Evaluation of Coulter® LH 750TM on Leukocyte Differential Count and Suspect Flags (Coulter® LH 750TM에 의한 백혈구 감별계산 및 Suspect Flags의 평가)

  • Automatic hematology analyzers provide the leukocyte differential count and the useful flags of the various hematological parameters. We compared LH 750 (Beckman Coulter Co., Miami, FL, USA) with manual method on differential leukocyte counts and evaluated the usefulness of the suspect flags, provided by this instrument. The comparison of leukocyte differential counts between two methods showed good correlation coefficient (r), which were 0.95 (neutrophil), 0.92 (lymphocyte), 0.82 (monocyte) and 0.95 (eosinophil). The frequency of the total flags displayed on LH 750 were 15.5%, which included immature granulocyte/left shift 63.5%, nucleated RBC 14.6%, platelet clumps 13.1%, variant lymphocyte 50% and blast 16.6%. This instrument showed higher positive predictive value in the flags such as platelet clumps 68.8%, immature granulocyte/left shift 61.5%, nucleated RBC 27.3%, variant lymphocyte 50% and blast 16.7%. In this study, the leukocyte differential counts of LH 750 showed good correlation with manual method and the suspect flags also showed a good performance for applying the criteria of re-examination in the clinical laboratory.

  • A total of 108 strains of MRSA (Methicillin-resistant Staphylococcus aureus) clinical isolates was collected from $121^{st}$ general hospital (U.S. military hospital), Korean healthcare facility from January to March in 2005 and Wonju Christian hospital in 1999. Antimicrobial susceptibility test by Vitek System and MIC test using oxacillin and cephalothin stripes by E-test were executed. PCR based detection of mecA gene was performed on the all of MRSA clinical isolates, too. MRSA clinical isolates were characterized with antimicrobial resistance patterns, PCR based detection of mecA gene and validation of the multiplex PCR strategy of SCCmec among clinical isolates.

Effects of 2-methoxy-1,4-naphthoquinone (MQ) on MCP-1 Induced THP-1 Migration (MCP-1에 의해 유도된 THP-1 유주에 미치는 2-methoxy-1,4-naphthoquinone (MQ)의 영향)

  • This study examined the effects of 2-methoxy-1,4-naphthoquinone (MQ) on the monocyte chemoattractant protein-1 (MCP-1)-induced migration of monocytes, which is an important phenomenon for the body defense and immune response. MQ is a major component extracted from Impatiens balsamina leaves, which have been used for many years in Asian medicine for the treatment of a range of diseases and pain. The cytotoxicity of MQ began to appear at a concentration of $10{\mu}M$, and approximately 50% cytotoxicity was confirmed at $100{\mu}M$. The MCP-1 induced migration of the THP-1 monocyte cell line increased after MQ treatment in a dose dependent manner and the largest increase was observed at $0.1{\mu}M$. The level of cAMP expression decreased after a treatment with $0.1{\mu}M$ MQ. The phosphorylation of extracellular signal-regulated kinases 1/2 (Erk1/2), a key signaling protein involved in the signaling pathway of C-C motif chemokine receptor 2 (CCR2), a receptor for MCP-1, was increased by the simultaneous treatment of $0.1{\mu}M$ MQ. These results show that MQ increases the MCP-1-induced migration of THP-1, decreases the level of cAMP expression, and increases the level of Erk1/2 phosphorylation.

The House Dust Mite Allergen, Dermatophagoides pteronyssinus Suppresses the Chemotactic Activity of Human Monocytes

  • House dust mite (HDM) is important in the pathogenesis of allergic diseases including asthma and atopic dermatitis. Dermatophagoides pteronissinus (Dp) is one of major HDM allergens. In this study, we investigated that Dp extract (DpE) affects on the chemotactic activity of monocytes isolated from the peripheral blood. DpE inhibited the migration of human monocytes in response to CC chemokines such as MIP-$1{\alpha}$, RANTES, HCC-4, MCP-1, and TARC. DpE did not alter the expression of CC chemokine receptors (CCRs) such as CCR1, CCR2, CCR3, CCR4, and CCR5. These results indicate that DpE blocks the chemotaxis of human monocytes and its mechanism is not involved in alteration of CCR expression. Better understanding of the effect of DpE on monocytes will enable elucidation of the role of Dp in the development of allergic diseases.

