The wrinkles correspond to the most obvious expression of skin ageing and are manifested by changes on the organization and dermal structure. In the extracellular matrix, decreased quantities of collagens and glycosaminoglycans as well as a deterioration of the fibrillary network is noted, result in a reduction of dermal thickness. In addition, the activity of the collagenases increases in contrast to the synthesis of collagen fibers. Nor are cells spared during the aging process. We thus studied and compared the contractile capacity as well as the synthesis capacity of normal human fibroblasts and human fibroblasts obtained from biopsies of forehead wrinkles. The capacity of the fibroblasts to be adhered to the collagen network and to maintain a three-dimensional structure of dermis was studied on a model of equivalent dermis. The metabolic activity was studied by evaluating the capacities of synthesis of collagen I, main component of dermis. Human fibroblasts resulting from the forehead wrinkle contract less the gel of collagen than the normal human fibroblasts and present an activity of biosynthesis of collagen I less important than normal human fibroblasts. These results show that fibroblasts with aging present a deceleration of their metabolic activity and lose their capacity of adhesion to collagen fibers thus limiting the possibility of organizing the dermal tissue. We investigated the potential of an active ingredient able to compensate for the reduction of the metabolic activity and to restore the contractile capacity of fibroblasts obtained from forehead wrinkles. This effect was compared with a reference molecule: the vitamin C.

MMP-2 and MMP-9, type IV collagenases which degrade basement membrane, have been known to play important roles in invasion and metastasis of tumor cells, In addition, they seem to be involved in cell differentiation, apoptosis, angiogenesis and immunity, etc. We immunohistochemically examined epithelial and stromal expressions of MMP-2 and MMP-9 in irritation fibroma, oral leukoplakia, and oral squamous cell carcinoma (OSCC) and have some results as follows: 1. Irritation fibromas, oral leukoplakias and OSCCs mostly showed increased expression of MMP-2 and MMP-9 in the epithelium and connective tissue compared with normal mucosa. 2. There was a significant difference in the epithelial expression of MMP-2 and MMP-9 between irritation fibroma and oral leukoplakia. 3. There was a significant difference in the epithelial and stromal expression of MMP-2 and MMP-9 between irritation fibroma and OSCC. 4. There was a significant difference in the stromal expression of MMP-9 between oral leukoplakia and OSCC. We concluded that rritation fibroma, oral leukoplakia and OSCC have somewhat different characteristics of MMP-2 and MMP-9 expressions, which perhaps result from different pathogenesis.
Purpose: Tetracycline and its chemically modified non-antibacterial analogues can inhibit certain host-derived tissue destructive collagenases such as matrix metalloproteinases. The purpose of this study was to evaluate clinical and microbiologic effects of the subantimicrobial dose of doxycycline(SDD) in conjunction with scaling and root planing. Materials and methods: A total of 30 patients with chronic periodontitis who were going to receive scaling and root planing were randomly allocated to receive either a doxycycline hyclate for 3 months or nothing. Clinical probing depth, clinical attachment level, gingival recession, and bleeding on probing were measured by one periodontist. After a periodontal examination, microbial samples were collected using sterile paper points. The effect of SDD in conjunction with scaling and root planing on alterations of the periodontal pathogens (Aggregatibacter actinomycetemcomitans, Tannerella forsythia, Porphyromonas gingivalis) were also assessed using l6S rRNA polymerase chain reaction. Results: During the treatment period, clinical parameters for both treatment group and control group were improved. After 3 months, reductions in probing depth and gains in clinical attachment level were significantly greater for the SDD group than control group. Microbial analysis showed that there was no alteration of the periodontal pathogens and no difference between the groups. Conclusion: This study suggested that the subantimicrobial dose of doxycycline as an adjunct therapy with scaling and root planing might be effective and safe in the management of chronic periodontitis.

