Histological and Histochemical Studies on the Epididymal Region and Deferent Ducts of the Drakes by the Age in Weeks (오리 부고환(副睾丸) 및 정관(精管)의 주령별(週齡別) 조직학적(組織學的) 및 조직화학적(組織化學的) 연구(硏究))

  • This study was made for the better information of the male reproductive system on the meat-type drake, Cherry Belly X White Golden. The epithelium of ductules of epididymal region and deferent duct were observed histologically and histochemically with the progress of their development. India-ink absorbability on the luminal epithelium was also investigated after the administration of India-ink. The results are as follows; 1. Rete testis and various round ductules in immature form appeared in epididymis within 6 weeks after hatching, and simple cuboidal and simple columnar epithelium were found in the epithelia of the ductules within 8 weeks after hatching. Larger ductules were found on epididymal surface which was in the developing stage near to the immature efferent ductule. From 10th to 20th week, various ductules appeared in epididymis, and developing form of efferent ductules were much more increased on epididymal surface. The luminal epithelium of the ductules were composed of ciliated simple columnar and pseudostratified ciliated columnar cells. At the same time, deferent duct appeared. From the 21th week, various ductules in epididymis became abruptly matured. Lumen of rete testis was lined by simple squamous or simple cuboidal epithelium, and that of efferent ductules, having many folds and being larger than any others were lined by pseudostratified ciliated columnar epithelium in which ciliated columnar cells, non-ciliated cells(clear cells) and basal cells were noted. Connecting tubules of star shaped lumen were composed of pseudostratified ciliated columnar epithelium in which ciliated columnar cells, nonciliated cells, and basal cells were observed. The luminal surface of epididymal ducts was smooth and has thick pseudostratified columnar epithelium which was composed of high columnar cells and basal cells. From 26th week after hatching, sperm pooling was started in various ductules. 2. From 4th to 10th week, simple cuboidal epithelium of deferent duct transformed to simple columnar epithelium with the progress of aging. At the basement of epithelium, clear round cells were noted. From 12th to 20th week, high columnar cells with enlongated nucleus were noted on the luminal border of deferent ducts, forming folds of pseuclostratified columnar epithelium. From 20th week, the deferent duct started to have septa in it's lumen and composed mainly of pseudostratified columnar epithelium, and round cells disappeared. From 20th week, the lumen diameter of deferent duct became wider with the progress of aging, but there was no difference among the values of lumen diameter in upper, middle, and lower part of deferent ducts. At 26th week, the pooling period of sperms in deferent ducts, the lumen diameter became rapidly widen, especially in the lower part of deferent ducts. Thickness of muscular layer of ductus deferens showed gradual growth within 24 weeks but did abrupt thickening from 26th week. 3. Saliva resistant PAS granules were dotted on the top of nucleus in efferent ductules epithelium but the amount of the granules were little in the connecting ductules's epithelium. The granules reactive to acid phosphatase were abundant in the some epithelial cells of efferent ductules and connecting ductules, especially above the nucleus of cells. The granules reactive to alkaline phosphatase were noted on the luminal border of efferent ductules. Parts of free border of efferent ductules and middle portion of deferent ducts were stained slightly by alcian blue technique. India ink granules were found mainly in the epithelium of efferent ductules but were few in that of connecting ductules.

  • The work was conducted with tole purpose of investigation on the development pattern of epididymis in accordance with the growth of meat-type cockerels. 1. Histological features of various ductules in epididymis of the cockerel on the age of weeks were as follow: within 10 weeks after hatching rete testis and connecting ductules were well developed but efferent ductules were observed in immature form. During 10th to 20th week, the lining epithelium of various ductules in epididymis was in the developing stage near to the mature form. From 21th week, various ductules were abruptly matured. Lumen of rete testis was lined by simple squamous or simple columnar epithelial cells and that of efferent ductules, having many folds and being larger than any others, were lined by ciliated pseudostratified columnar epithelium with ciliated columnar cells, clear cells and basal cells were noted. Luminal epithelium of connecting ductules was composed of ciliated low pseudostratified columnar epithelial cells, ciliated columnar cells, clear cells and basal cells. The luminal surface of epididymal ducts was pseudostratified columnar epithelium and which was composed of high columnar cells and basal cells. 2. In the India ink absorption test, India ink granules were noted above the nucleus of some cells in the efferent ductules and the connecting ductules at 7 hours after administration of India ink to the mature epididymis, but not absorbed in the other ductules. The granules reactive to acid phosphatase were most abundant in some epithelial cells of efferent ductules and connecting ductules, especially above the nucleus of cells. The granules reactive to alkaline phosphatase were noted on the luminal border of efferent ductules. The granules reactive to PAS were scattered in the epithelial cells of efferent ductules and connecting ductules.

