Protective Effect of Herbal Mixture Including Lycii Fructus on Hepatotoxicity Induced by Thioacetamide in Mice (구기자 복합물 약침액이 간기능 개선에 미치는 영향)

  • Objectives : This study investigated the hepatoprotective effect of herbal mixture including Lycii fructus (HML) in thioacetamide (TAA)-induced hepatotoxicity in mice. Methods : To confirm the liver protective effect, induced by TAA for 3 days injection at 100 mg/kg mice, HML were treated for 8 weeks at 300 mg/kg/day, 1000 mg/kg/day. Positive control was treated silymarin 50 mg/kg/day after TAA injection. The changes of mortality rate, clinical signs, organ weight, relative liver, blood chemistry and histopathological findings were analyzed after experiment. Results : Body weight gain was observed in all groups, but TAA treated group at 4th week and all treated groups decreased weight compared to the untreated group. As a result of organ weight measurement, organ weight gain due to hepatic injury was observed statistically significantly in TAA-treated group and TAA+Silymarin treated group, and the herbal mixture-treated group showed a tendency to decrease compared to the TAA treated group. Blood biochemistry showed that total cholesterol and very low density lipoprotein cholesterol decreased statistically in TAA+low-dose and high dose herbal mixture treated group compared to the TAA-treated group. Histopathological examination showed that liver abnormalities were not observed in untreated group, liver fibrosis was observed in liver injury with TAA treated and herbal mixture treated group. And, TAA+high dose herbal mixture group showed relaxation tendency on liver calcification compared to the TAA treated group. Conclusions : According to the above results, HML provided hepatoprotective effects on the hepatic injury by reduction of inflammatory responses.

Melanotrichia in Poodles : Clinical and Histopathological Findings (푸들에서 발생한 melanotrichia: 임상적 그리고 조직병리학적 특징)

  • A 5-year-old, female Poodle and her daughter, 2-year-old, were referred to Konkuk University Veterinary Teaching Hospital (KVTH) due to discoloration of hair on dorsum, and generalized alopecia following clipping. Some papules and pustules are found on patch area. Complete blood counts and serum chemistry profiles were not remarkable. Trichogram revealed that melanin-granule was pigmented on hair bulb and shaft in affected area and no melanin pigmentation was noted in non-patch area. And skin biopsy showed mild epidermal hyperplasia and mild inflammation in dermis of the patch area. This case report indicates that melanotrichia can be occurred by postclipping inflammation. However, the mechanism of melanocyte proliferation and rates of melanogenesis in response to postclipping remains to be unclear in these cases.

Diagnosis of pemphigus erythematosus in a Jindo dog (진도개에서 홍반천포창(pemphigus erythematosus)의 진단)

  • A 6-year-old female Jindo dog weighing 20 kg was presented to the Veterinary Medical Teaching Hospital of Seoul National University for the formation of crust and erosion of the nasal bridge and planum, and for alopecia of ear margin. Erythema, crusts, alopecia and erosions were present in the nasal planum and the bridge of the nose on physical examination. There were no abnormalities on CBC, serum chemistry, and basic dermatologic examination which included skin scraping, bacterial cultures, and fungal cultures. On cytology, the acantholytic cells were surrounded by non-degenerate neutrophils. Biopsy samples obtained from skin lesionswere diagnosedpemphigus erythematosus by histopathology and immunochemical staining. Therefore the dog was initially treated with oral prednisolone as an immunosuppressive dose for tour weeks, but clinical manifestations did not improve so far.

Pathological evaluation of renal changes induced by multiple nephropathogenic factors in SPF chickens II. Clinicopathological observation (신부전 요인에 의해 유발된 닭 신장변화의 병리학적 관찰 II. 임상병리학적 관찰)

  • Renal failure is one of the main causes of economic impacts in the poultry industry and complex syndrome with different severity of clinical signs caused by multiple nephropathogenic factors such as infectious bronchitis viral infection and excess salt and calcium in diet. To evaluate the correlation between severity of renal failure and the causative nephropathogenic factors, one-day-old specific pathogen free chickens were treated with either single causative factor or multiple causative factors described as above. Each group was designed as control for non-treated control, IB for infectious bronchitis virus (IB virus) infection, IBHNa for IB virus infection with high diet salt, IBHCa for IB virus infection with high diet calcium, IBHNC for IB virus infection with high diet salt and calcium, HNa for high diet salt, HCa for high diet calcium and HNC for high diet salt and calcium. Chickens were inoculated with IB virus at 1-day-old and remained on their respective diets until 21 day of age. Plasma $Na^+$, $Cl^-$, BUN, creatinine, calcium and uric acid values were examined. The results obtained were as follows ; IB virus and high dietary calcium combined treatment showed elevated plasma uric acid. BUN and creatinine values were not characteristic on chicken renal failure. But plasma uric acid values were increased according to renal lesion. Hypercalcemia and hyperuricemia did not induce urate deposition and mineralization in the kidney.

