Development of $^1H-^{31}P$ Animal RF Coil for pH Measurement Using a Clinical MR Scanner (임상용 MR에서 pH 측정을 위한 동물 실험용 $^1H-^{31}P$ RF 코일 개발)

  • Purpose : To establish a pH measurement system for a mouse tumor study using a clinical scanner, to develop the $^1H$ and 31P radio frequency (RF) coil system and to test pH accuracy with phantoms. Materials and Methods: The $^1H$ and the $^{31}P$ surface coils were designed to acquire signals from mouse tumors. Two coils were positioned orthogonally for geometric decoupling. The pH values of various pH phantoms were calculated using the $^1H$ decoupled $^{31}P$ MR spectrum with the Henderson-Hasselbalch equation. The calculated pH value was compared to that of a pH meter. Results: The mutual coil coupling was shown in a standard $S_{12}$. Coil coupling ($S_{12}$) were -73.0 and -62.3 dB respectively. The signal-to-noise ratio (SNR) obtained from the homogeneous phantom $^1H$ image was greater than 300. The high resolution in vivo mice images were acquired using a $^{31}P$-decoupled $^1H$ coil. The pH values calculated from the $^1H$-decoupled $^{31}P$ spectrum correlated well with the values measured by pH meter ($R^2$=0.97). Conclusion: Accurate pH values can be acquired using a $^1H$-decoupled $^{31}P$ RF coil with a clinical scanner. This two-surface coil system could be applied to other nuclear MRS or MRI.

Ulmus Macrocarpa Water Extract Prolongs Splenocyte Life Span (Ulmus Macrocarpa 열수 추출물에 의한 비장세포 수명 연장)

  • Ulmus macrocarpa has been used in Korean medicinal food material to physical disorder or tonic material. The purpose of the present study was to evaluate splenocyte life span expansion effects of Ulmus macrocarpa water extract (UMWE) in general cell culture condition. Splenocytes were handled in the presence of 100 μg/ml UMWE for several different time conditions. Live cells were detected with Hoechst 33,342 dye and cell survival molecules were identified through Western blot. Changes in level of cytokine synthesis were evaluated by ELISA. UMWE showed an effect on increased splenocyte survival. UMWE elevated slightly PI3K phosphorylation and ERK1/2 phosphorylation used at 48 hr and 96 hr. Moreover, Bcl-2 was elevated at 48 hr and 96 hr in UMWE-treated splenocytes. UMWE decreased caspase-3 level at 48 hr and 96 hr. ICAD protein increased at 48 hr culturing time. Hematopoietin IL-2 cytokine was down-regulated, however IL-4 hematopoietin cytokine was up-regulated in UMWE treated cell culture media. Increased IFN-γ levels were verified in the supernatant of UMWE-treated cells in all periods (48 hr and 96 hr). Increased patterns in the production of IL-12 cytokine occurred as compared with control after 48 and 96 hr in UMWE-treated-cell cultures. These results suggested that UMWE can prolong splenocyte life span by controlling various signal factors and cytokines.

  • The purpose of this research was to evaluate the clinical and the instrument of convergence utility of transient elastography (FibroScan(R):electromagnetic wave) in diagnosing and treating liver ailments through a comparison and an analysis between liver function blood test and transient elastography (FibroScan(R)) in patients with chronic hepatitis B virus infection. Of all the patients with chronic hepatitis B virus infection who visited clinic B in Daejeon City between July 1, 2015, and February 28, 2016, 75 who underwent a FibroScan(R) test were selected for this study. Their laboratory and liver function test results were compared for a correlation analysis before constructing an ROC (Receiver Operation Characteristic) curve. Aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels were 0.572 and 0.502, respectively, and showed highest correlation with fibrosis score, with statistical significance (p<0.000). Gamma glutamyltranspeptidase, total bilirubin, and alkaline phosphatase levels also showed relatively significant correlations in this order of sequence, while -fetoprotein and total protein levels did not show any statistically significant values. Albumin (-0.449) and platelet levels (-0.373) showed negative correlations with each other and no correlation with fibrosis score (p < 0.000). As liver fibrosis worsened, the accuracy of the ROC curve increased. At the F4 stage, which is the cirrhotic stage, the largest area under the curve was observed. FibroScan(R) showed significant correlation with the ALT (serum glutamic pyruvic transaminase) and AST (serum glutamic oxaloacetic transaminase) levels in the liver function test, which is a routine test for patients with chronic liver ailments. This implies that fibrosis correlates with liver inflammation severity.

