세포유전적 유형이 다른 무릇(Scilla scilloidise Complex)에서 유도된 캘러스 세포의 염색체 변이 (Chromosome Variation in Callus Cells Derived from Different Cytogenetic Type Plants of Scilla scilloides Complex)

  • 세포유전적 유형이 다른 무릇 (Scilla scilloidise Coplex)의 조직배양에서 유도된 캘러스에서 염색체 변이를 조사하였다. AA유형의 캘러스에서 심한 수적, 구조적 변이가 관찰된 반면, BB 유형의 캘러스 세포에서는 상염색체의 변이가 발견되지 않았다. 이질 4배체인 AABB유형에서는 두개의 hypoploid 세포만 관찰되었으나, B게놈의 진정4배체인 BBBB유형에서는 hypoploid 세포와 함께 hyperploid 세포가 관찰되었다. 캘러스 세포에서 상염색체의 안정성은 B염색체를 지닌 식물에서 유도된 캘러스에서 더 높게 나타났다. 이는 배양세포에서 B염색체가 상염색체의 안정성에 선택적으로 기능하기 때문인 것으로 여겨진다.

수도(水稻)에 대(對)한 심층추비효과(深層追肥効果)에 관(關)한 연구(硏究) - Zeolite 첨가(添加) Ball complex 비료(肥料)의 비효(肥効) - (Study on the Effect of Deep Fertilization on Paddy Field - Efficiency of Ball Complex Fertilizer Mixed with Zeolite -)

  • 수도(水稻)에 대(對)한 Zeolite 첨가(添加) Ball Complex 비료(肥料)의 심층(深層) 추비(追肥) 방법(方法)과 현행(現行) 표층(表層) 추비법(追肥法)과의 비효(肥効)를 비교(比較)했다. 대조구(對照區)와 Ball Complex 구(區)의 기비(基肥) 질소(窒素)를 동량(同量)으로 시비(施肥)하고 질소(窒素) 추비(追肥)는 대조구(對照區) 3회(回) 표층(表層) 시비(施肥)를 했고, Ball Complex 구(區)는 출수(出穗) 전(前) 35일(日)에 12~13cm 깊이로 4주간(株間)에 한개(個)씩 심층(深層) 추비(追肥)를 1회(回)했을 때 얻어진 결과(結果)를 요약(要約) 하면 다음과 같다. 1. Ball Complex 구(區) 수도체(水稻體)는 질소(窒素), 가리(加里)를 수확기(收穫期)까지 지속적(持續的)으로 흡수(吸收)하는데 비(比)해서 대조구(對照區) 수도체(水稻體)는 출수(出穗) 후(後) 부터 이들 양분(養分)의 흡수(吸收)는 급격(急激)히 감소(減少) 경향(傾向)을 보인다. 