Comparison of Gene Expression Patterns in Longissimus dorsi of Pigs between the High-parent Heterosis Cross Combination andrace×Large White and the Mid-parent Heterosis Cross Combination Large White×Meishan

  • In order to detect the molecular mechanism of heterosis in pigs, the mRNA differential display technique was performed to investigate the differences in gene expression of pig's Longissimus dorsi between the high-parent heterosis cross combination Landrace${\times}$Large White and the mid-parent heterosis cross combination Large White${\times}$Meishan. Three pig purebreds, Large White, Meishan, and Landrace and four types of reciprocal $F_1$ hybrids were analyzed using nine 3'-end anchored primers in combination with ten 5'-end arbitrary primers and nearly 7,000 reproducible bands were examined. The patterns of gene expression of each cross combination were analyzed and eight common patterns (fifteen kinds) were found. When the results from the two cross combinations were put together and compared, eight different typical expression patterns were observed, these indicated that the patterns of gene expression of these two cross combinations had obvious differences. Gene expression correlation and cluster analyses of the two cross combinations indicated that the gene expression of the mid-parent heterosis cross combination was correlated with maternal effect, but in the high-parent heterosis cross combination, paternal effect acted in the gene expression of the hybrids or the gene expression of the hybrids was biased towards one parent.

Protective Effect of Aminoglycosides and Their Combinations Against 2-Chloroethylethyl Sulfide Exposure

  • Exposure of splenocytes to 2-chloroethylethyl sulfide (CEES) resulted in the cell death, and the cytotoxicity of CEES was prevented by inhibitors of lysosomal hydrolases. Therefore, it has been postulated that the cytotoxicity of CEES may be partially due to the lysosomal labilization. This study, based on this mechanism, was undertaken to determine whether aminoglycoside antibiotics as inhibitors of lysosomal phospholipases and their combinations with other lysosome stabilizers can be useful as a treatment to reduce the CEES toxicity in mice. 2-Chloroethylethyl sulfide (20 mg/kg body weight) was injected ip into female ICR mice, and candidate compounds were administered ip before or after the CEES challenge. Kanamycin (40 mg/kg body weight) as effective as deferoxamine (100 mg/kg body weight) enhanced the survival rate after 5 days of intoxication from 10% of control to 50 - 60%. The most effective was found to be the combination of kanamycin, cycloheximide, deferoxamine and dextrose showing an almost full protection against 2LD50 of CEES. Consistent with the protection of the CEES toxicity, the decrease of body weight in mice intoxicated with CEES was effectively prevented by kanamycin or its combinations. It is suggested that kanamycin or its combination (kanamycin, cycloheximide, deferoxamine and dextrose) would be one of effective antidotes against the CEES poisoning in mice.

Hybridization Using a New Male-sterile Germplasm as the Female Parent in Chinese Jujube

  • This study identified a new male-sterile germplasm of Chinese jujube, named male-sterile No. 2 (JMS2), and achieved controlled hybridization using that germplasm as the female parent. The anthers of JMS2 before flower bud opening became shrunken, dingy yellow and much smaller than normal ones, and they changed to brown after anthesis. No pollen was observed in anthers of JMS2 and its male-sterile trait remained stable over different years. A total of 1,642 fruits were obtained from ten intra- and interspecific cross combinations via controlled hybridization from 2008 to 2012 using JMS2 as the female parent. Of those, 27.3% produced seeds, on average (0-72.6%). The rate of fruit with seed (RFS) was significantly different between cross combinations or years. Compared to other cross combinations, the RFS in the combination of JMS2 ${\times}$ 'Xingguang' (a Chinese jujube cultivar with high resistance to jujube witches' broom disease) and JMS2 ${\times}$ 'Xing16' (a sour jujube genotype) remained high in different years and reached means of 48.7 and 58.1%, respectively. Finally, 150 plantlets were regenerated from immature embryos, and 51 of them were randomly selected and identified to be authentic hybrids using amplified fragment length polymorphism (AFLP) markers. This is the first report of hybrids obtained from a cross between Chinese jujube and sour jujube.

Effect of Coning Combinations on Working Performances of Wavy Mechanical Face Seal (코닝 조합이 물결 프로파일이 가공된 미케니컬 페이스 실의 작동 성능에 미치는 영향)

  • Non-contact type mechanical face seals installed in mechanical systems prevent leakage of working fluid using thin working fluid film between stator and rotor. For that purpose, various kinds of surface profiles, grooves and conings have been applied on seal surfaces of stator and rotor to generate hydrodynamic and hydrostatic pressure. The thickness distribution of working fluid film is one of important factors which affect the working performances of mechanical face seal, and it is strongly affected by the surface height profiles of stator and rotor. Therefore, appropriate design of surface height profiles of stator and rotor is necessary to optimize the working performances and life of mechanical face seal. In this study, numerical analysis using finite volume method was conducted to estimate the working performances of wavy mechanical face seals which have 36 coning combinations. As results, minimum thickness of working fluid film, leakage volume of working fluid and friction torque in static equilibrium condition of mechanical face seal, and stiffness of working fluid film were obtained. The results show that the working performances of mechanical face seal were affected by the coning combinations which can change the thickness distribution of working fluid film and pressure distribution in sealing region of mechanical face seal.

