Biological Detoxification of Lacquer Tree (Rhus verniciflua Stokes) Stem Bark by Mushroom Species

  • The stem bark of Rhus verniciflua (RVSB) has been used in herbal medicine to treat diabetes mellitus and stomach ailments for thousands of years in Korea, despite its content of the plant allergen, urushiol. A new biological approach for the removal of urushiol from RVSB using mushrooms is described. All mushroom species (11 sp.) employed in this study were able to grow on RVSB, although the growth rate (mm/day) was lower than the control (sawdust). The components of urushiol congeners [C15 triene (m/z 314), C15 diene (m/z 316), C15 monoene (m/z 318), and C15 saturated (m/z 320)] were purified by HPLC and identified by GC-MS. A C15:3 (3-pentadecatrienly catechol) was found to be most abundant in RVSB. Urushiol analogues decreased remarkably from 154.15 to 10.73 mg/100 g (approximately 93%) by Fomitella fraxinea, whereas Trametes vercicolor showed only a 1.46% degradation capacity despite its 2 fold higher growth rate. Similarly, laccase activity was found to be high for F. fraxinea and low for T. vercicolor. Moreover, approximately 98% detoxification was accomplished by F. fraxinea cultivated on RVSB supplemented with 20%(w/w) rice bran. These findings suggest that mushrooms can be used in the detoxification of RVSB.

Measurement of Atmospheric Dry Deposition and Size Distribution of Particulate PCBs in 1999 at Seoul

  • Ambient particle size distributions of PCBs and their dry deposition fluxes were measured at a site in Seoul to quantify dry deposition fluxes of PCBs and size characteristics of PCBs in the air, and to estimate ambient concentrations of gaseous PCBs and dry deposition fluxes. The dry deposition plate was used to measure dry deposition fluxes of particulate mass and PCBs and a cascade impactor and rotary impactor were used to measure ambient particle size distributions for small ($D_p<9{\mu}m$) and large ($D_p>9{\mu}m$) particles, respectively. Six sample sets were collected from April to July 1999. The fluxes of particulate total PCBs (the sum of 43 congeners) ranged from 160 to $607ng\;m^{-2}day^{-1}$. The size distribution of total PCBs was bimodal with two peaks in small particle size ($D_p{\sim}0.6\;and\;6{\mu}m$, respectively) and, thus, mass concentration being dominant in small particles. The mean particulate PCBs concentration was $6.9{\mu}g$ PCBs/g. The concentrations of PCB homologues in the gas phase were estimated based on the particle/gas partition coefficient ($K_p$) with the measured values of particulate PCBs in this study and they were comparable to those observed in other previous studies. Dry deposition fluxes were estimated by calculating dry deposition velocities.

Comparison the quantification method of PCBs in waste transformer oils (절연유 중 폴리염화비페닐류의 정량법 비교)

  • Polychlorinated biphenyls (PCBs) were commercially produced as complex mixtures beginning in 1929. The PCBs manufactured commercially are known by a variety of trade names including; Aroclor (USA), Phenoclor (France), Kaneclor (Japan), Sovol (USSR) and so on. PCBs are a class of 209 congeners that were widely used in a wide variety of applications, including dielectric fluid in transformers and large capacitors; heat transfer fluids; hydraulic fluids; lubricating and cutting oils; and as additives in pesticides, paints, adhesives, sealants, and plastics. The quantification methods of peak matching and coefficient comparison were compared using the Aroclor 1242, 1248, 1254, 1260 standards. Also, six transformer oils were analyzed as a main source of polychlorinated biphenyls (PCBs) emission into the environment. The transformer oils contained the pure and mixed of Aroclor 1242, Aroclor 1254, and Aroclor 1260. The analytical results using two quantification methods showed the little difference between the measured results.

Studies on the Free Fatty Acids of Domestic Butter (국산 버터 중의 유리지방산에 관한 연구)

  • Free fatty acids of two brands of domestic butter were isolated directly by a modified silicic acid column chromatography, and were analyzed by gas liquid chromatography. $C_{18}$ FFA congeners $(C_{18_0},\;C_{18:1},\;C_{18:2}\;and\;C_{18:3})$ were the predominant components (52.83% in brand A and 47.50% in brand B), followed by $C_{16}$FFA (29.39% in brand A and 30.52% in brand B) and $C_{14}$FFA (11.85% in brand A and 13.76% in brand B). The other FFA were present as minor components (0.29-3.87%). Concentrations of four FFA $(C_4,\;C_6,\;C_{10}\;and\;C_{12})$FFA which would be expected to contribute strongly to hydrolytic rancidity off-flavors were below individual threshold level, except $C_4FFA$ (56 ppm) in butter B.

