콜레스테롤과 비타민 $D_2$ 첨가 사료로 유도된 흰쥐의 고지혈증에 대한 Taurine의 효과 (Effect of Taurine on Hyperlipidemic Rats Fed Cholesterol and Vitamin $D_2$ containing Diet)

  • Antihyperlipidemic effect of taurine was investigated in the hyperlipidemic rats induced by feeding a diet supplemented with cholesterol (1.5% in diet), vitamin $D_2$(1.25 million IU/kg of diet) and cholic acid (0.5% in diet). The rats were fed the diet containing 1% and 3% of taurine for 8 weeks. The contents of the cholesterol and triglycerides in the serum and liver of the hyperlipidemic rats were increased as compared with those of the control group. Feeding taurine resulted in decreases in total cholesterol and triglyceride levels. The HDL-cholesterol level in serum was decreased in the hyperlipidemic rats, but by administration of taurine its level was increased. In the aorta of the animals, total cholesterol and triglycerides contents were reduced significantly by treatment with taurine. The contents of calcium in the heart of hyperlipidemic rats were greatly increased as compared with those of the control group. Treatment of taurine produced significant decreases in calcium contents in the heart muscle of the animals. These results showed that the hyperlipidemic states in this model of rats were reversed by treatment of taurine.

어유의 Eicosapentaenoic Acid의 식이섭취가 성숙쥐의 혈청 및 간의 지질조성에 미치는 영향 (Effect of Dietary Eicosapentaenoic Acid on Serum and Liver Lipids Patterns of Male Rat)

  • The dietry effects of marine n-3, plant n-3 and plant n-6 fatty acid on serum lipids levels, liver phospholipid fatty acid composition in rat were investigated. Four groups of male Sprague-Dawley rats, 30 weeks old, were fed on one of 4 different experimental diets for 4 weeks. The diets were composed of 15% fat(w/w) of either concentrated EPA oil(20:5, n-3 : 65%), fish oil(20:5, n-3 : 19%, 22:6, n-3 : 18%), perilla oil(18:3, n-3 : 60%) or corn oil(18:2, n-6 : 49%). Blood was initially taken before experimental feeding and also taken after 2 weeks and 4 weeks feeding the diet respectively and then examined for the levels of serum lipids. Rats were sacrificed at 4 weeks after the diet for the analysis of liver phospholipid fatty acid. EPA feeding remarkably decreased the serum levels of triglyceride, total cholesterol, HDL-cholesterol and total phospholipid than any other oil feeding. Fish oil feeding decreased serum HDL-cholesterol level comparable to the effect of EPA feeding and decreased total cholesterol and phospholipid less than but close to the effect of EPA feeding. Perilla oil feeding did not change serum levels of triglyceride and phospholipid, but it decreased serum total cholesterol a lot and HDL-cholesterol a little. Corn oil feeding did not affect triglyceride and total cholesterol while it increased serum level of HDL-cholesterol and total phospholipid. Serum HDL-cholesterol level was increased only in corn oil group. But contrary to the result of serum total phospholipid, liver phospholipid level found to be higher in fish oil and EPA groups than in perilla oil and corn groups. The fatty acid composition of liver phospholipid, phosphatidylcholine(PC) and phosphatidyl ethanolamine(PE) turned out to be affected by dietary fatty acid. 18:2 of liver PC was the lowest in FO group following CO group. The ratio of 20:4/18:2 was lower in PO group than in EPA group in consequence of higher 18:2 and lower 20:4 in PO group and vise versa in EPA group. In the liver PC and PE, similar trends in the ratios of n-6/n-3 and 20:4/18 were found showing higher ratios with CO and EPA group over FO and PO group. EPA group showed the lowest level of 20:5 and lower level of 20:6 than group. Fish oil was more efficient than EPA oil and PO in lowering the ratio of n-6/n-3 in consequence of the highest 22:6, and the lowest 18:2 in liver phospholipid. But PO lowers the ratio or 20:4/18 more than FO. In conclusion, EPA oil was more effective in lowering serum lipids than FO and PO. Reviewing the dietary effect of fatty acid on eicosanoids composition in rats, it is considered that more possibility was with FO than PO in the effectiveness of atherosclerosis prevention and more with PO than with EPA oil. It was also found that FO showed more effective than EPA oil for atherosclerosis prevention. It was hardly found that CO had any effect on lowering serum lipids and on eicosanoids composition in liver phospholipid for the prevention of atherosclerosis.

