Objectives: Effectiveness and conditions of vapor-phase hydrogen peroxide (VPHP) system on decontamination of Geobacillus stearothermophilus(GS) spores, Escherichia coli (E.coli) and Enterobacteria phage felix01 (felix01) were determined. Methods: The VPHP system was designed to vaporize 35% (w/w) solution of hydrogen peroxide, continuously to inject and withdraw VPHP. The system and VHP 1000ED (Steris) were operated such that dehumidification and conditioning were initiated without samples in the chamber. Then the samples were loaded into and removed. Coupons (glass, anodizing, silicon, viton) with GS spores ($1{\times}10^6$ colony forming unit/mL [CFU/mL]), E.coli ($1{\times}10^7$ CFU/mL) and felix01 ($1{\times}10^7$ plaque forming unit/mL[PFU/mL]), and Biological Indicator (BI) with GS spores ($1{\times}10^6$ CFU/mL) on stainless steel coupons were used. The tested samples were sonicated and vortexed, and then were plated for enumeration, followed by incubation at $55^{\circ}C$, 24 hr for GS spores, and at $37^{\circ}C$, 24 hr for E.coli and felix01. BI analysis in broth culture was only qualitative. Results: The efficacy of the VPHP system on decontamination was almost equivalent to that of VHP 1000ED. The conditions for complete decontamination with the VPHP system was as follows: concentration; 700~450 ppm, relative humidity; approximately 55%, and temperature; $34{\sim}32^{\circ}C$. When comparing the decontamination efficiency among different kinds of coupons, glass was the most effective, however, all kinds of coupons were decontaminated completely after 60 min exposure in both systems. Conclusion: The VPHP system can be recommended as an alternative system for traditional system using ethylene oxide, formaldehyde or chlorine dioxide.
Culture conditions of yellow pigment in Monascus sp. were studied. According to the studies of culture conditions optimum condition was found to be pH 4.5, 3 days of incubation with 3% of sucrose as carbon source, 0.2 % of yeast extract as nitrogen source and 75m1 of medium in the 500m1 erlenmyer flask by rotary shaking (rpm 180) at 180 r.p.m. Effective levels of inorganic compounds were found to be 0.25 % of potassium phosphate monobasic and 0.1 % of Magnesium sulfate.
The ovaries of Korean Native cows or heifers were obtained from an abattoir and kept on 20 to $25^{\circ}C$ and transported to laboratory within 2 hrs. The follicular oocystes were collected from 2~6mm follicles in diameter and classified into 3 grades by the morphology of cumulus cells attached. The oocytes were matured in vitro(IVM) for 24 hrs. in TCM-199 supplemented with $23{\mu}g/ml$ FSH, $10{\mu}g/ml$ LH, $1{\mu}g/ml$ estradio-17 ${\beta}$ and granulosa cells at $39^{\circ}C$ under 5% $CO_2$ in air. They were fertilized in vitro(IVF) by incubation for 12 hrs. of epididymal spermatozoa pretreated with heparin, and then the zygotes were co-cultured in vitro(IVC) with oviductal epithelial cells for 7 to 9 days. Assessment of maturation revealed that 93.0%(147/158) of grade I oocytes had expanded of cumulus cells, which was higher(p<0.05) than the 79.4%(85/107) of grade II oocytes. Compared to epididymal sperm(32.9%), the insemination with frozen and thawed sperm resulted in slightly lower(20.5%), but not significant, development to morulae and blastocysts from grade I oocytes. Co-culture of bovine IVF embryos with oviductal epithelial cells improved the development to transferable embryos significantly(38.1%), compared to co-culture with granulosa cells(20.0%). When VF bovine embryos were vitrified at blastocyst, the post-thaw survival rate was obtained higher resulf for 1 min. equilibration time(82.6%) or 2 min.(73.9%) than 3 min.(18.2%) in EFS solution.

This study is conducted to investigate the effects of anaerobic treatments of swine manure slurry alone and combination of livestock manure slurry and fruit pomace on biogas production. Anaerobic co-digestion was evaluated in mesophilic tank reactors for 96 day-incubation period. The organic matter loading of anaerobic digestion was 1 kg of volatile solids(VS) per $1m^3{\cdot}day$. The highest methane production was achieved from the combination of swine manure slury and mandarin pomace(70:30) treatment, whereas the lowest daily and cumulative methane yields was observed in swine manure slurry alone treatment. More than two-fold increase in bio-gas and methane production was obtained by combination of livestock manure slurry and mandarin pomace treatment, compared to the swine manure slurry alone treatment. The co-digestion of livestock manure and fruits pomace has advantages to enhance the production of methane gas, compared to digestion of swine manure slurry alone.