Ex Vivo ${1}^H$ MR Spectroscopy: Normal gastric and cancer tissue (정상 위 조직과 위암 조직의 시험관 내 수소자기공명분광)

  • Purpose: In this study, we attempted to ascertain the proton magnetic resonance spectroscopy (${1}^H$ MRS) peak characteristics of human gastric tissue layers and finally to use the metabolic peaks of MRS to distinguish between normal and abnormal gastric specimens. Materials and Methods: Ex-vivo ${1}^H$ MRS examinations of thirty-five gastric specimens were performed to distinguish abnormal gastric tissues invaded by carcinoma cells from normal stomach-wall tissues. High-resolution 400-MHz (9.4-T) ${1}^H$ nuclear magnetic resonance (NMR) spectra of two gastric layers, a proper muscle layer, and a composite mucosasubmucosa layer were compared with those of clinical 64- MHz (1.5-T) MR spectra. Three-dimensional spoiled gradient recalled (SPGR) images were used to determine the size and the position of a voxel for MRS data collection. Results: For normal gastric tissue layers, the metabolite peaks of 400-MHz ${1}^H$ MRS were primarily found to be as follows: lipids at 0.9 ppm and 1.3 ppm; alanine at 1.58 ppm; N-acetyl neuraminic acid (sialic acid) at 2.03 ppm; and glutathione at 2.25 ppm in common. The broad and featureless featureless spectral peaks of the 64-MHz MRS were bunched near 0.9, 1.3, and 2.0, and 2.2 ppm in human specimens without respect to layers. In a specimen (Borrmmann type III) with a tubular adenocarcinoma, the resonance peaks were measured at 1.26, 1.36 and 3.22 ppm. All the peak intensities of the spectrum of the normal gastric tissue were reduced, but for gastric tumor tissue layers, the lactate peak split into 1.26 and 1.39 ppm, and the peak intensity of choline at 3.21 ppm was increased. Conclusion: We found that decreasing lipids, an increasing lactate peak that split into two peaks, 1.26 ppm and 1.36 ppm, and an increasing choline peak at 3.22 ppm were markers of tumor invasion into the gastric tissue layers. This study implies that MR spectroscopy can be a useful diagnostic tool for gastric cancer.

Fortified Antioxidative Potential by Chrysoeriol through the Regulation of the Nrf2/MAPK-mediated HO-1 Signaling Pathway in RAW 264.7 Cells (생쥐 대식세포에서 HO-1 발현 유도를 통한 chrysoeriol의 항산화 효과)

