Subcellular Distribution of Heavy Metals in Organs of Bivalve Modiolus Modiolus Living Along a Metal Contamination Gradient

  • Concentration and distribution of Fe, Zn, Cu, Cd, Mn, Pb, Ni among subcellular fractions (cellular membrane structures and cytosol) and Zn, Cu, Cd among cytoplasmic proteins in the kidney and digestive gland of mussel Modiolus modiolus living along a polymetallic concentration gradient were studied. It was found in the kidney of M. modiolus from contaminated sites that the Fe percent increased in the "membrane" fraction, whereas Zn, Pb, Ni and Mn percent increased in the cytosol compared to the kidney of the control mussel. Note kidney cytosol of M. modiolus from clean and contaminated sites sequestered major parts of Cu and Cd. In the digestive gland of M. modiolus from contaminated sites Fe, Zn, Cd, Mn, Ni percent increased in the "membrane" fraction, whereas Cu, Pb percent increased in the cytosol compared to digestive gland of control mussel. Gel-filtration chromatography shows kidney of M. modiolus contains increased metallothionein-like protein levels irrespective of ambient dissolved metal concentrations. It was shown that the metal detoxification system in the kidney and digestive gland of M. modiolus was efficient under extremely high ambient metal levels. However, under complex environmental contamination in the kidney of M. modiolus, the metal detoxification capacity of metallothionein-like proteins was damaged.

  • The aim of this study is to apply shape optimization technique for the repair of aging airframe components, which may extend fatigue life substantially. Free-form optimum shapes of a cracked part to be reworked or replaced are investigated with the objective to minimize the peak local stress concentration or fatigue-damage. Iterative non-gradient method, which is based on an analogy with biological growth, is employed by incorporating the robust optimization method to take account of the stochastic nature of the loading conditions. Numerical examples of optimal hole shape in a flat plate are presented to validate the proposed method. The method is then applied to determine the reworked or replacement shape for the repair of a cracked rib in the rear assembly wing body of aircraft.

산지 내 오염물질 확산의 2차원 수치해석 (Numerical Analysis of the Two-Dimensional Pollutant Dispersion Over Hilly Terrain)

  • Numerical prediction of the pollutant dispersion over a two-dimensional hilly terrain is presented. The dispersion model used in the present work is based on the gradient diffusion theory and the finite-volume method on a non-orthogonal boundary-fitted grid system. The numerical model is validated by comparing the results with the available experimental data for the flat-floor dispersion within a turbulent boundary-layer. The numerical error analysis is performed based on the guideline of Kasibhatla et al.(1988) for the elevated-source dispersion in the flat-floor boundary layer having a power-law velocity and linear eddy-diffusivity profile. The influences of the two-dimensional hilly terrain on the dispersion from a continuously released source are numerically investigated by changing the emission locations and heights. It is found that the distributions of ground-level concentration are strongly influenced by the source location and the emission height. Hence, the terrain amplification factor is greatly enhanced when the pollutant source is located within a flow separation region. Dispersion from a source of short duration is also simulated and the duration time of the pollutant is compared at several downstream locations on a hilly terrain. The results of the numerical prediction are applied to the evaluation of environmental impacts due to the automobile exhausts at the seashore highway with a parallel mountain range.

Relationship Between Acrylamide Concentration and Enzymatic Activity in An Improved Single Fibrin Zymogram Gel System

  • Based on the zymography analysis, Bacillus sp. DJ-4 (screened from Doen-Jang, a Korean traditional fermented food) secretes seven extracellular fibrinolytic enzymes (EFEs; 68, 64, 55, 45, 33, 27, and 13 kDa) in culture broth. These seven EFEs were analyzed by newly applied SDS-fibrin zymography combined with gradient polyacrylamide (SDS-FZGP). This improved gel system was used with a 5-20% acrylamide gradient in a fibrin zymogram gel for the separation of proteins with molecular masses from below 10kDa to over 100kDa on one gel plate. Using this system, high molecular weight bands (HMWBs) were clearly and sharply resolved. We also examined the relationship between an acrylamide concentration and the enzymatic activity of EFE using densitometric analysis.

  • The pressure tubes, which contain high temperature heavy water and fuel, are within the core of a CANDU nuclear reactor, and are thus subjected to high stresses, temperature gradient, and neutron flux. Further, it is well known that pressure tubes of cold-worked Zr-2.5Nb materials result in hydrogen diffusion, which create fully-hydrided regions (frequently called Blister). Thus a proper investigation of hydrogen diffusion within zirconium-alloy nuclear components, such as CANDU pressure tube and fuel channels is essential to predict the structural integrity of these components. In this respect, this paper presents numerical investigation of hydrogen diffusion to quantify the hydrogen concentration fur blister growth of CANDU pressure tube. For this purpose, coupled temperature-hydrogen diffusion analyses are performed by means of two-dimensional finite element analysis. Comparison of predicted temperature field and blister with published test data shows good agreement.

