Molecular Mapping of Resistant Genes to Brown Planthopper, Bphl and bph2, in Rice

  • This study was carried out to map Bphl and bph2 gene in Mudgo and Sangju13 (Oryza sativa L.) respectively conferring resistance to brown plan-thopper (BPH) and to establish the marker-assisted selection (MAS) system. Bulked seedling (grown for 20 days) test was conducted with the 73 F4 lines derived from a cross between Nagdongbyeo and Mudgo for Bphl and with 53 BC3F5 lines derived from the Milyang95/Sangju13 cross for bph2. Bph1 was mapped between RG413 and RG901 on chromo-some 12 at a distance of 7.5 cM from RG413 and 8.4 cM from RG90l. A recessive gene bph2 was located near RZ76 on chromosome 12 at a distance of 14.4 cM. Bphl and bph2 were linked to each other with a distance of about 30 cM. An RFLP marker, RG413 linked to Bphl, was converted to an STS marker to facilitate the marker-assisted selection. BPH resistant genotypes could be selected with 92% accuracy in a population derived from a line of NM47-B-B.

  • A defective lambda transducing phase carrying acetyl CoA carboxylase gene (fabE) from Escherichia coli chromosome (72 min on the current linkage map) has been isolated. A restriction map of the chromosomal region from defective transducing phage was established by digestion with combination of the restriction enzymes. No cleavage site for the enzyme EcoRI was found in this region. Restriction fragments were cloned from defective transducing phage into high copy number plasmid vector pACYC184 to generate hybrid plasmids which were capable of complementation of fabE temperature sensitive mutation. We show here that the fabE gene is located on a 3.4 megadalton Bam HI-SalI fragment with a HindIII site, which lies within the 7.4 megadalton BglIIfragment, by complementation analysis.

Treatment and Survival in Patients with Chronic Myeloid Leukemia in a Chronic Phase in West Iran

  • Background: CML includes 30% of all leukemias, and occurs from childhood to old age. The present study was a retrospective analysis of chronic phase CML patients registered to a Hematology Clinic in Kermanshah, Iran, with checking of treatment options. Materials and Methods: Between 2002 and 2014, 85 CML patients referred to our hematology clinic were enrolled in our study. We surveyed age, sex, B-symptoms, splenomegaly, Sokal score, Hasford score, treatment and survival in all patients. Philadelphia chromosome analysis was conducted for each patient by conventional cytogenetics. We compared treatment in the patients with three drugs, imatinib, hydroxyurea (HU) and interferon alpha (IFN-${\alpha}$). Results: The mean age of the patients at diagnosis was $47.5{\pm}14.5years$ (range, 23-82 years), with 43 (50.6%) being male. Some 13 (15.3%) were referred to our clinic for the first time with B-symptoms and 44 patients (51.8%) had splenomegaly. The Sokal score for 77 (90.6%) was low, 4 (4.7%) was intermediate and 4(4.7%) was high, but Hasford (Euro) scores for all patients were low. The 5-year survival rate for treated patients with imatinib, imatinib plus HU and imatinib plus HU plus IFN-${\alpha}$ was 90.5%, 81.1% and 55.6%, respectively Conclusions: The results show that imatinib therapy alone provides better survival in CML patients compared to HU or IFN-${\alpha}$. Combinations of IFN-${\alpha}$ and/or HU with imatinib probably reduce survival.

Clinico-Hematological Profile and Risk Stratification in Patients with Essential Thrombocythemia: Experience from Pakistan