Effect of Varying Levels of Aflatoxin, Ochratoxin and Their Combinations on the Performance and Egg Quality Characteristics in Laying Hens

  • A 50 day feeding trial was conducted with White Leghorn (WL) laying hens, 42 weeks old, to determine if feeding of varying levels of aflatoxin (AF), ochratoxin A (OA) or their combinations has any effect on their performance and egg quality parameters. Feeding of $T_4$, $T_7$, $T_8$, $T_9$ and $T_10$ caused significant reduction in feed intake of hens. Hen day egg productions were significantly reduced at all the levels of toxins except 0.5 ppm of AF. Maximum reduction in egg production was noticed at 2 and 4 ppm of AF and OA, respectively. Average body weight and egg weight were not affected by toxin feeding. The feed efficiency in terms of net feed efficiency and feed consumed per dozen egg produced was significantly reduced at higher levels of both the toxins and their combinations. Feed consumption for production of 1 kg egg mass remained uninfluenced due to aflatoxin feeding whereas significant increase in the value of the same was noticed at 4 ppm level of OA and combination of 1 and 2 ppm of AF and 2 and 4 ppm of OA ($T_9$ and $T_10$), respectively. Various levels of OA (1-4 ppm) and all the combination of two toxins ($T_8$, $T_9$ and $T_10$) significantly altered the shape index of eggs in laying hens. The shell thickness was significantly reduced by higher level of AF (2 ppm), OA (2 and 4 ppm) and their combination. Albumen index, Haugh Unit and yolk index remained unchanged due to incorporation of toxins in the diet. It is concluded that AF, OA either singly or in combination at higher levels could depress the performance in terms of egg production and feed efficiency significantly. The egg quality parameters i.e. shape index and shell thickness were also significantly affected.

  • In general, herbicides have been classified according to selectivity, mobility. time of application, methods of application, mode of action and chemical property and structure. However, there was no generally accepted classification system for practical use in the field. The primary processes affected by the majority of herbicides are the growth process through cell elongation and/or cell division, the photosynthetic process specifically the light reaction, the oxidative phosphorylation and the integrity of the membrane systems. The usual approach in the study of the mechanism by which herbicides kill or inhibit the growth of plants is to initially determine the morphological phototoxicity systems, The mechanism by which a herbicide kills a plant or suppresses its development is actually the resultant effect of primary and secondary(or side) effects. In most instances, the death of the plant is due to the secondary effects. To induce the desired response, a herbicide must be able to gain entry into the plants and once inside, to be transported within the plant to its site(s) of activity in concentrations great enough. Obstacles to the entry and movement of herbicides in plants are generally classified by leaf and soil obstacles, translocation obstacles and biochemical obstacles, and these obstacles are also strongly influenced by plant species and by environmental factors such as light, temperature, rainfall and relative humidity. And hence, in most instances, results obtained from laboratory or greenhous vary from those of field experiment. Author attempted to classify herbicides from the field experiment using the two-dimensional ordination analysis to obtain practical information for selecting effective herbicides or to choose effective herbicide combinations for increasing herbicidal efficacy or reducing the chemical cost. Based on this two-dimensional diagram, desired herbicides or combinations were selected and further investigated for the interaction effects whether these combinations are synergistic, additive or antagonistic. From the results, it was concluded that these new approach could possibly be give more comprehensive informations about effective use of herbicide than any other systems.

Variation in Fruit Characteristics of 3x Progenies Obtained from a Cross between 4x and 2x Grape Cultivars (4배체와 2배체 포도 품종간 교배를 통해서 생성된 3배체 후대개체에서 과실특성의 변이)

  • This study investigated the variation in fruit characteristics between different triploid progenies derived from a cross between tetraploid and diploid grape cultivars, with the aim of developing high quality, labor-saving triploid grapes. No differences were found in fruit quality traits such as soluble solids content and titratable acidity, whereas significant variation was noted in cluster weight induced by a one-time application of GA3 (100 ppm) on the triploid progeny of different cross combinations. The cluster weight was higher in progeny obtained from 'Kyoho' ${\times}$ 'Sekirei' than for offspring derived from other cross combinations; deviation in the mean cluster weight was also small for 'Kyoho' ${\times}$ 'Sekirei' progeny. These results suggest that specific cross combinations, such as 'Kyoho' ${\times}$ 'Sekirei', can be used to efficiently breed new, labor-saving seedless grape cultivars.

