Therapeutic Effect of Total Ginseng Saponin on Skin Wound Healing

  • In this study, we investigated the effects of total ginseng saponin (TGS) on the cutaneous wound healing process using histological analysis. A total of 24 ICR mice, 5-weeks-old, were used for all in vivo experiments. Mice were divided into control and TGS-treated groups and four equidistant 1-cm full-thickness dorsal incisional wounds were created. The wounds were extracted at days 1, 3, 5, and 7 post-injury for histomorphometrical analysis including wound area and contracture measurements, keratinocyte migration rate, and calculation of infiltrating inflammatory cells. The results showed that the wound area was smaller and keratinocyte migration rate was higher in the TGS-treated group than that of the control group from days 3 to 7. Inflammatory cells in the TGS-treated group at days 1 and 3 were reduced compared to the control group. Wound contraction in the TGS-treated group was greater than in the control group on days 3 to 5, and collagen deposition in the TGS-treated group was higher than in the control group during wound healing. The results indicate a beneficial effect of TGS when used to treat skin wounds.

  • Lachnospiraceae bacterium KGMB03038 (=KCTC 15821) belonging to the class Clostridia in phylum Firmicutes, was isolated from a stool sample of a healthy Korean. Herein, we report the complete genome sequence of strain KGMB03038 analyzed using the PacBio Sequel platform. The genome comprises of 3,334,474 bp with G + C content of 47.8%, which includes 3,099 predicted protein-coding genes, 12 ribosomal RNAs, 54 transfer RNAs, and 4 ncRNAs. Genome analysis revealed that strain KGMB03038 possesses a number of genes involved in hydrolysis of carbohydrates, including mono-, di-, and oligo-saccharides, and biosynthesis of various amino acids.

  • Ruminococcus sp. KGMB03662 was isolated from fecal samples obtained from a healthy Korean. The whole-genome sequence of Ruminococcus sp. KGMB03662 was analyzed using the PacBio Sequel platform. The genome comprises a 2,707,502 bp chromosome with a G + C content of 43.09%, 2,484 total genes, 2,367 protein-coding gene, 14 rRNA genes, and 53 tRNA genes. In the draft genome, genes involved in the hydrolysis enzyme, fatty acid biosynthesis, fatty acid metabolite, antibiotic biosynthesis, and antibiotic resistance have been identified. Those genes of KGMB03662 may be related to the regulation of human health and disease.

Arthroscopic Removal of Large Cartilage Fragment in a Dog with Osteochondritis Dissecans of Shoulder Joint

  • An 11-month-old, 19.5 kg, intact male Border collie was referred with intermittent left forelimb lameness to the Gyeongsang Animal Medical Center. The symptom was first discovered about 6 months ago, and it has gotten worse for the last 10 days with non-weight bearing on the left forelimb. During the physical examination, the patient showed painful reaction when the left shoulder was abducted. On radiographic assessment, a radiolucent line and some osteophytes were found in both humeral heads. Based on patient's clinical signs and radiographic findings, osteochondritis dissecans (OCD) was very suspicious. So, we decided to perform an arthroscopic surgery on left shoulder for definitive diagnosis and treatment because the right forelimb revealed no clinical signs. During arthroscopic technique, we found a large OCD flap on the caudo-central area of humeral head, and observed severe synovitis over a wide range on posterior area of the articular capsule. The large OCD flap was removed by a grasping forceps, and many joint mice were removed either. Curettage was performed using a curette on the articular surface until hemorrhage occurred, and articular capsule flushed with a lactated-ringer's solution. The patient was discharged on the same day without any specific abnormal status. Antibiotic, anti-inflammatory and analgesic drugs were administered. Mild lameness on left forelimb was observed in 2 weeks after surgery, but after 4 weeks, the patient showed complete normal gait without any lameness. Although surgical removal of OCD flap with arthroscopic was previously reported, We would like note that a large OCD flap can also be removed by arthroscopic surgery in this report.

