Four phenylpropanoid glycosides and a monoterpene glycoside were isolated from the flower of Clerodendrum trichotomum. Structures of the isolated compounds were identified as acteoside (1), martynoside (2), leucosceptoside A (3), isoacteoside (4) and neohancoside A (5) by spectroscopic analysis. Compounds 1-4 were isolated from the flower of C. trichotomum for the first time. Compound 5 was first obtained from genus Clerodendrum as well as family Verbenaceae. The antioxidant activity of compounds 1-5 were evaluated by the DPPH free radical scavenging assay. Compounds 1-4 exhibited strong antioxidant activity.

Seven flavonoids were isolated from the flower of Clerodendrum trichotomum. Their structures were identified as apigenin (1), genistein (2), chrysoeriol (3), genistein 7-O-glucoside (4), kaempferol 3-O-glucoside (5), isorhamnetin 3-O-glucoside (6) and apigenin 7-O-glucoside (7) on the basis of spectral data. These compounds were isolated from C. trichotomum for the first time. The antioxidant activity of compounds 1-7 were evaluated by the ORAC (oxygen radical absorbance capacity) assay, and the ORAC values were expressed as relative trolox equivalent. All isolated compounds exhibited antioxidant activity.

Clerodendrum trichotomum has been widely used as a traditional medicine for the treatment of numerous diseases, including pain management. However, there have been extremely limited pharmacological and phytochemical studies on Clerodendrum trichotomum up to now. In this study, we investigated the effects of the methanolic extract of Clerodendrum trichotomum (MCT) on nociceptive pain in mice. Our finding demonstrate that MCT treatment significantly extended the latency time in both the tail-immersion test and hot-plate test. Additionally, MCT treatment reduced acetic acid-induced writhing motions. These results suggest that MCT possesses strong anti-nociceptive activities against thermal and chemical nociception. In the formalin test, mice fed with MCT exhibited reduced licking time during both the early and late phases, thereby confirming the therapeutic potential of MCT in central and peripheral nociception. Furthermore, in combination tests using naloxone, the MCT-mediated anti-nociception was slightly reduced, indicating that MCT might act as a partial opioid receptor agonist. Based on these results, MCT may be a valuable candidate for the development of anti-nociceptive agents.

Clerodendrum trichotomum is a folk medicine exhibiting anti-hypertension, anti-arthritis, and anti-rheumatism properties. However, little is known about whether the material might prevent hyperuricemia and associated inflammation. In this study, we explored whether C. trichotomum leaf extract (CTE) prevented hyperuricemia induced by potassium oxonate (PO) in mice. CTE (400 mg/kg body weight) significantly reduced the serum uric acid (UA), blood urea nitrogen (BUN), and serum creatinine levels and increased urine UA and creatinine levels. CTE ameliorated PO-induced inflammation and apoptosis by reducing the levels of relevant proteins in kidney tissues. Also, CTE ameliorated both UA-induced inflammatory response in RAW 263.7 cells and UA-induced cytotoxicity in HK-2 cells. In addition, liver transcriptome analysis showed that CTE enriched mainly the genes for mediating positive regulation of MAPK cascade and apoptotic signaling pathways. Together, the results show that CTE effectively prevents hyperuricemia and associated inflammation in PO-induced mice.
This research is the palynotaxonomic revision to clarify palynological descriptions and relationships of taxa in Korean Verbenaceae. To do this, morphology, size, sculpture and aperture of pollen grain were investigated with scanning electron microscope. As a result, palynological key as to 5 genera 10 species drawn up. Palynological descriptions of Callicarpa dichotoma, Callicarpa mollis, Callicarpa chejuensis, Vitex chinensis and Caryopteris incana are near completion at first. Vervena officinalis are newly described and descriptions of Callicarpa japonica, Clerodendrum trichotomum, Vitex rotundifolia and Caryopteris divaricata are corrected. Palynological data of Korean Verbenaceae is useful to intrageneric relationships. Namely, as to genus Callicarpa, C. dichotoma and C. japonica are considered to be more advanced than C. mollis and C. chejuensis. As to genus Caryopteris, C. divaricata is more advanced than C. incana.