Combinatorial Antitumor Activity of Oxaliplatin with Epigenetic Modifying Agents, 5-Aza-CdR and FK228, in Human Gastric Cancer Cells

  • Epigenetic silencing is considered to be a major mechanism for loss of activity in tumor suppressors. Reversal of epigenetic silencing by using inhibitors of DNA methyltransferase (DNMT) or histone deacetylases (HDACs) such as 5-Aza-CdR and FK228 has shown to enhance cytotoxic activities of several anticancer agents. This study aims to assess the combinatorial effects of genesilencing reversal agents (5-Aza-CdR and FK228) and oxaliplatin in gastric cancer cells, i.e., Epstein-Barr virus (EBV)-negative SNU-638 and EBV-positive SNU-719 cells. The doublet combinatorial treatment of 5-Aza-CdR and FK228 exhibited synergistic effects in both cell lines, and this was further corroborated by Zta expression induction in SNU-719 cells. Three drug combinations as 5-Aza-CdR/FK228 followed by oxaliplatin, however, resulted in antagonistic effects in both cell lines. Simultaneous treatment with FK228 and oxaliplatin induced synergistic and additive effects in SNU-638 and SNU-719 cells, respectively. Three drug combinations as 5-Aza-CdR prior to FK228/oxaliplatin, however, again resulted in antagonistic effects in both cell lines. This work demonstrated that efficacy of doublet synergistic combination using DNMT or HDACs inhibitors can be compromised by adding the third drug in pre- or post-treatment approach in gastric cancer cells. This implies that the development of clinical trial protocols for triplet combinations using gene-silencing reversal agents should be carefully evaluated in light of their potential antagonistic effects.

A Study on the Enhancement of Protein Quality by Food Combinations in Korean Diet (한국식단의 식품배합을 통한 단백질의 질적상승효과)

  • A study on the balance of essential amino acids in Korean diet and further investigation on quality of protein through optimum combination of foods using the least square method based on FAO/WHO (1973) provisional score patterns were carried out The results of the optimum gross combination are summarized as follows: 1) The first limiting amino acids of the Korean diet was found to be methionine and cysteine based on both whole egg and FAO/WHO (1973) provisional score patterns. 2) Weight ratios between rice and beef, chicken, pork, rabbit meat are 7 : 3, 4 : 1, 4 : 1, and 3 : 1. The amino acid score and the least amino acid are 100(phe+tyr), 99(thr), 111(trp), and 109(leu) for each combination. This result indicates that pork and rabbit meat can be high quality protein sources for the rice eating people. 3) Weight ratios between rice and anchovy, saury, ark shell, pollack, cuttle fish, tuna and herring are 2 : 1, 5 : 1, 3 : 2, 3 : 1, 3 : 1, 4 : 1, and 3 : 1. 4) Weight ratios between potato and rice, wheat flour are 8 : 1 and 24 : 1. Lysine, the limiting amino acid of rice and wheat flour can be complemented by combining these with potato. 5) It is possible to apply this idea on typical Korean dishes. For example, in case of soybean-sprout rice the optimum combinations of rice : soybean-sprout : pork and rice : soybean-sprout : beef are 4 : 1 : 1 and 11 : 1 : 5. It is of utmost importance to pursue further more desirable combinations with more complex food items and transform the result obtained by the computer analysis into more practical terms for practical use.

Influence of Driving Routes and Seasonal Conditions to Real-driving NOx Emissions from Light Diesel Vehicles (주행 경로 및 계절의 변화가 소형 경유차의 실제 주행 시 질소산화물 배출량에 미치는 영향)

  • The objective of this study is to compare NOx emissions from light duty diesel vehicles measured from on-road tests that conducted under various driving routes and seasonal conditions. We measured real-driving NOx emissions using PEMS, portable emissions measurement system, under the urban, rural and motorway road traffic conditions. On-road tests were repeated at summer, fall and winter season. The accumulated driving distance is more than 1,200 km per each vehicle. Route average NOx emission factors were compared among nine route-season combinations. The emission characteristics of each combinations were investigated using time series mass emission rates and vehicle operation-based emission rates and activities, which is based on U.S. EPA's MOVES model. Most concerned route-season combination is "urban road condition at summer", which shows two to eleven times higher NOx emissions than other combinations. The emission rates and activities under low speed operating conditions should be managed in order to reduce urban-summer NOx. From a NOx control strategy perspective, the exhaust gas recirculation, EGR, is observed to be properly operated under wide range of vehicle driving conditions in Euro-5 vehicles, even if the air conditioner turns on. In high power demanding conditions, the effect of overspeeding could be more critical than that of air conditioner activation.