Taxonomic Status of Acheilognathus sp. (Cyprinidae) found in the Geum River, Korea (금강에서 발견된 Acheilognathus sp. (Cyprinidae)의 분류학적 위치)

  • To examine the taxonomic status of Acheilognathus sp. specimens from the Geum River, morphological and genetical characteristics of A. sp., A. yamatsutae and A. majusculus were investigated and compared in detail. Specimens of A. sp. could be distinguished from the other two species by the combination of some morphological characters such as nuptial color, vertebrae, gillrakers and etc. Males of A. sp. had red bands on the outer margin of dorsal and anal fins and a white band on the outer margin of ventral fin in breeding season. A. sp. had larger maximum body length and somewhat more vertebrae than A. yamatsutae, and had fewer gillrakers than A. majusculus. A. sp. appeared as a monophyletic group with A. majusculus and A. cyanostigma based on genetic analysis. In addition, it had even more close relationship with other congeners than A. yamatsutae. Therefore it is presumed that A. sp. from the Geum River may be a distinct species in genus Acheilognathus.

Depositional Characteristics of Atmospheric PBDEs on Pine Needles, Bark and Soil (대기 중 폴리브롬화디페닐에테르의 소나무 잎, 소나무 껍질 및 토양으로의 침착 특성)

  • Objective: This study was carried out in order to determine the depositional characteristics of pine needles, pine bark, and soil used as a passive air sampler (PAS) for atmospheric polybrominated diphenyl ethers (PBDEs). Methods: All three media were sampled from the same site. The PBDE concentrations were analyzed by HRGC/HRMS, and the lipid contents were measured using the gravimetric method by n-hexane extraction. Results: The total PBDE concentration was the highest in soil (22,274.57 pg/g dry), followed by pine bark (20,266.39 pg/g dry), and then pine needles (7,380.22 pg/g dry). Pine needles contained the highest lipid contents (21.31 mg/g dry), whereas soil (10.01 mg/g dry), and pine bark (4.85 mg/g dry) contained less. There were poor correlations between lipid content and total PBDE concentrations in the media ($R^2$=0.8216, p=0.2814). Congeners BDE 47, 99, 183, 196, 197, 206, 207 and 209 showed peak concentrations. Among these, BDE 206, 207, and 209 are highly brominated PBDEs that exist as particulates in ambient air. They accounted for 81.2% [69.2 (pine needles) - 89.0% (tree bark)] of the concentration and therefore are noted as the main congener of the total PBDEs. Conclusions: It can therefore be concluded that for reducing error by improper sampling, the same species of media should be recommended for use as a PAS for atmospheric PBDEs due to the differences in depositional characteristics.

The genetically healthy terrestrial orchid Liparis krameri on southern Korean Peninsula

  • Neutral genetic diversity found in plant species usually leaves an indelible footprint of historical events. Korea's main mountain range (referred to as the Baekdudaegan [BDDG]), is known to have served as a glacial refugium primarily for the boreal and temperate flora of northeastern Asia. In addition, life-history traits (life forms, geographic range, and breeding systems) influence the within- and among-population genetic diversity of seed plant species. For example, selfing species harbor significantly less within-population genetic variation than that of predominantly outcrossers. A previous study of two Liparis species (L. makinoana and L. kumokiri) emphasizes the role of the abovementioned factors shaping the levels of genetic diversity. Liparis makinoana, mainly occurring on the BDDG and self-incompatible, harbors high levels of within-population genetic diversity (expected heterozygosity, HeP = 0.319), whereas there is no allozyme variation (HeP = 0.000) in L. kumokiri, which is self-compatible and mainly occurs in lowland hilly areas. To determine if this trend is also found in other congeners, we sampled five populations of L. krameri from the southern part of the Korean Peninsula and investigated the allozyme-based genetic diversity at 15 putative loci. The somewhat intermediate levels of within-population genetic variation (HeP = 0.145) found in L. krameri are most likely due to its occurrence in mountainous areas that, despite being outside of the main ridge of the BDDG, still served as refugia, and a self-incompatible breeding system. Management strategies are suggested for L. krameri and L. makinoana based on the levels and distribution of genetic diversity and inbreeding.

