Plant regeneration from protoplasts-derived from embryogenic callus of Citrus (감귤 embryogenic callus 원형질체 배양에 의한 식물체 재분화)

  • This study describes conditions for plant regeneration from protoplasts-derived from embryogenic callus of satsuma mandarin. Plants were generated via somatic embryogenesis. Protoplasts isolated directly from nucellar callus induced from immature ovule of satsuma mandarin cv. Okitsu (Citrus unshiu Marc.) were cultured in 0.6M $BH_3$ medium. Cell division and plating efficiency were affected by protoplast culture method. The liquid over solid method was the most effective for formation of microcalli. Most of microcalli grew rapidly and transferred onto embryoid formation medium. Optimum embryoid formation medium was MT medium containing 1.5 g/L malt extract, 0.146 M sucrose and the medium for plantlet regeneration was MS medium containing 0.09M sucrose, 1.0 mg/L $GA_3$. No differences were noticed in growth habits and leaf characters such as shape, thickness, and colour between protoplast-derived plants and nucellar seedlings. This plant regeneration system from protoplasts-derived from embryogenic callus provides an alternative way for producing new scion and rootstock cultivar from citrus species which can not be crossed.

Effect of Humidity on the Storage Life of Satsuma Mandarin (저장습도가 온주밀감의 저장에 미치는 영향)

  • The storage effects of satsuma mandarin(Citrus unshiu Marc. var. miyagawa) by humidity control during storage; 90% relative humidity (RH) and 85% RH at $3^{circ}C$, and room temperature were investigated. After 98 days' storage, weight losses were 3.40% for 90% RH, 6.92% for 85% RH, and 11.87% for room temperature storage. Decay ratio was increased rapidly from 3.87% on 98 days' to 48.75% on 126 days' storage for 90% RH. Soluble solids and flesh ratio were declined gradually, but the differences were not significantly. Firmness of fruits was continuously reduced during storage, especially on room temperature storage by the softening of the fruits. Acid content and vitamin C were gradually reduced during storage. Coloration was continuously progressed on room temperature, compared to cold storage. In order to keep freshness of the fruits, optimum storage period of early variety of Satuma mandarin was regarded for 100 days at $3^{circ}C$, 85% RH on the basis of sensory evaluation and chemical compositions.

Changes in Pectin of Satsuma Mandarin during Ripening and Storage (온주밀감의 성숙과 저장 중 펙틴 함량의 변화)

  • Changes in firmness and pectin contents during maturation and sotrage of Stsuma mandarin (Citrus Unshiu Marc. var. miyagawa) were investigated. Firmness of fruits was decreased quickly from 1,176.8g-force to 503.6g-dorce. Satsuma mandarin was stored at 3$^{\circ}C$ and 85%${\pm}$5% relative humidity. Firmness were decreased from 538.9g-force to 336.9g-force during storage. Alcohol-insoluble solid(AIS) of peel and flesh were decreased quickly from 27.04g/100g to 12.30g/100g, and from 2.67g/100g to 1.91g/100g during maturation of fruits. During storage of fruits, AIS of peel was decreased from 14.32g/100g to 12.06g/100g . During maturation of fruits, water soluble pectin (WSP) of peel were increased from 420.82mg/100g to 601.62mg/100g as wet basis. Hexametaphosphate soluble pectin (HMP) was also increased from 450.17mg/100g to 577/53mg/100g. hydrochloricacid soluble pectin (HSP) was decreased from 1938.30 mg/100g to 695.14mg/100g. During storage of fruits, HMP and HSP of peel was decreased from 507.82mg/100g to 695.14mg/100g. During storage of fruits, HMP and HSP of peel was decreased from 527.82mg/100g to 275.47mg/100g , and from 672.28mg/100g to 351.36mg/100g, respectively. WSP of peel was increased from 543.70mg/100g to 584.31mg/100g. Total pectin substance (TPS) of peel was decreased from 2809.79 mg/100g to 1874.29mg/100g during maturation, and from 1723.80mg/100g to 1211.14mg/100g during storage, respectively. Composition ratio of pectin was in order of WSP>HSP>HMP.

