Cytogenetic Analysis of Bagrid Catfish, Pseudobagrus fulvidraco(Teleostomi : Siluriformes) (동자개, Pseudobagrus fulvidraco(Teleostomi : Siluriformes)의 세포유전학적 연구)

  • The chromosome numbers of bagrid catfish, Pseudobagrus fulvidraco was 52, nine pairs (No. 1 to 9) were metacentrics with the range of relative length 2.89~6.22 and arm ratio 1.09~1.58 ; thirteen pairs (No. 10 to 22) were submetacentrics with the range of relative length 2.88~5.88 and arm ratio 1.80~3.65 ; and all other pairs (No. 23 to 26) were acrocentrics with the range of relative length 2.63~3.30 and arm ratio 9.01~10.67, and fundamental number was 104. Heteromorphic sex chromosomes were not found. There was not exist significant difference in resultant erythrocyte measurements and parameters between female and male (p<0.05). The mean sizes of cell and nucleus, were $11.03{\times}9.67{\mu}m$ and $4.18{\times}3.66{\mu}m$ respectively. The number of erythrocytes of both females and males were $6{\sim}7{\times}10^5/ml$. Gill tissues from diploid individuals had cells with one or two nucleoli.

Pathway Analysis of Metabolic Syndrome Using a Genome-Wide Association Study of Korea Associated Resource (KARE) Cohorts

  • Metabolic syndrome (MetS) is a complex disorder related to insulin resistance, obesity, and inflammation. Genetic and environmental factors also contribute to the development of MetS, and through genome-wide association studies (GWASs), important susceptibility loci have been identified. However, GWASs focus more on individual single-nucleotide polymorphisms (SNPs), explaining only a small portion of genetic heritability. To overcome this limitation, pathway analyses are being applied to GWAS datasets. The aim of this study is to elucidate the biological pathways involved in the pathogenesis of MetS through pathway analysis. Cohort data from the Korea Associated Resource (KARE) was used for analysis, which include 8,842 individuals (age, $52.2{\pm}8.9years$ ; body mass index, $24.6{\pm}3.2kg/m^2$). A total of 312,121 autosomal SNPs were obtained after quality control. Pathway analysis was conducted using Meta-analysis Gene-Set Enrichment of Variant Associations (MAGENTA) to discover the biological pathways associated with MetS. In the discovery phase, SNPs from chromosome 12, including rs11066280, rs2074356, and rs12229654, were associated with MetS (p < $5{\times}10^{-6}$), and rs11066280 satisfied the Bonferroni-corrected cutoff (unadjusted p < $1.38{\times}10^{-7}$, Bonferroni-adjusted p < 0.05). Through pathway analysis, biological pathways, including electron carrier activity, signaling by platelet-derived growth factor (PDGF), the mitogen-activated protein kinase kinase kinase cascade, PDGF binding, peroxisome proliferator-activated receptor (PPAR) signaling, and DNA repair, were associated with MetS. Through pathway analysis of MetS, pathways related with PDGF, mitogen-activated protein kinase, and PPAR signaling, as well as nucleic acid binding, protein secretion, and DNA repair, were identified. Further studies will be needed to clarify the genetic pathogenesis leading to MetS.

Generation and DNA Characterization of High-lysine Mutants by Biochemical Selection from Callus Culture of 'Hwayeongbyeo'

  • Lysine is the first essential amino acid for optimal nutrient quality in rice grain. For the narrow genetic diversities of lysine contents in rice, somaclonal variation was the source of mutation in our breeding program. Biochemical selection was conducted using 1 mM S-(2-aminoethyl) cysteine followed by two passages of 5 mM lysine plus threonine in the callus subculture medium. The lysine contents in endosperm of all progenies recovered from the biochemical selection were higher than those of their donor cultivar 'Hwayeongbyeo'. These elevated lysine levels of mutants were successfully transmitted to $M_4$ generation. The lysine contents in endosperm varied 3.85 to $4.80\%$ compare to their donor cultivar 'Hwayeongbyeo' was $3.85\%$. Three of high-lysine germplasms, Lys-l, Lys-2 and Lys-7 were selected by biochemical selection and rapid screening methods. DNA analysis showed that a new insertion of Tos 17 which mapped to rice chromosome 11 on the high-lysine mutant, Lys-2.

Analysis of deleterious genes in natural and experimental populations of Drosophila melanogaster (Drosophila melanogaster 자연 및 실험집단내의 유해유전자 분석)

  • The genetic variabilities of second chromosomes of Drosophila melanogaster concealed in Busan natural and experimental populations have been analyzed by the Cy//Pm method. The experimental population was composed of D. melanogaster which had the lethal-free second chromosome, collected Sasang natural population in 1982. The frequencies of deleterious genes were estimated to be 14.3∼25.4% in Busan natural population and 26.5∼27.2% in experimental population. The allelism rates in lethal genes isolated from the natural and experimental populations were calculated to be about 0.76% and 9.76∼14.17%, respectively. The value of elimination by the frequencies of deleterious genes and allelism rate was 0.0106and the effective population size estimated to be about 430 flies at the 6570 days population.

Identification and expression of leuD Gene in Rice (Oryza sativa L.) (벼(Oryza sativa L.)의 leuD 유전자)

  • A rice OsLeuD gene for small subunit of 3-isopropylmalate isomerase (IPMI) (EC 4.2.1.33) has been isolated. OsLeuD gene is located on 109.3 cM of chromosome 2. OsLeuD gene was expressed abundantly in metabolically active organs including leaves and developing seeds, indicating that OsLeuD gene expression is developmentally regulated. The cDNA of OsLeuD gene was coded for 257 amino acids which showed 58% and 48% homology to small subunits of IPMI in OsLeuD genes of cyanobacteria and green sulfur bacteria, respectively. The molecular character of OsLeuD is closely related to those of photosynthetic bacteria rather than those of eukaryotes including fungi and yeast. This suggests that OsLeuD gene in chromosomal genome of plants may possibly be originated from chloroplast genome.