Clerodendrum trichotomum (CT) leaves and stems have been used in folk medicine for their anti-hypertension, arthritis, rheumatism, and anti-inflammatory properties. This study was performed to evaluate the potential of CT as an anti-oxidant and anti-inflammatory agent. CT leaves were extracted using 70% ethanol (EtOH). Then, using this extract, a hexane, chloroform ($CHCl_3$), ethyl acetate (EtOAc), and n-butanol (BuOH) fraction was prepared. The polyphenol contents were higher in the EtOAc fraction ($78.08{\mu}g/mg$) and BuOH fraction ($77.54{\mu}g/mg$) compared to the other fractions. Also, these two fractions exhibited strong 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging activities. Xanthine oxidase inhibitory activities were higher in the $CHCl_3$ fraction ($IC_{50}=4.43{\mu}g/ml$) and EtOAc fraction ($IC_{50}=5.69{\mu}g/ml$). Moreover, the EtOAc fraction effectively inhibited nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells ($IC_{50}=18.87{\mu}g/ml$). Thus, we investigated the effects of the EtOAc fraction on the expression of pro-inflammatory cytokines, inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX-2) in LPS-stimulated RAW 264.7 cells. The treatment of the EtOAc fraction ($100{\mu}g/ml$) effectively decreased the levels of the tumor necrosis factor ${\alpha}$ ($TNF-{\alpha}$) and interleukin-6 (IL-6), and the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). These results suggest the potential for CT extract and fractions as promising anti-oxidant and anti-inflammatory agents.
Since the main symptoms of COVID-19 involve the respiratory system, the infection rate of this disease is predicted to be higher in patients with other respiratory conditions such as allergic rhinitis. In such a situation, it will be meaningful to conduct research on an allergy treatment that has fewer side effects and can effectively reduce allergy symptoms. Here, we prepared experimental samples under various fermentation conditions with mixed extracts of six medicinal plants. To examine the anti-allergic efficacy of these samples, an egg albumin-induced allergic rhinitis animal model experiment, a serum histamine and IgE experiment, and a COX and LO inhibitory activity experiment were conducted. As a result of animal experiments, OVA+SP-4 showed superior efficacy compared to OVA+SP-1 in nasal rubbing and sneezing experiments and had anti-allergic efficacy similar to that of OVA-cetirizine. The serum histamine concentration of OVA+SP-4 was also 1.3 times higher than that of the OVA+cetirizine group, showing a high histamine reduction ability, and IgE showed the same trend. An analysis of COX inhibitory efficacy also confirmed that COX-1 and COX-2 inhibitory efficacy is high, and the longer the fermentation time, the higher the antiallergic efficacy. The composition proposed by this study is expected to have a significant effect on sustainable allergy prevention and treatment in the future by applying it to human patients.

Five species of plants (Clerodendrum trichotomum Thunb., Angelica dahurica (Fisch. ex Hoffm.) Benth. & Hook. f. ex Franch. & Sav., Caryopteris incana (Thunb. ex Houtt.) Miq., Lonicera japonica Thunb., and Parasenecio auriculatus var. matsumurana Nakai) native to the clean area of Mt. Jiri were selected. The collection period was from May to September 2021, and the five species plants were collected in their native habitats with flowers in full bloom. The collected plants were extracted with 70% EtOH, and 17 kinds of polyphenol components were analyzed. Next, flowers, leaves, stems, and roots were separated from plants, extracted with 70% EtOH for each part and experiments were conducted on DPPH, ABTS, total polyphenols, and total flavonoids. The results are as follows. 1. It was found that there were a total of 8 kinds of polyphenols contained in 5 species of plants that are native to Mt. Jiri. Among the polyphenol components, chlorogenic acid was contained in 4 species of plants, and caffeic acid was contained in 2 species of plants. 2. As a result, the DPPH radical scavenging activity was the best in the stem of P. auriculata and the C. trichotomum was good regardless of the specific part. It was found that the activity-scavenging activity was good in the flowers of A. dahurica and the leaves of L. japonica. 3. The highest ABTS radical scavenging activity was C. trichotomum Thunb., whose EC50 value was 38.73~66.28ppm. Next, the leaves and stems of L. japonica Thunb., A. dahurica and P. auriculata, and the leaves and stems of C. incana appeared in that order. 4. The highest total polyphenol content was 154.83mg GAE/g in the leaves of C. trichotomum, followed by about 130mg GAE/g in the flowers of C. trichotomum and P. auriculata. The lowest was 26.27mg GAE/g in the stems of A. dahurica.