  • Chrysoeriol is a widespread flavone, and it is usually found in alfalfa, which has been used as a traditional medicine to treat dyspepsia, asthma, and urinary system disorders. Recently, analysis has been conducted on the anti-inflammatory activity of chrysoeriol, but information on its antioxidative capacity is limited. In this study, the antioxidative potential of chrysoeriol against oxidative damage and its molecular mechanisms were evaluated by analysis of the cell viability, reactive oxygen species (ROS) formation, and Western blots in the RAW 264.7 cell line. Chrysoeriol significantly scavenged lipopolysaccharide (LPS)-induced intracellular ROS formation in a dose-dependent manner, without any cytotoxicity. Heme oxygenase-1 (HO-1), a phase II enzyme that exerts antioxidative activity, was also potently induced by chrysoeriol treatment, which corresponded to the translocation of nuclear factor-erythroid 2 p45-related factor 2 (Nrf2) into the nucleus. Moreover, mitogen-activated protein kinase (MAPK) and phosphoinositide 3-kinase (PI3K) were analyzed due to their important role in maintaining cellular redox homeostasis against oxidative stress. As a result, chrysoeriol-induced HO-1 upregulation was mediated by extracellular signal - regulated kinase (ERK), c-Jun $NH_2$-terminal kinase (JNK), and p38 phosphorylation. To identify the antioxidative potential exerted by HO-1, tert-butyl hydroperoxide (t-BHP)-induced oxidative damage was applied and mitigated by chrysoeriol treatment, which was confirmed by the HO-1 selective inhibitor and inducer, respectively. Consequently, chrysoeriol strongly strengthened the HO-1-mediated antioxidative potential through the regulation of the Nrf2/MAPK signaling pathways.

Effect of Curcuma longa Hot Water Extract on Activity of Neuronal Cells Related to Oxidative Stress (산화적 스트레스와 관련하여 신경세포의 활성에 미치는 강황 열수추출물의 영향에 대한 연구)

  • The aim of this study is to screen a therapeutic agent with a cognitive function. The inhibitory effect of $Curcuma$ $longa$ hot water extract (CLWE) on the angiotension-converting enzyme and acetylcholinesterase derived from rabbit lungs and neural cells (PC12), as well as its antioxidant effect, was investigated in this study. Thus, for the first time, the direct scavenging effect of CLWE on DPPH radicals, superoxide anions, hydroxyl radicals, lipid peroxidation, reducing power, and the protective effect of DNA oxidation related to oxidative stress was evaluated in vitro. In addition, it was observed that CLWE especially exhibited a scavenging effect on reducing power and superoxide anions in this study. CLWE showed a protective effect on DNA oxidation produced by hydroxyl radicals. Furthermore, CLWE inhibited the activity of angiotensin-converting enzymes above 0.25%. Additionally, the extract inhibited oxidative stress and inducible nitric oxide in neuronal cells. Therefore, these results demonstrated that CLWE has antioxidant activity and neuronal cell protective effects, suggesting that it may have great potential as a natural source for human health.

Estimation of the Flash Point for n-Pentanol + n-Propanol and n-Pentanol + n-Heptanol Systems by Multiple Regression Analysis (다중회귀분석법을 이용한 n-Pentanol + n-Propanol계 및 n-Pentanol + n-Heptanol계의 인화점 예측)

  • The flash point is one of the most important properties for characterizing the fire and explosion hazard of liquid solutions. In this study, the flash points of two flammable binary mixtures, n-pentanol + n-propanol and n-pentanol + n-heptanol systems were measured using a Seta flash closed cup tester. The flash point was estimated using the methods based on Raoult's law and multiple regression analysis. The measured flash points were also compared with the predicted flash points. The absolute average errors (AAE) of the results calculated by Raout's law were $1.3^{\circ}C$ and $1.3^{\circ}C$ for the n-pentanol + n-propanol and n-pentanol + n-heptanol mixtures, respectively. The absolute average errors of the results calculated by multiple regression analysis were $0.4^{\circ}C$ and $0.3^{\circ}C$ for the n-pentanol + n-propanol and n-pentanol + n-heptanol mixtures, respectively. According to the AAE, the calculated values based on multiple regression analysis were better than those based on Raoult's law.

