Characterization of Plasmids from Multiple Antibiotic Resistant Vibrio sp. Isolated from Molluscs and Crustaceans

  • This study investigated the role of plasmids and their relationship with the multiple antibiotic resistance of 30 Vibrios sp. isolated from molluscs and crustaceans sampled from the Kerala coastal waters of India. The biochemical identification and antibiotic resistance profiles were determined, followed by the plasmid profiles, conjugation and transformation efficiencies. The results showed a considerable difference in the level of bacterial resistance to various antibiotics; while all 30 strains were found to be MAR Vibrios sp. and their resistance patterns varied. All the strains were resistant to amoxycillin, ampicillin and carbeniciliin. 87% were resistant to rifampicin; 74% to cefuroxime; 67 to streptomycin; 53% to norfloxacin and ciprofloxacin and 47% to furazolidone and nalidixic acid. In addition to their antibiotic resistance, the plasmid DNA of the MAR Vibrios strains isolated from the molluscs and crustaceans was also studied. Nine strains isolated from crustaceans and molluscs were found to harbor 1-3 plasmids with sizes varying from 5. 98 kb to 19. 36 kb. The average transformation efficiency was about $5{\times}10^{-8}$ and the conjugation efficiency varied from $2.1{\times}10^{-3}$ to $10^{-9}$. A further study of antibiotic resistance patterns may be useful to test the extent of drug resistance in seafoods and help to devise a nationwide antibiotic policy.

  • Lockheed Martin has integrated and installed numerous AIS systems around the world as part of several VTS installation contracts and technology demonstrations for customers. Each of these systems incorporates specific customer requirements addressing differing objectives. This paper describes the capabilities and features of Lockheed Martin's current implementation of the Universal AIS system, and the experiences Lockheed Martin has had with earlier AIS and ADS (automated dependent surveillance) system installations. Specific examples are provided for an amphibious landing exercise with the US Army; installations at a Middle-East Oil port; technology demonstrations in New York Harbor, USA; river systems in New Orleans, Louisiana, USA; and coastal applications in the Gulf of Suez. The paper closes a brief overview of the most recent activity, the Turkish Straits VTMIS project that requires Universal AIS installations. The evolution of the technology and lessons learned will be discussed in the context of these examples. The perspective will be on Lockheed Martin's experience working with different customers to integrate AIS technology into a larger VTS or Port Information System. It will also discuss, from the practical side, how AIS information is used by a variety of maritime organizations with differing needs and information objectives.

A Study on the Charge of Water Quality in the Vicinity of Mokpo Harbor due to the Discharges from Yongsan River Estuary Weir and Yongam-Kumho Sea Dike (영산강 하구둑과 영얌-금호방조제 방류에 의한 목포항 주변 수역의 수질변화에 관한 연구)

  • It is essential for port planning, coastal zone management and environmental impact study to analyze the variation of current and water quality due to the development of water area and discharged water from the estuary barrage and basin, etc. Mokpo sea area has downstream from a long river and two large basins, the Yongsan river and Yongam-Kumho basins, discharging much of water through water gates for the purpose of flood and prohibition of salt intrusion to the inland fresh water area. In this study, the numerical calculation were carried out for the analysis of diffusion characteristics due to discharging operation, adopting the results of tidal current simulation. ADI method is applied to the governing equation for the movement of sea water and diffusion and 6-point method to the advection terms of diffusion equation. As the results of this study, it is known that the discharging operation causes increasing and/or decreasing of current velocity and enlarging and/or depressing of pollutant diffusion limits depending on the distance from the discharging gates and the modes of discharging operation. To utilize these result, the linked gate operation and the method increasing exchange of sea water must be considered.

