Effects of Inorganic Nutrients and Heavy Metals on Growth and Pigmentation of the Green Alga, Ulva pertusa Kjellman (녹조 구멍갈파래(Ulva pertusa Kjellman)의 생장 및 색소조성에 미치는 무기 영양염류 및 중금속의 영향)

  • Differential growth of Ulva pertusa Kjellman was observed in response to different photon irradiances and seawater. Growth rate of U. pertusa cultivated in the seawater collected from the East Sea was significantly higher than that in the seawater collected from the Yellow Sea. Optimal growth was found at $100{\mu} molm^{-2} s^{-1}$ in both cases. Chlorophyll contents of U. pertusa grown in the east sea water were higher than that cultivated in the west sea water at irradiances lower than $60{\mu} molm^{-2} s^{-1}$. At irradiances higher than $100{\mu} molm^{-2} s^{-1}$, there was no difference in chlorophyll content in between the two different sea waters with tendency that pigmentation decreased with increasing photon irradiances. Nitrate concentration in the west sea water was 2-fold higher than that in the east sea water, while phosphorus concentrations (0.03 ppm) were similar. Concentrations of $Cu^{2+}$ and $Pb^{2+}$ were 0.004 and 0.003 ppm respectively which are far below environmental standard concentrations (0.02 ppm for $Cu^{2+}$ and 0.1 ppm for $Pb^{2+}$). Taking those data into account, we have done laboratory investigations into the effects of inorganic nutrients and heavy metals on U. pertusa. As nitrate concentration increased from 0.5 to 5 ppm, growth rate of U. pertusa increased, but different concentrations of phosphorus did not cause any differential effect. On the other hand, chlorophyll contents increased with increasing phosphorus concentrations. Copper of U. pertusa be toxic decreased the growth and pigmentation as the concentration increased, whereas lead showed no such effect. Concentrations of $Cu^{2+}$ employed in the present study were much higher than those in ambient seawater. Intermittent soaring of $Cu^{2+}$ level as observed in natural seawater could, however, seriously damage the growth behaviour of U. pertusa.

Determination of Greening and Shelf Life of Potato Based on Washing and Storage Temperature Conditions (감자의 세척유무 및 저장온도에 따른 녹화 및 상품성 유지 기간 구명)

  • Potato greening determines the shelf life and affects the marketability of this tuber. Various stresses during handling and storage interact to affect the tuber's physiological status and can affect the rate of greening. This study investigated the effects of storage temperature on tuber greening and shelf life in unwashed and washed potatoes of the cultivar Superior. Physiological and biochemical changes were examined during 15 days at room temperature ($23{\pm}2^{\circ}C$) under cool-white fluorescent light after storage for 1 month at different temperatures ($4^{\circ}C$, $20^{\circ}C$). Hunter a values were negative (-) for washed potatoes after 3 days (-0.8) and 15 days (-2.5) at room temperature following 1 month of storage at $4^{\circ}C$ while positive (+) values were observed for unwashed potatoes after 15 days at room temperature. The Hunter ${\Delta}E$ values of washed potatoes previously stored at $4^{\circ}C$ for 1 month increased after 3 days at room temperature compared with those of unwashed potatoes. The total chlorophyll content of washed potatoes was higher than that of unwashed potatoes. The highest correlation was observed between the Hunter ${\Delta}E$ value and Hunter a value (-0.93506), while a positive correlation coefficient (0.89806) was observed between greening criteria and Hunter ${\Delta}E$ value by using colorimetry. We conclude, therefore, that there is a biosynthetic link between temperature-induced chlorophyll accumulation and tuber greening in storage.

Effect of a Combined Treatment with Uniconazole, Silver Thiosulfate on Reduction of Ozone Injury in Tomato Plant (Uniconazole 과 Silver Thiosulfate 의 복합처리가 토마토의 오존피해경감에 미치는 효과)

  • Studies were conducted to determine the combined effect of uniconazole [(E) -1-(4-chlorophenyl)-4, 4-demethyl 2-(1,2,4 triazol-1-yl)-1-penten-3-ol] and silver thiosulfate $[Ag {(S_2O_3)}^3\;_2-]$ (STS) on reduction of ozone injury in tomato plants(Lycopersicon esculentum Mill. 'Pink Glory'). Plants were given a 50ml soil drench of uniconazole at concentrations of 0, 0.001, 0.01 and 0.1 mg/pot at the stage of emerging 4th leaf. Two days prior to ozone fumigation, STS solution contained 0.05% Tween-20 was also sprayed at concentrations of 0, 0.3 and 0.6 mM. Uniconazole at 0.01 mg/pot and STS at 0.6 mM were effective in providing protection against ozone exposure(20h at 0.2ppm) without severe retardation of plant height and chemical phytotoxicity, respectively. Combined treatment with uniconazole, STS significantly reduced ozone injury at the lower concentration than a single treatment with uniconazole or STS. Uniconazole treatment reduced plant height, stem elongation and transpiration rate on a whole plant level and increased chlorophyll concentration. STS did not give any effect on plant growth and chlorophyll content but increased transpiration rate in non-ozone-fumigated plants. Ethylene production in the leaves of ozone-fumigated plants was decreased by uniconazole and STS pretreatment, but there was no protective effect on epinasty of leaves in uniconazole-treated plants. STS increased ethylene production in non-ozone-fumigated plants, but it significantly reduced the degree of epinasty and defoliation of cotyledons when plants were exposed to ozone. Uniconazole slightly increased superoxide dismutase and peroxidase activities. But STS showed little or no effects on such free radical scavengers. Day of flowering after seeding was shortened and percentages of fruit set were increased by uniconazole treatment. STS was highly effective on protecting reduction of fruit set resulting from ozone fumigation. These results suggest that combined use of uniconazole and STS should provide miximum protection against ozone injury without growth retardation resulting in yield loss.