Peeling damage of trees is usually caused by Cervidae such as deer, roe deer because of the lack of food in forests. However, it happens as part of the developing of antlers in Jeju Island when the roe deer try to remove the Velvet-the skin of the antlers. The research area is the Hannam experimental forest (400 m up to 500 m above sea level) of Korea Forest Research Institute in Jeju Island, and the survey was carried out along the 6 km long of forest road with 5 m width on both sides. Twenty five tree species (total 267 stands) are damaged by peeling; 18 (134 stands) deciduous broad-leaved species, 5 (71 stands) in evergreen broad-leaved species, 2 (62 stands) coniferous species. The most common damaged species are in order of Daphniphyllum macropodum, Cryptomeria japonica, Lindera erythrocarpa, Clerodendrum trichotomum, Zanthoxylum schinifolium. Mainly damaged trees are approximately 3~4 years old saplings, and they show the mean height $120.7{\pm}42.4cm$, diameter measured at 5 cm height $1.5{\pm}0.5cm$. The Lowest peeling beginning height is $22.1{\pm}10.1cm$, and the mean length of peeling is $27.5{\pm}10.6cm$. Once the peeling damage happens, the saplings are infected by fungi secondly, and are distorted or dead, therefore the future structure of warm-temperate forests could be in influenced in species. Warm-temperate forest landscape and species change related to the climate change is a rising issue in Jeju Island. However the changes caused by peeling damage also could be an important issue in the natural process of forest environment, afforestation, local nursery and sustainable forest management of Jeju Island.

In order to develop a new depigmenting agent, extracts were obtained from 60 native plants and their antimelanogenic activities were screened by evaluating the inhibitory effect on tyrosinase which is a major enzyme responsibles for the melanin synthesis. The extracts of Trichosanthes kirilowii fruits, Phyllostachys bambusoides inner films (BIF), Clerodendrum trichotomum leaves, and Acer okamotoanum leaves showed relatively high inhibitory effect on tyrosinase and their $IC_{50}$ values were $50{\sim}100{\mu}g/mL$. The extract of BIF inhibited melanin synthesis of B16F10 melanoma cells by 52%, which was the highest among those of various extracts. Furthermore, the effect of BIF extract is 10% higher than that of arbutin (42%), a popular depigmenting agent in Korea. Ten compounds having antimelanogenic activity were isolated from the BIF extract by solvent extraction and chromatography. These compounds were identified as phenolic derivatives: SM701, SM702, SM703, and BPR211 were hydroquinone derivatives; SM707 a gallic acid derivative; SM704, SM705, SM706, SM708 and SM709 ferulic acid derivatives. The free radical scavenging activities of these compounds were measured and compared to those of hydroquinone and vitamin C. The $SC_{50}$ values scavenging 50% DPPH of SM702 and SM709 were $60{\sim}70{\mu}M$ similar to that of hydroquinone and those of SM701 and SM708 were $30{\sim}40{\mu}M$ slightly lower than that of vitamin C. These results suggest the presence of components having high antioxidant activity in the BIF extract. The SM709, identified as 1,2-O-diferulylglycerol, inhibited the activities of tyrosine hydroxylase and dopa oxidase by 18 and 60%, respectively. The SM709 also inhibited the melanin synthesis of B16F10 melanoma cells by 62% and this was the highest antimelanogenic activity among those obtained from the various purified compounds. Therefore, antimelanogenic activity of the BIF extract was concluded to be due to both inhibition of DOPA oxidase and antioxidant activity.
