Synthesis of Novel N-(2-Hydroxyphenyl)arylsulfonamides as Selective HDAC Inhibitory and Cytotoxic Agents

  • Based on the finding that the 2-aminobenzamido group of MS-275 plays a crucial role in inhibiting HDACs through chelation of zinc existing at the active site of HDAC enzymes, novel N-(2-hydroxyphenyl)arylsulfonamide derivatives were synthesized for their potential ability to inhibit HDACs and evaluated for anticancer activity against human breast cancer cell line (MCF-7). Although the synthesized arylsulfonamides have failed to significantly inhibit total HDACs activity, phenyl carbamate-linked arylsulfonamide 10 and benzyl thiocarbamate-linked arylsulfonamide 15 exhibited good anticancer activities, which were only 4.3- and 3.6-fold lower anticancer activities, respectively, than MS-275 that is undergoing phase II clinical trials. These results suggest that these compounds may act as a selective HDAC inhibitor and probably N-(2-hydroxyphenyl) sulfamoyl group may play an important role in interacting with HDAC enzymes through chelation of zinc ion.

  • Concanavalin-A, lectin extracted from Korean native plant “Banha”(Pine-llia Termata) and their conjugation products with tyrosine and 5-iodo-6-aminouracil were labelled by radioactive iodiae-125, and these labelled compounds were subjected to a hemagglutination test in vitro. The labelling procedures were presented with respect to labelling yield and the trends on the conjugation reaction of lectin with various amino acids were discussed. The preliminary results on the hemagglutination tests against cancer tissues were presented and the potential clinical applications of these labelled compounds were discussed.

  • Isovitexin, a marker compound with various pharmacological activities, in Lespedeza cuneata, was analyzed using high performance liquid chromatography coupled with UV (HPLC/UV). There are no previous reports on using L. cuneata as the source material for the quantification of isovitexin. In this study, we developed an optimized method using HPLC-UV analysis, which was validated using various parameters. Our method demonstrated high specificity, and good separation of the chromatographic peak was achieved. Parameters such as linearity (r2≥0.9997), precision, and accuracy indicated that our proposed analytical method had good reliability and sensitivity. These results demonstrate the utility and convenience of our method for rapidly quantifying isovitexin in L. cuneata extracts.

The inhibitory effects of glabridin on human platelet aggregation and thrombus formation

  • Glycyrrhiza glabra is a well-known medicinal herb that grows in various parts of the world and glabridin is a major chemical compound that is found in the root extract of Glycyrrhiza glabra. Glabridin is a natural compound known to have antioxidant, anti-inflammatory, anti-atherogenic, anti-osteoporotic and skin-whitening. In this study, we investigated if glabridin inhibited platelet aggregation and thrombus formation. We observed that glabridin inhibited collagen-induced platelet aggregation and suppressed signal transduction molecules such as phosphatidylinositol-3 kinase (PI3K), Akt, glycogen synthase kinase-3α/β (GSK-3α/β), SYK, cytosolic phospholipase A2, and p38 expression. In addition, glabridin suppressed thromboxane A2 generation and thrombin-induced clot retraction. Taken together, glabridin showed strong antiplatelet effects and may be used to block thrombosis- and platelet-mediated cardiovascular diseases.

Effectiveness of Clinical Examination for Detection of Respiratory Tuberculosis (호흡기 결핵 환자에서 임상 검사의 유용성)

  • To find out the effectiveness of clinical examination for the diagnosis of respiratory tuberculosis, a 78 respiratory tuberculosis patient,s group was matched by sex and age with 78 control healthy subjects. In the result of blood chemistry, mean values of $123.5{\pm}62.04mg/dL$ in glucose, $429.01{\pm}150.77IU/L$ in LDH, and $44.51{\pm}43.76IU/L$ in ${\gamma}$-GTP, were higher than that of the controls (healthy subjects), and $3.51{\pm}0.68mg/dL$ in albumin was lower than that of the controls. In the result of the haematology examination, mean values of $12.52{\pm}3.27g/dL$ in hemoglobin, $36.72{\pm}7.28%$ in hematocrit, and $24.61{\pm}12.36%$ in lymphocyte, were lower than that of the controls, $9.23{\pm}5.25%$ in monocyte $78.30{\pm}37.35mm/hr$ in ESR, and $48.45{\pm}35.15U/L$ in ADA were higher than that of the controls. For the comparisons of the tuberculosis patients values from normal reference values, 22.2% in glucose, 22.4% in LDH, 25.0% in ${\gamma}$-GTP, 35.4% in albumin, 88% in ESR, and 88.6% in ADA, showed abnormal values. We concluded that the values of glucose, ${\gamma}$-GTP, albumin, WBC, RBC, hemoglobin, hematocrit, lymphocyte, monocyte, ADA, and the ESR were useful in tuberculosis diagnosis.