Effects of Electrical Stimulation on Wound Healing and Skin Mast Cells in Streptozotocin-Induced Diabetic Rats (스트렙토조토신 유도 당뇨 흰쥐에서 전기자극이 상처치유와 피부 비만세포에 미치는 영향)

  • The aim of this study was to investigate the effect of electrical stimulation on healing of impaired wound and alteration of mast cells in experimental diabetic rats. Thirty male Sprague-Dawley rats were divided into three groups : incision (control), diabetes+incision (diabetes) and diabetes + incision + electrical stimulation (D/ES). Diabetes was induced in rats by streptozotocin (STZ) injection (60 mg/kg, one time) and 20 mm length incision wounds were created on the back after shaving hair. The electrical stimulation rats were treated with a current intensity of 30~50 V at 120 pps and $140{\mu}s$ for 10 days from 3 days after STZ injection. The lesion and adjacent skin tissues were fixed with 10% buffered formalin, embedded with paraffin. For wound healing analysis, hematoxylin-eosin (HE) and picrosirius red staining were performed. Mast cells (MC) were stained with toluidine blue (pH 0.5) and quantified at ${\times}200$ using a light microscope. The density of keratinocyte proliferation and microvessels in skin tissues were analyzed using a computerized image analysis system on sections immunostained with proliferative cell nuclear antigen (PCNA) and ${\alpha}$-smooth muscle actin (${\alpha}$-SMA), respectively. The results showed that the wound healing rate, collagen density and neoepidermis thickness, density of PCNA-positive cells and density of ${\alpha}$-SMA-positive vessels were significantly higher in D/ES rats than in diabetic rats. The density of MCs and degranulated MCs in D/ES rats were also significantly higher than those in diabetic rats. These findings suggest that the electrical stimulation may promote the tissue repair process by accelerating collagen production, keratinocyte proliferation and angiogenesis in the diabetic rats, and MCs are required for wound healing of skin in rats.

Comparison of Post-hoc among Petty Merchants of Traditional Markets

  • The aim of this study undertook to ascertain the health levels and risk among petty merchants at some traditional markets by way of physical examination and laboratory tests. Data was collected through physical examination and laboratory tests on blood pressure, glucose, cholesterol, triglyceride, LDL cholesterol, and HDL cholesterol. The data was analyzed by Frequency analysis, T-test, One-Way ANOVA using the PASW statistics 18.0 program. By blood pressure on sex, male ($1.89{\pm}0.6$), female ($1.48{\pm}0.6$) appeared (p<0.001). Male levels were higher than in female levels. Additionally TG appeared male levels ($193.4{\pm}129.4$), female levels ($142.7{\pm}80.6$). Male levels were higher than in female levels (p<0.006). By blood pressure as the age increased in more than 70 (2.17) exhibited a statistically significant difference. These merchants did not practice health behaviors nor take institutional health management benefit. Therefore governmental level community based health management services are recommended for them.

Clinical Use of PFA®-100 in Pre-surgical Screening for Platelet Function Test (수술 전 혈소판 기능 검사를 위한 PFA®-100의 임상적 이용)

  • The Platelet Function Analyzer (PFA)$^{(R)}$-100 measures the ability of platelets activated in a high-shear environment to occlude an aperture in a membrane treated with collagen and epinephrine (CEPI) or collagen and ADP (CADP). The time taken for the flow across the membrane to stop (closure time, CT) is recorded. The aim of this study was to assess the potential of the PFA$^{(R)}$-100 as a primary clinical screening tool using the wide spectrum of clinical samples assessed for platelet function as well as to perform the optimal algorithm for the use of PFA$^{(R)}$-100. We established the reference interval in 460 hospital inpatients defined as having normal platelet function based on classical laboratory tests. The reference interval by using the range $5^{th}$ and $95^{th}$ percentile was 84~251 seconds for males CEPI-CT and 85~249 seconds for females CEPI-CT. A total of 1,200 inpatients were enrolled to identify impaired hemostasis before surgical interventions. The abnormal group showing prolonged CEPI-CT was 303 cases (18.9%). Only 3 cases had both abnormal CEPI-CT and CADP-CT. Several factors including sample errors, drugs, hematologic abnoralities were contributed to unexpected prolonged CEPI-CT for screening test. The von Willebrand factor (vWF:Ag) assay was performed only in one patient to verify the algorithm for the use of PFA$^{(R)}$-100. The PFA$^{(R)}$-100 was sensitive and rapid method for primary screening test of platelet dysfunction, so we can substitute it for the bleeding time in routine clinical practice.

