Toxic Activities of the Oxidant Chromate in Culture Cells (산화성 크롬의 배양세포에서의 독성작용)

  • The ROS-producing potency of chromium compounds of several oxidation states were determined in the H4 cells. $K_2Cr_2O_7$ as Cr (VI), synthetic Cr (V) compounds and Cr (III) as TPP produced high level of ROS. However, ROS values of Cr-picolinate as Cr (III), CrCl$_2$, CrCI$_2$, were almost equal to the control. The effects of physiological antioxidants compounds which react with free radicals were examined for their effects on chromate-induced production of reactive oxygen species (ROS) in A549 cells after the addition of $K_2Cr_2O_7$. The compounds used were vitamin C (ascorbate), vitamin E ($\alpha$-tocopherol), superoxide dismutase (SOD) and catalase. The preincubation of ascorbate (200uM) with A549 cells for 20hr resulted in a significant reduction of hexavalent chromate(100uM) induced ROS. However, there is no effects of preincubation of the cells with vitamin E succinate (10 and 20uM, 20hr) on the ROS production. Also, the effects of Cr (VI) on the cell cycle of A549 cells was measured by adding the DNA intercalating agent, propidium iodide. S phase of the cell cycle was increased by the chromium (VI) compounds up to 20uM indicating toxicity or possible mitogenic action of the cell. The shoulder in Go/G1 phase at 20uM Cr (VI) with 24 hr treatment indicates apoptosis.

Reduction of Hexavalent Chromium by Leachate Microorganisms in a Continuous Suspended Growth Culture (연속배양 체제에서의 침출수 미생물에 의한 6가 크롬이온의 환원)

  • Reduction of hexavalent chromium to its trivalent form by leachate microorganisms was studied in batch and bench-scale continuous stirred tank reactor. The inoculum was a culture of microorganisms in leachate and capable of providing up to 90% chromate reduction during 72 h batch assay with $20mg\;Cr(VI)\;L^{-1}$ in minimal media containing different levels of leachate (10 to 60%) and glucose (50 to 200 mM). Addition of glucose increased the efficiency of chromate reduction, but adverse effect was observed with increase of leachate probably due to the competitive inhibition between chromate and sulfate ions. The continuous culture experiment was conducted for 124 days using synthetic feed containing different levels of chromate (5 to $65mg\;L^{-1}$) at room temperature. With a hydraulic retention time of 36 h, chromate reduction efficiency was mostly 100% when Cr(VI) concentrations in the reactor were in the range of 5 to $50mg\;L^{-1}$ Specific rate of Cr(VI) removal was calculated as $3.492mg\;g^{-1}\;protein\;h^{-1}$ during the period of 101~124 days from the start-up which showed 81.2% of average reduction efficiency. The results indicate the potential application of using leachate microorganisms for detoxification of hexavalent chromium in various chromium-contaminated wastewater from landfill or tannery sites.

Development of the Limit Switch Box for a Ship and Its Performance Evaluation against Salt Water

  • A limit switch box is used for an indicator of a valve actuator. This device indicates an opening and closing of a valve or a throttle in the valve actuator. In a ship, equipments require safety and robustness because of a rough environment and a specific condition during a voyage. However, the limit switch box has been used in an indoor environment generally. This study developed a new limit switch box which can be used at an outdoor environment. This study designed the new limit switch box. The housing of the limit switch box was made by an aluminum die cast method with surface painting after anodizing or chromate coating. In order to evaluate the endurance of the housing, the endurance tests against salt water have been conducted. Experiment results showed that the proposed device provides a reliable performance against salt water.

Insulation Coating for non-oriented Silicon Steel Sheerts (무방향성 전기강판의 절연피막 코팅재)

  • Good appearance of insulation coating is required for non-oriented silicon steel sheets, The property is influenced by both the chemical composition of coating solution and the species of resin. The composition of inorganic-organic coating was studied to obtain good surface quality for non-oriented silicon steel. The greenish degree of coating surface depended on $Cr^{3+}$ content in the coating layer, which was satisfied when chromate content was more than 54.60 wt. %, in the coating solution. The homogeneous pattern and roughness of the coating surface depended on spreading property of the resin. Surface appearance of the coating could be improved by using resin with good spreading property at the chemical composition of chromate 59.00 wt.%.. resin 34.23wt.%, and etyhylene glycol 6.67 wt.% without colloidal sillica.
