Activity Profiles of Linear, Cyclic Monomer and Cyclic Dimer of Enkephalin

  • The cyclic dimers of enkephalin were isolated as minor components during the solution synthesis of the corresponding cyclic monomers. The ratio of cyclic dimer to monomer was approximately 1:4 from the percent of yields. In the receptor binding assay of two cyclic dimmers, ($Tyr_2-C[D-Glu-Phe-gPhe]_2$ 6, $Tyr_2-C[D-Asp-Phe-gPhe-rLeu]_2$ 8), both analogs exhibited the high preference for ${\delta}$ receptor compared to monocyclic counterparts. In the nociceptive activity, both showed about 5 times less potent than the cyclic monomers. The repeated synthesis of 14-membered cyclic analog, Tyr-C[D-Glu-Phe-gPhe-D-rLeu] 14, which was known as having three distinct cis-trans isomers, gave rise to apparently different conformational analog arousing only trans isomer. In the receptor binding assay, it showed tremendously high selectivity toward ${\mu}$ receptor $({\delta}/{\mu}=160)$.

Synthesis and Molecular Structure of Calix[4]arene Butanoate 1,2-Alternate Conformer

  • Three conformational isomers of calix[4]arene butanoate were isolated from the reaction of calix[4]arene and butanoyl chloride in the presence of NaH and their structures were determined by NMR spectra as 1,2-alternate 2a, partial cone 2b and 1,3-alternate conformer 2c, respectively. The crystal structure of 2a has been determined by X-ray diffraction method. The crystals are monoclinic, space group C2/c, a=18.435 (4), b=13.774 (2), c=16.941 (3) Å, β=116.23 (1)°, Z=4, V=3858.8 (12)Å3, Dc=1.21 g cm-3, Dm=1.21 g cm-3. The molecule is in the 1,2-alternate conformation. It has two-fold symmetry axis along the line connecting between C (7AA') and C (7BB') parallel to the b axis of crystal lattice.

초임계 이산화탄소에서 리파아제-효소를 이용한 부탄올 구조이성질체의 에스테르화 반응의 분자 동역학 연구 (Molecular Dynamics Simulation Study of Lipase-catalyzed Esterification of Structural Butanol Isomers in Supercritical Carbon Dioxide)

  • 부탄올의 구조이성질체(n-, iso-, sec-, tert-butanol) 와 n-부티르산에 대한 리파아제 효소.촉매 에스테르화 반응이 초임계 이산화탄소 조건 하에서 수행되었다. 본 실험은 교반속도 150 rpm, 반응 온도 323.15 K, 반응 압력 150 bar의 조건으로 고압반응기에서 5 h 동안 수행하였다. 실험에 사용된 리파아제는 Candida Antarctica lipase B (CALB)이다. 실험 결과는 HP-INNOWax 컬럼을 이용하여 FID (Flame Ionization detector)가 장착된 기체 크로마토그래피(Gas Chromatography, GC)를 이용하여 분석하였다. 반응 후 생성물의 전환률과 반응의 경향성을 분자동역학 시뮬레이션을 이용하여 예측된 결과와 정성적으로 비교하였다. 경쟁적인 저해반응이 포함된 Ping-Pong Bi-Bi 메커니즘을 기초로 하여, 반응의 각 단계를 적용하여 구조 최적화를 하였고 이를 이용해 전이상태의 에너지를 구하여 반응의 경향성을 예측하였다. 생성되는 에스테르 이성질체의 구조적 선호도는 분자동역학 시뮬레이션을 통하여 분석하였다. 이러한 방법의 개발은 앞으로 컴퓨터를 이용한 효소 반응의 설계에 유용하게 사용될 수 있을 것이다.

Synthesis and Molecular Structure of p-tert-butylcalix[4]arene Hexanoate

  • Two conformational isomers of p-tert-butylcalix[4]arene hexanoate were prepared from the reaction of-p-tert-butylcalix[4]arene and hexanoly chloride in the presence of AlCl3 in CH2Cl2 and their structures were determined by NMR spectra and X-ray diffraction as a cone and a 1,3-alternate conformer, respectively. The crystal of cone conformer (C68H96O8·(CH3)2CO) is triclinic, P, a=15.066(1) , b=16.063(1) , c=16.365(1) , α=79.75(2)o, β=109.95(2)o, γ=80.32(0)o, V=3602.7(4) 3, Z=2. The intensity data were collected on Simens SMART diffractometer/CCD area detector. The structure was solved by direct method and refined by least-squares calculations to a final R value of 0.144 for 4638 observed reflections. The molecular conformation is distorted symmetric cone with the flattening A and D phenyl rings. The crystal of 1,3-alternate conformer (C68H96O8·2CHCl3) is orthorhombic, Pca21, a=34.586(5) , b=10.207(3) , c=20.394(4) , V=7199(3) 3, Z=4. The intensity data were collected on an Enraf-Noninus CAD-4 Diffractometer with a graphite monochromated Mo-K radiation. The structure was solved by direct method and refined by least-squares calculations to a final R value of 0.152 for 2241 observed reflections. The molecule has a pseudo mirror symmetric 1,3-alternate conformation.