Structural and Functional Analysis of a Forkhead Gene, fkhF, in a Filamentous Fungus Aspergillus nidulans (사상성 진균 Aspergillus nidulans에서 forkhead 유전자인 fkhF의 구조와 기능 분석)

  • Genome analysis of a model filamentous fungus, Aspergillus nidulans, revealed that there are six putative forkhead genes. Among them, fkhF (AN8949.2) showed A. nidulans-specific. fkhF gene is located in chromosome VII and composed of 2,337 bp coding region for 778 amino acid. Since little is known about the involvement of the forkhead proteins in the developmental process of the filamentous fungi, including A. nidulans, we generated a deletion mutant of fkhF gene and analyzed. Deletion of fkhF resulted in less-dense conidiophore formation in a solid culture. However, the sexual developmental process or cleistothecia formation was normal. Furthermore, fkhF deletion mutant produced conidiophores and conidia under the submerged culture, suggesting that the fkhF gene is involved in repression of inappropriated induction and maturation of asexual developmental process but not in sexual development.

  • Due to the rapid increase of electricity demand, consideration of environmental constraints in optimal power flow (OPF) problems is increasingly important. In Algeria, up to 90% of electricity is produced by thermal generators (vapor, gas). In order to keep the emission of gaseous pollutants like sulfur dioxide (SO2) and Nitrogen (NO2) under the admissible ecological limits, many conventional and global optimization methods have been proposed to study the trade-off relation between fuel cost and emissions. This paper presents an efficient decomposed Parallel GA to solve the multi-objective environmental/economic dispatch problem. At the decomposed stage the length of the original chromosome is reduced successively and adapted to the topology of the new partition. Two subproblems are proposed: the first subproblem is related to the active power planning to minimize the total fuel cost, and the second subproblem is a reactive power planning design based in practical rules to make fine corrections to the voltage deviation and reactive power violation using a specified number of shunt dynamic compensators named Static Var Compensators (SVC). To validate the robustness of the proposed approach, the algorithm proposed was tested on the Algerian 59-bus network test and compared with conventional methods and with global optimization methods (GA, FGA, and ACO). The results show that the approach proposed can converge to the near solution and obtain a competitive solution at a critical situation and within a reasonable time.

  • A simple stereo matching algorithm using population-based incremental learning(PBIL) is proposed in this paper to decrease the general problem of genetic algorithms, such as memory consumption and inefficiency of search. PBIL is a variation of genetic algorithms using stochastic search and competitive teaming based on a probability vector. The structure of PBIL is simpler than that of other genetic algorithm families, such as serial and parallel ones, due to the use of a probability vector. The PBIL strategy is simplified and adapted for stereo matching circumstances. Thus, gene pool, chromosome crossover, and gene mutation we removed, while the evolution rule, that fitter chromosomes should have higher survival probabilities, is preserved. As a result, memory space is decreased, matching rules are simplified and computation cost is reduced. In addition, a scheme controlling the distance of neighbors for disparity smoothness is inserted to obtain a wide-area consistency of disparities, like a result of coarse-to-fine matchers. Because of this scheme, the proposed algorithm can produce a stable disparity map with a small fixed-size window. Finally, an alterative version of the proposed algorithm without using probability vector is also presented for simpler set-ups.

Genetic Identification on Natural Population of Triploid Crucian Carp, Carassius auraus in Korea (자연산 3배체 붕어 (Carassius auratus) 클론 집단에 대한 유전학적 동정)

  • Natural clonal stock of triploid crucian carp, Carassius aurahs was identified and its cytogenetic, molecular genetic and morphological traits were studied. Cytogenetic analysis of the clonal crucian carp revealed that they were natural triploidy, evidenced by 1.5-fold increases of cell size, DNA content, and chromosome number. Multi-locus DNA fingerprinting using $(GATA)_4$ probe showed that they had an identical fingerprint profile, indicating the clonal propagation of the population. External morphology and morphometric characteristics of triploid individuals were much uniform compared to those of diploids. Natural triploid crucian carp was proven to be all-female in this study.

Expression analysis and characterization of rice oligopeptide transport gene (OsOPT10) that contributes to salt stress tolerance

  • Knock-out of a gene by insertional mutagenesis is a direct way to address its function through the mutant phenotype. Among ca. 15,000 gene-trapped Ds insertion lines of rice, we identified one line from selected sensitive lines in highly salt stress. We conducted gene tagging by TAIL-PCR, and DNA gel blot analysis from salt sensitive mutant. A gene encoding an oligopeptide transporter (OPT family) homologue was disrupted by the insertion of a Ds transposon into the OsOPT10 gene that was located shot arm of chromosome 8. The OsOPT10 gene (NP_001062118.) has 6 exons and encodes a protein (752 aa) containing the OPT family domain. RT-PCR analysis showed that the expression of OsOPT10 gene was rapidly and strongly induced by stresses such as high-salinity (250 mM), osmotic, drought, $100\;{\mu}M$ ABA. The subcellular localization assay indicated that OsOPT10 was localized specifically in the plasma membrane. Overexpression of OsOPT10 in Arabidopsis thaliana and rice conferred tolerance of transgenic plants to salt stress. Further we found expression levels of some stress related genes were inhibited in OsOPT10 transgenic plants. These results suggested that OsOPT10 might play crucial but differential roles in plant responses to various abiotic stresses.