  • 최근 경제의 발전과 근무형태의 변화로 인한 여가활동이 점차 증가함에 따라 해안지역에 관광인프라 건설이 늘어나는 추세이다. 본 연구는 동해안 지역의 국제적 성장 가능성과 지역의 자원 개발 잠재력을 토대로 한 광역관광권 개발사업의 일환으로 제안된 울산과 포항사이의 양포항에 복합해양관광휴양단지 구상안에 대한 주변수역의 변화양상을 파악하고자 하였다. 이에 신뢰성 있는 수치도형 구현을 위해 3차원 수치모델을 구축하고 이를 바탕으로 해양관광휴양단지 조성시 발생하는 파동 및 유동장의 변화, 표사 및 해저지형 등의 해양환경변화를 예측하여 구상단계에서 기초자료로 활용하고자 하였다.

Benthic dinoflagellates in Korean waters

  • The occurrence of benthic dinoflagellates, many of which are known to be toxic, is a critical concern for scientists, government officers, and people in the aquaculture, dining, and tourism industries. The interest in these dinoflagellates in countries with temperate climate is increasing because tropical or subtropical species introduced into temperate waters by currents are able to survive the winter season in the new environment owing to global warming. Recently, several species from the benthic dinoflagellate genera Amphidinium, Coolia, Ostreopsis, Gambierdiscus, and Prorocentrum have been reported in the waters of the South and East Sea of Korea. The advent of the benthic dinoflagellates in Korean waters is especially important because raw or slightly cooked seaweeds, which may harbor these benthic dinoflagellates, as well as raw fish, which can be potentially intoxicated by phytotoxins produced by some of these benthic dinoflagellates, are part of the daily Korean diet. The recent increase in temperature of Korean coastal waters has allowed for the expansion of benthic dinoflagellate species into these regions. In the present study, we reviewed the species, distribution, and toxicity of the benthic dinoflagellates that have been reported in Korean waters. We also provided an insight into the ecological and socio-economic importance of the occurrence of benthic dinoflagellates in Korean waters.

  • Unlike in the past, port space in modern society not only serves to meet port functions but also other functions such as housing, leisure, and sightseeing. As a result, more and more people are turning into a dense space. Port space, on the other hand, is also a space vulnerable to disasters which can be attributed to geographical and environmental characteristics. Despite the sensitivity if the issue, in Korea disaster prevention and urban planning have not been clearly established. Therefore, there is a need to provide safety and disaster preventive measures in urban planning considering the characteristics of harbor space.

Characteristic Distributions of Nutrients and Water Quality Parameters in the Vicinity of Mokpo Harbor after Freshwater Inputs (담수 유입에 따른 목포항 주변해역의 영양염 및 수질인자 분포 특성)

  • The Mokpo coastal waters receive discharges from three artificial lakes(Youngsan, Youngam, Geumho) and other terrigenous freshwater inflows(streams, sewage treatment effluent, fresh groundwater), which exhibit very high concentrations of nutrients and/or organic matters. To understand spatial distributions of nutrients(DIN, DIP, DSi) and other water quality parameters(Chl-a, water temperature, salinity, DO, COD, SS), field surveys were conducted at 10 stations in the Mokpo harbor and adjacent estuaries on May, July, September, and November 2008 within 10 days following discharge events from artificial lakes. In this study, the freshwater flow rate influxed by the operation of sea dike sluice had significant influence on water qualities of the Mokpo coastal waters, although nutrient concentrations in other freshwater sources such as streams, sewage treatment effluent, and fresh groundwater were much higher. As a result of statistical analysis, DIN, COD, and Chl-a had a negative correlation with salinity. Therefore it was shown that discharge extents, time, and nutrients from the Youngsan lake were major impact factors dominating the spatial characteristics of nutrients and other water quality parameters in the Mokpo harbor and adjacent waters. However, despite non-discharge from the Youngsan Lake on September of this investigated period, it was observed that the nutrient addition was taking place in the lower layer of the estuary suggesting nutrient supply through different pathways. This result has emphasized the need to implement the combined assessment about the cumulative impacts on the Youngsan Estuary environment and ecosystem due to freshwater inputs derived from the artificial lakes as well as other terrigenous inflows, or benthic releases.

