면역계 질환 치료제로서의 인삼의 생리활성 성분의 개발 연구

  • 인삼이 면역계 질환의 일종인 과민성 반응에 대한 치료약으로서의 가능성을 연구하기 위해서 in viva와 in vitra실험을 수행하였다. in vitra실험으로서는 hyaluronidase의 활성을 지표로 삼았으며, in viva실험에서는 mouse이각, rat등피부, 복강 비만세포, 그리고 guinea pig의 ileum에서 면역반응에 대한 아답타겐의 작용을 관찰하였다. in uitra 실험에서는 hyaluronidase와 histamine 분비를 항알레르기 작용의 지표로 사용하였다. hyaluronidase의 경우 그 활성은 Cacl2로 활성화 시키기 전이나 후에 모두 아답타겐(100, 50, 및 10 mg/ml)에 의해서 현저하게 억제되었다. 한편 rat 복강 비만 세포에서의 histamine유리 시험에서는 아답타겐(0.5, 1, 5mg/ml)이 histamine유리가 증가되었다. Guinea pig의 ileum 평활근에서는 아답타겐이 평활근 자극 작용없이 항히스타민 효과률 나타내었다. in uiua 실험의 첫 단계로 우리는 혈관 투과성에 미치는 아답타겐의 효과를 mouse 이각과 rat 등피부에서 관찰하였다. Mouse이각에서의 실험에서는 hoistamine, serotonin, 그리고 LTC4로 challenge하여 관찰한 결과 histamine과 serotonin의 경우, 혈관투과성 항진은 아답타겐(50-400mg/m1)에 의해서 유의성있게 억제되었으나, rat 등 피부에서는 mouse에서와는 달리 48-hr PCA가 증가 되었고, histamine에 의한 혈관투과성 항진은 아답타겐에 의해 영향을 받지 않았다. 반면에 compound 48/80과 serotonin에 의한 혈관투과성 항진은 mouse 의 경우에서처럼 아답타겐에 의해 억제되었다.

Quantification of Caffeoylquinic Acids and the Effect of the Cacalia hastata var. orientalis Extract on Acetylcholinesterase Activity and Peroxynitrite Concentration (민박쥐나물 추출물의 Caffeoylquinic Acid 정량 및 Acetylcholinesterase 활성과 Peroxynitrite 농도에 대한 효과)

  • To find the anti-Alzheimer's activity of Cacalia hastata var. orientalis (Compositae), caffeoylquinic acids were analyzed in this plant by HPLC and the inhibitory activities of the extract on cholinesterases including acetylcholinesterase (AChE) and butyrylcholinesterse (BChE) and peroxynitrite ($ONOO^-$) were assayed. Sum of caffeoylquinic acids in the 80% MeOH extract was quantitatively higher in leaves (35.8% of the extract) than in stems (3.7%). The compound, 3,5-dicaffeoylquinic acid, was contained in the highest amount in the leaf (44.32 mg/g dry weight). The $IC_{50}s$ of the 80% MeOH extract were shown to be $67.45{\mu}g/ml$ for AChE, and $8.59{\mu}g/ml$ (for $ONOO^-$), respectively, suggesting that the leaves would have the anti-Alzheimer's activity due to the high content of caffeoylquinic acids.

Inhibitory effects of Polygoni cuspidati rhizoma on Mast Cell-Mediated Anaphylactic Reaction

  • Polygoni cuspidati Rhizoma (PCRH) has been used in treatment of menoxenia, skin burn, gallstone, hepatitis, hyperlipidaemia, favus athlete's foot, supperative dermatitis and inflammation. However, its effect in experimental models remains unknown. In this present study, the effect of PCRH for stability on mast cell was analyzed. Two g/kg PCRH inhibited the compound 48/80-induced anaphylaxis by 75%. In addition, PCRH inhibited the tumor necrosis factor-$\alpha$ and interleukin-8 secretion as compared with the phorbol 12-myristate 13-acetate plus calcium ionophore A23187 stimulated human mast cell line, HMC-1 cells. These results suggested PCRH may inhibit mast cell-mediated anaphylactic reaction.

Study on the Function of Hyunsamchungpye-eum on Anti-inflammatory and Anti-allergic Action (현삼청폐음이 항염 및 항알러지 작용에 관한 연구)

  • The purpose of this research was to investigate effects of Hyunsamchungpye-eum(HSCPE) on the anti-inflammatory/anti-allergic reaction in vivo and in vitro. HSCPE reduced the acute hind paw edema induced by histamine, the permeability of evans blue into peritoneal cavity. HSCPE inhibited the passive cutaneous anaphylaxis reaction in rat, the lethal anaphylaxis and degranulation of peritoneal mast cells induced by compound 48/80 in mice, HSCPE did not affect the Arthus reaction, but decreased the delayed type hypersensitivity induced by SRBC, contact dermititis induced by DNFB. These results suggest that HSCPE have an anti-inflammatory and anti-allergic action.

