The purpose of this study was to compare the adaptation to tooth structure of light - cured glass ionomer cement with that of self -cured glass ionomer cement. In this study, class V cavities were prepared on the buccal surfaces of 10 extracted human premolar teeth, and teeth were randomly assigned 2 groups of 5 teeth each. The cavities of self-curing glass ionomer cement group were restored with the Fuji n. and the cavities of lightcuring glass ionomer cement group were restored with the Fuji II LC. The surfaces of glass ionomer cements were applied with All-Bond 2 adhesive, and cured with visible light. The restored teeth were stored in 100% relative humidity at $37^{\circ}C$ for 24 hours. And then. the roots of the teeth were removed with the tapered fissure bur and the remaining crowns were sectioned occlusogingivally through the center of glass ionomer restorations. Adaptation at tooth-restoration interface was assessed occlusally. axially, and gingivally by scanning electron microscope. The results were as follows : 1. On the occlusal margin, the group of self - curing glass ionomer cement showed closer adaptation to both enamel and dentin than the group of light-curing glass ionomer cement showing 5/lm gap between cement and tooth structure. 2. On the axial wall. the group of light-curing glass ionomer cement showing 5-$7{\mu}m$ gap between cement and dentin showed closer adaptation to dentin than the group of self -curing glass ionomer cement showing 10-$15{\mu}m$ gap between cement and dentin. 3. On the gingival margin, the group of light-curing glass ionomer cement showing 2-$5{\mu}m$ gap between cement and dentin(X 1200) showed closer adaptation to dentin than the group of self-curing glass ionomer cement showing 20pm gap between cement and dentin(X 600). 4. The group of self -curing glass ionomer cement showed closer adaptation on the occlusal margin than on the gingival margin, and the group of light-curing glass ionomer cement showed similar adaptation on both occlusal and gingival margins.
The purpose of this study was to evaluate the effect of a desensitizer on dentinal bond strength in cementation of composite resin inlay. Fifty four molar teeth were exposed the occlusal dentin. Class I inlay cavities were prepared and randomly divided into six groups. Control group: no agent, Group 1 : Isodan, Group 2 : One-step, Group 3 : All-Bond SE, Group 4 : Isodan + One-step, Group 5 : Isodan + All-Bond SE. Desensitizing agent and dentin bonding agents were applied immediately after the completion of the preparations. Impressions were then made. The composite resin inlays (Tescera, Bisco) were fabricated according to the manufacturers' guidelines. Cementation procedures followed a standard protocol by using resin cement (Bis-Cem, Bisco). Specimens were stored in distilled water at $37^{\circ}C$ for 24 hours. All specimens were sectioned to obtained sticks with $1.0{\times}1.0\;mm^2$ cross sectional area. The microtensile bond strength (${\mu}TBS$) was tested at crosshead speed of 1 mm/min. The data was analyzed using one way ANOVA and Tukey's test. Scanning electron microscopy analysis was made to examine the details of the bonding interface, 1. Group 1 showed significantly lower ${\mu}TBS$ than other groups (p<0.05). 2. There was no significant difference between the ${\mu}TBS$ of Group 3 and Group 5. 3. The ${\mu}TBS$ of Group 4 showed significantly lower than that of Group 2 (p<0.05). In conclusion, a desensitizer (Isodan) might have an adverse effect on the bond strength of composite resin inlay to dentin.
Saprolegniasis is one of the most devastating oomycete diseases in freshwater fish which is caused by species in the genus Saprolegnia including Saprolegnia parasitica. In this study, we isolated the strain of S. parasitica from diseased rainbow trout in Korea. Morphological and molecular based identification confirmed that isolated oomycete belongs to the member of S. parasitica, supported by its typical features including cotton-like mycelium, zoospores and phylogenetic analysis with internal transcribed spacer region. Pathogenicity of isolated S. parasitica was developed in embryo, juvenile, and adult zebrafish as a disease model. Host-pathogen interaction in adult zebrafish was investigated at transcriptional level. Upon infection with S. parasitica, pathogen/antigen recognition and signaling (TLR2, TLR4b, TLR5b, NOD1, and major histocompatibility complex class I), pro/anti-inflammatory cytokines (interleukin $[IL]-1{\beta}$, tumor necrosis factor ${\alpha}$, IL-6, IL-8, interferon ${\gamma}$, IL-12, and IL-10), matrix metalloproteinase (MMP9 and MMP13), cell surface molecules ($CD8^+$ and $CD4^+$) and antioxidant enzymes (superoxide dismutase, catalase) related genes were differentially modulated at 3- and 12-hr post infection. As an anti-Saprolegnia agent, plant based lawsone was applied to investigate on the susceptibility of S. parasitica showing the minimum inhibitory concentration and percentage inhibition of radial growth as $200{\mu}g/mL$ and 31.8%, respectively. Moreover, natural lawsone changed the membrane permeability of S. parasitica mycelium and caused irreversible damage and disintegration to the cellular membranes of S. parasitica. Transcriptional responses of the genes of S. parasitica mycelium exposed to lawsone were altered, indicating that lawsone could be a potential anti-S. parasitica agent for controlling S. parasitica infection.
