  • 암세포가 특정 항암제에 의해 내성을 획득하면 구조가 상이한 타 항암제에도 교차내성을 나타내는 이른바 암세포의 다약제 내성이 암 화학요법에 있어서 가장 심각한 문제가 되고 있다. 본 연구에서는 다약제 내성 암세포주인 CL02 세포를 이용하여 cellulose 용해성 점액세균인 Sorangium cellulosum의 60여종의 균주를 대상으로 다약재 내성 암세포에 유효한 항암물질을 탐색하는 과정에서, 균주 JW1006의 대사산물에서 강한 증식억제 활성을 발견하고 그 활성 본체로서 macrolide계 화합물인 Disorazoles $A_1$$A_2$를 분리하였다 Disorazoles $A_1$$A_2$는 인체기원의 암세포에 대해 모두 강한 세포독성($IC_{50}$ <0.04 ng/$m\ell$)을 나타낼 뿐 아니라 다약제내성 세포주인 CL02와 cisplatin내성 세포주인 CP70에 대해서 감수성 세포주와 동일한 활성을 나타내어 다약제 내성을 극복하는 우수한 활성 물질임을 확인하였다

Luteolin Sensitizes Two Oxaliplatin-Resistant Colorectal Cancer Cell Lines to Chemotherapeutic Drugs Via Inhibition of the Nrf2 Pathway

  • Oxaliplatin is a first-line therapy for colorectal cancer, but cancer cell resistance to the drug compromises its efficacy. To explore mechanisms of drug resistance, we treated colorectal cancer cells (HCT116 and SW620) long-term with oxaliplatin and established stable oxaliplatin-resistant lines (HCT116-OX and SW620-OX). Compared with parental cell lines, $IC_{50}$s for various chemotherapeutic agents (oxaliplatin, cisplatin and doxorubicin) were increased in oxaliplatin-resistant cell lines and this was accompanied by activation of nuclear factor erythroid-2 p45-related factor 2 (Nrf2) and NADPH quinone oxidoreductase 1 (NQO1). Furthermore, luteolin inhibited the Nrf2 pathway in oxaliplatin-resistant cell lines in a dose-dependent manner. Luteolin also inhibited Nrf2 target gene [NQO1, heme oxygenase-1 (HO-1) and $GST{\alpha}1/2$] expression and decreased reduced glutathione in wild type mouse small intestinal cells. There was no apparent effect in Nrf2-/- mice. Luteolin combined with other chemotherapeutics had greater anti-cancer activity in resistant cell lines (combined index values below 1), indicating a synergistic effect. Therefore, adaptive activation of Nrf2 may contribute to the development of acquired drug-resistance and luteolin could restore sensitivity of oxaliplatin-resistant cell lines to chemotherapeutic drugs. Inhibition of the Nrf2 pathway may be the mechanism for this restored therapeutic response.

  • 암세포가 특정 항암제에 의해 내성을 획득하면 구조가 상이한 타 항암제에도 교차내성을 나타내는 이른바 암세포의 다약제 내성(MDR)이 암 화학요법에 있어서 가장 심각한 문제가 되고 있다. 본 연구에서는 다약제 내성 암세포주인 CL02 세포를 이용하여 점액세균의 대사산물을 대상으로 다약재 내성 암세포에 유효한 항암물질을 탐색하는 과정에서, Myxococcus stipitatus로 동정된 JW117 균주의 대사산물에서 우수한 활성을 발견하고 그 활성본체로서 polyene계 화합물인 phenalamides $A_1$, $A_2$, $A_3$를 분리하였다. Phenalamides $A_1$, $A_2$, $A_3$는 인체기원의 암세포에 대해 모두 강한 세포독성($IC_{50}$/ : 0.23~0.57 $\mu\textrm{g}$/ml)을 나타낼 뿐 아니라 다약제내성 세포주인 CL02와 cisplatin내성 세포주인 CP70에 대해서도 감수성 세포주와 동일한 활성을 나타내어 다약제 내성을 극복하는 우수한 활성 물질임을 확인하였다.

