Characterization of Denitrifying and Dissimilatory Nitrate Reduction to Ammonium Bacteria Isolated from Mud Crab Culture Environment

  • Microbial community plays important roles in the culture environment of mud crab Scylla serrata. One of the environmental management efforts for the cultivation of S.serrata is by stabilizing microorganisms involved in nitrogen cycle process. The availability of dissolved inorganic nitrogen in its culture environment under a recirculating system closely relates to the nitrogen cycle, which involves both anaerobic and aerobic bacterial activities. Anaerobically, there are two major nitrogen compound degradation processes, i.e., denitrification and dissimilatory nitrate reduction to ammonium (DNRA). This study aimed to identify denitrifying and DNRA bacteria isolated from the recirculating cultivation of S. serrata. The water samples were collected from anaerobic filters called close filter system, which is anaerobically conditioned with the addition of varying physical filter materials in the recirculating mud crab cultures. The results showed that three denitrifying bacterial isolates and seven DNRA bacterial isolates were successfully identified. The phylogenetic analysis based on 16S rRNA gene of the denitrifying bacteria revealed that HIB_7a had the closest similarity to Stenotrophomonas daejeonensis strain MJ03. Meanwhile, DNRA bacterial isolate of HIB_92 showed a 100% similarity to Bacillus sonorensis strain N3, Bacillus vallismortis strain VITS-17, Bacillus tequlensis strain TY5, Geobacillus sp. strain DB24, Bacillus subtilis strain A1, and Bacillus mojavensis strain SSRAI21. This study provides basic information denitrifying and DNRA bacterial isolates identity which might have the potential to be applied as probiotics in aquaculture systems in order to maintain optimal environmental conditions.

  • To facilitate low-pressure selective catalyst reduction (L.P SCR), a high exhaust-gas temperature of a four-stroke diesel engine for a ship's generator is required. This study aimed at reducing the exhaust-gas temperature by adjusting the valve open-close timing and fuel injection timing to satisfy the operating conditions of L.P SCR and prevent accidents associated with the generator engine due to high temperature. To lower exhaust-gas temperature, the angle of the camshaft was adjusted and the shim of the fuel injection pump was added. As a result, the maximum explosion pressure increased and the average of the turbocharger outlet temperature dropped. Considering the heat loss from the turbocharger outlet to the SCR inlet, the operation condition for L.P SCR was satisfied with 290 ℃. The study demonstrates that safe operation of a diesel generator can be achieved by lowering the exhaust-gas temperature.

  • Purpose: By applying an ultrasonic mechanical device to the liquid fertilizer storage in the pig dropping treatment plant, the initial odor of the odor source is reduced, and the air dilution drainage of the complex odor is fundamentally recognized to facilitate odor treatment on the mechanical and chemical biological treatment devices at the rear. Research design, data and methodology: The odor concentration on the site boundary was measured to confirm the state of reduction. In order to prevent the spread of odor from the collection of the pig dropping treatment plant, it was measured by installing an ultrasonic generator inside the installation wall after installing the sealing wall. Results: The average value of the March and April measurement data remained close to neutral at 8.2 after 8.6 treatment before pH treatment, decreased 97.3% from 462 mg/L before SS treatment to 10.5 mg/L after treatment, and the composite odor was reduced by 85% from 20 to 3 before treatment. It was confirmed that ammonia (NH3) was reduced by 99% from 5.8 ppm to 0.09 ppm, and general bacteria were also reduced by 99% from 3,200 CFU/mL to 57 CFU/mL Conclusion: Applying the ultrasonic air ejector hybrid system and zigzag air complex module development product to resource circulation centers or sewage treatment facilities is thought to reduce inconvenience to residents due to odors caused.

Coicis Semen Reduces Staphylococcus aureus Persister Cell Formation by Increasing Membrane Permeability