레이저빔에 의한 계면경사 Ni-Cr/steel 재료 제조에 관한 연구 (A study on the Fabrication of Graded-Boundary Ni-Cr/Steel Material by Laser Beam)

  • For a development purpose of thick metal / metal Graded-Boundary Materials(GBM), a basic research on the fabrication of Ni-Cr/steel GBM was carried out by a laser beam and its mechanical properties and thermal characteristics were investigated. In order to produce a compositionally graded boundary region between substrate steel and added Ni-Cr alloy, a series of surface alloying treatments was performed with a high power CO$_2$ laser beam. Ni-Cr sheet was placed on a low carbon steel plate(0.18%C), and then a CO$_2$ laser beam was irradiated on the surface to produce a homogeneous alloyed layer. On this first surface-alloyed layer, another Ni-Cr sheet was placed and then the CO$_2$ laser beam was irradiated again to produce second surface-alloyed layer. Sequential repetitions of laser surface alloying treatment 4 times resulted in a graded-boundary region with the thickness of about 1.4mm. Simultaneous concentration profiles of different kinds of alloying elements(Ni and Cr) showed from 42%Ni, 45%Cr and 13%Fe on surface region to 0%Ni, 0%Cr and 99%Fe in substrate region. Also a thermal conductivity gradient resulted in graded-region and its value changed from 0.03㎈/cm s$\^{C}$ in surface region to 0.1㎈/cm s$\^{C}$ in substrate region. Microstructural observation showed that any visible root porosities and solidification shrinkage cracks were not formed in graded region between alloyed layer and substrate region during rapid cooling.

Effect of Electrolyte Concentration on Surfactant-Enhanced Electrokinetic Removal of Phenanthrene

  • Surfactant-enhanced electrokinetic (EK) process was investigated to remove polycyclic aromatic hydrocarbons (PAHs) from low-permeable soils. Phenanthrene and kaolinite were selected as a representative PAH and a model soil, respectively. A nonionic surfactant Tergitol 15-S-12 was applied to improve the solubility of phenanthrene and sodium chloride was used as an electrolyte at the various concentrations from 0.001 to 0.1M. The addition of electrolyte affected both the removal efficiency and operation cost. When electrolyte was introduced, the electrical potential gradient became low and thus power consumption was reduced. However, as electrolyte concentration increased, the electroosmotic flow also decreased, so the removal efficiency of contaminant decreased. Therefore, the removal efficiency and power consumption should be considered simultaneously to determine the iptimum surfactant concentration, so a relatively lower concentration of electrolyte than certain value is desired.

  • Finite-difference analysis was conducted to study the natural convection of a stably stratified salt-water solution in an annulus due to lateral heating. The main purpose of this study is to examine in detail the multi-layered flow structure. Calculation was thus made for R $a_{\eta}$=2*10$^{5}$ and 6.5*10$^{5}$ . Formation of layered flow structure, merging process of layers, the corresponding temperature and concentration distributions, Nusselt number variations with time are examined. Numerical results show that in each layer, the temperature profile looks 'S`-shaped and the concentration profile is uniform due to the convective mixing. The formation of the roll and the layer is governed by natural convection due to the temperature gradient and the merging process of the layer by diffusion of the concentration.ation.

헬륨$\cdot$공기흡합기농도분포에 관한 실험적 연구 (An experimental study on the concentration distribution of helium and air mixture in the direct injection type engine)

  • This study has been conducted by experiments for distribution of concentration of helium gas, which is jetted into stationary atmosphere at the normal temperature and pressure. It is able to obtain the data for concentration of helium and air mixtures by the use of hot wire probe which has fast response. At an up stream, the concentration gradient which is attained is steep. At a down stream, the mixing time of helium and air is gradually shortened with the lapse of time in front of a jet. The arrival frequency of a jet in an unsteady area is mostly constant from 0% to 100% up to 80mm, but the time which is reaching to 100% is gradually to lengthen as a descending downstream. After starting a jet and the point of 90%, the mixing time is especially to lengthen. This reason comes from the turbulent intensity which causes for mixing of helium and air. This time difference which causes according to lengthen a jet should be considered in the design of combustion chamber.

마이크로 유동에서 PtOEP/PS 박막을 이용한 용존 산소 농도장의 정량적 가시화 (Quantitative Visualization of Dissolved Oxygen Concentration Field in Micro Flows using PtOEP/PS Membrane)

  • It is highly needed to measure the dissolved oxygen (DO) concentration field in water for a variety of purposes such as biological, industrial, environmental monitoring and medical application. Application of PSP (Pressure Sensitive Paint) which is sensitive to oxygen concentration has been carried out to measure DO concentration field using PtOEP/PS film and intensity based method under the UV-LEDs illumination. A micro round water jet having 100% of DO was obliquely impinged on to a PtOEP/PS film coated plate placed in a 0% of DO water container. DO concentration fields on the impinging plate were quantitatively visualized with a $2.94\;{\mu}m$ of spatial resolution. Through pixel-by-pixel calibration, uncertainty of each pixel by different sensitivity, different dye concentration and non-uniformity of illumination was removed. It is demonstrated that the high DO concentration region was coincided with the impingement area. The DO concentration gradient due to DO diffusion was affected by Reynolds number.
