A Study on Characteristics of Water Pollution in Woopo-Mokpo Wetlands (우포·목포늪 수질오염특성 연구)

  • We could know that the Woopo and Mokpo wetlands, which have the typical colony structure in the agricultural area of korea, had a characteristic which the pollution sources of water are scattered widely and it is caused by sewage, livestock wastewater and non point sources. As the result of an examination of water, the concentration of T-N was measured highly within five grade in the lake standard. If we build natural domestic treatment system in Taehap-myon, lbang-myon and Yuo-myon, which are located at the upper stream of wetlands, we can remove SS, BOD, COD and T-N largely.

  • The purpose of this study was to investigate the microstucture and hardness, corrosion of pure Ti alloy, which is widely used as partial denture frame work these days, depending on the cooling method, followed by casting. The first group was bench cooling at room temperature($18^{\circ}C$), the second group was slowly cooled in the furnace from $700^{\circ}C$ to room temperature, and third. rapidly cooled in $0^{\circ}C$ water. The microstructure of each specimen observed by means of photomicrograph taken by electron microscope, in add to the physical characteristics of each specimen were obtained using the rockwell Hardnest Number. the characteristics of corrosion. The results were obtained as follows: 1. From Potentiodynamic plot. we conclude furnace-cooled specimen had the best stabiltity of passive film and that air-cooled specimen showed similar characteristics. The density of electric current of quenched specimen was the highest, which formed kind of unstable passive film. 2. Specimen cooled at room temperature (air cooling) had the highest value of hardness of 81.26HRB, specimen cooled at ice-water, $0^{\circ}C$, had the value of 78.42HRB, and specimen furnace-cooled at $700^{\circ}C$ had lowest value of 77.1HRB. 3. Quenching treated micro-structure formed martensite structure by and large. In case of air cooling, we could see $\alpha$-structure widmanstatten formed overall. In furnace cooling, widmanstatten structure and various shape $\alpha$-structures forming colony with direction were detected.

Structural damage identification with output-only measurements using modified Jaya algorithm and Tikhonov regularization method

  • The absence of excitation measurements may pose a big challenge in the application of structural damage identification owing to the fact that substantial effort is needed to reconstruct or identify unknown input force. To address this issue, in this paper, an iterative strategy, a synergy of Tikhonov regularization method for force identification and modified Jaya algorithm (M-Jaya) for stiffness parameter identification, is developed for damage identification with partial output-only responses. On the one hand, the probabilistic clustering learning technique and nonlinear updating equation are introduced to improve the performance of standard Jaya algorithm. On the other hand, to deal with the difficulty of selection the appropriate regularization parameters in traditional Tikhonov regularization, an improved L-curve method based on B-spline interpolation function is presented. The applicability and effectiveness of the iterative strategy for simultaneous identification of structural damages and unknown input excitation is validated by numerical simulation on a 21-bar truss structure subjected to ambient excitation under noise free and contaminated measurements cases, as well as a series of experimental tests on a five-floor steel frame structure excited by sinusoidal force. The results from these numerical and experimental studies demonstrate that the proposed identification strategy can accurately and effectively identify damage locations and extents without the requirement of force measurements. The proposed M-Jaya algorithm provides more satisfactory performance than genetic algorithm, Gaussian bare-bones artificial bee colony and Jaya algorithm.

Transferable R plasmid of Streptococci Ioslation from Diseased Olive Flounder (Paralichthys olivaceus) in Jeju (제주도 양식넙치병어에서 분리된 연쇄상구균의 약제내성 전이성 plasmid)