The purpose of this in vitro study was to evaluate dentin bonding by two different dentin bonding systems(DBS) using acetone based primer or adhesive [All Bond 2(AB2), One Step(OS)] when they were applied by wet or dry bonding technique. Morphology of resin-dentin interface and hybrid layer thickness(HLT) were investigated using Confocal Laser Scanning Microscope(CLSM) and compared to shear bond strength(SBS). 72 extracted sound human molars were randomly divided into 4 groups of 18 teeth each - Group 1.(AW); AB2 by wet bonding. Group 2(AD); AB2 by dry bonding. Group 3.(OW); OS by wet bonding, Group 4.(OD); OS by dry bonding. In 6 teeth of each group, notch-shaped class V cavities(depth 2mm) were prepared on buccal and lingual surface at the cementoenamel juction(12 cavities per group). To obtain color contrast in CLSM observation, bonding resins of each DBS were mixed with rhodamine B and primer of AB2 was mixed with sodium fluorescein. Prepared teeth of each group were treated with AB2, OS, respectively according to the manufacturer's instructions except for dentin surface moisture treatment after acid etching. In group 1 and 3, after acid etching, excess water was removed with wet tissue(Kimwipes), leaving consistently shiny, visibly hydrated dentin surface. In group 2 and 4, dentin surface was dried for 10 seconds at 1 inch distance. The treated teeth were then packed with composite resin(${\AE}$litefil) and light-cured. 12 microscopic samples($60{\sim}80{\mu}m$ thickness) of each group were obtained after longitudinal section and grinding(Exakt cutting and grinding system). Morphological investigation of resin-dentin interface and HLT measurement using CLSM were done. For measurement of SBS, remaining 12 teeth of each group were flattened occlusally to remove all enamel and grinded to 500 grit SiC(Pedemet Specimen Preparation Equipment). After applying DBS on the exposed dentin surface, composite resin was applied in the shape of cylinder, which has 5mm diameter, 1.5mm thickness, and light cured. SBS was measured using Instron with a crosshead speed of 0.5mm/min. It was concluded as follows, 1. HLT of AW(mean: $2.59{\mu}m$) was thicker than any other group, and followed by AD, OW, OD in descending order(mean; 2.37, 2.28, $1.92{\mu}m$). Only OD had statistically significant differences(p<0.05) to AW and AD. 2. There were intimate contact of resin and dentin at the interface in wet bonding groups, but gaps or irregular interfaces were observed in dry bonding groups. 3. The length, diameter, density of resin tags were various even in the same group without significant differences between groups and lots of adhesive lateral branches were observed. 4. There were no statistically significant difference of SBS between AB2 and OS, but SBS of wet bonding groups were significantly higher(p<0.05) than dry bonding groups. 5. There were no consistent relationships between HLT and SBS.