등숙기(登熟期)에 있어서 수도체(水稻體)의 질소(窒素), 가리(加里) 일당(日當) 흡수량(吸收量)은 대조구(對照區)가 주당(株當) 각각(各各) 0.45mg, 0.68mg에 불과(不過)하나 Ball Complex 구(區)는 각각(各各) 4.8mg 7.0mg 이다. 2. 물질(物質) 생산 속도(速度)는 각(各) 처리구(處理區) 수도(水稻)의 양분(養分) 흡수(吸收) 양상(樣相)이 잘 반영(反映)되어 대조구(對照區)는 출수(出穗) 직후(直後) 최고(最高) 값을 나타 내였다가 등숙기(登熟期)에는 떨어지지만 Ball Complex 구(區)는 오히려 등숙기(登熟期)에 크다. 3. Bali Complex 심층(深層) 추비구(追肥區)는 무효(無効) 분얼(分蘖)이 적어 유효(有効) 경율(莖率)이 높기 때문에 주당(株當) 수수(穗數)는 대조구(對照區) 수도(水稻) 보다 많다. 4. Ball Complex 심층(深層) 추비(追肥)는 수당(穗當) 완전입수(完全粒數), 등숙율(登熟率), 천입중(千粒重) 모두 다 크다. 5. 수량(收量)은 10a당 대조구(對照區)가 423kg이고 Ball Complex 구(區)는 528kg로서 25%가 많았다. 6. 심층(深層) 추비(追肥) 수도체(水稻體)의 형태적(形態的) 특징(特徵)은 지엽(止葉), 제(第)2엽(葉)의 엽신장(葉身長), 엽(葉) 면적(面積)이 표층(表層) 추비(追肥) 수도체(水稻體) 보다 크다. 7. Ball Complex 비료(肥料)의 심층(深層) 추비(追肥)는 등숙기간중(登熟期間中) 질소(窒素), 가리(加里)의 흡수(吸收)가 많아서 엽신중(葉身中)의 질소(窒素) 농도(濃度)를 높이 유지(維持)하기 때문에 수도체(水稻體)의 지엽(止葉), 제(第)2엽(葉)의 엽신(葉身) 신장(伸長)을 촉진(促進)하여 엽(葉) 면적(面積)이 넓어지고 엽신중(葉身重)이 무겁고 단위(單位) 동화능(同化能)을 높여 광합성(光合成) 능력(能力)을 향상(向上) 시킨다. 또한 표층(表層) 추비(追肥) 수도체(水稻體) 보다 하엽(下葉) 고사(枯死)가 적고 생엽수(生葉數)가 많아 수도체(水稻體) 군(群)의 엽면적(葉面積) 지수(指數)의 감소(減少)를 적게 하므로 본시험(本試驗)에서는 현행(現行) 표층(表層) 추비법(追肥法) 보다 더 증수(增收)가 가능(可能)하였다. 8. 이상(以上)과 같은 결과(結果)로 보아서 Ball Complex 비료(肥料)는 현존(現存) 비료(肥料)의 시비(施肥)보다 추비효과(追肥効果)가 컸으며, 추비(追肥) 방법(方法)은 심층(深層) 추비(追肥)가 현행(現行) 표층(表層) 추비법(追肥法) 보다 훨씬 좋았다.