Effect of Herbal Mixture including Lycii Fructus (HML) on Stress Relief and Melatonin in Caffeine and Light induced Stress Animal Model (Caffeine 및 광자극 유발 스트레스 동물모델에서 구기자 복합물이 스트레스 완화 및 혈중 멜라토닌에 미치는 영향)

  • Objectives : In the present study, we investigated whether or not an herb mixture including Lycii fructus(HML) has attenuation effects on light stress in rats. Methods : Light stress was induced by exposure to 300 lx of constant lifht for 24 h and 1mg/kg of caffeine was injected into the rat. Rats were orally treated with distilled water of HML for 21 days at 100mg/kg/d. The changes of mortality rate, clinical signs, body, blood chemistry, melatonin and IL-2 expression were analyzed after experiment. Results : Normal weight changes were observed in all treatment groups. In addition, no changes were observed in dead animals and general symptoms. At the time of autopsy, the appearance of all groups and gross examination of major organs were observed, and no gross findings were observed. Blood biochemical tests showed that total cholesterol and HDL cholesterol levels were statistically significantly decreased in the group treated with HML compared to the stress-induced group. Melatonin was measured in the blood, but there was no significant difference in the group treated with HML compared to the group with stress-induced group, but it tended to increase. IL-2 was measured in the blood, no significant change was observed in the group treated with HML compared to the stress-induced group. Conclusions : According to the above results, HML maybe help with hematological changes expressed by caffeine and light-induced stress.

Health Impairment and Concentration of Hippuric Acid in Urine of Workers Exposed to Toluene (톨루엔 폭로근로자의 요중 마뇨산 농도 및 건강장해)

  • The relationships between subjective symptoms of toluene exposed workers and concentration of their urinary excretion of hippuric acid were investigated. The exposed groups of 146 workers exposed to toluene and the control groups of 47 workers have never been exposed to toluene in Ulsan area were selected and studied. Hippuric acid was measured by HPLC, and counts of blood cells and liver function test were also performed. The mean value of urinary hippuric acid concentration of the control group was 0.322($\pm$0.267) g/L, while that of the exposed group was 1.260($\pm$0.395) g/L. As the concentration of hippuric acid had statistical proximity in $0.1\%$ level, WBC, GOT, and GPT didn't have any proximity(P>0.05). The exposed group showed lower level of leucocyte counts 6522.40(1710.3) than the control group 6891.50(1483.7). The exposed group showed higher level of GOT(25.75), GPT(27.09) than GOT(23.75), GPT(25.21) of the control group. Dried skin was the highest complained symptom of toluene exposed workers, the second strained eye, the third poor auditory function, and the fourth was headache.

Anti-obesity Effects of Mulberry Leaf and Yacon Extract in Obese Rats

  • We evaluated the effect of anti-obesitic activity of MYE (mulberry leaf + Yacon tuber) extracted from Morus alba as muberry leaf and Smallanthus sonchifolia as yacon. 1%, 3%, or 5% of MYE were treated to Sprague-Dawley rats exposed to a high-fat diet. MYE treated rats were suppressed weight during four weeks, and they lost weight significantly after six weeks. Common blood chemistry panels related to liver function revealed significant improvement in the MYE-treated groups. The expression of leptin as indicators for obesity was decreased in perirenal fat. Such results indicate that MYE could be a promising candidate for the improvement of obesity. In addition, MYE effected on deceased glucose metabolism, reducing the activities of glucose-6-phosphate dehydrogenase (G-6-PDH) and glucokinase related to glycogen synthesis. The fatty liver was observed in high-fat diet-treated rats, resulting from increased number of adipose cells and Ito cells. However, this pathologic change was significantly improved by administration of MYE. MYE have significant effects on antioxdative function and glycometabolism against high fat diet. Thereby, it seems that MYE prevent fatty liver by high-fat diet. Thus it is suggested that MYE would be worth being developed as an biofunctional food to prevent undesirable effects caused by obesity.