성소의 제거와 고콜레스테롤 식이 급여가 흰쥐의 혈장과 간의 콜레스테롤, 중성지방 수준과 혈소판 응집성 및 간 조직에 미치는 영향에 있어서 암.수의 차이 (Gender Difference in the Effects of Gonadectomy and Hypercholesterol Diet on Plasma and Liver Cholesterol and Triglyceride bevels, Platelet Aggregation and Liver Tissue in Sprague Dawley Rats)

  • Gender differences in the effects of gonadectomy and high cholesterol diet on body weight and body cholesterol were investigated by using Sprague Dawley rats. Body weight, plasma and liver levels of cholesterol and triglyceride and platelet aggregation were examined in ovariectomized(OVX) or orchidectomized(ODX) rats with their intacts after feeding diet with or without 0.5% cholesterol. Body weight was significantly increased(p < 0.01) in OVX rats and significantly decreased(p < 0.01) in ODX rats compared to their respective intact rats, and cholesterol diet significantly(p < 0.05) decreased body weight in gonadectomized rats. Liver lobes from rats fed cholesterol diet were opaque and larger than those from rats find control dict, resulting in a significant increase(p < 0.01) in LW/BW ratio. Plasma and liver levels of total cholesterol were significantly increased (p < 0.01) in female rats regardless ovariectomy when find 0.5% cholesterol diet, but those levels in male rats were increased only when they were orchidectomized(p < 0.0l). Plasma HDL-cholesterol was significantly decreased(p < 0.05) in both sexes when find cholesterol diet. HDL-cholesterol were higher in female than male rats regardless treatments(p < 0.05). Liver triglyceride was significantly increased(p < 0.05) in both sexes when find cholesterol dict. Plasma level of triglyceride was not different among groups except significant decrease(p < 0.05) in cholesterol find ODX rats. Maximum platelet aggregation in female rats was significantly lower(p < 0.05) than male, but ovariectomy and cholesterol diet caused an increase te the level of male rats. Microscopic examination showed cholesterol diet caused a lipid accumulation in liver. Results indicate that intact female rats have higher response to hypercholestcrolemic diet than intact male rats and orchidectomy causes male rats more responsive to hypercholesterolemic diet. However, ovariectomy causes an increase female food efficiency ratio to the level of male rats, significantly increasing body weight.

고콜레스테롤 식이 투여 흰쥐에서 옥수수 펩타이드 섭취가 혈중 지질 성상에 미치는 효과 (Effects of Corn Peptide Consumption on Plasma Lipid Profiles in Cholesterol-Fed Rats)

  • In this study, the effects of corn peptide consumption on plasma lipid profiles were investigated in high cholesterol diet-fed rats. Male Sprague-Dawley rats (n = 21) were fed with corn peptide-free (control) diet, diets containing 2% or 5% com peptide for 5 weeks. Hypercholesterolemia was induced by adding l% cholesterol and 0.5% cholic acid to all diets. No difference was found in food intake and body weight gain among groups. The corn peptide treated groups showed significant improvement in the plasma level of HDL-cholesterol (p<0.05) compared to the control group, while the plasma total cholesterol and LDL-cholesterol were not affected.5oio corn peptide supplemented diet reduced plasma level of triglycerides (p<0.05) The atherogenic index was decreased in the corn peptide treated groups. These results suggest that consumption of corn peptide may lead to an amelioration of metabolic syndrome as well as a reduction of cardiovascular disease and hyperlipidemia through increasing the level of HDL-cholesterol, and decreasing the level of triglycerides in plasma.

Chitooligosaccharide가 생쥐의 간 기능에 미치는 영향 (Effects of Chitooligosaccharides on Liver Function in the Mouse)

  • Chitin의 탈아세틸화로 얻어지는 화합물인 chitosan과 chitooligosaccharide는 다양한 생리활성 기능을 나타내므로 생물의학적인 응용에 많은 관심이 집중되고 있다. 본 연구는 수용성 chitooligosaccharide가 생쥐의 간 기능에 미치는 영향을 조사하기 위하여 수행되었다. 6주 동안 3% cholesterol를 식이한 생쥐에서는 혈장 cholesterol 함량이 증가하였으나, 1% chitooligosaccharide와 3% cholesterol을 함께 식이한 생쥐에서는 혈장 cholesterol 함량이 23% 정도 감소하였다. 그러나 혈장내 HDL-cholesterol과 간내 cholesterol 함량에는 유의한 차이가 없었다. 한편, 15% ethanol을 8주간 투여한 생쥐에서는 inflammation, necrosis, 지방구의 축적, GPT의 활성 증가 등 간 상해가 관찰되었다. 그와 반면에 0.5% chitooligosaccharide와 15% ethanol을 함께 투여한 생쥐에서는 ethanol에 의해 유발된 간 상해가 현저히 억제되어 지방구의 축적이 관찰되지 않았고, 혈장의 GPT 활성이 25%정도 감소하였다. 따라서, chitooligosaccharide가 생쥐에서 혈장 cholesterol 함량을 감소시키고, 알코올성 지방간을 예방하는 등 부분적인 기능이 있음을 보여주었다.