고리 원자력 발전 단지 사고 발생에 따른 방사능 물질 확산 가능성의 계절적 특성 연구 (Numerical Estimates of Seasonal Changes of Possible Radionuclide Dispersion at the Kori Nuclear Power Plants)

  • To establish initial response scenarios for nuclear accidents around the Kori nuclear power plants, the potential for radionuclide diffusion was estimated using numerical experiments and statistical techniques. This study used the numerical model WRF (Weather Research and Forecasting) and FLEXPART (Flexible Particle dispersion model) to calculate the three-dimensional wind field and radionuclide dispersion, respectively. The wind patterns observed at Gijang, near the plants, and at meteorological sites in Busan, were reproduced and applied to estimates of seasonally averaged wind fields. The distribution of emitted radionuclides are strongly associated with characteristics of topography and synoptic wind patterns over nuclear power plants. Since the terrain around the power plants is complex, estimates of radionuclide distribution often produce unexpected results when wind data from different sites are used in statistical calculations. It is highly probable that in the summer and autumn, radionuclides move south-west, towards the downtown metropolitan area. This study has clear limitations in that it uses the seasonal wind field rather than the daily wind field.

Visualization of Multicolored in vivo Organelle Markers for Co-Localization Studies in Oryza sativa

  • Eukaryotic cells consist of a complex network of thousands of proteins present in different organelles where organelle-specific cellular processes occur. Identification of the subcellular localization of a protein is important for understanding its potential biochemical functions. In the post-genomic era, localization of unknown proteins is achieved using multiple tools including a fluorescent-tagged protein approach. Several fluorescent-tagged protein organelle markers have been introduced into dicot plants, but its use is still limited in monocot plants. Here, we generated a set of multicolored organelle markers (fluorescent-tagged proteins) based on well-established targeting sequences. We used a series of pGWBs binary vectors to ameliorate localization and co-localization experiments using monocot plants. We constructed different fluorescent-tagged markers to visualize rice cell organelles, i.e., nucleus, plastids, mitochondria, peroxisomes, golgi body, endoplasmic reticulum, plasma membrane, and tonoplast, with four different fluorescent proteins (FPs) (G3GFP, mRFP, YFP, and CFP). Visualization of FP-tagged markers in their respective compartments has been reported for dicot and monocot plants. The comparative localization of the nucleus marker with a nucleus localizing sequence, and the similar, characteristic morphology of mCherry-tagged Arabidopsis organelle markers and our generated organelle markers in onion cells, provide further evidence for the correct subcellular localization of the Oryza sativa (rice) organelle marker. The set of eight different rice organelle markers with four different FPs provides a valuable resource for determining the subcellular localization of newly identified proteins, conducting co-localization assays, and generating stable transgenic localization in monocot plants.

Tomato Yellow Leaf Curl China Virus Impairs Photosynthesis in the Infected Nicotiana benthamiana with βC1 as an Aggravating Factor

  • Tomato yellow leaf curl China virus is a species of the widespread geminiviruses. The infection of Nicotiana benthamiana by Tomato yellow leaf curl China virus (TYLCCNV) causes a reduction in photosynthetic activity, which is part of the viral symptoms. ${\beta}C1$ is a viral factor encoded by the betasatellite DNA ($DNA{\beta}$) accompanying TYLCCNV. It is a major viral pathogenicity factor of TYLCCNV. To elucidate the effect of ${\beta}C1$ on plants' photosynthesis, we measured the relative chlorophyll (Chl) content and Chl fluorescence in TY-LCCNV-infected and ${\beta}C1$ transgenic N. benthamiana plants. The results showed that Chl content is reduced in TYLCCNV A-infected, TYLCCNV A plus $DNA{\beta}$ (TYLCCNV A + ${\beta}$)-infected and ${\beta}C1$ transgenic plants. Further, changes in Chl fluorescence parameters, such as electron transport rate, $F_v/F_m$, NPQ, and qP, revealed that photosynthetic efficiency is compromised in the aforementioned N. benthamiana plants. The presense of ${\beta}C1$ aggravated the decrease of Chl content and photosynthetic efficiency during viral infection. Additionally, the real-time quantitative PCR analysis of oxygen evolving complex genes in photosystem II, such as PsbO, PsbP, PsbQ, and PsbR, showed a significant reduction of the relative expression of these genes at the late stage of TYLCCNV A + ${\beta}$ infection and at the vegetative stage of ${\beta}C1$ transgenic N. benthamiana plants. In summary, this study revealed the pathogenicity of TYLCCNV in photosynthesis and disclosed the effect of ${\beta}C1$ in exacerbating the damage in photosynthesis efficiency by TYLCCNV infection.

원자력발전소의 출력감발모드를 위한 경보축약 규칙 (Practical Alarm Suppression Rules and their Implementation for Nuclear Power Plants)

  • It is necessary to adopt some logical techniques and methods of alarm processing for a large complex plant such as nuclear power plants in order to present the occurred alarm messages properly and concisely. Among such alarm processing techniques, the alarm suppressing function is a strong tool to avoid alarm flooding during the sudden transients of plant output power such as turbine trips, reactor trips and other incidents. Unless any suppression or representation technologies are used in an alarm message listing system, it cannot provide quick assistance to plant operators or supervisors during plant upsets because too many alarm messages are presented in an alarm list window. This paper presents the key suppression methods and analysis processes developed for implementing a suppressed alarm message listing function of an integrated alarm system called LogACTs which has been applied to a CANDU nuclear power plant. A simulation testing of the suppressing function conducted with the real plant alarm message list data has demonstrated an effective performance of the developed logics with the high suppression rate.

