Effect of Solvent Extraction on the Anti-complementary Activities of Green and Ripe Cucurbita moschata Duch

  • The edible part of cucurbita moschata Duch, which is commonly used as a Korean traditional medicine as well as a popular food source, was studied to isolate anti-complementary substance. Extracts of Cucurbita moschata Duch showed significant anti-complementary activities on the classical pathway of the complement system. Especially, the ripe Cucurbita moschata Duch had more activity than that of the complement system. Especially, the ripe Cucurbita moschata Duch had more activity than that of the green one in terms of the overall anti-complementary activity. Among the extracts of various organic solvents of the ripe Cucurbita moschata Duch, chloroform and ethyl-acetate extracts, which are non-polar solvent extracts, showed the strongest activities. These results suggest that the major difference in the solvent extraction for the anti-complementary substances depends on the change in the chemical composition such as the fatty acid with the degree of ripening.

Anticomplementary and Antitumor Activities of the Alkali Extract from the Mycelia of Lentinus edodes 1'11105 (Lentinus edodes IY105 알칼리 추출물의 보체계활성 및 항종양효과)

  • Alkali extract obtained from mycelia of Lentinus edodes IY105 was shown to have potent anticomplementary activity and alternative complementary activity in vitro. It was also shown to activate reticuloendothelial system of ICR mice in the carbon clearance. Amount of carbohydrates and protein of the extract were 17% and 42% respectively. It was found that carbohydrates were consisted of 5 monosaccharides and protein was consisted of 16 amino acids. Antitumor activity was observed in the alkali extract treated group of sarcoma 180 bearing ICR mice.

  • Studies on modified complement fixation (MCF) test of antiblackleg bovine serum were made and the results obtained were summarized as followings. 1. The most satisfactory antigen for the MCF test among various materials studied was found to be the vegetative cells of Cl, chauvoei grown in cooked meat medium (CMM) containing 0.5mM l-(alpha) alanine and 0.1mM manganese. The antigen was prepared by inoculating the spores of Cl. chauvoei, heated at $70^{\circ}C$. for 30 minutes, into the CMM followed by incubation at $37^{\circ}C$. for 15 hours. 2. An active component contained in the factor serum of fresh normal rabbit serum was found to be C'4 fraction. It was also shown that, furthermore, DEAE cellulose sieved C'4 fraction of the factor serum enhanced antibody titer and the highest antibody titer was resulted by the addition of 0.03 ml, of the factor serum to each tube. 3. More than four fold increases of antibody titer, in antiblbckleg bovine serum-antigen system, was made with the MCF test than that with the direct complement fixation test. 4. The MCF antibody titer of cattle vaccinated against blackleg was 128 until seven month and 64 for five months thereafter.

  • The extracorporeal circulation has been much improved recently, but has yet much complex problems such as the protein denaturation and the activation of the complement system by the exposure of the blood to the foreign surface, which may result in such as the postperfusion syndrome. We studied the changes of the plasma protein fractions by the electrophoresis and the complement consumption [C3, C4] by the immunodiffusion method in the patients undergoing cardiac operation from Mar. 1, 1986 to Aug. 31, 1986. The results were summarized as follows: 1. y-globulin fraction was decreased [p<0.02 by paired t-test, N=25], but a,-globulin was increased [p<0.001 by paired t test, N=25] after operation. 2. C3,C4 were significantly reduced [p<0.001 by paired t-test, N=14] postoperatively and normalized from 24 hours after operation. 3. The consumption of C3,C4 had significant linear correlation [correlation coefficient r=0.97] and C, was more markedly reduced comparing with C3, which probably means the complement activation by classical pathway in our bubble oxygenator group.

