Antioxidant Activity of Extracts from Dangyuja (Citrus grandis Osbeck) Fruits Produced in Jeju Island

  • Crude extract of young dangyuja (Citrus grandis Osbeck) fruit was investigated for its antioxidant activity as measured by 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity. Among the samples, including 4 Citrus species and various solvent-extracted-fractions of young dangyuja fruit, the water-extracted fraction (WF) and butanol-extracted fraction (BF) showed the greatest DPPH free radical scavenging activity. WF and BF were further examined for their antioxidant activities by three different in vitro assays. The cell viability tests using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) reduction assay showed that both fractions significantly reduced $H_2O_2$-induced cytotoxicity in HepG2 cells dose-dependently. Generation of the reactive oxygen species (ROS) was also reduced in cells pretreated with both fractions. In addition, BF showed a higher level of lipid peroxidation inhibitory capacity than WF in $H_2O_2$-treated HepG2 cells. Taken together, these results indicate that young dangyuja fruit can be used as an easily accessible source of natural antioxidants.

  • The antioxidant activities of the extracts of dangyuja (Citrus grandis Osbeck) leaves were evaluated. The highest phenolic content was obtained from the ethyl acetate fraction (EF) (202.1$\pm$0.8 mg GAE/g dried extract) and it exhibited the highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity. The cytoprotective effects of EF on oxidative damage induced by tert-butyl hydroperoxide (t-BHP) in a human hepatoma cell line, HepG2 cells, were investigated to understand the intracellular antioxidant mechanisms. Treatment of HepG2 cells with EF prior to oxidative stress was found to inhibit reactive oxygen species (ROS) generation, lipid peroxidation, and DNA damage in a dose-dependent manner. Gas chromatography-mass spectrometry (GC-MS) studies on EF resulted in tentative identification of 19 compounds representing 94.3% of the total content. Taken together, these results demonstrated that EF has excellent antioxidant activities and thus dangyuja leaves have great potential as a source for natural antioxidant which can be applied in food products.

Anti-inflammatory Effect of Unripe Fruit of Citrus grandis Osbeck in RAW 264.7 and HaCaT Cells (RAW 264.7 및 HaCaT Cell에서 당유자 미숙과의 염증억제 효과)

  • We investigated the anti-inflammatory activities of unripe fruit of Citrus grandis Osbeck growing at Jeju Island, through the evaluation of their inhibitory effect on the production of inflammatory markers (IL-6, iNOS, COX, TARC and MDC) in RAW264.7 murine macrophage cells and HaCaT human keratinocyte cells. Among the sequential solvent fractions obtained from crude extract, hexane and chloroform $(CHCI_3)$ fractions showed potential inhibitory activity on the mRNA expressions of IL-6, iNOS and COX-2 at the concentration of $100\;{\mu}g/ml$ in RAW264.7 cells. Also, EtOAc fraction showed inhibitory activity on the thymus and activation-regulated chemokine (TARC)/CCL17 and macrophage-derived chemokine (MDC)/CCL22 at the concentration of $50\;{\mu}g/ml$ in HaCaT cells. These results suggest that the unripe fruit of C. grandis may have anti-inflammatory activity through the suppression of inflammatory markers (IL-6, iNOS, COX, TARC and MDC).

  • Dangyuja (Citrus grandis Osbeck) is a native plant growing only on Jeju Island in Korea. In this study, antiinflammatory effect of dangyuja leaves on a murine macrophage cell line was investigated. RAW 264.7 murine macrophage cells were stimulated with lipopolysaccharide (LPS, $1{\mu}g/mL$) to induce expression of pro-inflammatory markers [interleukin (IL)-6 and inducible nitric oxide synthase (iNOS)]. The crude extract (80% MeOH Ex.) and solvent fractions (hexane, $CHCl_3$, EtOAc, BuOH, and $H_2O$ Ex.) were obtained from dangyuja leaves. The $CHCl_3$ fraction inhibited the nitric oxide (NO) and IL-6 production in a dose-dependent manner. Also, the $CHCl_3$ fraction inhibited mRNA expression and protein levels of iNOS in a dose-dependent manner. Furthermore, the $CHCl_3$ fraction inhibited LPS-induced nuclear factor (NF)-${\kappa}B$ activation and phosphorylation of mitogen-activated protein kinases (MAPKs: ERK, JNK, and p38). These results suggest that dangyuja leaves may inhibit LPS-induced production of inflammatory markers by blocking NF-${\kappa}B$ and MAPKs signaling in RAW 264.7 cells.