The purpose of this study was to observe the changes of acidity of resin cement(Time Line), glass ionomer cement(GC Fugi Lining LC), zinc phosphate cement(Fleck's zinc cement). zinc oxide eugenol cement(Sultan,Chemists.) in vivo and in vitro. Class I cavities with 3mm depth were prepared on the occlusal surfaces of 20 recently extracted human Mn. molar teeth and 20 human Mn. 3rd molar teeth in oral cavity. The prepared cavities were divided into 4 groups of each 5 teeth using the above 4 cavity liners. Each cement was mixed in accordance with manufacturer's direction at the room temperature of $23^{\circ}{\pm}5^{\circ}C$ and filled into the cavity in a width of 1 mm. The microelectrode of pH meter was inserted into the prepared cavity which was filled with mixed cement, and the acidity of cement was measured for 3 days from the beginning of cement mix in vitro and in vivo. The measured acidity was then statistically analyzed by ANOVA. The results were as follows. 1. In vitro, the pH of zinc oxide eugenol cement was statistically lower than that of the three other groups at 2min, 4min, 6min, 8min, 10min, 12min, 18min, 20min. (p<0.05). 2. The pH of zinc oxide eugenol cement in vivo was statistically higher than that in vitro at 16min,16min, 20min(p<0.05). 3. The pH of zinc phosphate cement in vivo was statistically higher than that in vitro at 4min, 20min(p<0.05). 4. In vitro and in vivo, there was no significant difference in the pH between the resin cement and the glass ionomer cement(p>0.05). 5. The initial acidity was not high, but almost neutral in all kinds of the cements.
Korean Native Pig (KNP) has a uniform black coat color, excellent meat quality, white colored fat, solid fat structure and good marbling. However, its growth performance is low, while the western origin Yorkshire pig has high growth performance. To take advantage of the unique performance of the two pig breeds, we raised crossbreeds (KNP ${\times}$ Yorkshire to make use of the heterotic effect. We then analyzed the liver transcriptome as it plays an important role in fat metabolism. We sampled at two stages: 10 weeks and at 26 weeks. The stages were chosen to correspond to the change in feeding system. A total of 16 pigs (8 from each stage) were sampled and RNA sequencing was performed. The reads were mapped to the reference genome and differential expression analysis was performed with edgeR package. A total of 324 genes were found to be significantly differentially expressed (${\left|log2FC\right|}$ > 1 & q < 0.01), out of which 180 genes were up-regulated and 144 genes were down-regulated. Principal Component Analysis (PCA) showed that the samples clustered according to stages. Functional annotation of significant DEGs (differentially expressed genes) showed that GO terms such as DNA replication, cell division, protein phosphorylation, regulation of signal transduction by p53 class mediator, ribosome, focal adhesion, DNA helicase activity, protein kinase activity etc. were enriched. KEGG pathway analysis showed that the DEGs functioned in cell cycle, Ras signaling pathway, p53 signaling pathway, MAPK signaling pathway etc. Twenty-nine transcripts were also part of the DEGs, these were predominantly Cys2His2-like fold group (C2H2) family of zinc fingers. A protein-protein interaction (PPI) network analysis showed that there were three highly interconnected clusters, suggesting an enrichment of genes with similar biological function. This study presents the first report of liver tissue specific gene regulation in a cross-bred Korean pig.
We investigated anti-hyperglycemic and anti-obese effects of Panax ginseng berry extract and its major constituent, ginsenoside Re, in obese diabetic C57BL/6J ob/ob mice and their lean littermates. Animals received daily intraperitoneal injections of Panax ginseng berry extract for 12 days. On Day 5, 150 mg/kg extract-treated ob/ob mice had significantly lower fasting blood glucose levels compared to vehicle-treated mice $(156{\pm}9.0\;mg/dl\;vs.\;243{\pm}15.8mg/dl,$ P<0.01). On Day 12, the extract-treated ob/ob mice became normoglycemic $(137{\pm}6.7\;mg/dl)$ and had significantly improved glucose tolerance. The overall glucose excursion during the two-hour intraperitoneal glucose tolerance test (IPGTT), calculated as area under the curve (AUC), decreased by $46\%$ (P<0.01) compared to vehicle-treated ob/ob mice. Glucose levels of lean mice were not significantly affected by the extract. The improvement in blood glucose levels in 150 mg/kg extracttreated ob/ob mice was associated with significant reduction in serum insulin levels of fed and fasting mice. Consistent with an improvement in insulin sensitivity, hyperinsulinemic euglycemic clamp study revealed a more than 2-fold increase in the rate of insulin-stimulated glucose disposal in treated ob/ob mice $(112{\pm}19.1\;vs.\;52{\pm}11.8{\mu}mol/kg/min$ for the vehicle group, P<0.01). In addition, 150 mg/kg extract-treated ob/ob mice, but not the lean mice, lost significant weight (from $51.7{\pm}1.9g\;on\;Day\;0\;to\;45.7{\pm}1.2$ on Day 12, P<0.01 compared to vehicle-treated ob/ob mice), associated with a significant reduction in food intake (P<0.05) and a very significant increase in energy expenditure (P<0.01) and body temperature (P<0.01). A 12-day treatment with 150 mg/kg Panax ginseng berry extract also significantly reduced plasma cholesterol levels in ob/ob mice. Additional studies demonstrated that ginsenoside Re, a major constituent of the ginseng berry, but not from the root, plays a significant role in anti-hyperglycemic action. This anti-diabetic effect of ginsenoside Re was not associated with body weight changes, suggesting that other constituents in the extract have distinct pharmacological mechanisms on energy metabolism. The identification of a significant anti-hyperglycemic activity in ginsenoside Re may provide an opportunity to develop a novel class of anti-diabetic agent.