  • Seventy-five streptococci were isolated from diseased olive flounder, Paralichthys olivaceus in Jeju. Their drug susceptibility and transferable multiple drug resistance were characterized. All isolates were resistant to flumequine (AR) and oxolinic acid (OA) and 26 isolates (34.7%) showed 4~6 multiple resistance of ampicillin (ABPC), AR, doxycycline (DOXY), erythromycin (EM), norfloxacin(NOR), OA and oxytetracycline (OTC) in various combinations. pST9 of a transferable R plasmid was detected from a multiple drug resistance strain, Streptococcus sp., ST9 originated from diseased flounder in Jeju, previously. We performed DNA hybridization to know the distribution of plasmid with the same DNA structure as pST9 in streptococci. Thirteen out of 60 isolates analyzed were positive in colony DNA hybridization and the part of bacteria isolated from raw meal was also hybridized with pST9. It suggested that raw meal is one of the origin of the resistance plasmid and R plasmid with DNA structure differing from pST9 is also involving in multiple drug resistance of the streptococci. In conjugation experiment, we found transferable R plasmid carrying OTC, DOXY and/or EM resistance determinant in the 13 resistance strains. all of the streptococci carrying the transferable R plasmid were similar in RAPD patterns. However, pST -type R plasmid was rare in S. iniae most frequently appearing in flounder farm.

Impact of a Recombinant Biocontrol Bacterium, Pseudomonas fluorescens pc78, on Microbial Community in Tomato Rhizosphere

  • Pseudomonas fluorescens pc78 is an effective biocontrol agent for soil-borne fungal diseases. We previously constructed a P43-gfp tagged biocontrol bacteria P. fluorescens pc78-48 to investigate bacterial traits in natural ecosystem and the environmental risk of genetically modified biocontrol bacteria in tomato rhizosphere. Fluctuation of culturable bacteria profile, microbial community structure, and potential horizontal gene transfer was investigated over time after the bacteria treatment to the tomato rhizosphere. Tagged gene transfer to other organisms such as tomato plants and bacteria cultured on various media was examined by polymerase chain reaction, using gene specific primers. Transfer of chromosomally integrated P43-gfp from pc78 to other organisms was not apparent. Population and colony types of culturable bacteria were not significantly affected by the introduction of P. fluorescens pc78 or pc78-48 into tomato rhizosphere. Additionally, terminal restriction fragment length polymorphism profiles were investigated to estimate the influence on the microbial community structure in tomato rhizosphere between non-treated and pc78-48-treated samples. Interestingly, rhizosphere soil treated with strain pc78-48 exhibited a significantly different bacterial community structure compared to that of non-treated rhizosphere soil. Our results suggest that biocontrol bacteria treatment influences microbial community in tomato rhizosphere, while the chromosomally modified biocontrol bacteria may not pose any specific environmental risk in terms of gene transfer.

Study on Iron-making and Manufacturing Technology of Iron Swords with Ring Pommel Excavated in Ipbuk-dong, Suwon (수원 입북동 출토 철제환두도의 제철과 제작기술 연구)

  • This study analyzed nonmetallic inclusions in iron swords with a ring pommel excavated in the Ipbuk-dong, Suwon. Scanning electron microscopy with energy dispersive spectroscopy(SEM-EDS) was used to estimate the iron-making temperature, and we compared the oxide with $SiO_2$ to investigate the heat-treatment technology in the production of iron swords with a ring pommel by investigating the artificial insertion of a slag former and the metallurgical structure. From the wustite observed in most of the specimens, it is judged that these swords were produced by heating and forging iron smelted at a low temperature using the solid reduction method. In addition, judging from the partial presence of $P_2O_5$, it is assumed that they were smelted directly with natural ore, not calcined. From the ratios of $CaO/SiO_2$ and $TiO_2/SiO_2$, it is judged that the raw material for iron-making was iron ore and that a calcareous slag former was not artificially inserted. The structure of the blade part on the front end was pure iron. From the high carbon content of the blade part on the ring pommel and the formation of a martensitic structure and pearlite colony, it is judged that they were tempered after carburizing and that the back, handle part, and ring pommel were unintentionally carburized. Judging from the structure of these specimens, it was noted that they were produced by applying artificial partial heat-treatment technology. This study attempted to present a more scientific analysis by using the method of interpretation through component analysis of nonmetallic inclusions appearing in one relic by the ratio of the oxide divided by $SiO_2$. It is judged that reinterpreting the arguments by the results of the existing analysis and research in this way can obtain different interpretations.