Alpha hydroxy acid (AHA) includes a group of organic acids found in natural foods such as sugarcane (glycolic acid), milk (lactic acid), apples (malic acid) and oranges (citric acid). Earlier studies demonstrated the effect of AHAs on the skin by diminishing the adhesiveness of the corneal layer and increasing the viable epidermal thickness. Recent data suggest that AHAs have some effects on the dermal component of skin and even affect the aging process of the skin. A previous study revealed increased collagen production by treatment with glycolic acid among AHAs in vitro. However, the mechanism of the regulation of collagen production by glycolic acid was unclear. In present study, we tried to demonstrate the effect of glycolic acid on human dermal fibroblasts and to unveil the mechanism of regulation of collagen production by glycolic acid in human dermal fibroblasts: proliferation of fibroblasts and collagen synthesis and degradation by collagenases in fibroblasts. Our results suggested that glycolic acid had no effect on proliferation and cytotoxicity of adult human dermal fibroblasts. However, glycolic acid not only induced the increase of the collagen synthesis in human dermal fibroblasts at lower concentration than 0.1 % but also inhibited MMP-2 activity of human dermal fibroblast in the range between 0.01 and 0.4% and MMP-9 activity of human dermal fibroblast in the range between 0.06 and 0.09%. In summary, our results suggest that glycolic acid may increase wrinkle reduction partially by both increase in collagen synthesis and decrease in collagen degradation.
Collagnase는 천연 collagen의 triple-stranded helix를 분해할 수 있는 protease로서 조직의 수복과 재생 과정에서 collagen의 재형성과 세포의 이동에 아주 중요한 역할을 하고, 숙주 감염시에는 collagen 기질을 빠르게 분해함으로써 감염을 돕는다. 본 연구에서는 일반가정에서 식용하는 김치로부터 collagenase를 생산하는 균주를 분리하여 Bacillus subtilis로 동정하였으며 이를 Bacillus subtilis JS-17이라 명명하였다. Bacillus subtilis JS-17이 생산하는 collagenase의 최적 생산 조건은 $1.5\%$ fructose, $1\%$ yeast extract, $0.5\%\;K_2HPO_4,\;0.4\%\;KH_2PO_4,\;0.01\%\;MgSO_4{\cdot}4H_2O,\;0.1\%\;citrate,\;0.1\%\;CaCl_2(pH\;7.0)$의 배지에서 $30^{\circ}C$, 200 rpm으로 72시간 동안 배양하는 것이다. 최적 조건에서 Bacillus subtilis JS-17이 생산하는 collagenase를 Amberlite IRA-900 column chromatography, Sephacryl S-300 HR column chromatography, DEAE-Sephadex A-30 column chromatography를 거쳐 분리 정제하고, 얻어진 정제 효소의 특성에 대하여 검토하였다. 정제된 collagenase의 비활성은 growth medium에서 192.1 units/mg였고, $1.1\%$의 수율로 얻어졌으며 분자량은 28 kDa이었다. 정제된 collagenase는 $55^{\circ}C$까지는 $100\%$의 활성을 유지하였고 $65^{\circ}C$에서도 $60\%$ 정도의 활성을 유지하였다. 또한 pH $6.0\~9.8$에서 $60\%$ 이상의 활성을 유지하였다. 정제된 collagenase는 metalloprotease inhibitor인 EDTA와 O-phenanthroline에 의해 효소 활성이 감소하였을 뿐만 아니라 Ammoninum persulfate, L-cysteine, N-ethylmaleimide, SDS, $NaN_3$, NaF, $KMnO_4$, PMSF에 대해서도 활성이 감소하였다. 정제된 collagenase를 여러 가지 기질에 대해 효소 활성을 비교한 결과 collagen (type I)에 대해 기질 특이성을 가지고 있었다.
