared with diseased and non-diseased soils. Reddish discoloration was most frequently observed on the epidermis of ginseng root and the pigments were accumulated in all epidermal cells of the diseased lesions. The lower the redox potential of the ginseng cultivated soil was, the more severe the rusty root was observed. Fe content in the diseased epidermis was 3 times higher than that of healthy one. Organic acids such as oxalic, malonic, succinic, and citric acids were also higher in the mss root than in the healthy one. Thin layer chromatogram of phenolic acid fractions obtained from the epidermal cells of the rusty root of ginseng exhibited 3 to 4 unidentified substances not found in the healthy root. Also lignification of the epidermal cells and the activity of phenylalanine ammonia lyase were greater in the rusty root than the healthy root. Colony formation and conidia production of F. solani, And mycelial growth and sclerotium formation of Sclerotinia sp. isolated from ginseng root were suppressed in a nutritionally minimal medium supplemented with water extract of rusty ginseng root epidermis. It is, therefore, suggested that rusty root of ginseng is caused by unfavorable rhizosphere environmental stress or stresses resulting abnormal metabolism in the root as a selfdefence mechanism of non-specific resistance responses.

Monascus anka 732Y3 was induced from Monascus anka KFCC 11832 (IFO 4478, ATCC 16360) by ultra-violet light irradiation. The growth of this new fungus is frequently more dependent on sexual propagation than asexual propagation, compared with that of its parental strain, M. anka KFCC 11832. Less conidia than those of M. anka KFCC 11832 were observed by a microscope. The optical density of the red pigments ($OD_{500}$) produced by M. anka 732Y3 was 157, which was about 10 times higer than that of M. anka KFCC 11832. Such high production of the red pigments by the mutant could be explained by the following observations.
Tolerance against mercuric chloride in mutants of Pyricularia oryzae Cavara induced by ultraviolet(UV) irradiation has been investigated. The tolerant isolates obtained using ultraviolet(UV) irradiation were maintained a high level of tolerance even after 15 times transfer to the chemical free media. Each isolate of mutants UM-1, UM-2, and UM-3 on the successive monoconidial culture has genetically homogeneous for tolerance. The tolerant isolates sporulated less and showed a higher percentage of germination of conidia on media without the chemical than the parental isolate. The xotins released from the parental and the tolerant isolates have been identified as piricularin and $\alpha$-picolinic acid by paper-chromatography. The tolerant isolates produced more piricularin and its virulence on seedlings of rice varieties were higher than the parental isolates. Both piricularin production and virulance on rice were highest in the UM-2 isolate.