마그네슘 및 칼슘이 혈청콜레스테롤 농도 저하에 미치는 영향 (III) - 인삼의 영향 - (Influence of magnesium and calcium on the serum cholesterol level lowering (III) - Influence of Korea Ginseng -)

  • 마그네슘, 칼슘 및 인삼을 첨가하여 사육한 토끼의 혈청콜레스테롤 농도에 미치는 영향을 조사하여 다음과 같은 결론을 얻었다. 1. 인삼분말의 급여로 체중증가는 별차가 없었으며 간장의 무게는 상당한 증가를 나타냈다. 2. Total cholesterol은 인삼 4mg을 첨가한 실험군에서 가장 낮았고, 마그네슘과 칼슘이 1:2로 급여한 군에서 낮게 나타났다. 3. Free cholesterol과 cholesterol ester도total cholesterol의 경우와 같이 인삼 4mg을 첨가한 것과 마그네슘과 칼슘을 1:2로 급여한 실험군에서 가장 낮았다. 4. Total cholesterol이 가장 낮은 실험군에서 LDL이 가장 낮았으며, HDL/T-CHOL의 비는 가장 높게 나타났다. 5. Triglyceride와 phospholipid에 있어서는 인삼 첨가로 TG의 양은 점점 감소되었으나 PL의 경우는 상당히 증가되었다. 6. 전해질의 경우는 마그네슘과 칼슘을 1:2로 첨가하고 인상 4mg을 급여한 실험군의 $Na^+$이 120mEq/l, $K^+$이 6.8mEq/l를 보여 인삼이 전해질의 이동에 크게 작용하고 있는 것으로 생각된다. 이상과 같은 결과로 인삼은 토끼의 혈청 전해질의 분포상태와 콜레스테롤의 농도에 영향은 주며, 콜레스테롤 농도를 저하시키는 작용을 함과 동시에 지방질을 감소시키고 인지질은 증가시키는 작용을 하는 것으로 추정된다.

식이섬유가 고콜레스테롤혈증 흰쥐의 지질농도에 미치는 영향 (Effect of Fiber on Lipid Concentration in Hypercholesterolemic Rats)

  • The study was conducted to investigate the effect of cholesterol and fiber on liver lipid metabolism in rats. Male Sprague Dawley rats were administrated 1% cholesterol and 0.25% sodium cholate to induce hypercholesterolemia and were fed diet containing three levels(0%, 5% and 10%) of cellulose and pec-tin respectively. The rats were sacrified after 5 weeks of feeding period. Net weight gain and feed ef-ficiency ratio were increased in 10% fiber groups as compared to those of 5% fiber groups. Feed intake did not show significancy by fiber kinds and levels. Liver and kidney weights tended to be decreased in proportion to level of fiber. Heart weight was lowered in 10% fiber groups as compared to that of 5% fi-ber groups. Liver triglyceride concentration was significantly increased in pectin groups. Total-free-cholesterol and cholesteryl ester concentrations in liver were significantly decreased by fiber and were decreased in proportion to level of fiver. Phospholipid concentration was significantly decreased by fiber and were decreased in proportion to level of fiber. Phospholipid concentration was significantly decreased in 5% cellulose group. the results indicate that 10% pectin may have beneficial roles in hypercholester-olemia.