Altered Proteome of Extracellular Vesicles Derived from Bladder Cancer Patients Urine

  • Proteomic analysis of extracellular vesicles (EVs) from biological fluid is a powerful approach to discover potential biomarkers for human diseases including cancers, as EV secreted to biological fluids are originated from the affected tissue. In order to investigate significant molecules related to the pathogenesis of bladder cancer, EVs were isolated from patient urine which was analyzed by mass spectrometry based proteomics. Comparison of the EV proteome to the whole urine proteome demonstrated an increased number of protein identification in EV. Comparative MS analyses of urinary EV from control subjects and bladder cancer patients identified a total of 1,222 proteins. Statistical analyses provided 56 proteins significantly increased in bladder cancer urine, including proteins for which expression levels varied by cancer stage (P-value < 0.05). While urine represents a valuable, non-invasive specimen for biomarker discovery in urologic cancers, there is a high degree of intra- and inter-individual variability in urine samples. The enrichment of urinary EV demonstrated its capability and applicability of providing a focused identification of biologically relevant proteins in urological diseases.

Antioxidative Activity of Cornus officianalis Extracts Obtained by Four Different Extraction Techniques (산수유 추출방법에 따른 항산화 기능 분석)

  • Oxidative stress leads to damage in all components of the cell, including proteins, lipids, and DNA due to imbalance between reactive oxygen species production and cellular detoxification. Phytochemicals are well-known to contain antioxidants, and their physiological role has been intensively studied. The fruit of Cornus officianalis has been used in oriental medicine and has been reported to have many functions. In this study, four different extraction techniques were applied to extract functional components from the fruit of Cornus officianalis, and the content of loganin, which is an antioxidant having DPPH radical and hydrogen peroxide scavenging activity and reducing power, was analyzed in each extract. Extraction techniques employed in this study were heat extraction by water, 70% ethanol extraction, enzyme treatment, and combination of enzyme treatment and heat extraction by water. All extracts contained 11.8-18.0 mg/g loganin and showed antioxidation function assayed by measuring DPPH radical and hydrogen peroxide scavenging activity and reducing power. Among them, heat extraction was the most effective technique, showing a maximum amount of loganin (18.0 mg/g) and antioxidative activity at 100 mg/ml concentration. Each extract showed very low cytotoxicity up to at 500 mg/ml but 10-20% cytotoxicity at 1,000 mg/ml by in vitro MTT assay.

Feline adipose tissue-derived mesenchymal stem cells pretreated with IFN-γ enhance immunomodulatory effects through the PGE2 pathway

  • Background: Preconditioning with inflammatory stimuli is used to improve the secretion of anti-inflammatory agents in stem cells from variant species such as mouse, human, and dog. However, there are only few studies on feline stem cells. Objectives: This study aimed to evaluate the immune regulatory capacity of feline adipose tissue-derived (fAT) mesenchymal stem cells (MSCs) pretreated with interferon-gamma (IFN-γ). Methods: To assess the interaction of lymphocytes and macrophages with IFN-γ-pretreated fAT-MSCs, mouse splenocytes and RAW 264.7 cells were cultured with the conditioned media from IFN-γ-pretreated MSCs. Results: Pretreatment with IFN-γ increased the gene expression levels of cyclooxygenase-2, indoleamine 2,3-dioxygenase, hepatocyte growth factor, and transforming growth factor-beta 1 in the MSCs. The conditioned media from IFN-γ-pretreated MSCs increased the expression levels of M2 macrophage markers and regulatory T-cell markers compared to those in the conditioned media from naive MSCs. Further, prostaglandin E2 (PGE2) inhibitor NS-398 attenuated the immunoregulatory potential of MSCs, suggesting that the increased PGE2 levels induced by IFN-γ stimulation is a crucial factor in the immune regulatory capacity of MSCs pretreated with IFN-γ. Conclusions: IFN-γ pretreatment improves the immune regulatory profile of fAT-MSCs mainly via the secretion of PGE2, which induces macrophage polarization and increases regulatory T-cell numbers.