Genic Vadadon and Speciation of Fishes of the Genus Moroco(Cyprinidae) (버들치속(잉어과) 어류의 유전적 변이 및 종분화)

  • Surveys of electrophoretic variation in isozymes and general proteins encoded by 26 loci were conducted to assess species recognition and to estimate the degree of genic variation and species divergence for seven species of the genus Moroco inhabiting in Korea and Japan. Estimates of the average calculated heterozygosity per species of M semotilus, M sp., M percnurus, M lagowskii, M oxycephalus, M steindachneri and M jouyf are low: 0.021, 0.019, 0.051, 0.031, 0.023, 0.046, and 0.007, respectively, and observed heterozygosities are 0.038, 0.022, 0.060, 0.027, 0.025, 0.042, and 0.002, respectively. Allozyme analyses show these species to be distinct genetically with the lafter four species being more closely related one another than any one of them is to the rest of the species. However, these four species (M. lagowskii, M. oxycephalus, M. steindachneri and M jouyi), had unique genetic markers in each species to be recognized as valid species. These results contrast to the previous report of Chung et of. (1986) mainly due to their error in analyzing the isozyme pallems, particularly in MDH and PGI analyses. The genetic distances among M semotilus, M sp., and M percnurus are near the high end of the scale of such estimate for freshwater fish congeners. Based on estimated divergent time of these species of the genus Moroco (5 to 0.6 million years) it is assumed that they are speciated during late Pliocene to middle Pleistocene epoch prior to migration to Korean and Japanese waters through Paleo Amur River system.

Genetic and Biochemical Characterization of the Biphenyl Dioxygenase from Pseudomonas sp. Strain B4

  • Biphenyl dioxygenase (BPDO), which catalyzes the first step in the bacterial degradation of biphenyl and polychlorinated biphenyls, was characterized in Pseudomonas sp. B4. The bphA locus containing the four structural genes encoding BPDO were cloned and sequenced. A regulatory gene as well as a putative regulatory sequence were identified upstream of this locus. A transposase-like gene was found within a 1-kb region further upstream, thereby suggesting that the bphA locus may be carried on a transposable element. The three components of the BPDO enzyme have been separately overexpressed and purified from E. coli. The ferredoxin and terminal dioxygenase components showed biochemical properties comparable to those of two previously characterized BPDOs, whereas the ferredoxin reductase exhibited an unusually high lability. The substrate selectivity of BPDO was examined in vivo using resting cell assays performed with mixtures of selected polychlorinated biphenyls. The results indicated that para-substituted congeners were the preferred substrates. In vitro studies were carried out on a BPDO complex where the reductase from strain B4 we replaced by the more stable isoform from Comamonas testosteroni B-356. The BPDO enzyme had a specific activity of $0.26{\pm}0.02 {\mu}mol {min^-1}{mg^-1}\;of\;ISP_{BPH}$ with biphenyl as the substrate. The 2,3-, 4,4'-, and 2,4,4'-chlorobiphenyls were converted to single dihydrodiols, while 2,4'-dichlorobiphenyl gave rise to two dihydrodiols. The current data also indicated that 2,4,4'-trichlorobiphenyl was a better substrate than the 4,4'-dichlorinated congener.

Concentration of Polybrominated Diphenyl Ethers and Their Composition in Octopus minor Collected from Seosan Intertidal Zone (낙지(Octopus minor)에서의 브롬계화합물(Polybrominated diphenyl ethers, PBDEs)의 잔류농도와 조성특성)

  • Octopus(Octopus minor), benthic cephalopod, were collected from intertidal zone in Seosan, Choongnam and analyzed for polybrominted diphenyl ethers(PBDEs). PBDEs concentrations ranged from 29 to 109 ng/g lipid wt(mean; 54 ng/g) in mantle and from 12 to 89 ng/g lipid wt(mean; 48 ng/g) in internal organ. PBDEs concentrations in octopus internal organ were lower about two times than that in common squid collected in Yellow Sea, indicating Seosan is relatively less contaminated with PBDEs. Major congener was BDE 206, occupying 72% and 49% of total PBDE concentration in mantle and in internal organ, respectively, which is very different from PBDE composition in common squid with major congeners of BDE 47 and 99. From PBDE composition, octopus seemed to concentrate higher brominated BDE rather than lower brominated BDE. This PBDE composition in octopus may be affected by sediment with extremely high contribution of deca-BDE to total PBDE concentration.