Quantification of Environmental Characteristics on Citrus Production Area of Jeju Island in Korea (제주도 감귤 재배지역에 대한 환경특성의 정량화)

  • To analyze quantitatively environmental characteristics of cultivation area of citrus, Satsuma mandarin (Citrus unshiu Marc.), we made digital maps of environmental elements such as topography and climate. Elevation, degree of slope, and slope aspect were selected as elements of topological environment, and the annual mean air temperature, annual total precipitation, mean air temperature on January, extreme value of daily minimum air temperature, and the number of days below $-5^{\circ}C$ were selected as elements of climatic environments. The grid values of 8 environmental elements were extracted by shape of citrus farm area and analyzed distribution patterns. We can determine 3 agroclimatic criteria for growing Satsuma mandarin as over $14.5^{\circ}C$ of annual mean air temperature, over $-10.0^{\circ}C$ of extreme value of daily minimum air temperature, and less 5 days of below $-5^{\circ}C$ of daily minimum air temperature.

Physiochemical Characteristics and Sensory Evaluation of Greenhouse Satsuma Mandarin (시설온주밀감의 이화학적 특성과 관능평가)

  • To assess the physiochemical characteristics of greenhouse Satsuma mandarin (Citrus unshiu Marc. var, miyakawa) produced in Cheju, were analyzed chemical compositions for fruits, external and internal factors influencing the edible quality. Changes in organoleptic value according to brix and acid content were also evaluated. The ratios of sucrose : glucose : fructose in citrus juice were 2 : 2 : 1.5. Citric acid as the main acid in the juice represented about 70% of total organic acid. Most of the amino acids were found to be nonessential amino acids. Deep yellow color of the citrus peel showed a significant relationship(r=0.563) with brix/acid ratio of the citrus juice, indicating the ripeness of the fruit. Since the acid content showed highly negative statistical relationship(r= -0.882) with the pH value of the juice, the pH value appears to be a simple indicator for the sourness of the fruit in the field test. The fruits were divided into 9 groups based on the brix/acid contents for sensory evaluation. The palatability patterns of each group changed depending on the acid content. The optimal ranges of brix/acid content for acceptable taste were 11 and $0.5{\sim}1.0$, respectively.

Effect of Humidity and Gas Composition on Fruit Quality during Heat Treatment of Satsuma Mandarin ($Citrus$ $unshiu$ Marc.) (온주밀감의 산함량 감소를 위한 열처리 시 습도 및 가스조성의 차이가 과실품질에 미치는 영향)

  • This study was conducted to determine the effects of the humidity and gas composition on the quality of satsuma mandarins during their heat treatment, in an effort to reduce their acidity. To come up with different humidity and gas conditions, various plastic films were used. The fruits were wrapped with an 18-${\mu}m$ high-density polyethylene (HDPE) punched film, a 30-${\mu}m$ low-density polyethylene (LDPE) film, or a 100-${\mu}m$ LDPE film just before treatment at $30^{\circ}C$ for 55 hours. After heat treatment, the titratible acidity (TA) was significantly reduced while the soluble-solid content (SSC) showed no differences, which resulted in increased SSC/TA ratios in all the treatments. The fruits that were not wrapped with a film, however, which had low RH, developed higher ethanol and acetaldehyde contents than those wrapped with an 18-${\mu}m$ HDPE punched film, which had high RH and a similar gas composition. Among the films, the higher the $CO_2$ concentration in the film was, the more the ethanol and acetaldehyde contents increased, which induced more off-flavor. It was thus concluded that high humidity and normal gas composition are favorable conditions for heat treatment to reduce the acidity of satsuma mandarins.

Stress Effects on Photosynthesis of Greenhouse Plants as Measured by the Fluorescence Method (형광분석법에 의한 시설재배작물의 광합성 Stress 측정)

  • To assess the effects of plant stress by light, temperature, NaCl and soil moisture on photosynthetic activity, the fluorescence method was used for Chinese cabbage(Brassica perkinensis Rupr.), strawberry(Fragaria grandiflora Ehrh.) and citrus tree(Citrus unshiu Marc.). With decreasing the light intensity, Fv/Fm ratios of intact leaves of Chinese cabbage and strawberry increased significantly, indicating lower photochemical efficiency in PS II system, resulting in an inverse relationship with the photosynthetic activity. Chinese cabbage and strawberry that were grown at higher temperature had higher Fv/Fm ratios and photosynthetic activities, while those given high concentration of NaCl and having low soil moisture had lower values. Chinese cabbage more resistant to salt stress and requiring more water had a greater Fv/Fm ratio than strawberry. In citrus tree, Fv/Fm ratio was lower in the non-irrigated group than the irrigated one. From these results, the fluorescence method was found to be a useful tool which can be used to assess the degree of in vivo stress induced by various environmental factors.