  • This study was conducted in order to obtain the information of the histological changes in each of six segments of the epididymal ducts in Korean native goats. Thirty-two Korean native male goats were examined, dividing into seven groups, at 4 weeks intervals from 8 to 32 weeks of age. The results obtained were as follows. 1. The epididymal ducts showed histologically an abrupt growth at the age of 16 weeks being followed by almost full maturation at the age of 24 weeks. Diameter of the cauda was steadily larger than that of the caput and corpus of the epididymal ducts. 2. Spermatozoa in the lumen of epididymal ducts were first observable at the age of 16 weeks, thereafter showing sparse in the lumen of caput, whereas most dense in the lumen of cauda in the density of spermatozoa. 3. Ducts in the caput and corpus were lined by ciliated columnar epithelium until the age of 12 weeks, and later by pseudostratified ciliated columnar epithelium which was composed of ciliated columnar cells, clear cells and basal cells. Ducts of cauda epididymis were lined by simple ciliated columnar epithelia until 12 weeks of age and later by simple or pseudostratified ciliated columnar epithelium, and two types of ducts (small ducts with high epithelium and large ducts with lower epithelium) were noted. Nucleus of the epithelial cells in the caput were located in the base of cells but in the corpus and cauda, those were located in the mid part of cells. cilia were most developed in the epithelia of the corpus.

  • The morphological changes of the midgut epithelium during the metamorphosis of pine caterpillar are observed with light and electron microscope, being divided into 5 stages from the 8th instar larva to just after pupation. The midgut epithelium of 8th instar larva is composed of columnar cell, goblet cell, regenerative cell, and endocrine cell. The secretorials are arranged on the nuclear membrane in the columnar cell of the midgut epithelium in the 8th instar larva, and lysosomes are augmented in the apical portion. Cytoplasmic extrusions are observed in the apical surface of columnar cell but they have no cell organells. Nucleus, mitochondria, rER, Golgi complex, and free ribsomes are observed in the regenerative cell. Regenerative cells are differentiated into the form of goblet cell, and vacuoles are gradually increased in the cytoplasm. Just pupa stage, the materials, which appears to be mainly composed of Ca, are observed in the circular form and goblet cavity of regenerative cell are detached to lumen. As a result, it reflects the process of the degeneration of the midgut epithelium that lysosomes are gradually augmented in the columnar cell, that nuclear materials are removed to cytoplasm, and that cytoplasmic extrusions are observed in the apical surface. And though regenerative cells are differentiated into the form of the goblet cell, it is believed that goblet cavity is detached from regenerative cell to the lumen and midgut epithelium of pupa stage is formed.

Morphological and Absorptive Studies on Canal Epithelium of the Various Ducts in Rooster Epididymal Region (닭 부고환부(副睾丸部)의 각종(各種) 관상피(管上皮)의 형태(形態) 및 흡수(吸收)에 관한 연구(硏究))

  • Histological and histochemical studies were made on the lining epithelia of the various ducts in epidymis of the Rooster and absorptive function of the canal epithelial cells in the Rooster epididymis were also investigated after administration of India ink. The results obtained were summerized as follows; 1. Epihtelium lining the rate testis was mainaly composed of single later of cuboidal cells, and was partially composed of flattened squamous or low columnar cells. Efferential ductules were characterized by having many villous projections orrfolds which extened into the lumen, and were lined by stereociliated pseudostratified epithelium which consisted of manily ciliated columnar cells, a few scattered clear cells and basal cells. Connecting ductules were lined by ciliated pseudostriatified colummnar epithelium in which ciliated columnar cells, clear cells and basal cells were noted. Epididymal ducts were lined by pseudostratified epidhelium in which columnar and basal cells were noted. 2. PAS-granules, saliva resistant were noted mainly in the epithelial cells of efferential and connecting ductules. 3. Sudan black B stained heavily the granules in the epithelial cells of sufferential and connecting ductules. 4. The granules reactive to acid phosphatase most abundant in the epithelial cells of efferential ductules and were lesser amount in the epithelial cells of connecting ductules where as very few or no granules were seen in the rest of the ducts. 5. Alkaline phosphatase activity was most prominent but discontinuous in the luminal surface of the epithelium of efferential ductules and less marked in the connecting ductless. No enzyme activity was noted in the canal epithelium of epididyml duct. 6. India ink granules were most numerous in the epithlial cells of efferential ductules and were a few in connecting ductules. Very few or no granules of India ink were noted in the other types of the ducts. India ink granules in the epithelium increased gradually as the time after the administration of India ink (one up to twenty-nine hours) has proceeded. From those results it is suggested that epithelial cells of efferential and connecting ductules have active absorptive function, whereas the rest of duct system in the epididymis of the Rooster may be the mere pathway of the seminal fluid without significant modification of its constituents.

  • An intratesticular simple cyst is a rare lesion. The pathogenesis of such lesions is uncertain. Clinically, an intratesticular simple cyst may be difficult to differentiate from a cystic neoplasm. However, preoperative diagnosis is possible using ultrasonography (US). Testis-sparing surgery with simple enucleation of the cyst has a favorable outcome. We report an intratesticular simple cyst in a 48-year-old male who had a non-tender cystic mass in the right testis for 2 years. The diagnosis was made by preoperative US. The patient underwent a testis-sparing surgery. After simple excision of the cyst, histological examination revealed a simple cyst lined by ciliated cuboidal and columnar epithelium.