Effects of High-Intensive Exercise Duration on Hematologic Properties and Cytokines in Rats (고강도운동 지속시간이 rat의 혈액학적 조성과 사이토카인에 미치는 영향)

  • The effects of duration and time-dependent, high-intensity exercise on hematological properties and inflammation-related parameters in rats were studied. 20, 60, and 120 min of high-intensive exercise were performed daily for 8 weeks. None of the complete blood count (CBC) factors were affected by the exercise, except for the leukocyte concentration which, in the 20 min group, showed an increase of 47% compared to the control, but this was decreased after 60 min by 30% compared to the control. As exercise was performed for 60 min or longer, serum concentrations of $Fe^{++}$, unsaturated iron biding capacity (UIBC), and total iron biding capacity (TIBC) were significantly elevated in comparison to the control, where 20 min of exercise did not show any change. Both levels of interleukin-6 (IL-6), a pro-inflammatory cytokine, and interleukin-10 (IL-10), an anti-inflammatory cytokine in the serum, were elevated in response to the high-intensity exercise, however the rate of IL-6 increase was higher than the rate of exercise intensity increase, thus the offset of inflammation might be suggested. The concentration of nitric oxide (NO) in the serum became high in response to the exercise. Overall, the current observation suggests that inflammation-like responses to high-intensity exercise might be due to high circulation of blood flow and high oxygen requirement, resulting in systemic damages. However, under the current high-intensity exercise conditions, more than 20 min of exercise might not be suggested for health care purposes.

Evaluation of Human Papillomavirus Genotyping from Formalin-fixed Paraffin-embedded Specimens in Cervical Cancers (자궁경부암 파라핀 조직에서 인유두종바이러스 유전형 검사의 유용성 평가)

  • Cervical carcinoma is the second leading cause of cancer-related deaths in women around the world, and it is associated with the Human Papillomavirus (HPV) infection. HPV genotyping is important for vaccine policy, etiology, natural history, and epidemiology studies. The use of formalin-fixed paraffin-embedded (FFPE) tissues for HPV genotyping by reverse blot hybridization assays (REBA) has not been clearly confirmed in retrospective studies. The aim of this study was to evaluate the usefulness and efficiency of FFPE tissues from cervical cancers for HPV genotyping. HPV genotypes were detected in 52 FFPE tissues from cervical carcinoma specimens by REBA. HPV was detected in 32 (61.5%) of 52 specimens from FFPE, among which 27 (84.4%) harbored single infections and 5(15.6%) contained multiple infections. The HPV single infections (27) were analyzed by high-risk type 18(8), 58(6), 16(5), 33(1), 35(1), 39(1), 56(1) and low risk type 11(2), 6(1), 70(1). The HPV multiple infections (5) included 16/18(2), 18/52(1), 16/56(1), 16/18/33(1). Please consider being more specific here. Do you mean the analysis? Prevalence and Genotype Analysis of High Risk-human Papillomavirus Infection in Busan Women (부산지역 여성의 고위험군 인유두종 바이러스 유병률과 유전자형 분석)

  • The main cause of cervical cancer is the human papillomavirus (HPV), and HPV DNA has been reported in 99.7% of patients with cervical cancer. The worldwide prevalence is highest for the HPV 16 and 18 genotypes, but HPV 52 and 58 have the highest prevalence in Asian countries, including Korea. The purpose of this study was to obtain basic data for the prevention of cervical cancer by analyzing the prevalence of HPV and the genotypes of high risk-human papillomavirus (HR-HPV) infection in women in Busan, Korea. We analyzed 1,995 cases of HPV in women who visited a Busan obstetrics and gynecology hospital from January 2016 to December 2017. The prevalence of HPV among these women was 28.3% (565/1995), and the HR-HPV infection rate was 75.4% (426/565). The HR-HPV genotype with the highest prevalence was HPV-52 (63/565, 11.2%), followed by HPV-58 (56/565, 9.9%), HPV-53 (55/565, 9.7%), and HPV-16 (53/565, 9.4%). The HR-HPV infection rate of young women 18-39 years old was 60.3% (257/426), so this age group should undergo continuous monitoring. The cytological results revealed a high infection rate for HPV-16 in high grade squamous intraepithelial lesions (HSIL) and squamous cell carcinoma (SCC). However, further evaluation of more samples is needed to confirm the HR-HPV genotypes related to the development of cervical epithelial neoplasias.