Effects on the Storage Life of Satsuma Mandarin by the Pretreatment at various Temperatures (저장전 온도처리가 온주밀감의 저장에 미치는 영향)

  • The storage life of satsuma mandarin(Citrus unshiu Marc. var. miyagawa) by the various pretreatment of temperatures; non-treated, room temperature, $10,\;20^{\circ}C$ and $35^{\circ}C$ were investigated. The pretreated citrus fruits were stored at $4^{\circ}C$, 85% relative humidity. Weight loss of citrus fruits by the pretreatment at $35^{\circ}C$ for 24 hrs was the lowest among that of others. Decay ratio of $35^{\circ}C$ pretreated fruits was increased at initial stages of storage, but was maintained low level after that, compared to other treatments. After 115 days storage, firmness of fruits was lowered by the softening, and decayed fruits were occurred increasingly. Ethylene evolution was increased between $55{\sim}65$ days after storage, and the amount was increasing rapidly after 115 days. It seemed to be derived from decayed fruits and physiological activities. $CO_2$ content in fruit was decreased at initial stages of storage, but was increased between $55{\sim}100$ days during storage periods. Acid content, soluble solids, total sugar and vitamin C were reduced gradually during cold storage, but the difference among treatments was not so great. Pretreated fruits at $35^{\circ}C$ for 24 hrs before cold storage was effective on preventing from weight loss and respiration ratio. Optimum storage period of early variety of Satuma mandarin was regarded for 100 days on the basis of appeareance and taste.

Ethephon Mixed with Calcium Carbonate Accelerate Coloration of Satsuma Mandarin (Citrus unshiu Marc.) in the Plastic House (에스렐과 탄산칼슘에 의한 하우스 밀감의 착색 촉진)

  • This experiment was conducted to study the effect of ethephon(100mg.L$^{-1}$ ) and calcium carbonate (10000mg.L$^{-1}$ ) on the acceleration of peel coloration of ‘Miyagawa Early’ Satsuma Mandarin in the plastic house. The foliar application treatments were conducted once. twice or 3 time at intervals of 15 days from degreening of the fruit apex. Peel coloration of a value by spraying ethephon + clef-non was increased with treatment frequency. The a/b values ran similar to this trend. As a result of this, peel coloration was accelerated by foliar application of ethephon + clef-non. Glucose and fructose did not differ significantly among foliar application frequencies. Sucrose and total sugar increased as foliar application frequency increased, but there was no significant di(ference among foliar application frequency group. The brix values were higher than 12˚BX in all treatments. However, a distinct difference between the control Brix value and the values for the treatment groups was found only in the level 3 frequency group.

Effects of Ethychlozate on the Fruit Quality of Satsuma Mandarin(Citrus unshiu Marc. cv. Miyagawa Early) in the Plastic House (Ethychlozate 살포가 하우스 밀감의 품질에 미치는 영향)

  • This experiment was carried out to elucidate the effect of ethychlozate foliar application on the fruit quality of Miyagawa Early Satsuma Mandarin in the plastic house. Ethychlozate(100mg.L$^{-1}$ ) was applied to the fruit at 100, 115 and 145 days after full bloom stage. The fruit was divided into four frequency level groups, with the control group receiving no Ethychlozate treatment and the level 3 group receiving all three applications. Peel coloration by a value Increased as application frequency increased. The a/b value trend was seen to be similar to this, but the b value ran contrary to the a value trend. Glucose contents of the fruit juice did not differ statistically among the application frequency levels but the fructose level was seen to be higher with increasing application frequency. Sucrose showed a similar trend to fructose in that it had a tendency to increase as application frequency increased. As a result of this, total sugar increased naturally with application frequency. The average Brix level among all treatment groups was higher than the average Brix level in the control group but Brix level did not differ significantly among application frequency levels. Acidity had a tendency to increase as application frequency increase, but there was no statistical difference found.