  • Chemical plants with high-technological equipments and complex processes are continually increasing potential hazards that may result in huge industrial accidents. If an incident breaks out, it will have a hazardous effect on in-plant workers, neighboring inhabitants or the surrounding environment. Therefore, it is necessary to develop a safety management system that can pre-evaluate the safety and reduce the risk of a chemical plants. The safety management evaluation program developed in this study can implement an effective loss control management in chemical plants. It integrates important lists in safety management lists and legal measures to analyze elements systematically. It also includes incident case analysis and the required components in domestic laws.

Xanthomonas oryzae pv. oryzae triggers complex transcriptomic defense network in rice

  • High throughput transcriptome investigations of immunity in plants highlight the complexity of gene networks leading to incompatible interaction. To identify genes crucial to resistance against Xanthomonas oryzae pv oryzae, functional genetic analysis of selected differentially expressed genes from our microarray data set was carried out. A total of 13 overexpression vector constructs were made using 35S CaMV promoter which drive constitutive expression in rice. Most of the genes are developmentally expressed especially during maximum tillering stage and are commonly highly expressed in the leaves. When screened against Xoo strain K2, the transgenic plants displayed shorter lesion length compared with wild type Dongjin which indicates partial resistance. The levels of ROS continuously magnified after inoculation which indicates robust cellular sensing necessary to initiate cell death. Elevated transcripts levels of several defense-related genes at the downstream of defense signal network also corroborate the phenotype reaction of the transgenic plants. Moreover, expression assays revealed regulation of these genes by cross-communicating signal-transductions pathways mediated by salicylic and jasmonic acid. These collective findings revealed the key immune signaling conduits critical to mount full defense against Xoo.

MODELING OF NONLINEAR CYCLIC LOAD BEHAVIOR OF I-SHAPED COMPOSITE STEEL-CONCRETE SHEAR WALLS OF NUCLEAR POWER PLANTS

  • In recent years steel-concrete composite shear walls have been widely used in enormous high-rise buildings. Due to high strength and ductility, enhanced stiffness, stable cycle characteristics and large energy absorption, such walls can be adopted in the auxiliary building; surrounding the reactor containment structure of nuclear power plants to resist lateral forces induced by heavy winds and severe earthquakes. This paper demonstrates a set of nonlinear numerical studies on I-shaped composite steel-concrete shear walls of the nuclear power plants subjected to reverse cyclic loading. A three-dimensional finite element model is developed using ABAQUS by emphasizing on constitutive material modeling and element type to represent the real physical behavior of complex shear wall structures. The analysis escalates with parametric variation in steel thickness sandwiching the stipulated amount of concrete panels. Modeling details of structural components, contact conditions between steel and concrete, associated boundary conditions and constitutive relationships for the cyclic loading are explained. Later, the load versus displacement curves, peak load and ultimate strength values, hysteretic characteristics and deflection profiles are verified with experimental data. The convergence of the numerical outcomes has been discussed to conclude the remarks.

아파트단지 조경수목의 식재하자에 관한 연구 (Defects of Planting in Landscape Plants in Apartment Complex)

  • The purpose of this thesis was to provide the basic data for landscape architecture of apartment constructions and to minimize the users´ and builder´ loss in terms of finance and landscape due to defects in planting. After a review of literature related to defects of planting work, the researcher investigated the defects in planting of landscape plants on the basis of data from the drawing and defect-repair construction of 50 civil apartment complexes in Daegu and Kyongbook are from 1994 to 1998. The defect-ratio was analysed in terms of the species, shapes, and sizes of trees. It provides matters for consideration in terms of the design of the landscape planting on the apartment unit. It also provided the future directions for landscape architecture in apartment constructions with regard to the selection of the planting trees, etc., given statistics on defect occurrence. The causes of defect of the landscape planting trees were not studied accurately in this study due to the board range of researched area, the differences of the planting ground environment, the management ability, and the parameters of judging planting defects. It is recommended that those areas should be researched in the future.