Multidrug Resistance Reversal Activity of Methanol Extracts from Basidiomycete Mushrooms in Cancer Cells

  • Mushrooms have a long history of use in traditional medicine, and hundreds of novel constituents in mushrooms with miraculous biological properties have been identified recently. Although diverse effects for medicinal use of mushrooms such as anticancer activity are proven, their reversal activities of drug resistance in cancer cells was rarely reported so far. In the search for novel medicinal use of mushrooms, we tested the multidrug resistance (MDR) reversal activities of diverse mushrooms collected from Korea. Among, the mushroom extracts tested, Cantharellus cibarius (M02) and Russula emetica (M12) revealed MDR reversal activities of paclitaxel in the P-glycoprotein (Pgp)-positive HCT15 and MES-SA/dX5 cancer cells, but not in the Pgp-negative A549 and MES-SA cancer cells. In addition, these mushrooms also enhanced the cytotoxicity of doxorubicin, another well-kwown Pgp-associated anticancer drug against MES-SA/DX5 cells, but not against MES-SA cells. Meanwhile, the cytotoxicity of cisplatin, a well-known Pgp-non-associated anticancer drug, was not affected by the mushrooms all the cells tested. From these results, we suspected that some ingredients of M02 and M12 have Pgp-associated MDR reversal activities.

Hypoxia-Inducible Factor 1 Promoter-Induced JAB1 Overexpression Enhances Chemotherapeutic Sensitivity of Lung Cancer Cell Line A549 in an Anoxic Environment

  • The presence of lung cancer cells in anoxic zones is a key cause od chemotherapeutic resistance. Thus, it is necessary to enhance the sensitivity of such lung cancer cells. However, loss of efficient gene therapeutic targeting and inefficient objective gene expression in the anoxic zone in lung cancer are dilemmas. In the present study, a eukaryotic expression plasmid pUC57-HRE-JAB1 driven by a hypoxia response elements promoter was constructed and introduced into lung cancer cell line A549. The cells were then exposed to a chemotherapeutic drug cis-diamminedichloroplatinum (C-DDP). qRT-PCR and western blotting were used to determine the mRNA and protein level and flow cytometry to examine the cell cycle and apoptosis of A549 transfected pUC57-HRE-JAB1. The results showed that JAB1 gene in the A549 was overexpressed after the transfection, cell proliferation being arrested in G1 phase and the apoptosis ratio significantly increased. Importantly, introduction of pUC57-HRE-JAB1 significantly increased the chemotherapeutic sensitivity of A549 in an anoxic environment. In conclusion, JAB1 overexpression might provide a novel strategy to overcome chemotherapeutic resistance in lung cancer.

Sox12 Is a Cancer Stem-Like Cell Marker in Hepatocellular Carcinoma

  • Recent studies on molecular carcinogenesis suggest that the chemo-resistance of some cancers is largely due to presence of cancer stem cells (CSCs), which affect the chemotherapy outcome for hepatocellular carcinoma (HCC). However, currently no consensus on a CSC phenotype in HCC has been obtained. Here, we examined Sox12 as a novel CSC marker in HCC. Sox12+ versus Sox12- cells were purified from HCC cell lines. The Sox12+ cells were compared with Sox12- HCC cells for tumor sphere formation, chemo-resistance, tumor formation after serial adoptive transplantations in nude mice, and the frequency of developing distal metastasis. We found that compared to Sox12- HCC cells, Sox12+ HCC cells generated significantly more tumor spheres in culture, were more chemo-resistant to cisplatin, were detected in circulation more frequently, and formed distal tumor more frequently. Moreover, Sox12 appeared to functionally contribute to the stemness of HCC cells. Thus, we conclude that Sox12 may be a novel marker for enriching CSCs in HCC.

Effects of Parafibromin Expression on the Phenotypes and Relevant Mechanisms in the DLD-1 Colon Carcinoma Cell Line