  • The purpose of this research was to investigate the effect of Yonggak-san (YGS) on the anti-allergic reaction in vivo and in vitro. Administration of YGS(500 mg/kg) enhanced hemaggutination(HA)titer against SRBC. On the while, YGS inhibited hyaluronidase activity in vitro and passive cutaneous anaphylaxis reaction, lethal anaphylaxis and mortality induced by compound 48/80 in mice, YGS decreased Arthus reaction, acute hind paw edema induced by histamine. But YGS did not affect delayed type hypersensitivity induced by SRBC. These results suggest that YGS have anti-allergic action

The Beneficial Effect of Avocado on Skin Inflammation in a Mouse Model of AD-like Skin Lesions

  • Avocado, superfood, contains a variety of essential nutrients and phytochemicals. The purpose of this study was to explore whether avocado could modulate skin inflammation in vivo. We elucidated the pharmacological effects of avocado on compound 48/80- or histamine-induced scratching behaviors and 2, 4-dinitrochlorobenzene (DNCB)-induced atopic dermatitis (AD)-like skin lesions in mice. Additionally, we investigated the anti-inflammatory activity of avocado and its underlying mechanism including its effect on the expression levels of inflammatory-related genes and nuclear factor-κB (NF-κB) in DNCB-induced AD-like skin lesions. The findings of this study demonstrate that avocado attenuated AD-clinical symptoms including itching, eczematous, erythema and dryness and histamine levels in mice. Moreover, avocado suppressed both inflammatory cytokines expression as well as NF-κB and caspase-1 activation in AD-like skin lesions in mice. Taken together, these results demonstrate that avocado may be a potential candidate for treating skin inflammatory diseases like AD.

Anti-allergic Property of Chungjokupye-tang (정조구폐탕의 항알레르기 효과에 관한 연구)

  • The purpose of this research was to investigate the effects of Chungjokupye-tang(CJKPT) on the anti-allergic property. CJKPT decreased the hyaluronidase activity in vitro, and inhibite the anaphylaxis induced by compound 48/80 and suppress the degranulation of peritoneal mast cell. And CJKPT also decreased the passive cutaneous anaphylaxsis, arthus reaction and contact dermititis by DNFB. These results suggest that CJKPT have an anti-allergic property.

  • In the present study, the alcoholic extract of Terminalia arjuna (TA) and Arjunolic acid (AA) were studied fur its anti-asthmatic and anaphylactic activity. Treatment with TA (250 & 500 mg/kg) and AA (50 & 100 mg/kg) has shown significant protection against mast cell disruption in rats induced by compound 48/80. TA and AA also protected the guinea pig against histamine as well as acetylcholine induced bronchospasm. Both TA & AA exhibited better protection against histamine release than against acetylcholine release. Anti-asthmatic and anaphylactic activity may be possibly due to membrane stabilizing potential and inhibition of antigen induced histamine and acetylcholine release.

Effect of Ripe Fruits of Rubus coreanus on Anaphylactic Allergic Reaction and Production of Cytokines

  • The anaphylactic allergic reaction is involved in many allergic diseases such as asthma and allergic rhinitis. In this report, I investigated the effect of ripe fruits of Rubus coreanus Miq. (Rosaceae) (RFRC) on the anaphylactic allergic reaction and studied its possible mechanisms of action. RFRC inhibited compound 48/80-induced systemic anaphylactic reaction in mice. RFRC dose-dependently decreased the IgE-mediated passive cutaneous anaphylaxis. In addition, RFRC decreased the gene expression and production of tumor necrosis factor-$\alpha$ (TNF-$\alpha$) and interleukin (IL)-6 in phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore A23187 (A23187)-stimulated human mast cells. These findings provide evidence that RFRC could be a candidate as an anti-allergic agent.

Inhibitory Effects of the Extracts from Eleutherococcus senticosus Maxim. on Histamine-release from Rat's Mast Cell

  • Eleutherococcus senticosus Maxim. has been successfully used as an oriental medicine for various diseases including allergic disorders. Histamine is a major factor on various allergic responses and it is reported that histamine was released from mast cells by sensitization of allergens. In this study, ethanol extracts from E. senticosus Maxim. were prepared and the composition was analyzed by high performance liquid chromatography. The eleutheroside B as a primary effective component of E. senticosus was contained approximately 225 mg/kg in root bark extracts. The extracts were found to significantly inhibit compound 48/80-induced histamine release form mast cells in dose dependent manner. However the extracts had low cytotoxicity on the mast cells with MTT assay. These results showed that E. senticosus Maxim. extracts may be the effective materials on inflammatory disorders.
