For the sustained release formulation of recombinant human growth hormone (rhGH), dissociable rhGH aggregates were microencapsulated within poly(D,L-lactic-co-glycolic acid) [PLGA] microparticles. rhGH aggregates with 2 - 3 m Particle diameter were first produced by adding a small volume of aqueous rhGH solution into a partially water miscible organic solvent phase(ethyl acetate) containing PLGA. These rhGH aggregates were then microencapsulated within PLGA polymer phase by extracting ethyl acetate into an aqueous phase pre-saturated with ethyl acetate. The resultant microparticles were 2 - 3 m in diameter similar to the size of rhGH aggregates, suggesting that PLGA polymer was coated around the protein aggregates. Release profiles of rhGH from these microparticles were greatly affected by changing the volume of the incubation medium. The release rhGH species consisted of mostly monomeric form with having a correct conformation. This study reveals that sustained rhGH release could be achieved by microencapsulating reversibly dissociable protein aggregates within biodegradable polymers.
When reserpine-producing cell strains of Rauwolfia serpentina were transferred from the dark to the light irradiation, the production of reserpine was extremely enhanced whereas the cell growth was suppressed. In an incubation period of 20 days, the most effective culture condition for reserpine production was the combination of 8 days of dark culture and following 12 days of light culture. The time courses of both cell growth and reserpine production were measured in vitro in order to clarify the effect of wave length range of light on the biosynthesis of reserpine. Although the growth of cultured cells which had been incubated under continuous red, yellow, and green lights, respectively, was similar to that of the cultured cells subcultured in the dark. The cells cultured under red light irradiation produced less reserpine than dark-grown cultures. Both blue and near-ultraviolet light inhibited the growth of cultured cells. The production of reserpine was strikingly enhanced by blue light, but was strongly inhibited by near-ultraviolet light.

One Helicosporium strain, isolated from a wilted chestnut tree, evidenced in vitro antimicrobial activity against various types of bacteria and fungi, and generated a diffusible pigment. The antimicrobial compounds and the diffusible pigment of the Helicosporium sp. isolate were purified via solvent fractionation, column chromatography, and recycling preparative chromatography. Both the major antimicrobial compound and the diffusible pigment were identified as 2-methylresorcinol via nuclear magnetic resonance spectroscopy. Therefore, 2-methylresorcinol, a diffusible pigment generated by Helicosporium sp., appears to be an active antimicrobial principle. This pigment also exhibited considerable cytotoxicity against mammalian cells.
In this study, biological production of fumaric acid by Rhizopus oryzae KCTC 6946 using rotary biofilm contactor was investigated. In study of neutralizing agent on fumaric acid production, $Na_2CO_3$ was more effective than NaOH. After 24 hr of incubation with a rotating speed of 10 rpm at $35^{\circ}C$, biofilm was grown on and around the surface of the disks. The yield and volumetric productivity of rotary biofilm contactor were 33.8% and 0.595 g/L${\cdot}$h, respectively, with the optimum effective disk surface area of 1,583 $cm^2/L$.
Thousand actinomycetes and 50 soil samples were used for the isolation of microorganisms producing yeast cell wall lytic enzymes. Among 493 strains producing large clear zones on autolysed washed yeast (AWY), 117 strains were selected on living yeast cell agar plates. With the method of lytic activity, one strain (St-1702) was selected, which was temporarily identified as Streptomyces eurythermus. The optimal condition for enzyme production of this strain was partially determined as follows: incubation of the strain for 3 days at 30$\circ$C in the medium containing 2% freeze dried yeast cell, 1% glucose, 1% K$_{2}$HPO$_{4}$, 0.01% MgSO$_{4}$'7H$_{2}$O, 0.5% peptone, and 0.2% (NH$_{4}$)$_{2}$CO$_{3}$ with pH 7.0. The protoplast formation of yeast by using the enzyme produced by this strain was compared with commercial enzymes.
To examine whether the xylene component of BTX (benzene, toluene, xylene) mixture is cometabolized and residues are produced in soil, $\^$14/C-labeled-0-xylene was added to sandy loam in combination with unlabeled benzene and toluene. After 4 weeks of incubation in a sealed system connected to an oxygen reservoir, 55.1% of the radiocarbon was converted to $\^$14/CO$\sub$2/, 3.0% was to 95.8% radiocarbon recovery. Biomass incorporation of o-xylene radiocarbon which was detected by fumigation/extraction was usually low (5.6%), but 32.1% radiocarbon became associated with soil humus. Most of the numus-bound radiocarbon was found in humin fraction. In addition to o-xylene, p-xylene and toluene also showed similar results. The evidence shows that some of their reactive methylcatechol biodegradation intermediates attach to the humic metrix in soil in preference to mineralization and biomass incorporation.