  • In recent years, the mesenchymal stem cells (MSC) derived from various tissues have been widely tested for developing cell therapies, tissue repair and transplantation. Although there has been much interest in the immunomodulatory properties of MSC and their immunologic reactions following autologous, allogeneic and xenogenic transplantation of MSC in vivo, up to date, the expression of immunogenic markers, such as class I and II human leukocyte antigens (HLA), after differentiation of human umbilical cord blood (hUCB)-derived MSC has been poorly investigated and require extensive in vitro and in vivo testing. In this experiment, the expression of the HLA-ABC and HLA-DR on hUCB-derived MSC have been tested by immunocytochemical staining. The undifferentiated MSC were moderately stained for HLA-ABC but very weakly for HLA-DR. In order to investigate the inhibitory effect of allogeneic lymphocytes on proliferation of MSC, the MSC were cultured in the presence or absence of peripheral allogeneic lymphocytes stimulated with concanavalin A. The allogeneic lymphocytes did not significantly inhibit MSC proliferation. We conclude that hUCB-MSC expressed moderately class I HLA antigen while almost negatively class II HLA antigen. The MSC have an immunomodulatory effect which can suppress the allogeneic response of lymphocytes. These in vitro data suggest that allogeneic MSC derived from cord blood can be useful candidate for allogeneic cell therapy and transplantation without a major risk of rejection.

A Case Study of Electrical Resistivity and Borehole Imaging Methods for Detecting Underground Cavities and Monitoring Ground Subsidence at Abandoned Underground Mines (폐광산 지역의 공동 탐지 및 지반침하 모니터링을 위한 전기비저항탐사와 시추공영상촬영기법 적용 사례)

  • We employed electrical resistivity and optical borehole imaging methods to identify underground cavities and determine ground subsidence rate at the study area affected by land subsidence due to abandoned underground mines. At the study site 1, the anomalous zones of low resistivity ranging between 100 ohm-meter and 150 ohm-meter were observed and confirmed as an abandoned underground mine by subsequent borehole drilling and optical borehole imaging. Although the electrical resistivity survey was unavailable due to the paved surface of the study site 2, we were able to locate another abandoned underground mine with the collapsed mine shaft based on the distribution of the ore veins and confirmed it with borehole drilling. In addition, we measured vertical displacements of underground features indicating underground subsidence by conducting optical borehole imaging 6 times over a period of 43 days at the study site 2. The displacement magnitude at the deep segment caused by subsidence appeared to be 3 times larger than those at the shallow segment. Similarly, the displacement duration at the deep segment was 4 times longer than those at the shallow segment. Therefore, the combination of electrical resistivity and optical borehole imaging methods can be effectively applicable to detect and monitor ground subsidence caused by underground cavities.

  • An occurrence of inclined heterolithic stratification (IHS) is described from a tidal point bar in a 40-m-deep distributary of the macrotidai, Han River delta, Korea. The channel bank demonstrates a convex-upward profile with intermittent presence or wave-formed scarps and terraces near the low-water level. The vertical succession of IHS is approximately 25 m thick and dips into the channel with angles reaching up to $14^{\circ}C$. The IHS overlies 15 m of trough cross-bedded sand deposited in the channel bottom. Even though the channel as a whole is ebb dominated, the preserved cross bedding is predominantly flood directed because the mutually evasive nature of the ebb and flood currents causes the point bar surface to be flood dominated. The IHS itself consists of inter-stratified fine sand, sandy silt, and silt with an fining-upward textural trend. Seasonal discharge variations of the Han River are not obvious in the deposits, because tile large size, distal location, and energetic tidal environment of the studied channel reduces the impart of river-stage fluctuations.