  • Unlike resistant cells, persister cells resist antibiotics due to a decreased cellular metabolic rate and can transition back to normal susceptible cells when the antibiotic is removed. These persister cells contribute to the chronic symptoms of infectious diseases and promote the emergence of resistant strains with continuous antibiotic exposure. Therefore, eliminating persister cells represents a promising approach to significantly enhance antibiotic efficacy. Here, we found that Coicis Semen extract reduced Staphylococcus aureus persister cells at a concentration of 0.5 g/L. Linoleic acid and oleic acid, the major components of Coicis Semen extract, exhibited a comparable reduction in persister cells when combined with three antibiotics: ciprofloxacin, oxacillin, and tobramycin. Conversely, these effects were nullified in the presence of the surfactant Tween 80 (1%), suggesting that the hydrophobic characteristics of linoleic acid and oleic acids play a pivotal role in reducing the number of S. aureus persister cells. Considering the concentration-dependent effects of linoleic acid and oleic acid, the persister-reducing activity of Coicis Semen extract was primarily attributed to these fatty acids. Moreover, Coicis Semen extract, linoleic acid, and oleic acid increased the cell membrane permeability of S. aureus. Interestingly, this effect was counteracted by 1% Tween 80, indicating a close association between the reduction of persister cells and the increase in cell membrane permeability. The identified compounds could thus be used to eliminate persister cells, thereby enhancing therapeutic efficacy and shortening treatment duration. When used in conjunction with antibiotics, they may also mitigate chronic symptoms and significantly reduce the emergence of antibiotic-resistant bacteria.

Predictive Factors and Prognostic Relevance of Sunitinib-induced Subclinical and Overt Hypothyroidism in Korean Patients with Metastatic Renal Cell Carcinoma

  • Background: Sunitinib, commonly used for metastatic renal cell carcinoma (mRCC), often induces hypothyroidism, affecting 27 to 85% of patients. There are clues suggesting an association between sunitinib-induced hypothyroidism and improved survival outcomes. This study aims to identify the predictive factors of sunitinib-induced hypothyroidism and evaluate whether the occurrence of overt or subclinical hypothyroidism predicts tumor outcome in patients with mRCC. Methods: Patients administered to sunitinib for mRCC was included in this retrospective study. Log-rank test and Cox proportional hazards model were conducted to identify predictive factors of hypothyroidism and prognostic factors of progression-free survival (PFS) and overall survival (OS). Results: A total of 156 patients with mRCC treated with sunitinib were included. Predictive factors of sunitinib-induced hypothyroidism were female (odds ratio (OR), 2.77), sunitinib-induced hypertension (OR, 2.99) and dose reduction of sunitinib due to intolerance (OR, 3.57). Sunitinib-induced overt hypothyroidism was a significant prognostic factor in predicting PFS and OS (hazard ratio, 0.38 and 0.23, respectively). Thyroid hormone replacement did not have an influence on PFS and OS. Conclusions: Female patients, patients who experienced sunitinib-induced hypertension and sunitinib dose reduction are at higher risk of hypothyroidism and need close monitoring. Overt hypothyroidism is a strong prognostic factor of sunitinib treatment outcome in mRCC patients and thyroid hormone replacement does not have a negative effect on tumor outcome.

Kinetics and mechanism of chromate reduction by biotite and pyrite (흑운모 및 황철석에 의한 6가 크롬의 환원 반응속도와 반응기작)

  • The removal of chromate from aqueous solution using finely ground pyrite and biotite was investigated by batch experiments and the kinetics and the mechanism of chromate reduction were discussed. The chromate reduction by pyrite was about hundred times faster than that by biotite and was also faster at pH 3 than at pH 4. When pyrite was used, more than 90% of initial chromate was reduced within four hours at pH 4 and within 40 min. at pH 3. However, more than 400 hours was taken for the reduction of 90% of initial chromate by biotite. The results indicate that the rate of chromate reduction was strongly depending on the amount of Fe(II) in the minerals and on the dissolution rate of Fe(II) from the minerals. The reduction of chromate at pH 4 resulted in the precipitation of (Cr, Fe)(OH))$_3$$_{ (s)}$, which is believed to have limited the concentrations of dissolved Cr(III) and Fe(III) to less than expected values. When biotite was used, amounts of decreased Fe(II) and reduced Cr(Ⅵ) did not show stoichiometric relationship, which implying there was not only chromate reduction by ferrous ions in the acidic solution but also heterogeneous reduction of ferric ions by the structural ferrous iron in biotite. However, the results from a series of the experiments using Pyrite showed that concentrations of the decreased Fe(II) and the reduced Cr(Ⅵ) were close to the stoichiometric ratio of 3:1. It was because the oxidation of pyrite rapidly created ferrous ions even in oxygenated solutions and the chromate reduction by the ferrous ions was significantly faster than ferrous ion oxygenation.