Adenovirus which is an important infectious viral agent in respiratory and alimentary tract was investigated in Pusan, 1999. Fifteen cases of adenovirus were detected from stools and throat swabs of suspected patients. Two cases of enteric adenovirus were detected from a 5 years old boy and a 6-month-old boy. Thirteen cases of respiratory adenoviruses were detected from children aged under 10 years old and one adult. From respiratory specimens, 1 case of adenovirus type 2, 1 case of type 5, and 11 cases of type 3 were found. Enterotype 41 was detected from fecal preparations. Adenoviruses appeared mostly during winter months, January, February and December. Adenovirus showed a slowly progressive cytopathic effect on HEp-2 cells, Vero cells and BGM cells at 37$^{\circ}C$, in a 5-7% $CO_{2}$ incubation. An electron microscopic observation exhibited non-enveloped icosahedron with a diameter of 70nm. No significant differences on cytopathic effect and morphological features have been found from specimens of either alimentary tract or respiratory secretions.
To follow the metabolic fate of aglycone of ginseng saponins,in vitro and in vivo experiments were performed. Incubation of 20(S)-prtopanaxatriol (1) with rat liver S9 fraction afforded unique ocotillol derivatives, 20, 24-epoxysides (3 and 4). Also 20(S)-prtopanaxadiol (2) gave the corresponding epoxides (5). Healthy volunteers were taken with Sanchi Ginseng, which contains protopanaxatriol and protopanaxadiol saponins and no ocotillol saponins. From the alkaline hydrolysate of the urine samples of these volunteers,3 was detected as well as 1, and the ratio of 3/1 increased up to 2.0 at the maximum at 50 hrs. Biochemical significance of the ocotillol derivatives is discussed, since the main bioactive saponin in Panax vietnamensis is an ocotillol-type saponin, majonoside R2 (7).

(1R,9S)-(${\beta}$)-Hydrastine (HS) at 10-50 ${\mu}$M has been proven to have an inhibitory effect on dopamine biosynthesis in PC12 cells by the inhibition of tyrosine hydroxylase (TH) activity and TH gene expression. In the present study, therefore, the effects of HS on the basal and K$\^$+/-induced dopamine release, and Ca$\^$2+/ influx induced by high K$\^$+/ and caffeine in PC12 cells were investigated. The dopamine release by high K$\^$+/ (56 mM) was inhibited by co-incubation of 20 ${\mu}$M HS. Application of HS also significantly reduced the magnitude of the maintained Ca$\^$2+/ influx induced by K$\^$+/ depolarization. (omitted)
본 연구는 비섬유탄수화물(NFC)과 반추위분 해단백질(RDP)을 젖소사료에 다양하게 배합하여 NFC : RDP의 비율을 3.5, 3.0 및 2.5로 다양하게 조정하였을 때, in vitro 발효성상 및 젖소의 산유성적에 미치는 영향을 조사하여 적정 NFC : RDP비율을 결정하고자 실시하였다. In vitro 발효시험은 반추위 cannulae가 장착된 Holstein 젖소로부터 반추위액을 얻어 3, 6, 9, 12시간 동 안 배양하였다. 반추위내 pH는 NFC비율이 증가함에 따라 유의한 영향을 받지 않았고, 암모 니아질소농도는 배양 6시간까지 처리군간에 유의한 차이가 나타나지 않았으나, 9시간과 12 시간에서는 NFC : RDP비율이 증가함에 따라 유의하게 감소하였다(P < 0.05). 휘발성지방산생산 량은 acetate 및 propionate농도가 발효 3시간에 NFC함량이 증가함에 따라 유의하게 증가하였 으나(P < 0.05), 나머지 시간대에서는 유의차가 나타나지 않았다. Valerate 및 A : P비율은 NFC : RDP비율에 의하여 영향을 받지 않았고, iso- acids 및 총 휘발성지방산은 전반적으로 NFC함 량이 증가함에 따라 유의하게 증가하였으며(P < 0.05), 3.0비율구가 가장 높은 값을 나타내었다(P < 0.01). 한편, 사양시험은 18 두의 착유우를 공시하여 처리구당 6 두씩 배치하여 총 24주간 실시하였 다. 건물 및 에너지섭취량은 NFC : RDP비율이 증가함에 따라 유의하게 증가하였고(P < 0.01), 섬 유소섭취량은 NFC : RDP비율이 감소함에 따라 직선적으로 증가하였다(P < 0.01). 산유량은 3.5(32.7 kg)비율구가 다른 처리구보다 유의하게 높았다(P < 0.05). 유지방율은 3.0(3.79 %) 및 2.5(3.79 %)비율구가 3.5(3.48 %)비율구보다 유의하 게 높았고(P < 0.05), 유지방생산량은 처리구간 에 차이가 나타나지 않았다. 유단백질과 무지 고형분은 NFC : RDP비율이 증가함에 따라 직선 적으로 증가하였다(P < 0.01). 그러나 우유중 요소태질소농도는 NFC함량이 증가함에 따라 유 의하게 감소하였다(P < 0.05). 이상의 결과로부터 사료내 증가하는 수준의 NFC는 반추위발효, 질소이용효율 및 산유량을 향상시켰고, 사료중 NFC : RDP비율이 3.0이상일 때 반추위발효뿐만 아니라 산유성적이 최대화되는 것으로 나타났다.