Anti-hypertensive Effects of Artichoke Supplementation in Adults: A Systematic Review and Dose-response Meta-analysis of Randomized Controlled Trials

  • Despite controversies, no earlier study has systematically summarized findings from earlier studies on the effect of artichoke supplementation on blood pressure. Therefore, current systematic review and meta-analysis was done on the effect of artichoke supplementation on systolic blood pressure (SBP) and diastolic blood pressure (DBP) in adults. Five databases were searched from inception to January 2022 using relevant keywords. All randomized clinical trials investigating the impact of oral artichoke supplementation on any of the blood pressure parameters including SBP or/and DBP were included. Out of 1,507 citations, 7 trials that enrolled 472 subjects were included. Artichoke supplementation resulted in significant reduction in SBP (weighted mean difference [WMD], -2.01 mmHg; 95% confidence interval [CI], -3.78, -0.24; p = 0.026) and DBP (WMD, -1.45 mmHg; 95% CI, -2.81, -0.08; p = 0.038). Greater effects on SBP were detected in trials using ≤ 500 mg artichoke, lasted > 8 weeks, participants aged < 50 years' old and sample size ≤ 70. There was also a similar impact of artichoke on DBP. However, significant non-linear associations were found between artichoke supplementation dosage and study duration with both SBP (for dosage: pnon-linearity = 0.002, for duration: pnon-linearity = 0.016) and DBP (for dosage: pnon-linearity = 0.005, for duration: pnon-linearity = 0.003). We found a significant reduction in both SBP and DBP following artichoke supplementation in adults. It could be proposed as a hypotensive supplement in hypertension management.

Clinical Effectiveness of Cefoperazone(Cefobid) in Obstetrics-Gynecological Infection (부인과감염증(婦人科感染症)에 대(對)한 Cefoperazone(Cefobid)의 임상효과(臨床效果))

  • A total of 30 cases of gynecological infection were treated with Cefoperazone at the Department of Ob-Gy, Seoul Baik Hospital, Seoul, Korea from Sept. 1979 to Aug. 1980. Cefoperazone sodium was administered in a dose of 2g/day intramusculary or intravenously for 6 to 13 days. The effective rates of Cefoperazone based on clinical and bacteriological response were 100% in 9 cases of acute non-gonorrheal PID, 5 cases of postpartum endometritis, 2 cases of recurrent non-gonorrheal PID and 80% in acute gonorrheal PID(8 out of 10 cases). A case of recurrent gonorrheal PID and 2 out of 3 cases of postoperative infection responded also satisfactorily to Cefoperazone treatment. There were no drug-related abnormal findings in urinalysis, hematology and blood chemistry. Any particular side effects, except for one case of fever and rash which disappeared spontaneously without any treatment or discontinuation of the drug, were not noted during or after the treatment.

  • Nephrotic syndrome(NS) is characterized albuminuria, hypoalbuminemia, hyperlipidemia and edema. The thromboembolic complications of NS are reasonably common and have a broad range of clinical manifestations. The treatment of NS is empirical and symptomatic because the etiology and pathogenesis are unknown. We report treatment of patient who has been afflicted with NS for 20 years. He complained edema, dyspnea, Rt. hemiparesis, aphasia. We treated him with converting enzyme inhibitor, steroid, three kinds of herb-medicine for 45 days. The patient improved in clinical symptoms and routine chemistry, urine analysis test.

  • For quantitative measurement of reflected light from a clinical diagnostic scrip, a prototype of reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate re(leclance parameters. The strop was illuminated at $45^{\circ}C$ through optical fiber and the intensity of reflected light was determined at rectangulat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500,550,570 and 610nm) for blood glucose strip. For higher concentration than 300mg/dl about glucose, a saturation state of absorbance was observed at 500,550 and 570nm. The correlation between glucose concentration and parameters was the best at 610nm.

  • Objective : The purpose of this study was to know the clinical effect of Hydrotherapy based Ultasonic spa on Atopic Dermatitis and Allergic Rhinitis. Methods : Allery Test(MAST CLA Allergy system, inhalant panel), DITI, Bio Chemistry, Hematology were investigated in 20 patients(Atopic Dermatitis 10, Allergic Rhinitis 10) who visited Department of Ophthalmology & Otolaryngology & Dermatology, Semyung Oriental medicine hospital from November 2004 to January 2005. Results : 1. In atopic dermatitis patients, there was good in 7 cases(70%), excellent in 3 cases(30%). 2. In allergic rhinitis patients there was improved in 5 eases(50%), good in 2 cases(20%), unimproved in 3cases(30%). 3. In conclusion, there was good(doctor and patients ail) in 6(60%), unimproved in 3(30%) Conclusion : Hydrotherapy based Utasonic spa showed useful effect on Atopic Dermatitis and Allergic Rhinitis.