Ursodeoxycholic acid(UDCA) is a hydrophilic gall bladder acid and has been used as a effective drug for liver disease related to in1munity. This drug inhibits secretions of IL-2, IL-4, and $IFN-{\gamma}$ from T-cells and production of immunoglobulin from B-cells. Also it has been reported that UDCA inhibits production of IL-1 related to the progression of periodontal disease and activation of collagenases. The purpose of the present study was to elucidate the effects of UDCA on inhibition of periodontal disease progression using clinical, microbiological and histometrical parameters. Twelve pure bred, 16 month-old-beagle dogs were used in the study. After ligature-induced periodontal diseases were formed, experimental drugs were applied twice a day and then the results of clinical, microbiological, and histometrical parameters were measured at baselie(initiation of experiment) , 4weeks and 8weeks. The gel with UDCA(concentration 0.5%, 5% 3 dogs in each) was applied to experimental group, chlorhexidine to positive control group(3dogs) and the gel without UDCA(base) to negative control group. After induction of general anesthesia, the maxillary 2nd, 3rd premolars and 1st molar and the mandibular 2nd, 3rd, 4th premolars and 1st molar were ligated in one side selected randomly and were not ligated in the opposite side. The plaque index(PI), gingival index(GI), pocket depth(PD) and gingival crevicular fluid(GCF) volum were measured clinically. The PI and GI were measured at 3 buccal points of all experimental teeth and the GCF was measured only at the 3rd premolar in the maxilla and the 4th premolar in the mandible. In the microbiological study, the samples extracted from the 3rd premolar of the maxilla and the 4th premolar of the mandible at the center of buccal surface were analyzed aerobics, anaerobics and Streptococcus colony forming units, After clinical and microbiological examination at 8weeks, the dogs were sacrificed by carotid artery perfusion. The samples were fixed and sectioned including interproximal area, and the distance from cementoenamel junction(CEJ) to alveolar crest was measured. The results were that PI, GI and PD increased until 4 weeks and decreased at 8 weeks in three groups but the differences between all the groups were not significant. The 0.5% UDCA in non-ligated group showed remarkable decrease of GCF. The experimental group applied 5% UDCA decreased the number of aerobics and anaerobics. The distance from CEJ to alveolar crest was greater in the negative control group on both ligated and non-ligated sides, but the differences were not significant stastically.
배 경 : Matrix metalloproteinase(MMPs), 특히 주로 염증세포에서 분비되는 MMP-9은 여러 가지 급성폐손상 모델 및 급성호흡곤란증후군 환자에서 증가하고, 최근에는 주기적인 물리적 스트레스가 폐포대식세포 및 결체조직세포에서 MMP-9의 생성 및 활성을 증가시키는 것으로 보고된 바 있다. 따라서 본 연구에서는 기계환기로 인한 백서의 급성폐손상에서 MMP-9의 발현 및 MMP 억제제(MMPI)의 효과에 대해서 연구하고자 하였다. 방 법 : Sprague-Dawley 백서를 적은 일호흡량(tidal volume, $V_T$)과 적절한 호기말양압(positive end-expiratory pressure, PEEP)을 적용한 LVT군과 많은 일호흡량과 PEEP을 적용하지 않은 HVT군 및 동일한 조건에서 MMPI를 투여한 HVT+MMPI의 세 군으로 나누어 실험하였다. MMPI로는 CMT-3(chemically modified tetracycline-3)를 기계환기 3일 전부터 구강으로 투약하였다. 폐손상의 정도는 습건중량비와 급성 폐손상지수로 측정하였고, MMP-9의 발현은 면역조직화학염색으로 고찰하였다. 결 과 : 습건중량비, 급성 폐손상지수 및 MMP-9의 발현이 HVT 군에서 다른 두군에 비하여 유의하게 높았고(p<0.05), HVT+MMP군에서 HVT군에 비하여 폐손상의 정도 및 MMP-9의 발현이 현저하게 낮았다(p<0.05). 결과적으로 MMPI의 투여가 MMP-9의 발현을 저하시킴으로써 기계환기로 인한 폐손상의 정도를 유의하게 감소시키는 것으로 관찰되었다 결 론 : 많은 일호흡량과 PEEP을 적용치 않은 기계환기는 폐조직에서 MMP-9의 발현을 유의하게 증가시켜 폐손상을 유발하고, MMPI는 MMP-9의 작용을 억제함으로써 기계환기로 인한 폐손상의 정도를 유의하게 감소시키는 것으로 판단된다.