Antibacterial Activity by Chitosan Derivatives with Quaternary Ammonium Salt (4차 암모늄기를 가진 키토산 유도체의 항균성)

  • Chitosan is the deacetylation product of chitin. Chitosan was converted to N-alkyl chitosan derivatives using proper aldehyde having different alkyl chains, then chitosan derivatives with quaternary ammonium salt were producted by quaternization with methyl iodide. Their antibacterial activities aganst S. aureus(ATCC 6538P) and E. coli (ATCC 14339), gram-positive bacteria and gram-negative bacteria, respectively, as introduced alkyl length were evaluated by shake flask method using colony count. The antibacteiral activity was found to be increased as alkyl chains. hydrophobic goups. These antibacterial agents were more active against S. aureus than E. coli. It may be due to the different of structure of cytoplasmic membrane.

An Optimal Reliability-Redundancy Allocation Problem by using Hybrid Parallel Genetic Algorithm (하이브리드 병렬 유전자 알고리즘을 이용한 최적 신뢰도-중복 할당 문제)

  • Reliability allocation is defined as a problem of determination of the reliability for subsystems and components to achieve target system reliability. The determination of both optimal component reliability and the number of component redundancy allowing mixed components to maximize the system reliability under resource constraints is called reliability-redundancy allocation problem(RAP). The main objective of this study is to suggest a mathematical programming model and a hybrid parallel genetic algorithm(HPGA) for reliability-redundancy allocation problem that decides both optimal component reliability and the number of component redundancy to maximize the system reliability under cost and weight constraints. The global optimal solutions of each example are obtained by using CPLEX 11.1. The component structure, reliability, cost, and weight were computed by using HPGA and compared the results of existing metaheuristic such as Genetic Algoritm(GA), Tabu Search(TS), Ant Colony Optimization(ACO), Immune Algorithm(IA) and also evaluated performance of HPGA. The result of suggested algorithm gives the same or better solutions when compared with existing algorithms, because the suggested algorithm could paratactically evolved by operating several sub-populations and improve solution through swap, 2-opt, and interchange processes. In order to calculate the improvement of reliability for existing studies and suggested algorithm, a maximum possible improvement(MPI) was applied in this study.

Pathogenic bacteria causing rot in commercial soybean sprout cultivation

  • Soybean sprout pathogenic bacteria were isolated from the large, deep containers of a commercial factory. Over a period of one year, 40 pathogenic-like bacteria were isolated among a total of 732 isolates. In addition to bacteria previously reported to be associated with rotting, such as Pseudomonas putida and Erwinia carotovora, several other genera were also identified: Acinetobacter spp., Chryseobacterium spp., Klebsiella sp., Pantoea agglomerans, Bacillus sp. Fatty acid methyl ester (FAME) analysis using the Microbial ID (MIDI) system, and 16s rRNA sequence analysis, yielded identical results, confirming the identities of these microorganisms. Several types of selective media were not good for identification and determination of population structure in commercial environments, as colony type was not specific to the genus. There was no dominant bacterium, and we were not able to find the main bacterium responsible for soybean spout rot. Even though we did not identify a major target for controlling rot or screening for resistant cultivars, the results of this study indicated that bacterial rot of soybean sprout is endemic. In addition, it emerged that factory epidemics in summer are not caused by the bacteria isolated in this study.

Long-acting Recombinant Human Granulocyte Colony Stimulating Factor (rhG-CSF) with a Trimer-Structured Polyethylene Glycol

  • Mono PEGylated rhG-CSF (PEG-G-CSF) prepared by utilizing unique PEG was purified and characterized by cation-exchange chromatography. A unique, trimer-structured PEG was chosen for PEGylation of rhG-CSF among various PEG moieties. The in-vitro bioactivity, stability, and pharmacokinetics of mono-PEG-G-CSF were examined and compared to those of native rhG-CSF. Mono PEG-G-CSF exhibited reduced in-vitro bioactivity to native rhG-CSF but showed an excellent in-vivo bioactivity and stability. Furthermore, it showed markedly reduced clearance in rats, thereby increasing the biological half-life by about 4.5-fold compared to that of native rhG-CSF. The results suggest that this unique, trimer-structured 23 kDa PEG can provide advantages to improve the bioactivity of therapeutic proteins in clinical use.
