Effect of Decomposition on Nitrogen Dynamics in Soil Applied with Compost and Rye

  • Soil organic matter (SOM) plays an important role in the continuous production and environmental conservation in arable soils. In particular, the decomposition of organic matter in soil might promote soil organic matter and fertility due to the mineralization of N. In this study, to evaluate the effect of organic matter amendment on the C mineralization and N dynamic, $CO_2-C$ flux, extractable N and $N_2O$ emission were determined using closed chamber for 4 weeks at 10, 15, $20^{\circ}C$ of incubation temperature after the mixture of $2Mgha^{-1}$ rice straw compost and rye in sandy loam and clay loam. Regardless of soil texture, decomposition rates of rice straw compost and rye at $10{\sim}20^{\circ}C$ of incubation temperature ranged from 0.9 to 3.8% and 8.8 to 20.3%, respectively. Rye application in soil increased $NH_4-N$ and $NO_3-N$ content as well as the $N_2O$ emission compared to the rice straw compost. After incubation for 4 weeks, total C content in two soils was higher in rice straw compost than in rye application. In conclusion, application of rice straw compost and rye to soil was able to improve the soil organic matter and fertility. However, organic matter including the recalcitrant compounds like rice straw compost would be effective on the management of soil organic matter and the reduction of greenhouse gases in soil.

Effects of baicalein on hydrogen peroxide productions in RAW 264.7 mouse macrophages stimulated by poly-IC and lipoteichoic acid (바이칼레인(baicalein)이 poly-IC와 lipoteichoic acid로 자극된 마우스 대식세포 RAW 264.7의 hydrogen peroxide 생성에 미치는 영향)

  • Objectives : The aim of this study was to investigate the effect of baicalein (BA) on the production of hydrogen peroxide and nitric oxide (NO) in RAW 264.7 mouse macrophages stimulated with polyinosinic-polycytidylic acid (poly-IC) and lipoteichoic acid. Methods : RAW 264.7 co-stimulated with poly-IC and lipoteichoic acid were incubated with baicalein at concentrations of 25 and 50 μM. Incubation time is 16 h, 18 h, 20 h, 22 h, and 24 h. After incubation, The production of hydrogen peroxide in RAW 264.7 was measured with dihydrorhodamine 123 assay. Chrysin was used as a comparative material. NO production was evaluated by griess assay. Results : For 16 h, 18 h, 20 h, 22 h, and 24 h incubation, BA at the concentration of 25 and 50 μM significantly inhibited the production of hydrogen peroxide in RAW 264.7 stimulated by poly-IC and lipoteichoic acid (p <0.001). In details, production of hydrogen peroxide in 'poly-IC and lipoteichoic acid'-stimulated RAW 264.7 treated for 16 h with BA at concentrations of 25 and 50 μM was 82.36% and 77.24% of the control group treated with poly-IC and lipoteichoic acid only, respectively; the production of hydrogen peroxide for 18 h was 83.15% and 77.91%, respectively;production of hydrogen peroxide for 20 h was 82.88% and 77.82%, respectively; production of hydrogen peroxide for 22 h was 83.27% and 78.17%, respectively; production of hydrogen peroxide for 24 h was 83.54% and 78.35%, respectively. Additionally, BA at the concentration of 50 and 100 μM significantly inhibited NO production in lipoteichoic acid-induced RAW 264.7 (p <0.001). Conclusions : BA might have anti-oxidative activity related to its inhibition of hydrogen peroxide production in 'poly-IC and lipoteichoic acid'-stimulated RAW 264.7 macrophages.