Comparison of Hemoglobin Activity of Angelica gigas, Angelica sinensis and Angelica acutiloba roots (참당귀, 중국당귀, 일당귀의 헤모글로빈 활성도 비교)

  • Objectives : Angelicae gigas, A. sinensis and A. acutiloba are three types of plants used as Angelicae Radix (Dang-Gui). Many doctors of Korean medicine want to know the difference in clinical use of these three species. This study aimed to compare the hemoglobin-related activity of the extracts of Angelicae gigas, A. sinensis and A. acutiloba roots by measuring the intensity of binding oxygen to hemoglobin using Raman spectroscopy. Methods : Hemoglobin activity was measured by chemical analysis and Raman spectroscopy to compare the pharmaceutical efficacy of three Angelica root extracts. The oxygenated hemoglobin intensity, blood decursinol and acetylcholinestrase(AChE) concentration in mice were measured. In addition, the effects of three Angelica root extracts on oxygenated hemoglobin intensity, decursinol and AChE concentration in red blood cells (RBC) from human were also investigated. Results : The contents of decursin, decursinol and decursinol angelate, which affected physiological activity and RBC properties, were higher in the extract of A. gigas root than in those of A. sinensis and A. acutiloba roots. Moreover, oxygenated hemoglobin intensity in the A. gigas extract was higher than that of other two species in the blood of mice and human RBCs. Also, the blood decursinol and AChE concentrations of A. gigas root extract were higher than that of A. sinensis and A. acutiloba roots. Conclusions : These results suggest that A. gigas is more effective in treating disease related oxygen deficiency in RBC deformation under oxidative stress.

Ginseng-plus-Bai-Hu-Tang ameliorates diet-induced obesity, hepatic steatosis, and insulin resistance in mice

  • Background: Dietary fat has been suggested to be the cause of various health issues. Obesity, hypertension, cardiovascular disease, diabetes, dyslipidemia, and kidney disease are known to be associated with a high-fat diet (HFD). Obesity and associated conditions, such as type 2 diabetes mellitus and nonalcoholic fatty liver disease (NAFLD), are currently a worldwide health problem. Few prospective pharmaceutical therapies that directly target NAFLD are available at present. A Traditional Chinese Medicine, ginseng-plus-Bai-Hu-Tang (GBHT), is widely used by diabetic patients to control glucose level or thirst. However, whether it has therapeutic effects on fat-induced hepatic steatosis and metabolic syndrome remains unclear. Methods: This study was conducted to examine the therapeutic effect of GBHT on fat-induced obesity, hepatic steatosis, and insulin resistance in mice. Results: GBHT protected mice against HFD-induced body weight gain, hyperlipidemia, and hyperglycemia compared with mice that were not treated. GBHT inhibited the expansion of adipose tissue and adipocyte hypertrophy. No ectopic fat deposition was found in the livers of HFD mice treated with GBHT. In addition, glucose intolerance and insulin sensitivity in HFD mice was also improved by GBHT. Conclusion: GBHT prevents changes in lipid and carbohydrate metabolism in a HFD mouse model. Our findings provide evidence for the traditional use of GBHT as therapy for the management of metabolic syndrome.

A 90-Day Inhalation Toxicity Study of Ethyl Formate in Rats

  • Ethyl formate, a volatile solvent, has insecticidal and fungicidal properties and is suggested as a potential fumigant for stored crop and fruit. Its primary contact route is through the respiratory tract; however, reliable repeated toxicological studies focusing on the inhalation route have not been published to date. Therefore, the present study was conducted to investigate the safety of a 90-day repeated inhalation exposure in rats. Forty male and 40 female rats were exposed to ethyl formate vapor via inhalation at concentrations of 0, 66, 330, and 1,320 ppm for 6 hr/day, 5 days a week for 13 weeks. Clinical signs, body weights, food consumption, urinalysis, hematologic parameters, serum chemistry measurements, organ weights, necropsy, and histopathological findings were compared between the control and ethyl formate-exposed groups. Locomotor activity decreased during exposure and recovered afterward in male and female rats exposed to 1,320 ppm ethyl formate. Body weight and food consumption continuously decreased in both sexes exposed to 1,320 ppm ethyl formate from week 1 or 3 compared with the control values. The increases in adrenal weight and decreases in thymus weight were noted in both sexes exposed to ethyl formate at 1,320 ppm. Degeneration, squamous metaplasia of olfactory epithelium in the nasopharyngeal tissue, or both were noted in the male and female rats at 1,320 ppm and female rats at 330 ppm ethyl formate. Taken together, our results indicate that ethyl formate-induced changes were not observed in male and female rats at 330 and 66 ppm, respectively. This indicates that exposure to ethyl formate at concentrations below 66 ppm for 90 days is relatively safe in rats. This is the first report of a full-scale repeated inhalation toxicity assessment in rats and could contribute to controlling occupational environmental hazards related to ethyl formate.