Flavonoid Component Changes and Antioxidant Activities of Fermented Citrus grandis Osbeck Peel (당유자 과피 발효물의 플라보노이드 성분 변화 및 항산화 활성)

  • In this study, we investigated the change of antioxidant activity and flavonoid contents by fermentation of Citrus grandis Osbeck peel (CGP) using the Saccharomyces cerevisiae (KCCM35053), comparing to unfermented CGP. Total flavonoid content in the fermented Citrus grandis Osbeck peel (FCGP) was 3,768 g/100 g sample and higher than that of CGP. The antioxidant activities of FCGP was determined by DPPH, hydroxyl, alkyl radicals, and hydrogen peroxide scavenging assays. FCGP showed higher activities than CGP in all scavenging assays. The $IC_{50}$ values of FCGP were 261.3 ${\mu}g$/mL for DPPH; 1,474 ${\mu}g$/mL for hydroxyl; 90.9 ${\mu}g$/mL for alkyl and 1,195 ${\mu}g$/mL for $H_2O_2$ in respective scavenging assays. Flavonoid compositions of both samples were determined by liquid chromatography/mass spectrometry (LC/MS). In the spectrum FCGP was similar to CGP in the contents of neohesperidin, naringin and an unknown No. 7 compound, but some unknown compounds (No. 1, 2, 4, 5, 6) were higher than CGP in each flavonoid contents. Therefore, the fermentation of CGP could increase the contents of unknown compound and improved antioxidant activities.

Physiological activity of methanol extract and fractions from Citrus grandis Osbeck pericarp (당유자 과피 메탄올 추출물 및 분획물의 생리활성 검정)

  • Citrus grandis Osbeck pericarp is used as tea, herbal medicine, etc., but is not commercialized in various ways. So, in this study, we identified potential for use of Citrus grandis Osbeck as health functional foods, cosmetics and food preservatives. Methanol extract of Citrus grandis Osbeck pericarp was fractionated with hexane, dichloromethane, ethyl acetate and butanol, to quantitatively analyze total phenol and flavonoid, and investigate antioxidative and enzyme inhibitory activities. Total phenol and flavonoid contents were highest in ethyl acetate fraction, FRAP and ORAC results also revealed highest activity in proportion to total phenol content. DPPH radical scavenging activity revealed that ethyl acetate, butanol and dichloromethane fraction manifested highest activity without significant difference. However, dichloromethane fraction revealed higher TEAC value and tyrosinase inhibitory activity than ethyl acetate fraction, and hexane fraction manifested best results with superoxide radical scavenging activity and ${\alpha}-glucosidase$ inhibitory activity. Antimicrobial activity also revealed best effect in dichloromethane and hexane fractions. So, based on the following results, use of dichloromethane fraction as material of natural functional cosmetics, ethyl acetate fraction for health functional foods, and hexane fractions for pharmaceuticals and food preservatives, would be most practical for product development.