  • The physiological characteristics of cultures of very high cell mass (e.g. 10g cell mass/L), termed“ultrahigh cell density cultures”is reviewed. A close relationship was found between the length of the optical path (OP) in flat-plate reactors and the optimal cell density of the culture as well as its areal (g m$\^$-2/ day$\^$-1/) productivity. Cell-growth inhibition (GI) unfolds as culture density surpasses a certain threshold. If it is constantly relieved, a 1.0cm OP reactor could produce ca. 50% more than reactors with longer OP, e.g. 5 or 10cm. This unique effect, discovered by Hu et al. [3], is explained in terms of the relationships between the frequency of the light-dark cycle (L-D cycle), cells undergo in their travel between the light and dark volumes in the reactor, and the turnover time of the photosynthetic center (PC). In long OP reactors (5cm and above) the L-D cycle time may be orders of magnitude longer than the PC turnover time, resulting in a light regime in which the cells are exposed along the L-D cycle, to long, wasteful dark periods. In contrast, in reactors with an OP of ca. 1.0 cm, the L-D cycle frequency approaches the PC turnover time resulting in a significant reduction of the wasteful dark exposure time, thereby inducing a surge in photosynthetic efficiency. Presently, the major difficulty in mass cultivation of ultrahigh-density culture (UHDC) concerns cell growth inhibition in the culture, the exact nature of which is awaiting detailed investigation.

Lifetime Prediction of Geogrids for Reinforcement of Embankments and Slopes through Time-Temperature Superposition

  • The creep resistance of geogrids is one of the most significant long-term safety characteristics used as the reinforcement in slopes and embankments. The failure of geogrids is defined as creep strain greater than 10%. In this study, the accelerated creep tests were applied to polyester geogrids at various loading levels of 30, 50% of the yield strengths and temperatures using newly designed test equipment. Also, the new test equipment permitted the creep testing at or above glass transition temperature($T_g$) of 75, 80, $85^{\circ}C$. The time-dependent creep behaviors were observed at various temperatures and loading levels. And then the creep curves were shifted and superposed in the time axis by applying time-temperature supposition principles. The shifting factors(AFs) were obtained using WLF equation. In predicting the lifetimes of geogrids, the underlying distribution for failure times were determined based on identification of the failure mechanism. The results confirmed that the failure distribution of geogrids followed Weibull distribution with increasing failure rate and the lifetimes of geogrids were close to 100 years which was required service life in the field with 1.75 of reduction factor of safety. Using the newly designed equipment, the creep test of geogrids was found to be highly accelerated. Furthermore, the time-temperature superposition with the newly designed test equipment was shown to be effective in predicting the lifetimes of geogrids with shorter test times and can be applied to the other geosynthetics.

  • The need for total enterprise management that covers from accepting orders, design, cost, procurement, manufacture, and delivery has been recently raised in order for efficient project management of small and medium size plant construction enterprises. This trend is reflected in the introduction of EPM(Enterprise Project Management) solutions by some shipbuilding enterprises and large construction corporations. In step with the trend, small and medium size plant construction enterprises are enabled to provide knowledge base in terms of reducing cost and production period and managing portfolio for accepting orders and attaining contract by inducing customized EPM This study presents a design of customized EPM based on a case study of a company. It exhibits a collaborative/collaboration resource-supply model which can be universally applied to small and medium size job-order production enterprises. The research discusses that under the assumption that information is shared internally between departments on planning, supply, manufacture, and quality control in order to create collaborative environment and close participation is created externally between suppliers and orderers, productivity can be improved in cost reduction through planning efficient collaborative supply model.

A Study on the Efficiency of Horizontal Direction Deck-charge Blasting Method Using Electronic Detonator (전자뇌관을 이용한 수평방향 데크차지 발파공법의 효율성검토 연구)

  • In close to downtown construction, the main problem is complaints caused by blasting vibration and noise. To reduce blasting vibration and noise, deck-charge blasting method using electronic detonator can be more secure because there is no cut-off problem. And in this method it is possible to blast in horizontal direction. In this study, the efficiency of horizontal direction deck-charge blasting method using electronic detonator is compared to that of the existing blasting method. And the possibility of applying the construction site is evaluated. As a result, the reduction of blasting vibration, noise and secondary breaking has been determined, as well as large-scale blasting in the vibration criterion can be regulated by the overall increase in blasting efficiency.