The purpose of this study was to investigate the effects of flipped learning through EBSmath on Students' 'rate and ratio' learning. By increasing demands for change in education, an innovative teaching and learning paradigm, 'Flipped Learning', has been presented and drawing attentions. In South Korea, Flipped Learning is also highly recognized for its effectiveness by many scholars and various media. However, this innovative learning model has limitations in application and expansion due to the excessive burden of class preparation of teachers. As remote learning becomes more active, it would be possible to overcome the limitations of Filliped learning by using the platform provided by the Korea Educational Broadcasting System (EBS). EBSmath is an online learning module that is designed to assist students' self-directed learning. Thus, EBSmath would reduce teachers' burden to prepare mathematics classes for the application of Flipped Learning; and led to students' better understanding of mathematical concepts and problem solving. In this study, the effect of Flipped Learning through EBSmath on learning 'rate and ratio' was investigated. In order to scrutinize the effects of flipped learning, students' achievement and mathematical disposition were examined and analyzed. Students' achievement, specifically, was divided into two subcategories: concept understanding and problem solving. As a result, Flipped learning through EBSmath had a positive effect on students' 'rate and ratio' problem solving. In addition, a statistically significant change was identified in the 'willingness', which is subdomain of students' mathematical disposition.

The purpose of this study is to recognize kindergarten teachers' experiences of child violence for identifying the site's needs for the implementation of preventive education in early childhood and to provide basic data on child violence. To this end, nine teachers were interviewed in-depth. From the study results, first, child violence-related child behavior, which usually occurs in kindergartens, includes physical assault, aggression, verbal violence, threats and threats, and bullying. Second, teachers usually use direct intervention laws in cases of child violence, but were finding it difficult to intervene properly with many tasks or high teacher-to-child ratios. Teachers recognized the need for lower teacher-to-child ratios and placement of each class burden. Third, teachers were aware of the need for prevention education for child violence targeting infants, and instigated the following appropriate interactions immediately after problem behavior occurred: large group activities, specific multimedia education data and parent education. Fourth, teachers are concerned about the possibility of problem behavior being learned and imitated through education in the implementation of prevention education for children's violence. Therefore, this study proposed the need to develop various teaching methods that could be applied to infant education sites, focusing on the types of child violence-related problem behaviors that occur in kindergartens.
This study proposes a new method for categorizing the hydrological soil groups by considering the recent revision results of soil series. Also, the proposed method is evaluated by comparing the categorizing result with those based on existing three different methods. As an example, the proposed method is applied to Jeju Island to estimate the CN value, which is then compared with CN values estimated by applying the existing three different methods. Summaries of the results are as follow. (1) The revision result since 2007 shows that the soil texture has been changed in the 43 soil series, the drainage class in the 1 soil series, the permeability in the 15 soil series, and the impermeable layer in the 26 soil series. (2) The categorizing result of hydrological soil groups by applying the proposed method shows that the group B is the most dominant group covering up to 49.25%. On the other hand, one of the existing method of 1987 provides the group C as the most dominant group (46.43%). Method of 1995 defines the group B as the most dominant group (27.69%). The other method of 2007 distinguishes the group D (35.82%) to be the most dominant group. (3) Also, the CN value estimated by applying the proposed method to Jeju Island is found to be smaller than those based on existing three methods. This result indicates the possible overestimation of the CN value when applying the existing three methods.