  • Sedum Sarmentosum Bunge has been used as medical material and food in Korea. To upgrade it's value as a materials of beverage, I research and analysed nutritive substance and taste. Inorganic components content was included in the order of Ca, mg, P, K, Na, in line, and there were little Fe, Cu, Zn. It appeared much Ascorbic acid contents in May sample for March sample, but it was included less than other Green Vegetables. Chlorophyll contents decreased about 37.8% and 37.6% to the first each contents after seven days resulting measuring May sample and March sample in room temperature. There was much difference in appearance. Sedum Sarmentosum Bunge included a little polyphenol contents. Polyphenol contents decreased a little in five days in May sample and March sample. I measured phenol compounds of garlic acid, vani11ic acid, caffeic acid, p-coumaric acid, ferulic acid, most of consisted in corrected forms. Fragrance components were confirmed 9 kinds in March sample, 11 kind in May sample the common components in May sample and March sample were 7-octen-4-o1.

Spatial Distribution of Transparent Exopolymer Particles(TEP) and Their Relation to Carbon Species in the Euphotic Layer of the Northern East Sea (동해 북부해역 유광층에서 TEP 분포와 이산화탄소 인자와의 상호관련성)

  • Transparent exopolymer particles (TEP) are formed by aggregation of polysaccharide products excreted by phytoplankton and have sticky character like gel. They play important role in the production of marine snow in water column. To study the distribution pattern of TEP concentration and its role in carbon cycle in the surface ocean, we measured pH, Total alkalinity (TA), and chlorophyll-a in addition to physical characteristics of seawater within the surface water column. TEP concentrations ranged from nearly undetectable values to $338{\mu}g\;Xeq\;l^{-1}$. They were considerably lower than previously reported values from costal sites, but showed similar values observed in other oceanic region during phytoplankton bloom periods. The spatial distribution of TEP concentrations were similar to those of chlorophyll-a, which indicate that the production of TEP were closely related to phytoplankton. Calculated total dissolved inorganic carbon ($TCO_2$) from the pH and TA was normalized to 35 psu of salinity ($NTCO_2$) and showed negative linear relationship with temperature. Biological drawdown of $NTCO_2$ ($NTCO_{2bio}$) was estimated from the difference between theoretical $NTCO_2$ values and observed $NTCO_2$. In the warm region located south of $40^{\circ}N$ along the $132.5^{\circ}N$ meridional lines, $NTCO_{2bio}$ showed negative value and TEP concentrations were high. This suggested that negative $NTCO_{2bio}$ may be attributed to the biological processes. At the stations located between 44 and $46^{\circ}N$, TEP concentrations showed high concentration at the chlorophyll-a maximum layer within the water column while they showed low concentration in the surface layer. Carbon content of TEP constituted about 40% of $NTCO_{2bio}$ at the chlorophylla maximum layer. In this study, we could not observe any positive and negative relationship between TEP concentration and $NTCO_2$ or pH. It is obvious that we should consider the importance of TEP in the biological carbon cycling processes within surface layer.

Growth inhibition of hydrotrope-combined copper against Microcystis aeruginosa and evaluation of its toxicity (Microcystis aeruginosa에 대한 hydrotrope-combined copper의 생장억제 및 독성 평가)

  • Hydrotrope-combined copper (HCC) is a copper ($Cu^{2+}$)-based algicide, which is combined with a hydrotrope that keeps copper ion in solution to improve performance. This study assessed the growth inhibition effect of HCC against Microcystis aeruginosa which is one of the most common toxic cyanobacterium in eutrophic freshwater environment. Various HCC doses, ranging from 5.5 to $550{\mu}g/L$ as $Cu^{2+}$, were applied to either BG-11 or 1/4 diluted medium with low- or high-inoculum density of M. aeruginosa. Growth inhibition was monitored based on a decrease in chlorophyll-a content in culture medium during the incubation. Results showed that HCC significantly inhibited the growth of M. aeruginosa in a dose-dependent manner. In case of 1/4 diluted BG-11 medium, HCC dose as low as $5.5{\mu}g$ $Cu^{2+}/L$ completely inhibited the production of chlorophyll-a by M. aeruginosa. It was found that HCC did not induce any significant release of microcystin-LR from M. aeruginosa. Acute toxicity of HCC was tested using Daphnia magna, and the 24-h $EC_{50}$ value was 0.30 mg/L as $Cu^{2+}$ which was much higher than the actual inhibition dose. Ames test was performed using Salmonella enterica serovar Typhimurium TA100, and HCC showed no increase in the number of revertant colonies. The result suggested that HCC does not have any mutagenic potential in the aquatic environment. In addition, no genotoxic effect of HCC was also confirmed based on the SOS ChromoTest using Escherichia coli PQ37. Therefore, HCC could be used as a relatively safe and effective pre- and post-treatment agent to control hazardous algal blooming in aquatic environments.

