  • Disposal of waste containing heavy metals into the environment is a major threat to human health and can result in toxic or chronic poisoning in aquatic life. In the current study, metal-resistant Raoultella ornithinolytica was isolated from metal-contaminated samples collected from the Tanjaro River, located southwest of Sulaymaniyah, Iraq. R. ornithinolytica was identified by partial amplification of 16S rRNA. The uptake potency of heavy metals was assessed using inductively coupled plasma-optical emission spectroscopy (ICP-OES) and indicated that R. ornithinolytica removed 67, 89, 63.4, 55.6, 56.5, 65, and 61.9% of Cd, Pb, Cr, Ni, Zn, Co, and Fe, respectively. These removal rates were influenced by temperature, pH, and contact time; at 35℃ and pH 5 with a change in the incubation time, the reduction rate improved from 89 to 95% for Pb, from 36.4 to 45% for Cu, and from 55.6 to 64% for Ni. Gene analysis indicated that R. ornithinolytica contained pbrT, chrB, nccA, iroN, and czcA genes, but the pcoD gene was absent. Energy-dispersive X-ray spectroscopy (EDS) images showed evidence of metal ion binding on the cell wall surface with different rates of binding. Transmission electron microscopy (TEM) detected different mechanisms for metal particle localization; cell surface adsorption was the main mechanism for Pb, Zn, and Co uptake, while Cd, Ni, and Fe were accumulated inside the cell. The current study describes, for the first time, the isolation of R. ornithinolytica from metal-contaminated water, which can be used as an eco-friendly biological expedient for the remediation and detoxification of metals from contaminated environments.

  • As an idea of healing or fatigue recovery, a desire for massage therapy has increased for modern people who complain of pain such as knotted muscles or shoulder discomfort due to fatigue accumulation and stress. To resolve this, although a massage chair has been developed, it does not provide the most efficient massage as it is not suitable for Korean body types. In this paper, this research analyzed massage, assembly, massage intensity, Korean body types and functions of the massage for maximized efficiency to design critical functions. Also, we implemented the action control and algorithm of the massage chair for localization.

  • The present study was undertaken to evaluate the chemical characteristics of Asian dust (hereafter called "AD") particles with the aid of the most advanced micro-PIXE (Particle-induced X-ray emission) analytical technique. To this end, size-selected particles were sampled on a rural peninsula of Korea (Byunsan, 35.37N; 126.27E) during AD and non-AD periods in 2004. The coarse particle (> $2{\mu}m$) number density during an AD event were 170 times higher than those of the non-AD counterpart. The average net-count of silica in individual particles collected on AD event was roughly 11 times higher than that of non-AD counterpart. The X-ray net-counts of trace elements (Zn, Co, Mn, and V) were also considerably high in AD relative to the non-AD day. Particle classification based on the inter ratio analysis of elemental net-count suggests that a large portion of the coarse particles collected during AD event underwent chemical transformation to a certain degree. The visual interpretation of micro-PIXE elemental maps and elemental localization data in and/or on individual AD particles clarified the internal mixture of AD particles with sea-salt and artificial metallic particles.

  • We previously identified cancer-upregulated gene 2 (CUG2) as a commonly up-regulated gene in various human cancer tissues, especially in ovary, liver, and lung (Lee et al., 2007a). CUG2 was determined to be a nuclear protein that exhibited high proto-oncogenic activities when overexpressed in NIH3T3 mouse fibroblast cells. To identify other cellular functions of CUG2, we performed yeast two-hybrid screening and identified CENP-T, a component of CENP-A nucleosome complex in the centromere, as an interacting partner of CUG2. Moreover, CENP-A, the principle centromeric determinant, was also found in complex with CENP-T/CUG2. Immunofluorescent staining revealed the co-localization of CUG2 with human centromeric markers. Inhibition of CUG2 expression drastically affected cell viability by inducing aberrant cell division. We propose that CUG2 is a new component of the human centromeric complex that is required for proper chromosome segregation during mitosis.

  • Environmental quality regulations are used to establish the maximum permissible concentrations of heavy metals before a site can be deemed to be polluted or contaminated. This paper compares the results obtained by 0.IN HCl extraction and acid extraction method(HNO$_3$+HClO$_4$.HCl). The leaching efficiency of 0.1N HCl extraction is directly proportional to the final pH of leachate, due to the different solubilities of the heavy metals at different pH values, The severe differences between 0.1N HCl-extractable and total metal contents result mainly from the buffering effect of carbonates, present in sediment. samples. Application of sequential extraction experiments to some sediments collected from gully pot in Seoul illustrates a much stronger scavenging effect by Fe and Mn-hydroxides, carbonates and organic phases. As deduced from both sequential extraction and leaching experiments, the relative mobility of heavy metals is found to be: Mn>Zn>>Co>Cd>>Cu>Pb>Cr>Fe, in spite of large differences in heavy metal content and localization. Changes in the physicochemical environments (such as acidification) caused by a traffic accident may result in the severe environmental pollution of heavy metals of surrounding area( surface water and rivers).

  • Periodontal ligament cells may have a role in the regulation of hard and soft periodontal tissues, but their specific function has not yet to be determined. To evaluate further their role in periodontal regeneration, they were examined for osteoblast-like behavior. Periodontal ligament cells and gingival fibroblasts were primarily cultured from extracted premolar with non-periodontal diseases. Cells were cultured with DMEM at $37^{\circ}C$, 5% $CO_2$, 100% humidity incubator, and as a measure of cell characterization, it was examined that the morphology, alkaline phosphatase activity, collagen synthesis, and immunocytochemistry for osteonectin, osteocalcin, and collagen type I. Healthy periodontal ligament cells has more osteoblastic-like cell property in alkaline phosphatase activity. and collagen synthesis than gingival fibroblast. Immunocytochemistry localization explained that calcitonin were expressed in periodontal ligament cells only, and osteonectin and type I collagen were produced in both cells simultaneously. This results indicate that the growth characteristics of periodontal ligament cells and gingival fibroblasts exhibit some differences in proliferative rates and biochemical synthesis. The differences may help to calrify the role such cells play in the regenearation of periodontal tissues.

  • Acid-sensing ion channels (ASICs) are proton-gated cation channels widely expressed in the nervous system. Proton sensing by ASICs has been known to mediate pain, mechanosensation, taste transduction, learning and memory, and fear. In this study, we investigated the differential subcellular localization of ASIC2a and ASIC3 in heterologous expression systems. While ASIC2a targeted the cell surface itself, ASIC3 was mostly accumulated in the ER with partial expression in the plasma membrane. However, when ASIC3 was co-expressed with ASIC2a, its surface expression was markedly increased. By using bimolecular fluorescence complementation (BiFC) assay, we confirmed the heteromeric association between ASIC2a and ASIC3 subunits. In addition, we observed that the ASIC2a-dependent surface trafficking of ASIC3 remarkably enhanced the sustained component of the currents. Our study demonstrates that ASIC2a can increase the membrane conductance sensitivity to protons by facilitating the surface expression of ASIC3 through herteromeric assembly.
