Monitoring Culicine Mosquitoes (Diptera: Culicidae) as a Vector of Flavivirus in Incheon Metropolitan City and Hwaseong-Si, Gyeonggi-Do, Korea, during 2019

  • The flaviviruses are small single-stranded RNA viruses that are typically transmitted by mosquitoes or tick vectors and are etiological agents of acute zoonotic infections. The viruses are found around the world and account for significant cases of human diseases. We investigated population of culicine mosquitoes in central region of Korean Peninsula, Incheon Metropolitan City and Hwaseong-si. Aedes vexans nipponii was the most frequently collected mosquitoes (56.5%), followed by Ochlerotatus dorsalis (23.6%), Anopheles spp. (10.9%), and Culex pipiens complex (5.9%). In rural regions of Hwaseong, Aedes vexans nipponii was the highest population (62.9%), followed by Ochlerotatus dorsalis (23.9%) and Anopheles spp. (12.0%). In another rural region of Incheon (habitat of migratory birds), Culex pipiens complex was the highest population (31.4%), followed by Ochlerotatus dorsalis (30.5%), and Aedes vexans vexans (27.5%). Culex pipiens complex was the predominant species in the urban region (84.7%). Culicine mosquitoes were identified at the species level, pooled up to 30 mosquitoes each, and tested for flaviviral RNA using the SYBR Green-based RT-PCR and confirmed by cDNA sequencing. Three of the assayed 2,683 pools (989 pools without Anopheles spp.) were positive for Culex flaviviruses, an insect-specific virus, from Culex pipiens pallens collected at the habitats for migratory birds in Incheon. The maximum likelihood estimation (the estimated number) for Culex pipiens pallens positive for Culex flavivirus was 25. Although viruses responsible for mosquito-borne diseases were not identified, we encourage intensified monitoring and long-term surveillance of both vector and viruses in the interest of global public health.

항만과 도시기능의 연계 방안에 관한 연구 - 인천의 사례를 중심으로 - (Connection Strategies between Port and City functions - Focused on the case of Incheon -)

  • 본 연구는 항만과 도시기능이 충돌하는 현상을 보이는 인천의 사례를 통하여 항만과 도시기능의 연계 방안을 도출하는 것을 연구의 목적으로 하였다. 연구의 분석방법으로 다차원 척도분석을 사용하였으며, 유사성을 갖는 항만도시로 그룹핑 하기 위하여 총 5개 지표(컨테이너 물동량, 인구수, 도시면적, 정기취항 선사 수, RCI[Relative Concentration Index])를 사용하였다. 연구대상은 Containerisation International Yearbook (2010)을 통하여 제시된 18개 아시아 항만도시를 연구대상으로 선정하였다. 분석 결과, 동종 항만도시 군내에 연계지수인 RCI가 인천보다 상회하는 요코하마를 벤치마킹 대상으로 선정하였다. 요코하마는 인천에 비해 항만배후단지의 활용 측면에서 다양하며 '미나토미라이 21'과 같은 항만과 도시의 일관된 정책 사례를 펼쳐 항만과 도시기능의 연계성을 높인 것으로 파악되었다.

  • In this study, it analysis electric field of SMD(Surface Mounted Device) Inductor Core and it get electric field only exist inside of SMD core. Therefore electric fields do not affect any device and equipments. These results are very important to design data acquisition system(several test equipments such as temperature, impedance, and current test), because data acquisition system can place under the SND Inductor core. So, it can be decrease their test error due to electric field.

  • Eggs of parasite were found 73 of 1,459 in sand samples from public parks and playgrounds in Incheon for 5 years (from 2004 to 2008). Of the 1,459 sand samples examined, 17.8% of samples were found to be positive ova of Toxocara canis, 11.0% for Toxocaris leonina, 5.5% for Toxocara cati, 2.7% Ancylostoma caninum and 1.4% for Strongyloides spp. It was found mixed infestation with Toxocara canis and Toxocara cati 60.2%.

Investigation of Brucella canis infection in public animal shelters and kennels in Incheon

  • This survey was performed to investigate the seroprevalence of antibodies to Brucella canis in dogs from public animal shelters and breeding kennels in Incheon. A total of 402 dogs selected randomly were tested serologically by using immunochromatographic antibody test kit. None of 289 dogs in public animal shelters were sero-positive and 10 (8.9%) of 113 dogs in breeding kennels were sero-positive. 5 (4.4%) strains of Brucella canis were isolated from 10 sero-positive dogs' blood. Antimicrobial susceptibility test was carried out by Disk diffusion method. They were susceptible to tetracyclines, quinolones, aminoglycosides and combination amoxicillin with clavulanic acid.

인천광역시 유통 농산물의 최근 3년간 잔류농약 실태조사 (A Survey on the Residual Pesticides on Agricultural Products on the Markets in Incheon from 2016 to 2018)

  • BACKGROUND: This survey conducted on agricultural products in Incheon for the past three years from 2016 to 2018 to detect residual pesticides and to monitor the use of pesticides. METHODS AND RESULTS: Residual pesticides were analyzed for a total of 5,937 agricultural products in Samsan wholesale market, traditional market and large retailers in Incheon. Samples were analyzed by multi class pesticide method using GC-MS/MS, LC-MS/MS, GC-ECD/NPD, and UHPLC. In 59 cases (1.0%) residual pesticides were detected improperly and these cases exceeded maximum residue limits (MRLs). CONCLUSION: The ratios of violative agricultural products were similar each year at approximately 1.0% per year, but the residual pesticide detection rates with the limit included in the samples were shown to be gradually decreasing to 17.9%, 12.6%, and 11.2% annually. The frequency of violative residual pesticides was high in order of Diazinon, Chlorpyrifos, Iprodione, etc. Residual pesticide monitoring might be needed in the future continuously, as violations in agricultural products distributed in Incheon are shown at a similar level every year.