  • With the rapid increase in the elderly population, numerous problems arise related to the decreased cognitive function such as the declines of reaction time and concentration which are both resulting from ageing. Recently, ICT-based smart home and other residential services are being developed as a means of complementing human cognitive functions, but the elderly, who are in most cases digital-illiterate, find it hard to utilize these tools actively and if so it usually takes longer time and endeavor for the elderly to be familiarized with ICT devices. In this context, this study suggests a configuration of the holographic system in houses that complement the cognitive function of the elderly with the consideration of the interaction with the vision which is a critical function in the cognitive process. The method of this study consists of a literature review on topics including the characteristics of the cognitive function among the elderly, holographic technology and residential services; a configuration of residential services for different space types, depending on the aspects of the cognitive function among the elderly; and finally a suggestion of a holographic system configuration and application depending on various scenarios.

  • "Samsung Composite Shear Wall System" has its basis on "Samung Able System". "Samsung Able System" has some problems not only in the connection structure but also in waterproofing and soundproofing. We developed "Samsung Composite Shear Wall System" in order to solve these matters and expand PC system. "Samsung Composite Shear Wall System" is the compromise and complement of all the merits of full PC, half PC and in-situ concrete system.

C1qa deficiency in mice increases susceptibility to mouse hepatitis virus A59 infection

  • Background: Mouse hepatitis virus (MHV) A59 is a highly infectious pathogen and starts in the respiratory tract and progresses to systemic infection in laboratory mice. The complement system is an important part of the host immune response to viral infection. It is not clear the role of the classical complement pathway in MHV infection. Objectives: The purpose of this study was to determine the importance of the classical pathway in coronavirus pathogenesis by comparing C1qa KO mice and wild-type mice. Methods: We generated a C1qa KO mouse using CRISPR/Cas9 technology and compared the susceptibility to MHV A59 infection between C1qa KO and wild-type mice. Histopathological and immunohistochemical changes, viral loads, and chemokine expressions in both mice were measured. Results: MHV A59-infected C1qa KO mice showed severe histopathological changes, such as hepatocellular necrosis and interstitial pneumonia, compared to MHV A59-infected wild-type mice. Virus copy numbers in the olfactory bulb, liver, and lungs of C1qa KO mice were significantly higher than those of wild-type mice. The increase in viral copy numbers in C1qa KO mice was consistent with the histopathologic changes in organs. These results indicate that C1qa deficiency enhances susceptibility to MHV A59 systemic infection in mice. In addition, this enhanced susceptibility effect is associated with dramatic elevations in spleen IFN-γ, MIP-1 α, and MCP-1 in C1qa KO mice. Conclusions: These data suggest that C1qa deficiency enhances susceptibility to MHV A59 systemic infection, and activation of the classical complement pathway may be important for protecting the host against MHV A59 infection.

Mapping of the Complement C9 Binding Region on Clonorchis sinensis Paramyosin

  • Heliminthic paramyosin is a multifunctional protein that not only acts as a structural protein in muscle layers but as an immune-modulatory molecule interacting with the host immune system. Previously, we found that paramyosin from Clonorchis sinensis (CsPmy) is bound to human complement C9 protein (C9). To analyze the C9 binding region on CsPmy, overlapping recombinant fragments of CsPmy were produced and their binding activity to human C9 was investigated. The fragmental expression of CsPmy and C9 binding assays revealed that the C9 binding region was located at the C-terminus of CsPmy. Further analysis of the C-terminus of CsPmy to narrow the C9 binding region on CsPmy indicated that the region flanking 731Leu-780Leu was a potent C9 binding region. The CsPmy fragments corresponding to the region effectively inhibited human C9 polymerization. These results provide a precise molecular basis for CsPmy as a potent immunomodulator to evade host immune defenses by inhibiting complement attack.

  • Surveying and GIS have increasingly contributed to improve the quality of humane life through the development of its technology. While the surveying professions are now experiencing the crisis of identity. In this paper, we aim to study the concepts and presence of newly used terminologies, present state and future vision of surveying and GIS in Korea. To cope with various surveying consumers for expanding the market and their demands, enhancement of educational system, complement of laws legal system, construction of cooperative system in business are basically required.