  • Objectives : Effects of baicalein (BA) on oxidative stress in RAW 264.7 mouse macrophages stimulated with peptidoglycan (PG) and lipopolysaccharide (LPS) were investigated. Methods : RAW 264.7 co-stimulated with LPS and PG were incubated with BA at concentrations of 25 and 50 µM. Incubation time was 18 h, 20 h, 22 h, 24 h, and 26 h. After incubation, the production of hydrogen peroxide in RAW 264.7 was measured with dihydrorhodamine 123 assay. Additionally, RAW 264.7 stimulated with PG were incubated with BA at concentrations of 25 and 50 µM for 24 h. After incubation, NO production was evaluated by griess reagent assay. Results : BA significantly inhibited hydrogen peroxide productions (p <0.05). In details, production of hydrogen peroxide in 'LPS and PG'-stimulated RAW 264.7 treated for 18 h with BA at concentrations of 25 and 50 µM was 91.27% and 89.22% of the control group treated with LPS and PG only, respectively; the production of hydrogen peroxide for 20 h was 92.19% and 90.58%, respectively; production of hydrogen peroxide for 22 h was 91.69% and 89.89%, respectively; production of hydrogen peroxide for 24 h was 92.4% and 90.19%, respectively; production of hydrogen peroxide for 26 h was 91.7% and 89.04%, respectively. Additionally, BA at the concentration of 50 and 100 µM significantly inhibited NO production in PG-induced RAW 264.7 (p <0.05). Conclusions : BA might have anti-oxidative activity related to its inhibition of hydrogen peroxide production in 'LPS and PG'-stimulated RAW 264.7 macrophages.

Enrichment of $CO_2$-Fixing Bacteria in Cylinder-Type Electrochemical Bioreactor with Built-In Anode Compartment

  • Bacterial assimilation of $CO_2$ into stable biomolecules using electrochemical reducing power may be an effective method to reduce atmospheric $CO_2$ without fossil fuel combustion. For the enrichment of the $CO_2$-fixing bacteria using electrochemical reducing power as an energy source, a cylinder-type electrochemical bioreactor with a built-in anode compartment was developed. A graphite felt cathode modified with neutral red (NR-graphite cathode) was used as a solid electron mediator to induce bacterial cells to fix $CO_2$ using electrochemical reducing power. Bacterial $CO_2$ consumption was calculated based on the variation in the ratio of $CO_2$ to $N_2$ in the gas reservoir. $CO_2$ consumed by the bacteria grown in the electrochemical bioreactor (2,000 ml) reached a maximum of approximately 1,500 ml per week. Time-coursed variations in the bacterial community grown with the electrochemical reducing power and $CO_2$ in the mineral-based medium were analyzed via temperature gradient gel electrophoresis (TGGE) of the 16S rDNA variable region. Some of the bacterial community constituents noted at the initial time disappeared completely, but some of them observed as DNA signs at the initial time were clearly enriched in the electrochemical bioreactor during 24 weeks of incubation. Finally, Alcaligenes sp. and Achromobacter sp., which are capable of autotrophically fixing $CO_2$, were enriched to major constituents of the bacterial community in the electrochemical bioreactor.

Effect of a co-culture of scenedesmus dimorphus and nitrifiers on advanced wastewater treatment capacity (Scenedesmus dimorphus와 질산화 박테리아의 공배양이 하수고도처리능에 미치는 영향)

  • This study investigated the effect of a co-culture of Scenedesmus dimorphus and nitrifiers using artificial wastewater on the removal of ammonium, nitrate and phosphate in the advanced treatment. To test the synergistic effect of the co-culture, we compared the co-culture treatment with the cultures using S. dimorphus-only and nitrifiers-only treatment as controls. After 6 days of incubation, nitrate was removed only in the co-culture treatment and total amount of N removal was 1.3 times and 1.6 times higher in the co-culture treatment compared to those in the S. dimorphus- and nitrifiers-only treatments, respectively. In case of total amount of P, co-culture treatment removed 1.2 times and 12 times more P than the S. dimorphus -and nitrifiers-only conditions, respectively. This indicates that the co-culture improved removal rates for ammonium, nitrate, and phosphate. This further implies that there was no need for denitrification of nitrate and luxury uptake of P processes because nitrate and phosphate can be removed from the uptake by S. dimorphus. In addition, co-culture condition maintained high DO above 7 mg/L without artificial aeration, which is enough for nitrification, implying that co-culture has a potential to decrease or remove aeration cost in the wastewater treatment plants.