  • Background: Parafibromin is a protein encoded by the HRPT2 (hyperparathyroidism 2) oncosuppressor gene and its down-regulated expression is involved in pathogenesis of parathyroid, breast, gastric and colorectal carcinomas. This study aimed to clarify the effects of parafibromin expression on the phenotypes and relevant mechanisms of DLD-1 colon carcinoma cells. Methods: DLD-1 cells transfected with a parafibromin-expressing plasmid were subjected to examination of phenotype, including proliferation, differentiation, apoptosis, migration and invasion. Phenotype-related proteins were measured by Western blot. Parafibromin and ki-67 expression was detected by immunohistochemistry on tissue microarrays. Results: The transfectants showed higher proliferation by CCK-8, better differentiation by electron microscopy and ALP activity and more apoptotic resistance to cisplatin by DNA fragmentation than controls. There was no difference in early apoptosis by annexin V, capase-3 activity, migration and invasion between DLD-1 cells and their transfectants. Ectopic parafibromin expression resulted in down-regulated expression of smad4, MEKK, GRP94, GRP78, $GSK3{\beta}$-ser9, and Caspase-9. However, no difference was detectable in caspase-12 and -8 expression. A positive relationship was noted between parafibromin and ki-67 expression in colorectal carcinoma. Conclusions: Parafibromin overexpression could promote cell proliferation, apoptotic resistance, and differentiation of DLD-1 cells.

  • The presence of epidermal growth factor receptor (EGFR ) mutation is a prognostic and predictive marker for EGFR-tyrosine kinase inhibitor (TKI) therapy. However, inevitably, relapse occurs due to the development of acquired resistance, such as T790M mutation. We report a case of repeated responses to EGFR-TKIs in a never-smoked woman with adenocarcinoma. After six cycles of gemcitabine and cisplatin, the patient was treated by gefitinib for 4 months until progression. Following the six cycles of third-line pemetrexed, gefitinib retreatment was initiated and continued with a partial response for 6 months. After progression, she was recruited for an irreversible EGFR inhibitor trial, and the time to progression was 11 months. Although EGFR direct sequencing on the initial diagnostic specimen revealed a wild-type, we performed a rebiopsy from the progressed subcarinal node at the end of the trial. The result of peptide nucleic acid clamping showed L858R/L861Q.

Genetic Variants in the PI3K/PTEN/AKT/mTOR Pathway Predict Platinum-based Chemotherapy Response of Advanced Non-small Cell Lung Cancers in a Chinese Population

  • Objective: The PI3K/PTEN/AKT/mTOR signaling pathway has been implicated in resistance to cisplatin. In the current study, we determined whether common genetic variations in this pathway are associated with platinum-based chemotherapy response and clinical outcome in advanced non-small cell lung cancer (NSCLC) patients. Methods: Seven common single nucleotide polymorphisms (SNPs) in core genes of this pathway were genotyped in 199 patients and analyzed for associations with chemotherapy response, progression-free survival (PFS) and overall survival (OS). Results: Logistic regression analysis revealed an association between AKT1 rs2494752 and response to treatment. Patients carrying heterozygous AG had an increased risk of disease progression after two cycles of platinum-based chemotherapy compared to those with AA genotype (Adjusted odds ratio (OR)=2.18, 95% confidence interval (CI): 1.00-4.77, which remained significant in the stratified analyses). However, log-rank test and cox regression detected no association between these polymorphisms in the PI3K pathway genes and survival in advanced NSCLC patients. Conclusions: Our findings suggest that genetic variants in the PI3K/PTEN/AKT/mTOR pathway may predict platinum-based chemotherapy response in advanced NSCLC patients in a Chinese population.

키토산이 암세포성장에 미치는 효과 (Effects of Chitosan on Anti-tumor Activity in Mice)

  • 본 연구에서는 키토산이 암세포에 미치는 효과를 보기 위하여 암세포에 대한 세포독성효과, 키토산을 기존 항암제와 함께 사용하였을 때의 암세포에 대한 세포독성효과 및 암마우스에 대한 생명연장 효과의 변화를 관찰하여 암 치료 가능성을 조사하고자 하였다. 암세포인 K562세포나 Yac-1세포에 키토산을 단독으로 작용시켰을 때 암세포성장억제효과를, 기존항암제(mitomycin C, cisplatin, 5-fluorouracil)와 복합으로 작용시켰을 때 암세포성장억제효과의 상승효과를 보이고 암 마우스에서 키토산을 기존항암제와 복합으로 투여한 결과 생명연장효과를 보였다. 따라서 이러한 결과들은 앞으로의 추가적인 연구결과들이 있게 되면 임상에서의 암 치료에 키토산의 이용가능성을 시사한다 하겠다.