  • This article reports a spontaneous thoracic teratoma in a laying hen (Deklb brown warren). In this case, a 40 days old hen was submitted for necropsy as part of an investigation into a flock problem suspected Marek's disease to Chonbuk Veterinary Service Laboratory in 1983. On the gross finding, mass was In the cranial subpulmonary cavity and attached to the vertebral column. It contained fully developed contour feathers. Histologically, feathers were arised from feather follicles complete with arrector plumi muscle, nerve, vessel and mucous gland composed with simple tall columnar epithelium. The outer surface of mass was lined by keratinized or nonkeratinized stratified squamous epithelium. It converted to pseudostratified ciliated columnar epithelium in some area. There were lymphocyte infiltration around gland tissue and eosinophilic intranuclear inclusion bodies in nonkeratinized epithelium. This thoracic teratoma was composed of ectodermal origin, squamous epithelium and nerve, and endodermal origin, mucous gland. This case in laying hen has never been reported in the literature in the world.

  • The epithelium of the hindgut in the german cockroach, Blattella germanica Linne, was observed with electron microscope. The epithelium of the ileum, which is located at the anterior hindgut, is composed of a single layer of squamous and cuboidal cells. The liminal surface of the epithelium is lined with the cuticular intima. The epithelial cells contain cell organelles expected to be found in absorptive cells, and some epithelial cells have numerous lamelated crystals, the "spherites". The rectal epithelium of posterior hindgut is composed of rectal pads which are covered with cuticular intima on the luminal side. The rectal pads are composed of columnar absorptive cells and basal cells. The apical plasma membrane of columnar cell is made of microvilli, where mitochondria associated with some of the microvilli. The lateral plasma membrane is highly infolded and space is an uniform width of approximately 200$\\AA$. Well developed mitochondria are found closely associated with the infoldings and this is referred to as the "mitochondrial-scalariform complex". A septate junction is found near the apical zone between the columnar absorptive cells, whereas many desmosomes and intercellular spaces are formed between the columnar cells. Basal cells are bowl-shaped where the convex surface is inlaid into the basal surface of the columnar cells while the concave surface faces the basal lamina. The cytoplasm of the basal cell is electron dense and contains well developed cell organelles. The basal sheath is located between the basal membrane and basal lamina, providing barrier between the epithelium and the hemolymph. The epithelium is surrounded by the subepithelial space and muscles. The subepithelial space, which is composed of fibrous connective tissue, is innervated by many tracheoles and axons.

Development on the abomasum of fetuses and neonates in Korean native goats (한국재래산양의 태아 및 신생아의 제4위 발달에 관하여)

  • The development of abomasum in fetuses between 60, 90, 120 days of gestation and neonates of Korean native goats was investigated by light, scanning and transmission electron microscopy. The results obtained were summarized as follows ; 1. The abomasum wall appeared to be differentiated into the epithelium, lamina propria, muscle layer, and serosa at 60 days of gestation. The epithelium was stratified columnar and these nuclei were located near the apical two thirds portion of the cell at 60 days of gestation, and then transformed into simple columnar epithelium with the flat basal nuclei. 2. The inner circular and outer longitudinal muscle layers were observed at 90 days of gestation and the blood vessels had become quite well developed as various arterioles, venules and capillaries of different size during this age. 3. Gastric pits were seen at 90 days of gestation and continued gradually to increase depth during gestation. 4. The mucous, parietal and chief cells appeared in epithelium at 90 days of gestation and continued gradually to increase in number during gestation. In 120 days fetuses and neonates, muscle layer had become very thickeness. 5. Scanning electron microscopically, the inner surface of the abomasum already consisted of wavy spiral folds which had many fine wrinkles at 60 days of gestation. In 90 day old fetuses, each spiral fold was enlarged and its surface was tended to be split into many straight longitudinal ridges and among these ridge were found shallow grooves, At 120 days, the subdivided swellings of ridges were progressively complicated in shape. In the neonates, the inner surface was flat and holed with many gastric pits. 6. Transmission electron microscopically, the epithelium was straified columnar and these nuclei were irregular shape at 60 days fetus. The parietal, chief and mucous cells were observed in 90 day old fetuses and continued gradually to increase in number during gestation. 7. The development of the abomasum was relatively slow at early stages, it was accelerated greatly in the last of gestation.

Histological and immunohistochemical studies on the colo-rectum of the chicken embryos (닭 태자의 직결장에 대한 조직학적 및 면역조직화학적 연구)

  • Histological changes and ontogeny, distribution and relative frequency of bovine SP-1/chromogranin(BCG)-, serotonin-, human pancreatic polypeptide(HPP)- and glucagon-immunoreactive cells were investigated in the colo-rectum of the chicken embryos. The pseudostratified columnar-like epithelium was observed in 10 days of incubation to 15 days of incubation, thereafter these epithelium were differentiated to simple columnar epithelium and goblet cells were detected for the first time on 19 days of incubation. BCG and serotonin-immunoreactive cells were observed for the first time on 15 days of incubation respectively, thereafter these cells were increased with ages. However no HPP and glucagon-immunoreactive cells were found in this study.