Effects of Using Carbon Dioxide on Heart Rate and Arterial Blood Pressure during Laparoscopic Ovariohysterectomy in Dogs (개에서 복강경을 이용한 난소자궁절제술시 $CO_2$가스에 의한 기복증이 심박수와 동맥혈압에 미치는 영향)

  • Cardiovascular changes caused by $CO_2$ pneumoperitoneum during laparoscopic ovariohysterectomy (LOVH) were measured in nine healthy mixed breed dogs ($16.7{\pm}4.6kg$). The dogs were premedicated with the combination of atropine, acepromazine, and butorphanol. General anesthesia was induced with propofol and maintained with isoflurane in oxygen. Controlled ventilation maintained partial pressure of end-tidal $CO_2$ between 35-45 mmHg. The $CO_2$ pneumoperitoneum was maintained at a constant pressure of 12 mmHg and the dog was placed in the $15^{\circ}$ Trendelenburg position as LOVH was performed. Dorsal pedal artery was catheterized for measurements of heart rate (HR) and invasive arterial blood pressure (IBP). Prior to the intraperitoneal insufflation, baseline measurements of HR and IBP were made every minute for a total of 10 min. Then, measurements of HR and IBP were made every 5 min following intraperitoneal insufflation and were also made every 5 min following desufflation for a total of 10 min. The $CO_2$ pneumoperitoneum during LOVH resulted in a significant (P < 0.05) increase in systolic arterial blood pressure at the time of the onset of insufflation. In addition, diastolic and mean arterial blood pressure increased significantly (P < 0.05) at the time of the onset of insufflation and 5 min following insufflation. The mean heart rate did not change significantly during LOVH. Although IBP showed sharp initial rise following the $CO_2$ pneumoperitoneum, the changes were within physiological acceptable limits in these healthy, ventilated dogs.

Effects of Intraperitoneal Bupivacaine Instillation on Postoperative Pain Behavior after Laparoscopic Ovariohysterectomy in Dogs (개에서 복강경을 이용한 난소자궁절제술시 복강 내 Bupivacaine의 점적투여가 술 후 통증행동에 미치는 영향)

  • The present study was aimed to investigate the effect of intraperitoneal bupivacaine instillation on postoperative pain after laparoscopic ovariohysterectomy (LOHE) in dogs. Twelve female German shepherd dogs (17-30 kg) were divided into two groups. The treatment group received 4.4 mg/kg of instilled intraperitoneal bupivacaine diluted to 0.25% with an equivalent volume of saline after pneumoperitoneum, but the control group received 1.76 ml/kg of 0.9% saline. Two blind observers measured the extent of dog's pain and sedation by using dynamic interactive visual analogue scale (DIVAS) preoperatively and 0.5, 1, 2, 4, 6, and 12 h postoperatively. At each designated time, blood cortisol, glucose, and creatine kinase (CK) concentrations were also measured. Based on the repeated-measures ANOVA, there were significant differences in time-dependent postoperative changes in patterns of DIVAS-pain score between two groups. In addition, the treatment group had significantly lower DIVAS-pain scores at 1, 2, 4, and 6 h postoperatively compared to the control group. DIVAS-sedation score and biochemical measures including cortisol, glucose, and CK did not show any significant differences between two groups. No complications associated with bupivacaine administration were observed. Thus, instilled bupivacaine intraperitoneally may be an effective method on relieving behavioral expressions associated with postoperative pain after laparoscopic ovariohysterectomy in dogs.

Clinical Application of Mesenchymal Stem Cells in a Dog with Intervertebral Disc Disease (추간판 탈출증이 발생한 개에서 동종의 지방유래 줄기세포의 임상적 적용)

  • A 5-year old, intact male, Cocker spaniel dog was referred with paraplegia and loss of deep pain perception. Physical, neurological examinations, radiography, and computed tomography were evaluated. Based on the clinical examinations, the dog was diagnosed with severe disc herniation ($L_2$ to $L_3$ intervertebral disc space). On the next day of presentation (6 days after loss of deep pain perception), hemilaminectomy was performed. After decompression of spinal cord and removal of herniated disc materials, $1{\times}10^6$ canine allogenic adipose tissue-derived mesenchymal stem cells (MSCs) diluted by $50{\mu}l$ saline were directly applied to the injured site of the spinal cord. Ten weeks of follow-up after surgery, full recovery of deep pain perception and motor function were evaluated in both hind limbs. Based on the result, we suggest that the transplantation of allogenic adipose tissue-derived MSCs to dogs with spinal cord injuries could be a considerable method to expect better clinical outcomes in veterinary practice.