In Vitro Evaluation of Antimicrobial Activity of Lactic Acid Bacteria against Clostridium difficile

  • Clostridium difficile infection (CDI) has become a significant threat to public health. Although broad-spectrum antibiotic therapy is the primary treatment option for CDI, its use has evident limitations. Probiotics have been proved to be effective in the treatment of CDI and are a promising therapeutic option for CDI. In this study, 4 strains of lactic acid bacteria (LAB), namely, Lactobacillus rhamnosus (LR5), Lactococcuslactis (SL3), Bifidobacterium breve (BR3), and Bifidobacterium lactis (BL3) were evaluated for their anti-C. difficile activity. Co-culture incubation of C. difficile ($10^6$ and $10^{10}$ CFU/ml) with each strain of LAB indicated that SL3 possessed the highest antimicrobial activity over a 24-hr period. The cell-free supernatants of the 4 LAB strains exhibited $MIC_{50}$ values between 0.424 mg/ml (SL3) and 1.318 (BR3) mg/ml. These results may provide a basis for alternative therapies for the treatment of C. difficile-associated gut disorders.

Non-invasive Transcutaneous pCO2 Gas Monitoring System for Arterial Blood Gas Analysis

  • Monitoring the carbon dioxide concentration in arterial blood is vital for the evaluation and prevention of pulmonary disease. Yet, domestic pure arterial blood carbon dioxide sensor technologies are not being developed, instead all sensors are imported. In this paper, we develop a real time monitoring system for arterial blood partial pressure of carbon dioxide($pCO_2$) gas from the wrist by using a carbon micro-heater. The micro-heater was fabricated with a thickness of 0.3 ${\mu}m$ in order to collect the carbon dioxide under the skin. The micro-heater has been designed to perform temperature compensation in order to prevent damage to the skin. Two clinical trials of the system were undertaken. As a result, we demonstrated that a portable, transcutaneous carbon dioxide analysis($TcpCO_2$) device produced domestically is possible. In addition, this system reduced the analysis time significantly. Carbon films could reduce the unit price of these sensors by replacing the gold film used in foreign models. Also, we developed a real time monitoring system which can be used with optical biosensors for medical diagnostics as well as gas sensors for environmental monitoring.

  • A rapid assay for interferon based on reduction of cytopathic effect was developed with HEp-2 and Vesicular Stomatitis Virus. The number of manipulations and the lengths of the various incubation steps were reduced to minimum. The assay is simple to perform and can be completed with 22-24 hr. Moreover, it was precise method than CPE-reading method.

  • Cephalosporin-C deacetylase (CAH) catalyzes the deacetylation of cephalosporin derivatives. A novel gene encoding the CAH from Bacillus sp. KCCM10143 was cloned and sepuenced. The uncleotide sequence contained an open reading frame encoding a polypeptide consisting of 217 amino acids and a molecular weight of 24 kDa which was in good agreement with the value obtained by sodium dodecylsulfate-polyacrylamide gel electrophoresis. An expression plasmid was constructed by inserting the CAH gene into the region of the pTrc99A expression vector. An active from of the CAH protein was expressed in the soluble fraction obtained after cell disruption. in fermentation using a 5-1 jar fementer, the transformant E. coli JM109 (pDST654) produced 4.12 U of CAH per ml of culture during 16 h of incubation.

Effect of Heme Oxygenase Induction by NO Donor on the Aortic Contractility

  • Carbon monoxide (CO) binds to soluble guanylate cyclase to lead its activation and elicits smooth muscle relaxation. The vascular tissues have a high capacity to produce CO, since heme oxygenase-2 (HO-2) is constitutively expressed in endothelial and smooth muscle cells, and HO-1 can be greatly up-regulated by oxidative stress. Moreover, the substrate of HO, heme, is readily available for catalysis in vascular tissue. Although the activation of heme oxygenase pathway under various stress conditions may provide a defence mechanism in compromised tissues, the specific role of HO-1-derived CO in the control of aortic contractility still remains to be elucidated. The present study was done to determine the effect of HO-1 induction on the aortic contractility. Thus, the effects of incubation of aortic tissue with S-nitroso-N-acetylpenicillamine (SNAP) for 1 hr on the aortic contractile response to phenylephrine were studied. The preincubation with SNAP resulted in depression of the vasoconstrictor response to phenylephrine. This effect was restored by HO inhibitor or methylene blue but not by NOS inhibitor. The attenuation of vascular reactivity by preincubation with SNAP was also revealed in endothelium-free rings. $AlF4^--evoked$ contraction in control did not differ from that in SNP-treated group. These results suggest that increased production of CO was responsible for the reduction of the contractile response to phenylephrine in aortic ring preincubated with SNAP and this effect of SNAP was independent on endothelium.
