Asterias pectinifera-Derived Collagen Peptides Mixed with Halocynthia roretzi Extracts Exhibit Anti-Photoaging Activities during Exposure to UV Irradiation, and Antibacterial Properties

  • Asterias pectinifera, a species of starfish and cause of concern in the aquaculture industry, was recently identified as a source of non-toxic and highly water-soluble collagen peptides. In this study, we investigated the antioxidant and anti-photoaging functions of compounds formulated using collagen peptides from extracts of Asterias pectinifera and Halocynthia roretzi (AH). Our results showed that AH compounds have various skin protective functions, including antioxidant effects, determined by measuring the scavenging activity of 2,2-diphenyl-1-picrylhydrazyl radicals, as well as anti-melanogenic effects, determined by measuring tyrosinase inhibition activity. To determine whether ethosome-encapsulated AH compounds (E(AH)) exert ultraviolet (UV)-protective effects, human dermal fibroblasts or keratinocytes were incubated with E(AH) before and after exposure to UVA or UVB. E(AH) treatment led to inhibition of photoaging-induced secretion of matrix metalloproteinase-1 and interleukin-6 and -8, which are associated with inflammatory responses during UV irradiation. Finally, the antibacterial effects of AH and E(AH) were confirmed against both gram-negative and gram-positive bacteria. Our results indicate that E(AH) has the potential for use in the development of cosmetics with a range of skin protective functions.

Translocation of $^14$C-assimilates During Grain Filling and Influence of Defoliation and Emasculation on Grain Weight in Oats (연맥 등숙기 동안 $^14$C물질의 전유와 절엽 및 제영이 종실중에 미치는 영향)

  • $^14$C-sucrose was labeled on detached panicles and $^14{CO}_2$ on flag leaves or panicles of intact plants to study grain sink activity in spring oats cultivar Pennfield. Defoliation and emasculation experiment was conducted to study source-sink relationship during grain filling. Specific activity of groat rose up to 15 days after anthesis and declined rapidly to 18 days. Daily gain of groat wt. matched closely with specific activity of. groat during grain filling. Primary groats were higher in specific activity of groat than secondary groats.$^14{CO}_2$ exposure on panicle was three times higher in specific activity of groat than $^14{CO}_2$exposure on flag leaf. In the defoliation and emasculation experiment, groat wt. of Pennfield oats decreased as ratio of source/sink decreased. Half number of spikelets with half leaf area was no different in groat wt. compared to control, but normal number of spikelets with zero leaf area was decreased 16% in groat wt., indicating a significant compensation by green area on panicle.

The comparison of lesion localization methods in breast lymphoscintigraphy (Breast lymphoscintigraphy 검사 시 체표윤곽을 나타내는 방법의 비교)

  • Purpose Breast lymphoscintigraphy is an important technique to present for body surface precisely, which shows a lymph node metastasis of malignant tumors at an early stage and is performed before and after surgery in patients with breast cancer. In this study, we evaluated several methods of body outline imaging to present exact location of lesions, as well as compared respective exposure doses. Materials and Methods RANDO phantom and SYMBIA T-16 were used for obtaining imaging. A lesion and an injection site were created by inserting a point source of 0.11 MBq on the axillary sentinel lymph node and 37 MBq on the right breast, respectively. The first method for acquiring the image was used by drawing the body surface of phantom for 30 sec using $Na^{99m}TcO_4$ as a point source. The second, the image was acquired with $^{57}Co$ flood source for 30 seconds on the rear side and the left side of the phantom, the image as the third method was obtained using a syringe filled with 37 MBq of $Na^{99m}TcO_4$ in 10 ml of saline, and as the fourth, we used a photon energy and scatter energy of $^{99m}Tc$ emitting from phantom without any addition radiation exposure. Finally, the image was fused the scout image and the basal image of SPECT/CT using MATLAB$^{(R)}$ program. Anterior and lateral images were acquired for 3 min, and radiation exposure was measured by the personal exposure dosimeter. We conducted preference of 10 images from nuclear medicine doctors by the survey. Results TBR values of anterior and right image in the first to fifth method were 334.9 and 117.2 ($1^{st}$), 266.1 and 124.4 ($2^{nd}$), 117.4 and 99.6 ($3^{rd}$), 3.2 and 7.6 ($4^{th}$), and 565.6 and 141.8 ($5^{th}$). And also exposure doses of these method were 2, 2, 2, 0, and $30{\mu}Sv$, respectively. Among five methods, the fifth method showed the highest TBR value as well as exposure dose, where as the fourth method showed the lowest TBR value and exposure dose. As a result, the last method ($5^{th}$) is the best method and the fourth method is the worst method in this study. Conclusion Scout method of SPECT/CT can be useful that provides the best values of TBR and the best score of survey result. Even though personal exposure dose when patients take scout of SPECT/CT was higher than another scan, it was slight level comparison to 1 mSv as the dose limit to non-radiation workers. If the scout is possible to less than 80 kV, exposure dose can be reduced, and also useful lesion localization provided.

  • We synthesized N-Hydroxysuccinimidyl (NHS) azidobenzoate as a cross-linking reagent for immobilization of peptide onto the solid surface. Thin polystyrene(PS) films spin-coated with a NHS azidobenzoate solution were exposed with ultraviolet light at 245nm$(3.3mW/cm^2)$ for 5 min. The NHS active ester groups became covalently attached to the polymer via photogenerated, highly reactive nitrene intermediates derived from NHS azidobenzoate. Using this technique, it is demonstrated that well-defined surface regions can be functionalized with a minimum observable feature size of 1mm for UV exposure. Through reaction of this functionalized PS surface with primary amine-containing biomolecules, biological molecule had been immobilized on the polymer surface.

