Structural Stability of High-Temperature State of Bacteriorhodopsin: A Model of Multi-state Membrane Proteins

  • A state of bacteriorhodopsin at high temperature was studied by various spectral measurements. The stability measurements indicated that the onset temperature of the denaturation was 70$^{\circ}C$ in the dark and 60$^{\circ}C$ under illumination. The reactivity of hydroxylamine with the Schiff's base also significantly increased in the temperature range between 60 and 70$^{\circ}C$. A spectral band at about 470 nm appeared in the temperature range higher than 60$^{\circ}C$. The circular dichroism spectra in the visible region started to change from a bilobed exiton type to a positive band at about 60$^{\circ}C$, suggesting that the two-dimensional configuration of bacteriorhodopsin molecules changed from crystalline to amorphous. All the measurements suggested a new state between 60 and 70$^{\circ}C$ in which bacteriorhodopsin is stable only in the dark.

Stability Enhancement by the Interaction of Diffusion Flames (다수 비예혼합 화염의 안정화 특성)

  • The stability of turbulent nonpremixed interacting flames is investigated in terms of nozzle configuration shapes and kind of fuels. Four nozzle arrangements - cross 5, matrix 8, matrix 9 and circle 8 nozzles - are used in the experiment. There are many parameters affecting flame stability in multi-nozzle flames such as nozzle separation distance, fuel flowrates and nozzle configuration etc. Key factors to enhance blowout limit are the nozzle configuration and the existence of center nozzle. Even nozzle exit velocity equal 204 m/s, flame is not extinguished when there is not a center nozzle and s/d=15.3∼27.6 in matrix-8 and circular-8 configurations. At these conditions, recirculation of burnt gas is related with stability augmentation. Fuel mole fraction measurements using laser induced fluorescence reveal lifted flame base is not located at the stoichiometric contour.

  • Traditional vacuum cleaner adoptsa highly rotating centrifugal impeller for generating suction region at lower pressure. The rotational speed is very high at 35,000 to 40,000 rpm and accessory structure such as a circular brush at the base plate of the cleaning devices is equipped for gathering dusts. Highly rotating impeller is effective for low pressure generation but causes noise problems. Recently, the unwanted noise is removed by installation of air-pump, instead of a centrifugal impeller, and the internal flows of the modified system are estimated in numerical and experimental approach, respectively.

  • A new type of piexoelectric membrane traveling wave motor has been designed and forbricated. The small motor is composed by the stator which is the combination of annular/circular membrane and metal elastic base using as the common electrode at some time and the rotor which is placed on the metal elastic body. Thus the motor structure is simple and easy to fabricate. The material of a piezoelectric membrane is fabricated by sol-gel method or wear-down method. A piezoelectric traveling wave motor has been fabricated with the stator diameter 8mm The total thickness of the stator is 350$\mu$m. Under the alternative excitation voltage 10-12V the revolving speed of the rotor is more than 100RPM.

A new free-living marine nematode species of the genus Phanoderma Bastian, 1865(Enoplida: Phanodermatidae) from the East Sea, Korea

  • A new species of free-living marine nematode is described from intertidal sediments of the East Sea, Korea. Phanoderma koreense sp. nov. is characterized by the presence of well-developed pharyngeal and cephalic capsule, six inner labial sensilla present as minute papillae around with circular groove, long and slender spicules with 4-5 serrated distal end, located at the base of the precloacal supplement, a series of eight to nine stout and short setae on the ventral cloacal region and conico-cylindrical tail with two pairs of blunt setae. In this study, we provide taxonomic descriptions and illustrations of a new species by differential interference contrast microscope and a pictorial key to the valid species of Phanoderma Bastian, 1865. This is the first record of the genus Phanoderma in the East Sea, Korea.