  • Background: Essential thrombocythemia (ET) is a Philadelphia chromosome-negative myeloproliferative neoplasm characterized by sustained thrombocytosis and megakaryocytic hyperplasia. It is an uncommon hematological malignancy which primarily affects elderly individuals. The rational of this study was to determine its clinico-hematological profile along with risk stratification in Pakistan patients. Materials and Methods: In this retrospective cross sectional study, 21 patients with ET were enrolled from January 2011 to December 2014. Data was analyzed with SPSS version 21. Results: The mean age was $56.7{\pm}19.0years$ (range 18-87) and the male to female ratio was 1:1.1. Of the total, 62% of patients were above 50 years of age. Overall 61.9% were diagnosed incidentally and were asymptomatic. In symptomatic patients, major complaints were weakness (19%); erythromelalgia (14.2%), transit ischemic attack (9.5%) and gastrointestinal bleed (4.7%). The mean hemoglobin count was $11.7{\pm}2.4g/dl$ with a total leukocyte count of $13.3{\pm}8.1{\times}10^9/l$ and platelets count of $1188.8{\pm}522.2{\times}10^9/l$. Serum lactate dehydrogenase, serum creatinine and uric acid were $454.3{\pm}127.8$, $1.2{\pm}0.5$ and $7.4{\pm}3.4$ respectively. According to risk stratification, 57.1% were in high risk; 23.8% in intermediate risk while 19.1% in low risk group. Conclusions: ET in our patients in Pakistan, unlike in the West, is seen in a relatively young population. Primarily patients were asymptomatic and risk stratification revealed predominance of high risk disease in our setting.

  • Bacillus stearothemophilus isolated from soil was identified to express multiple extracellular xylanases. Two HindIII restriction fragments of 5.4 and 6.4 kb from B. stearothermophilus genomic DNA were cloned into pBR322 to obtain recombinant plasmids pMG0l and pMG02, respectively, which enabled E. coli HBlOl cells to produce $\beta$-D-xylosidase activity. By subcloning into pUC18 and Southern blotting, the loci of the $\beta$-D-xyiosidase genes were elucidated to be on non-homologous DNA fragments of 2.2 kb from pMGOl(pMG1) and 1.0 kb from pMG02(pMG2), respectively. The two enzymes produced in E. coli cleaved xylobiose, xylotriose, xylotetrose and xylotetrose to produce xylose as a major end product. The gene on pMG1, distinct from that on pMG2 was observed to encode a bifunctional protein that displayed both P-D-xylosidase (EC.3.2.1.37) and a-L-arabinofuranosidase activities (EC.3.2.1.55).

Studies on the Karyotypes and Comparative DNA Values in Several Korean Cyprinid Fishes (한국산 담수어류(잉어과) 몇종의 핵형과 DNA 상대량에 관한 연구)

  • Karyological characteristics and relative DNA values were investigated in four species of the family Cyprinidae of Korean fresh-water. The results obtained were as follows; 1) The two Hemibarus species, H. longirostris and H. labeo, had the aiploid chromosome number of 50. The compliment of former consisted of 7 metacentric, 14 submets- and subtelocentric and 4 telo- or acrocentric pairs, whereas latter had 9, 11 and 5 pairs respectively, Total arm numbers (AN) were 92 and 90. 2) In the case of two species of genus Moroce, M. oxycephalus and M. lagowskii, their diploid number and karyotype showed in identical pattern for each other (2n=50); 6 pairs of metacentrics, 14 pairs of submeta- and subtelocentrics and 5 pairs of telo- or acrocentrics. The AN of both species were 90. 3) Relative DNA values of all species were measured as about 60% that of gold fish. From the above observation, the cytogenetical character was discussed with special concern to the phylogenetic significance.

Studies on the Cleavage Inhibidng Activity in the Cytoplasm of Growing Follicular Oocytes in Mammals (성장중인 포유동물 여포난자 세포질의 난할억제효과에 관하여)

  • In order to determine whether maturation inhibiting activity(MIA) in the cytoplasm of growing follicular oocytes would suppress the cleavage of the embryonal cells, the growing oocytes were fused with the 2 or 4 cell blastomeres and then examined for the nuclear phase of the fused giant cells 24 hr after culture. A significant number of the giant cells(60%) composed of growing mouse oocyte and 2 cell mouse blastomere(1/2) in interphase has contained 2 nuclei 24 hr after culture and most of the giant cells (90%) composed of the growing oocyte and 4 cell blastomere(1/4) also contained 2 nuclei after culture. The unfused blastomeres or the isolated blastomeres cultured without fusion treatment cleaved one cell cycle under the same culture condition. In contrast, the nucleus of the growing oocytes was disintegrated and the chromosome condensed when fused with 2 cell blastomere in mitosis. The growing rat oocytes also suppressed the nuclear disintegration of the mouse embryonal cells during culture. The data presented here showed that MIA in the growing mammalian oocyte inhibited the cleavage of the embryonal cells in interphase stage, but not in milosis stage.

