The essential oils from the roots of Codonopsis lanceolata and the cultivated callus were analysed and compared by gaschromatography-mass spectrometry. In the experimental study of cell culture, it appeared that 2,4-dichlorophenoxy acetic acid in the culture medium induced higher production of essential oils in the callus than indole acetic acid. The growth of callus was inhibited by illumination of the light. The production of essential oil in cultured cells was increased by the addition of biosynthetic precursors. The essential oils from the roots of Codonopsis lanceolata and the cultured callus showed different compositions. Tetradecanoic acid, 1,1,-dimethoxyl 4-methoxy phenol, 9,12-octadecanoic acid and hexadecanoic acid were identified as main components of the cultured callus oil.

In general, stepwise hot steaming process is known to be effective in improving its biological activities; however, not much employed in processing Codonopsis lanceolata due to its hardness. The complex processed C. lanceolata showed highest free radical scavenging acitivity as 45.21%. Total phenol and flavonoid content were of complex processed C. lanceolata higher than conventional extract and alone steaming process. It was showed the lower melanogenesis rate on melanin production test by B16F10 cells as 27.46%. High inhibitory of tyrosinase was also measured as 28.61% by adding steamed Codonopsis lanceolata extracts by high pressure extraction of 1.0 $mg/m{\ell}$. And anti-wrinkle activity were 39.08%. In comparing phenolic acids profiles in the extract, in general higher amounts of polyphenol were obtained possibly by easy release of active components during thermal processing, which results in better antioxidant activities than that of general extract. This findings can also be supported by result that the extract by steaming process showed better activities than the general extraction extract.
The chromosome number was identified and fluorescence in situ hybridization(FISH) mapping of 5S and 45S rDNAs were conducted for C. minima and C. lanceolata in the genus Codonopsis from Jeju island. In this study, we have confirmed that the somatic metaphase chromosome number determined as 2n=2x=16 was the same as the findings from the previous studies. While the conventional staining method makes it rather difficult to distinguish satellite chromosomes due to high degree of variability, FISH analysis produced the exact number and location of 5S and 45S rDNAs. Both species in the genus Codonopsis have a pair of 5S rDNA and their gene loci were observed on chromosome 3. Although two pairs of 45S rDNAs (one on chromosome 1 and the other on chromosome 8) were identified in both species, the 45S rDNA signals on chromosome 8 in C. minima were significantly weaker than those on chromosome 1. In addition, the 45S rDNA signals on chromosome 1 in C. lanceolata showed that the chromosome is non-homologus. In this study, we have determined cytogenetic characteristics of C. minima and C. lanceolata according to their gene replication patterns.
This study was conducted to develop optimal control tactics of the northern root-knot nematode, Meloidogyne hapla, using cultural method and biological agents {Bacillus thuringiensis (Bt), Paecilomyces lilacinus and plant extract (Huhjunl)} in the fields of Codonopsis lanceolata. Germination of C. lanceolata was susceptible to fosthiazate, but not to Bt or a plant extract. In pot assay, the inhibitory effect of two microbial agents, Bt and Paecilomyces lilacinus, on M. hapla were significant, but less than that of fosthiazate. The plant extract also had significantly inhibitory effect on M. hapla. In field assay, treatments of P lilacinus and fosthiazate resulted in maximal yields and qualities of C. lanceolata. The effect of the plant extract on the yields of C. lanceolata was also better than no treatment. The nematode-occurring condition of the fields before transplanting had significant effect on development of C. lanceolata; nematode-occurring field type gave less yields than nematode-free field type. These results suggest that a cultural control technique using paddy field, microbial pesticides using Bt or P lilacinus, and the plant extract are the promising control tactics against M. hapla in C. lanceolata fields. As a field manual to decrease economical damage of C. lanceolata due to M. hapla, this study suggests that C. lanceolata can be cultured directly in paddy field or in upland field after nematode control using microbial agents or the plant extract.

This study was done to investigate the antidiabetic effect of ethanol extract from Codonopsis lanceolata root in Streptozotocin (STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose 45mg/kg.b.w. dissolved in citrate buffer(pH4.5). The ethanol extract of Codonopsis lanceolata root was orally administrated once a day for 7 days. The contents of serum glucose, triglyceride(TG) and total cholesterol were significantly decreased(p<0.05) in Codonopsis lanceolata root treated group compared to the those of STZ-control group. Also the content of hepatic glycogen and activities of glucose-phosphate dehydrogenase(G-6-PDH) and glucokinase(GK) were significamtly increased(p<0.05). These results indicated that ethanol extract of Codonopsis lanceolata root would have antidiabetic effect in STZ-induced diabetic rats.
Seed of Codonopsis lanceolata exhibits low germination due to impermeable seed coat. Prolonged seed dormancy can be overcome by the application of gibberellins, as it promotes growth of the embryo and weakens the structures surrounding of embryo. The effects of photoperiod, sugar and gibberellin concentration were investigated at constant temperature for 12 days and 22 days in vitro and invivo conditions respectively. The rate of germination of seeds of Codonopsis lanceolata in wet filter paper in both complete dark and light treatments was significantly lower than that of seed treated with $GA_3$. It clearly indicates that there is significant coat imposed dormancy in the seed of Codonopsis lanceolata. The rate of germination in vivo condition was lower than that of the in vitro condition supplemented with $GA_3$ Thus, the physical dormancy due to impermeable seed coat and low level of endogenous gibberellins in the seed was the cause of poor germination rate in Codonopsis lanceolata.
The immunomodulatory effect of Codonopsis lanceolata based on the production of cytokines and the activation of macrophage was studied. The mRNA expression of nitric oxide synthase (iNOS) was gradually induced after 24 hr treatment of Codonopsis lanceolata, and NO production was a maximum after 24 hr treatment with 1 mg/mL. RAW 264.7 cell on in vitro treatment with Codonopsis lanceolata induced mRNA of cytokines such as interleukin-1(IL-1)${\beta}$, interleukin-6(IL-6), tumor necrosis $factor(TNF)-{\alpha}\;and\;interferon(IFN)-{\gamma}$; $IL-1{\beta}$ and IL-6 mRNA were gradually induced up to 24 hr, $TNF-{\alpha}\;mRNA$ was regularly induced up to 24 hr, and $IFN-{\gamma}\;mRNA$ level was a maximum within 1 hr. These results suggest that Codonopsis lanceolata exerts as an effective immunomodulator and enhances antitumor activity of macrophages.