  • It has been observed that local increase in melanin synthesis or uneven distribution can cause local hyperpigmintation or spot. Pigmentary disorders are caused by various factors, including inflammation, imbalance of hormones, and genetic disorder. Recently the harmfulness of Ultraviolet radiation is increasing due to destruction of ozone layer. Excessive exposure to UV radiation caused post-inflammatory pigmentation. Most women want to avoid uneven skin pigmentation. To satisfy this desire many cosmetic companies have been developing melanogenesis inhibitors and finding promising active agents for use in cosmetic preparations for skin whitening. In cosmetic preparations, many inhibitors such as kojic acid, arbutin, ascorbic acid, and licorice extracts6 have been used as whitening purpose. Plant extracts having an inhibitory effect on melanin formation may be a good choice for cosmetic purpose because of their relatively lower side effects. Therefore, we screened 285 plant extracts for their inhibitory activity in tyrosinase. Of the plant extracts, ramulus mori extracts showed potent tyrosinase inhibition activity. We also identified the active compound in the extract.

  • Free radicals and reactive oxygen species (ROS) caused by UV exposure or other environmental facts play critical roles in cellular damage and aging. The extract of tinged autumnal leaves of maple tree(Acer palmatum) has proven to be a powerful antioxidant. The Acer palmatum extract is very effective on the stabilization of biological membranes( containing unsaturated fatty acid). We studied photoprotective effect of the extract against UVB-induced cytotoxicity in human keratinocytes. The extract improved cell viability comparing to control after UVB irradiation. In the determination test of pro inflammatory cytokines the extract decreased expression of interleukin 1 a and 6, which play an important role in inflammation and skin erythema caused by UV. We also studied property of varying cosmetic formulations on the percutaneous absorption of the extract. After 24 hour in vitro penetration study, the content of the extract was more highly detected in skin residue part. This result showed the extract had relatively high compatibility of skin in our emulsion system. On human skin, after appling the product containing the extract we obtained a good result of antiwrinkle effect by skin visiometer. In conclusion, the Acer palmatum extract is a photoprotective and very effective in stressed and aged skin care. And we can predict the extract mainly affects on the skin cell and tissue in our emulsion system.

Development of Biosphere Assessment Modeling Strategy for Deep Geological Disposal in Generic Site of the Korean Peninsula

  • As part of the safety case development for generic disposal sites in Korea, it is necessary to develop generic assessment models using various geosphere-biosphere interfaces (GBIs) and potentially exposed groups (PEGs) that reflect the natural environmental characteristics and the lifestyles of people in Korea. In this study, a unique modeling strategy was developed to systematically construct and select Korean generic biosphere assessment models. The strategy includes three process steps (combination, screening, and experts' scoring) for the biosphere system conditions. First, various conditions, such as climate, topography, GBIs, and PEGs, were combined in the biosphere system. Second, the combined calculation cases were configured into interrelation matrices to screen out some calculation cases that were highly unlikely or less significant in terms of the exposure dose. Finally, the selected calculation cases were prioritized based on expert judgment by scoring the knowledge, probability, and importance. The results of this study can be implemented in the development of biosphere assessment models for Korean generic sites. It is believed that this systematic methodology for selecting the candidate calculation cases can contribute to increasing the confidence of future site-specific biosphere assessment models.

Effect of High Pressure Carbon Dioxide on Inactivation of Leuconostoc sp. (고압 이산화탄소에 의한 Leuconostoc sp.의 살균 효과)

  • Inactivation of Leuconostoc sp. isolated from kimchi using carbon dioxide under pressure was investigated in terms of operating parameters in order to evaluate its feasibility as a novel nonthermal process. Inactivation rates increased with increasing pressure, temperature and exposure time, but with decreasing working volume. Microbial reduction of 3 log cycles was achieved within 150 min under a $CO_2$ pressure of $60\;kg/cm^2$ at 30^{\circ}C$. It was confirmed that microbial inactivation by the high pressure $CO_2$ was governed essentially by the characteristic of $CO_2$ mass transfer and thus penetration of $CO_2$, into cells was a rate limiting step to determine efficiency of the inactivation process. The experimental results suggested that the high pressure $CO_2$ treatment could be used as one of the effective nonthermal methods for preserving foods.

Toxicity Evaluation of Effluent Gases from a Residental Fire by Rats (연립 주택 화재시 유독가스 방출 특성과 Rats를 이용한 독성평가)

  • It has long been recognized that exposure to fire-induced toxic gases is a fatal hazard confront-ing people in fires. In this study, an indoor fire experiment was conducted in an unoccupied residential building located in An-san city, Kyoung-gi province, and the composition of effluent gases, which include CO, $CO_2$, $O_2$, $SO_2$, NO and $NO_2$, were measured by a gas analyzer. A group of lab rats were exposed to the toxic gases released from fire, and the blood samples of the rats were gathered every 2 minutes. A toxicity evaluation was conducted by analyzing the concentrations of Glucose, AST(GOT), ALT(GPT), CBC Count and CO(carboxy)-Hb in the blood samples. Shown from the analysis is the does-response relationship between the CO concentration that rats were exposed to and the CO-Hb concentration in rat blood.