Effect of Freshwater Discharge from a Water Reservoir on the Flow Circulation in the Semi-Closed Harbor (유수지로부터의 담수 방류가 항 내 해수순환에 미치는 영향)

  • To investigate the effect of freshwater discharge on the seawater circulation in the semi-closed harbor, a 3-D hydrodynamic model was applied to the International Ferry Terminal (IFT). The model run is conducted for 45 days (from May 15 to June 30, 2020), and the reproducibility of the model for time-spatial variability of current velocity and salinity was verified by comparison with model results and observation data. There are two sources of freshwater towards inside of the IFT: Han River and water reservoir located in the eastern part of IFT. In residual current velocity results, the two-layer circulation (the seaward flow near surface and the landward flow near bottom)derived from the horizontal salinity gradient in only considering the discharge from a Han River is more developed than that considering both the Han River and water reservoir. This suggests that the impact of freshwater from the reservoir is greater in the IFT areas than that from a Han River. Additionally, the two-layer circulation is stronger in the IFT located in southern part than Incheon South Port located in northern part. This process is formed by the interaction between tidal current propagating into the port and freshwater discharge from a water reservoir, and flow with a low salinity (near 0 psu) is delivered into the IFT. This low salinity distribution reinforces the horizontal stratification in front of the IFT, and maintains a two-layer circulation. Therefore, local sources of freshwater input are considered to estimate for mass transport process associated with the seawater circulation within the harbor and It is necessary to perform a numerical model according to the real-time freshwater flow rate discharged.

Pollutant Content of the Sediments in the Drain Channel of Paved Catchments in Coastal Areas (연안역 불투수지역 배수구 퇴적물의 오염특성에 관한 연구)

  • In this study, the particle size distribution and pollutants content of sediments collected from different sites in the coastal area of Chungnam province were analyzed. The sediment samples were collected from different parking lots near beaches, harbors, museum and bridges. The particle size distribution analysis showed that the particle is generally $106-500{\mu}m$ and the effective size $D_{10}$ ranges from 40 to $60{\mu}m$ while the $D_{50}$ and $D_{60}$ ranges from 200 to $810{\mu}m$ and 235 to $1005{\mu}m$, respectively. For particle size $D_{10}$, there was no significant difference in all sites. However, for $D_{50}$ and $D_{60}$, the range is large. Sediment analysis implicates that as the particle size decreases, the pollutant content increases. This is because smaller particles have higher specific surface area resulting to have more adsorption capacity. Particles from tires, emission gas from vehicles and dust particles belongs to smaller particles. For particle sizes less than $63{\mu}m$, the analysis showed that as the particles become coarser, the concentration of VS, $COD_{cr}$, TN, and TP is at least 2 to 14 times higher. Cu and Pb were detected in all sites and shows a higher concentration with smaller particle size. Cu concentration are almost the same for all sites but in the case of Pb, the sediments from Sinjindo has higher concentration of up to 2 to 3 times as compared to those collected from the other sites. In the Beach site as well as in the Seocheon Ocean Museum, Cd was contained only in fine particles. However, in Daechon Harbor and Sinjindo Bridge sites, Cd was detected in all the sediment particle size.

Development of a Dynamic Deformable Rubber Membrane Parapet to Cope with the Long Term Sea Level Rise and the Abnormal Waves (장기해수면 상승 및 이상파랑에 대비한 동적 가변형 고무막체 파라펫 개발)

  • It's been reported that the global warming effect has invoked the ever increasing typhoon intensity and long-term sea level rise which jointly cause severe wave overtopping over breakwaters or shore dykes. A simple measure to cope with this undesirable change may be just to increase the crest height of the dykes and breakwaters. This is surely effective to prevent wave overtopping, but it also decreases the seaward visibility of coastal waterfront. In this paper, a dynamic deformable rubber membrane parapet which not only reduces wave overtopping in storm period but also secures seascapes in normal days is presented. Several optimal configurations of the parapet are proposed. Through numerical analyses using a nonlinear finite element model and hydraulic experiments, the air controlled expansion and contraction of the parapets, their behavior against wave overtopping and structural stability are investigated.