Evaluation of biological activity for Dangyuja (Citrus grandis) leaves and investigation of optimal concentrations extracted by alternative ethanol concentrations (에탄올 농도별 당유자 잎의 최적추출조건 및 생리활성 평가)

  • TheCitrus grandis Osbeck is a special product in the Jeju island. The product has been as a remedy for liver damage and hang over. This study demonstrates how to investigate and compare the antioxidant, phenol content, tyrosinase and ${\alpha}$-glucosidase inhibitory activity, antimicrobial, and alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activity with the C. grandis leaves extracted in different ethanol concentrations. From the yield, a 20% ethanol extract demonstrated the highest results among the other extracts. The distilled water extract showed the most abundant in a total phenol content and highest ABTS radical scavenging activity and reducing power assay. In the DPPH radical scavenging activity, ${\alpha}$-glucosidase and tyrosinase inhibitory assay (used ${\text\tiny{L}}$-tyrosine as substrate), the 80% ethanol extract exhibited a higher value than other extracts. The 60% ethanol extract showed prominent activities in the tyrosinase inhibitory (used ${\text\tiny{L}}$-dopa as substrate), ADH and ALDH activity assay. In the minimum inhibitory concentration (MIC) assay, 60% and 80% ethanol extracts inhibited the bacterial growth almost similarly. Moreover, the gram-positive bacteria was more restrained than the gram-negative bacteria. The resultsrevealed that the distilled water and 80% ethanol extract showed a relatively higher antioxidant activity compared to other extracts. The 60 ~ 80% ethanol extracts demonstrated potential tyrosinase, ${\alpha}$-glucosidase inhibitory, antimicrobial, ADH and ALDH activities. Therefore, the C. grandis is suggested to be considered as a functional material for various proposes.

Induction of Apoptosis by Citrus grandis Osbeck Peel (CGP) Extract in HL60 Cells (당유자 과피 추출물에 의한 HL60 세포의 Apoptosis 유도)

  • In the present study, we investigated the anti-proliferation activity of Citrus grandis Osbeck peel (CGP) in HL60 (human promyelocytic leukemia) cells. It was found that 80% ethanol extract of CGP could inhibit the cell growth in a dose-dependent manner ($250{\sim}1,000{\mu}g/mL$), which was associated with morphological changes and apoptotic cell death such as depolarized mitochondrial membrane, formation of apoptotic bodies and increased populations of apoptotic sub-G1 phase. The results indicate that CGP extract inhibits the growth of HL60 cancer cells by the induction of apoptosis, which may be mediated by its ability to change the Bcl family proteins and increase the activation of caspase-3 and PARP. Therefore, it is suggested that CGP has the potential to provide a remarkable natural defense against the proliferation of HL60 cells.

Antioxidant Activity of Ethanol Extract from Different Parts of 5 Types of Imported Citrus (Citrus limon, Citrus paradisi Macfad, Citrus. grandis Osbeck × Citrus.   • 수입귤 6종(레몬, 자몽, 청자몽, 오렌지, 만다린, 라임) 부위별 에탄올 추출물의 항산화 활성에 관하여 총 폴리페놀 함량 측정, 총 플라보노이드 함량 측정, DPPH radical 소거 활성 실험을 실시하였다. 수입귤 6종(레몬, 자몽, 청자몽, 오렌지, 만다린, 라임)의 각 부위별 생리활성 실험에서 총 폴리페놀 함량(mg/g)은 청자몽 중피(252.14mg/g)와 청자몽 외피(224.60mg/g), 청자몽 과피(213.13mg/g), 자몽 중피(197.13mg/g) 순으로 모두 150mg/g 이상의 양호한 폴리페놀 함량을 나타내었다. 수입귤 6종(레몬, 자몽, 청자몽, 오렌지, 만다린, 라임)의 각 부위별 총 플라보노이드 함량(mg/g)은 자몽 중피(147.24mg/g)와 청자몽 중피(147.08mg/g), 청자몽 과피(127.74mg/g), 청자몽 외피(122.5mg/g) 순으로 모두 120mg/g 이상의 매우 양호한 플라보노이드 함량을 나타내었다. 수입귤 6종(레몬, 자몽, 청자몽, 오렌지, 만다린, 라임)의 각 부위별 항산화 활성(EDA, %)은 레몬 과피(83.21%)를 시작으로 라임 전체(80.23%), 레몬 과육(77.02%), 레몬 전체(69.85%), 만다린 과피(68.86%), 라임 과육(64.61%) 순으로 EDA 50% 이상 값을 나타내어 양호한 항산화 활성을 보였다.