Mapping quantitative trait loci controlling low-temperature germinability in rice

  • Low-temperature germination is one of the major determinants for stable stand establishment in the rice direct seeding method in temperate regions and at high altitude areas. Quantitative trait loci (QTL) controlling low-temperature germinability in rice were identified using 96 introgression lines (ILs) derived from a cross between Oryza rufipogon and the Korean japonica cultivar, 'Hwaseongbyeo'. The germination rate at $15^{\circ}C$ was measured to represent low-temperature germination and used for QTL analysis. The germination rate at $15^{\circ}C$ for 7 days of Oryza rufipogon and Hwaseongbyeo was 93.3 and 28.7%, respectively, and that of progenies ranged from 0 to 48%. A linkage map was constructed using 135 simple sequence repeat (SSR) markers. Five putative QTLs associated with low-temperature germination were detected on chromosomes 1, 3, 4, 10 and 11. The QTL, qltg10 on chromosome 10 accounted for 19.2% of the total phenotypic variation for low-temperature germinability. Four additional QTL, accounted for 10.4 - 15.1% of the total phenotypic variation. The O. rufipogon alleles in all detected QTLs loci increased the low-temperature germination rate. No QTL associated with low temperature germinability has been detected near the qltg10 QTL in this study suggesting that qltg10 is a new QTL. The locus, qltg10 is of particular interest because of its independence from undesirable height and maturity effects. The DNA markers linked to the QTL for low temperature germinability would be useful in selecting lines with enhanced low temperature germinability in rice breeding program.

Complete genome sequence of Flavivirga eckloniae ECD14T isolated from a seaweed Ecklonia cava (감태(Ecklonia cava)에서 분리한 Flavivirga eckloniae ECD14T의 유전체 서열 분석)

  • The genome of Flavivirga eckloniae $ECD14^T$ isolated from a seaweed Ecklonia cava was sequenced. The genome comprises a single circular 5,665,358 bp chromosome with a G + C content of 33.9%, 4,647 total genes, 4,595 protein-coding genes, 44 pseudo genes, and 52 RNA genes. CRISPER genes and sequences were not found and there were some phage remnants and transposons. This strain contains alginate lyase and ${\beta}$-glucosidase genes responsible for the degradation of seaweed polysaccharides.

Chracteristics of Mulberry Fruits on Daeseongppong, Daebungppong, Daeokppong and Shingwangppong(Morus Spp.) (대성뽕, 대붕뽕, 대옥뽕 및 신광뽕 오디의 과실적 특성)

  • Daeseongppong, Daebungppong, Daeokppong were bred, and Shingwangppong was selected among the recommended mulberry varieties of the grown silkworm, in order to meet the increasing the interest about the mulberry fruit, The major characteristics of the mulberry fruits were also studies. The chromosome number of Daeseongppong, Daebungppong and Daeokppong are 2n=56 recognized autotetraploid, and that of Shingwangppong is 2n=42 as a triploid. Seed forming rate of Daeseongppong, Daebungppong, and Daeokppong are more than 58%, but that of the Shingwangppong is very low with 6.7%. Concerning the single fruit weight, Daeseongppong is 4.05g, Daeokppong 3.38g, Daebungppong 2.99g which belonged to big fruit group. However Shingwangppong is medium as 2.28g. The sugar contents of Shingwangppong, Daeokppong, Daebungppong, Daeseongppong are 14.8%,13.7%,13.1% and 12.7% respectively. The sugar/acidity rate of them are above 21. The fruit maturity of Shingwangppong is early, those of Daebungppong, Daeokppong are medium and that of Daeseongppong is late. Yielding Potentiality of Daebungppong is high, those of Daeseongppong, Shingwangppong are comparatively high, and that of Daeokppong is moderate. In the case of cold hardiness, those of Shingwangppong, Daeokppong, Daebungppong are comparatively good, that of Daeseongppong is weak slightly.