In this work, preparation and characterizations of hybrid membranes containing sulfonated poly(arylene ether sulfone) (SPES) and sulfonated poly(vinylidene fluoride-co-hexafluoropropylene) (SPVdF-co-HFP) (20, 30 or 40 wt%) were carried out. The structure of hybrid membranes was confirmed using X-ray diffraction (XRD) analysis and the Fourier transform infrared (FT-IR) spectroscopy. The prepared SPAES/SPVdF-30 membrane exhibits higher ionic conductivity of 68.9 mS/cm at $90^{\circ}C$ and 100% RH. Besides, the other studies showed that the hybrid membrane has good oxidation stability, thermal stability, and mechanical stability. Thus, we believe that the prepared hybrid membrane is suitable for the development of membranes for fuel cell applications.
A series of laboratory experiment was conducted to find out the effects of physico-chemical, microbiological and commercial inhibitors on the odorous gas evolution in the fermentation of livestock feces. The results obtained were summarized as follows. 1. The rate and amount of evolution of gas were the highest at 7 days after incubation, thereafter gradually decreased until 24 days after incubation. 2. The rate and amount of gases were evolved in order of $CO_2>N_2O>CH_4>NH_3>N_2S$, respectively. 3. The highest amount of methane gas was evolved from the poultry feces, those of carbon dioxide and nitrous oxide were evolved from the pig feces, and that of hydrogen sulfide was dominantly evolved from the cattle feces. 4. Negative correlation were obtained between the total amounts of $NH_3$ and $CH_4$, $CO_2$ and $CH_4$, $N_2O$ and $CH_4$, $N_2O$ and $CH_4$, while positive correlations were obtained between the amounts of $CO_2$ and $N_2O$, $CO_2$ and $NH_3$, and $NH_3$ and $N_2O$, respectively. 5. There was no significantly inhibiting effect obtained that the application of commercial gas inhibitor as VK 88. On the other hand there was significantly inhibiting effect obtained when application of fertile paddy soil and photosynthetic bacteria to the fermentation of livestock feces.

An in situ experiment was conducted to find out whether Tween 80 improves rice straw digestion through increased adhesion of major fibrolytic bacteria. Rice straw was sprayed with various levels of Tween 80 non-ionic surfactant or SDS ionic surfactant 24 h before incubation in the rumen of Holstein steers. Dry matter (DM) disappearance and adhesion of F. succinogenes, R. flavefaciens and R. albus on rice straw after in situ incubation were measured by real-time PCR. Application of Tween 80 increased DM disappearance, which was more noticeable at an application level of 1% compared to lower application levels. Application of SDS resulted in an opposite response in DM disappearance with highest reduction in DM disappearance at 1% level. In a subsequent in situ experiment, higher Tween 80 was applied to rice straw in an attempt to find the optimum application level. Tween 80 at 2.5% gave better DM disappearance than 1% with a similar result at 5%. Therefore, an adhesion study was carried out using rice straw treated with 2.5% Tween 80. Our results indicated that Tween 80 reduced adhesion of all three major rumen fibrolytic bacteria to rice straw. Present data clearly show that improved DM disappearance by Tween 80 is not due to increased bacterial adhesion onto substrates.
To investigate the production and characteristics of thermostable levan sucrase from Rahnella aquatilis ATCC 33071, the levan sucrase gene from R. aquatilis was cloned and expressed in Escherichia coli without induction system. Expression of levansucrase gene in E. coli had no notable or detrimental effect on the growth of host strain, and the recombinant levan sucrase exhibited levan synthesis activity. Levansucrase was secreted to the periplasm in E. coli, and addition of $0.5\%$ glycine yielded further secretion of levansucrase to the growth medium and resulted in an increase of total levansucrase activity. Furthermore, the cellular levansucrase was evaluated for the production of levan by using toluenepermeabilized whole-cells. The levansucrase was thermostable at $37^{\circ}C$. The molecular size oflevan was $1{\times}\;10^{6}$ Da, as determined by HPLC, and the degree of polymerization of levan varied with incubation temperatures: Low incubation temperature was preferable for the production of high-molecular size levan. The present study demonstrated that the mass production of levan and levan oligosaccharides can be achieved by glycine supplementation to the growth medium or by toluenepermeabilized whole-cells.

BACKGROUND: Coal ashes generated from thermal power plants have been known as beneficial materials for agricultural use because of their nutrient elements. However, there is limitation to recycle them due to their alkalinity. The objective of this study was to evaluate the effectiveness or safety of the coal ashes for their heavy metals on agricultural recycling when adjusted to pH of 5 with sulfuric acid. METHODS AND RESULTS: Concentration of hydrogen which is needed to adjust pH of coal ash was estimated by using a buffering curve and then the amount of sulfuric acid was changed by the estimation before incubation. Each of fly ash (FA) and bottom ash (BA) was collected from both thermal plants of Yeongdong (YD) and Yeongheung (YH). The pH values of coal ashes increased to 4.76 (from 4.34) after incubation with sulfuric acid for 56 days, closer to the targeted pH. Coal ashes also increased the contents of available phosphorus by 2-fold (165 mg/kg) and 11-fold (1,137 mg/kg) for YDBA and YDFA, respectively, compared to the control. CONCLUSION: The utilization of coal ash with its acidity regulation would be very beneficial to agriculture sector and further suggest promising environmental safety against heavy metals.

This study was conducted to investigate effect of corn flake and corn ground on nutrient digestibility and fermentation characteristics of rumen in Hanwoo. The animals used were three Hanwoo cows implanted with ruminal fistula. Corn were categorized in 2 groups based on the corn processing method: Ground and Flake. The rumen digestibility of dry matter, starch, nitrogen free extract and non fiber carbohydrates were increased in flake compared to ground from 3 to 24 hours of incubation(P<0.05). The pH of rumen was lower in the flaked treatment than ground treatment at 3 hours after incubation, but average pH was no significantly difference between treatments. The average acetic acid, propionic acid and butyric acid were significantly increased in the flaked treatments compared to the ground treatment (P<0.05). Thus, flake processing can improve the carbohydrate availability of corn in the rumen and increase feed value.