과루지실탕(瓜蔞枳實湯) 및 그 가미방(加味方)이 고지혈증(高脂血症)에 미치는 영향(影響) (A study of the effects of Gualouzhishiltang and Gualouzhishiltangjiawei on hyperlipidemia)

  • In order to study the effects of Gualouzhishiltang and Gualouzhishiltangiiawei on hyperlipidemia white rats were ind uced by lipid metabolism disorder by the oral-administration of cholesterol. The effects of gualouzhishitang and gualouzhishiltangjiawei on lipid in serum were measured by the quantity of total-chohestarol and of H.D.L-cholesterol and of triglyceride and of phospholipid and of ${\beta}$-lipoprotein in serum The obtaind results are summarized as follow 1. The level of serum total-cholesterol, H.D.L-cholesterol, triglyceride, phospholipld, ${\beta}$-lipoprotem decresed in the S1-group and S2-group 2. The level of serum total-cholesterol and phospholipid showed significant value on 3,4 week of S1-group and S2-group. 3. The level of serum H.D.L-cholesterol showed significant value on 3,4 week of S1-group and on 4 week of S2-group. 4. The level of serum triglyceride showed significant value on 4 week of S1-group 5. The level of serum ${\beta}$-lipoprotein showed significant value on 4 week of S1 and S2-group. According to the above experimental results eualouzhishilta ng and Gualouzhishiltangjiawei are aseumed to have a curative effect against hyperlipidemia thus hyperlipidemia and phlegrnretention have relation each other.

흡연 여대생의 혈청 지질, Cu, Zn, Ceruloplasmin 농도 및 Ferroxidase 활성도 (Levels of Serum Lipids, Copper, Zinc, Ceruloplasmin and Ferroxidase Activity in Smoking College Women)

  • This study was done to compare serum levels of lipids, trace minerals, ceruloplasmin and ferroxidase activity between female college smokers and nonsmokers. Serum levels of lipids, Cu, Zn, ceruloplasmin and ferroxidase activity were determined in 33 smokers and 42 nonsmokers residing in the Seoul area. Serum levels of TG, total cholesterol and HDl-cholesterol were measured using test kits. Serum Cu Zn were determined by the AAS. Serum ceruloplasimin level and ferroxidase activity were determined by spectrophotometric methods. All data were statistically analyzed by the SAS PC package program. Mean and standard errors were calculated for each item, and the significance of the difference between two groups was evaluated by Student's t-test at p〈0.05. There was no significant difference in levels of total cholesterol, TG, VLDL-cholesterol, LDL-cholesterol, LDL/HDL and atherogenic index, but HDL-cholesterol level was significantly lower(p$\ll$0.05) in female college smokers than in nonsmokers. Serum zinc level showed no significant difference, whereas copper level was significantly higher in smokers, consequently yielding higher Cu/Zn ratio in smoking college women. Ceruloplasmin level was 32.6mg/dl in smoking college women, which was significantly higher(p$\ll$0.05) than that of 28.6mg/dl in nonsmokers. However, cerulopasmin ferroxidase activity, 0.34U/mg in nonsmokers was higher(p$\ll$0.05) than that of 0.29U/mg in smokers. From these results, it is confirmed that cigarette smoking itself has a very detrimental effect on health and mineral nutritional status although the smoking history is relatively short among college women(pack-years=1.7). The effect of seamustard on blood lipid profiles and glucose level of rats fed diet with different energy composition

  • Recently, Korean people are consuming seaweeds almost 3.5 times more now than three decades ago. It is well known that seaweeds contain lots of soluble dietary fiber in addition to micronutrients such as ${\beta}$-carotene, iodine and some bioactive components. Seaweeds are considered to be effective for preventing chronic diseases including obesity, diabetes mellitus, atherosclerosis, cancer or constipation. This study was conducted to investigate the effect of seamustard intake on body weight gain, blood glucose level and lipid profiles in rats fed diets with different energy nutrient composition. Male Sprague-Dawley rats (average initial weight 103.7g) were divided into groups for two experiments as follows; Control, M2.5 & M5 groups (Exp. I) and M5, M10, HCM5, HCM10, HFM5 & HFM10 groups (Exp. II). The rats were fed diet and water ad libitum for 4 weeks. In general, there was no significant difference in blood glucose and triglyceride concentration among groups. In Exp. I, serum LDL-cholesterol level of rats fed diet with 5% seamustard powder (M5) was significantly lower than that of control group, while HDL-cholesterol level, TC/LDL ratio and weight of adrenal gland were higher. In Exp. II, food intake, body weight gain and EER of high fat diet with 10% seamustard group (HFM10) were the lowest among groups. Except gastrocnemius muscle, all organ weights of HFM10 group were the lowest. Fecal cholesterol excretion and serum LDL-cholesterol concentration of HFM10 group were the highest, while serum HDL-cholesterol level was the lowest among groups. Interestingly, HDL-cholesterol concentration was the highest in HCM5 group among groups. From these results, it was suggested that seamustard intake might be more effective for body weight control, but not for improving blood lipid profiles in high fat diet than in high carbohydrate diet.