Li+ and Li+I-Li+ ions Solvated by 1,4-dioxane: An ion Mobility Spectrometry-Mass Spectrometry Study

  • Electrospray ionization (ESI) and ion mobility spectrometry-mass spectrometry (IMS-MS) were employed to investigate the solvated structures of ionic species in the lithium iodide electrolyte solution in the gas phase. The Li+I-Li+ triple ion and single standalone Li+ ions solvated by 1,4-dioxane were successfully generated and observed by ESI-MS under the influence of dioxane vapor at the inlet region. Under the present experimental condition, (1,4-dioxane)m·Li+ complex ions (m = 1, 2, and 3) and a (1,4-dioxane)·Li+I-Li+ complex ion were observed, which were further examined by IMS to investigate their structures. The presence of multiple structural isomers was confirmed, which accounts for the endothermic conformational transition of 1,4-dioxane from a chair to a boat to achieve bidentate O-donor binding to Li+ and Li+I-Li+. Further structural details critical for the ion-solvent interactions were also examined and discussed with the help of density functional theory calculations.

  • $25^{\circ}C$의 1 : 4 dioxane-물의 혼합용액속에서 파라-치환된 phenyl N-(p-chlorobenzoyl)chloroformimidate (I) 유도체들의 가수분해 반응속도 상수를 측정하고 반응속도식, 치환기 효과$(\rho\>{\rho}^+)$, 생성물 분석 및 분자궤도 함수의 계산 결과로부터 pH3.0 이하에서는 azocarbonium 이온(II)이 생성되는 $S_N1$반응 메카니즘으로 무촉매 반응이 일어나며, pH 4.0이상에서는 전이상태(III)를 지나는 $S_N2$반응 메카니즘을 통하여 염기 촉매반응이 일어남을 제안 할 수 있었다. 4가지 peri planar형태 이성질체들의 상대적인 안정도는 각각 (E-ap) > (Z-ap) > (E-sp) > 및 (Z-ap)이었고, (E-ap)형태의 가장 안정한 입체구조는 benzimidochloroformyl group면에 대하여 Y-치환 phenyl group이 수직$(90^{\circ})$을 이루었으며 (I)의 활성화된 azomethine탄소 원자에 대하여 물분자는 시그마 공격에 의하여 친핵성 반응이 일어난다.

强酸性 溶液中에서 Cinnamonitrile의 加水分解 反應메카니즘 (Mechanism on the Hydrolysis of Cinnamonitrile in Strong Acid)

  • Cinnamonitrile의 산-가수분해 반응속도상수를 25$^{\circ}$C, HClO$_4$ 1 ~ 5M의 센산성 용액속에서 UV분광법으로 측정하고 Bunnett관계식에 적용하여 ${\omega}$ = 9.8, ${\omega}^*$ = 0.42 및 ${\phi}$=1.6등의 hydration parameter를 구하였다. 이는 질소원자에 양성자화가 이루어진 짝산에 대하여 속도결정단계에서 친핵체로 물분자가 첨가된 다음에 양성자 전달체로 작용한다는 것을 시사한다. Cinnamonitrile 분자의 궤도함수를 CNDO/2방법으로 계산한 바, 형태이성체의 안정도는 (E)-planar>(Z)-planar이였으며 음하전의 크기는 $C_8({\beta}){\ll}N$이였고 전이상태에서 물분자는 짝산의 양하전이 큰 $C_7({\alpha}$)원자에 대하여 ${\sigma}$접근함을 알았다. 이상의 결과로 부터, 센산성속에서 cinnamonitrile의 가수분해반응은 특정 산-촉매작용을 수반하는 A-2형의 산-가수분해 반응메카니즘에 의하여 진행됨을 알았다.

Subcellular partitioning-dependent functional switching of Arabidopsis photoreceptor phytochrome B in response to brassinosteroids

  • Many organisms control their physiology and behavior in response to the local light environment, which is first perceived by photoreceptors that undergo light-dependent conformational changes. Phytochromes are one of the major photoreceptors in plants, controlling wide aspects of plant physiology by recognizing the light in red (R) and far-red (FR) spectra. Higher plants have two types of phytochromes; the photo-labile type I (phyA in Arabidopsis) and photo-stable type II (phyB-E in Arabidopsis). Phytochrome B (phyB), a member of the type II phytochromes in Arabidopsis, shows classical R and FR reversibility between the inter-convertible photoisomers, Pr and Pfr. Interestingly, the Pr and Pfr isomers show partitioning in the cytosol and nucleus, respectively. In the over 50 years since its discovery, it has been thought that the type II phytochromes only function to mediate R light. As described in the text, we have now discovered phyB has an active function in FR light. Even striking is that the R and FR light exert an opposite effect. Thus, FR light is not simply nullifying the R effect but has an opposing effect to R light. What is more interesting is that the phyB-mediated actions of FR and R light occur at different cellular compartment of the plant cell, cytosol and nucleus, respectively, which was proven through utilization of the cytosolic and nuclear-localized mutant versions of phyB. Our observations thus shoot down a major dogma in plant physiology and will be considered highly provocative in phytochrome function. We argue that it would make much more sense that plants utilize the two isoforms rather than only one form, to effectively monitor the changing environmental light information and to incorporate the information into their developmental programs.