Hard Ticks as Vectors Tested Negative for Severe Fever with Thrombocytopenia Syndrome in Ganghwa-do, Korea during 2019-2020

  • This study aimed to characterize the seasonal abundance of hard ticks that transmit severe fever with thrombocytopenia syndrome virus from April to November 2019 and 2020 on Ganghwa-do, Incheon Metropolitan City, Korea. The ticks were collected at grassland, grave site, copse and mountain road using a collection trap method. The ixodid hard ticks comprising three species (Haemaphysalis longicornis, H. flava, and Ixodes nipponensis) collected were 6,622 in 2019 and 3,811 in 2020. H. longicornis was the most frequent (97.9% in 2019 and 96.0% in 2020), followed by H. flava (2.0% and 3.0% in 2019 and 2020, respectively) and I. nipponensis (less than 0.1%). Our study demonstrated that seasonal patterns of the tick populations examined for two years were totally unsimilar. The hard ticks tested using RT-qPCR were all negative for severe fever with thrombocytopenia syndrome virus.

The monitoring of heavy metals (lead and cadmium) in honeys consumed in Incheon region

  • This study was conducted to investigate the contents of lead (Pb) and cadmium(Cd) in honeys (n=60) from April to May in 2004 and to provide the scientific basis for heavy metal regulations of Korea Food Code. The samples were digested with acids, then analyzed for the contents of Pb and Cd by an inductively coupled plasma spectrometer (ICP). The contents of Pb and Cd [minimum-maximum (mean)] were 0.189-1.82 mg/kg (0.568) and not detected (ND)-0.016 mg/kg (0.0008) in domestic acacia honeys (n=20), ND-1.702 mg/kg (0.329) and ND-0.243 mg/kg (0.013) in domestic wild flower honeys (n= 18), ND-0.322 mg/kg (0.073), ND-0.027 mg/kg (0.002) in imported honeys (n=13), ND-3.754 mg/kg (0.671) and ND-0.658 mg/kg (0.073) in foreign honeys (n=9) brought by Korean travellers, respectively. According to the results, foreign honeys brought by Korean travellers were detected with the highest level of Pb and Cd. Therefore, we recommend that heavy metals of domestic and foreign honeys should be continuously monitored. It is also thought that these results could be the important references to establish the standard of Pb and Cd in honey.

Studies on mycotoxins using LC-MS/MS for the forage produced in Incheon

  • The purpose of this study was to investigate the contamination level of representative mycotoxins that have adverse effects on livestock by using LC-MS/MS method and to utilize the results as basic data for the establishment of quality control system for feed, and to provide information on production and storage. A total of nine mycotoxins, including aflatoxin $B_1$, aflatoxin $B_2$, aflatoxin $G_1$, aflatoxin $G_2$, ochratoxin A, fumonisin $B_1$, fumonisin $B_2$, deoxynivalenol (DON), zearalenone (ZEN) were simultaneously analyzed in LC-MS/MS under ESI positive mode. Fumonisin $B_1$ and fumonisin $B_2$ were detected from 3 cases of 75 forage produced in Incheon area, the detection rate was 4.0%. The detection concentration was 0.01~0.02 mg/kg, which was lower than the domestic recommended limit. Fumonisins were detected in a slightly different manner from the results of mycotoxin studies reported in Korea, which is attributed to the high temperature and dry summer weather of the year. The result of LC-MS/MS method performance of 9 mycotoxins, the recovery of DON was quite low as $41.53{\pm}3.91%$ that is not suitable for simultaneous analysis. This is probably due to that the extract solution used in this study was not suitable for the extraction of DON, along with the characteristics of a very dry forage. For the study of mycotoxins in Incheon area forage for the first time, further investigation is needed for the safe supply of livestock products.

액체크로마토그래피를 이용한 벌꿀 중 테트라싸이클린계 항생물질의 정량분석 및 잔류조사 (Determination and survey of tetracyclines residue in honey by high performance liquid chromatography)

  • Oxytetracycline, tetracycline, chlortetracycline and doxycycline in honey were separated by solid phase extraction (SPE) and determined with high performance liquid chromatography (HPLC) with UV/Visible detector. Analysis was carried out using following conditions: XTerra $C_8$ column $(3.9\times150mm\;i.d. 5{\mu}m)$, mobile phase composed of 0.01M oxalic acid : methanol : acetonitrile (820 : 80 : 100, v/v/v), isocratic pump at a flow rate of 0.9 ml/min. and $50{\mu}l$ of injection volume, UV/Visible detector with wavelength of 360nm. The calibration curves of four tetracyclines showed linearity $(\gamma^2>0.999)$ at concentration range of $100\~1,000 ng/ml$. The recoveries in fortified honey represented more than $70\%$ with low coefficient of variation $(<10\%)$ for concentration range of four tetracyclines. The detection limits for oxytetracycline, tetracycline, chlortetracycline and doxycycline were 13.8, 14.6, 26.2 and 24.9ng/g in acacia honey. respectively. We also monitored tetracyclines residue in domestic honey [n : 38, acacia (20), wild flower (18) ] and foreign honey [n=22, legally distributed (13), illegally distributed (9)] using modified Charm II screening and HPLC confirmation methods. Seven of the 60 samples $(11.7\%)$ were suspect positive using modified Charm II screening test. Chlortetracycline residue was found in one foreign honey (illegally distributed) tested at concentrations of 0.22 ppm. Conclusively, for more effective control of tetracyclines used in beekeeping should be further survey for residues in honey and also national guidelines (maximum residue limit : MRL) and methods should be obligatory.