Copolyester of 3-Hydroxybutyrate and 3-Hydroxyvalerate Produced by Methylobacterium sp. GL-10 (Methylobacterium sp. GL-10이 생산하는 3-Hydroxybutyrate와 3-Hydroxyvalerate의 Copolyester)

  • - The further study for the identification of the previously reported pink-pigmented facultative methylotrophic bacterium (PPFM) GL-10 was carried out. The PPFM GL-10 was Gram nagative, rod, and motile by a single polarly inserted flagellum. The colonies were smooth, pink, circular, along with convex with entire margin. The isolate could utilize C1 compounds and a variety of multicarbon substrates as sole carbon and energy source. The isolate was obligately aerobic, and exhibited both catalase and oxidase activities. The deoxyribonucleic acid base composition was 65-67 mol% guanine plus cytosine. The isolate was mostly identical with Methylobacterium extorquens and named Methylobacterium sp. strain GL-10. Methylobacterium GL-10 accumulated a copolyester of 3-hydroxybutyrate and 3-hydroxyvalerate (poly-3HB/3 HV) when grown in nitrogen-free culture media containing sodium propionate as substrate at the second polyester accumulation stage. The composition of copolyester, as determined from $^1h$ NMR spectra, was 23 mol% of 3-hydroxyvalerate (3HV).

  • The drawbead is one of the most important factors in sheet metal forming for automotive parts. So clarifying the friction characteristics between sheets and drawbead is essential to improve the formability of sheet metal. Therefore in this study, drawbead friction test was performed at various bead materials(FC300, HC891, FCD550, HD700, HK600, HK700, SKD11) and surface treatment of beads(Base, induction hardening, Cr plating, ion nitriding, Toyoda diffusion process, TiCN, TiN, CrN). Circular shape bead has been used for the test. The results show that friction and drawing characteristics were mainly influenced by surface treatment.

Dynamic Analysis of Asphalt Concrete Pavement Structure

  • A new solution for the dynamic analysis of as asphalt concrete pavements under moving loads has been developed. The asphalt concrete pavement can be modeled in elastic or viscoelastic medium of multi-layered structure. The subgrade can be modeled as either a rigid base or a semi-infinite halfspace. The loads may be constant or arbitrary circular loads into one direction. The method utilizes the Complex Response Method of transient analysis with a continuum solution in the horizontal direction and a finite-element solution in the vertical direction. This proposed method incorporates such important factors as wave propagation, inertia and damping effects of the medium as well as frequency-dependent asphalt concrete properties. The proposed method has been validted with the full-scale field truck test, which was conducted on instrumented asphalt concrete section on a test track at PACCAR Technical Center in Mount Vernon, Washington. Comparison with field strain data from full-scale pavement tests has shown excellent agreement.   • Anthracnose of statice (Limonium sp. var. Misty Blue and L. sinuatum Mill. cv. Early Blue) occurred up to 20% in vinyl-houses at Buan, Imsil and Namwon from July to November, 1993. All isolates from the anthracnose symptoms were identified as Glomerella cingulata (Stonem.) Spauld. & Schrenk and its anamorph was Colletotrichum gloeosporioides (Pens.) Sacc. Pathogenicity of the isolates was ascertained on the statice host by artificial inoculation. The symptoms on leaves appeared as circular brown spots in early stage and developed to zonate spots. The spot aggregated and diseased leaves were blighted. The symptom also developed on the stems of severely infected plants and the infected branches were blighted. A lots of acervuli developed on the lesions of stems. Acervuli on necrotic lesions were setose, rounded and elongated. Setae variable in length, 1~4 septate, brown, slightly swollen at the base and tapered to the apex on which conidia were occasionally borned. Statice was revealed as the new host of G. cingulata.

Design of NC Controller with personal computer (PC를 이용한 NC 장치의 설계)

  • In this study, we designed a model of NC controller with IBM-PC as a host CPU and intelligent servo controller for 2 axes that can be expaned to 4 axes. OS software was developed with C language at the base of mode selection technic for 9 NC operating modes. Servo controller design was based on the application of interpolator IC(3701) and position controller IC(3702) that permits low cost and high performance. For connection of two systems, parallel I/O communication was implemented. Finally, auto interpolation program test was executed for linear and circular paths resulting 1 LSB accuracy.