Microprogation And Environment Conditions Affecting On Growth Of In Vitro And Ex Vitro Of A. Formosanus Hay

  • The goal of this research was to develop the effectiveness of in vitro culture method for A. formosanus and study the environment in vitro conditions affecting on growth. The first series of experiments were examined to investigate the response of three different basal media, MS (Murashige and Skoog, 1962), Knudson (KC; Knudson, 1946) and modified hyponex on growth and multiplication during in vitro culture. Multiple shoot proliferation was induced in shoot tip explants on Hyponex (H3) media supplemented with BA (1 mg1$\^$-1/) or TDZ (1-2 mg1$\^$-1/). Addition of activated charcoal (1%) to the TDZ containing medium promoted rapid shoot tip proliferation (11.1 shoots per explant) but the same medium had an opposite effect resulting in poor proliferation in the nodal explants. However, the regenerated shoots had slow growth rate and failed to elongate. This problem was overcome by transferring the shoot clumps to a hormone free H3 media supplemented with 2% sucrose and 0.5% activated charcoal. Using bioreactor culture for scaling up was also shown the best way for multiple shoot induction and growth of this plant. The second series of experiments was studied to investigate the effect of physical environment factors on growth of in vitro plantlets. The Anoectochilus formosanus plantlets were cultured under different air exchange rate (0.1, 0.9, 1.2h$\^$-1/), without sucrose or supplement 20g.1$\^$-1/ (photoautotrophic or photomixotrophic, respectively), and different photosynthesis photon flux (40, 80, 120 ,${\mu}$mol.m$^2$.s$\^$-1/- PPF). Under non-enrichment CO$_2$ treatment, slow growth was observed in photoautotrophical condition as compared with photomixotrophical condition on shoot height, fresh weigh and dry weight parameters; High air exchange (1.2.h-l) was found to be inadequate for plant growth in photomixotrophical condition. On the contrary, under CO$_2$, enrichment treatment, the plant growth parameters were sharply (visibly) improved on photoautotrophic treatments, especially on the treatment with air exchange rate of 0.9.h-1. The growth of plant in photoautotrophic condition was not inferior compared with photomixotrophic, and the best growth of plantlet was observed in treatment with low air exchange rate (0.9.h-1). Raising the PPF level from 80 to 120${\mu}$mol.m$\^$-2/.s$\^$-1/ decreased the plant height, particularly at 120${\mu}$mol.m$\^$-2/.s$\^$-1/ in photoautotrophic condition, fresh weight and dry weight declined noticeably. At the PPF of 120${\mu}$mol.m$\^$-2/,s$\^$-1/, chlorophyll contents lowed compared to those grown under low PPF but time courses of net photosynthesis rate was decreased noticeably. Light quality mainly affected morphological variables, changes of light quality also positively affected biomass production via changes in leaf area, stem elongation, chlorophyll content. Plant biomass was reduced when A. formosanus were grown under red LEDs in the absence of blue wavelengths compare to plants grown under supplemental blue light or under fluorescent light. Stem elongation was observed under red and blue light in the present experiment. Smaller leaf area has found under blue light than with other lighting treatments. Chlorophyll degradation was more pronounced in red and blue light compared with white light or red plus blue light which consequent affected the photosynthetic capacity of the plant. The third series of experiment were studied to investigate the effect of physical environment factors on growth of ex vitro plants including photosynthesis photon flux (PPF), light quality, growing substrates, electrical conductivity (EC) and humidity conditions. In the present experiments, response of plant on PPF and light quality was similar in vitro plants under photosynthesis photon flux 40${\mu}$mol.m,$\^$-2/.s$\^$-1/ and white light or blue plus red lights were the best growth. Substrates testing results were indicated cocopeat or peat moss were good substrates for A. formosanus growth under the greenhouse conditions. In case of A. formosanus plants, EC is generally maintained in the range 0.7 to 1.5 dS.m-1 was shown best results in growth of this plant. Keeping high humidity over 70% under low radiation enhanced growth rate and mass production.

Application of Chlorophyll Fluorescence Parameters for the Detection of Water Stress Ranges in Grafted Watermelon Seedlings (수박접목묘의 건조스트레스 범위 탐지를 위한 엽록소형광 지수의 적용)

  • This study was carried out to quantify the drought stress in grafted watermelon seedlings non-destructively by using chlorophyll fluorescence (CF) imaging technique rather than the visual judgment. Six-day old watermelon seedlings were grown under uniform irrigation for 3 days, and then given drought stress. Afterward, the sensor for the measurement of water content in plug tray cell unit was used to classify the drought-stress level into nine groups from D1 (53.0%, sufficient moisture state) to D9 (15.7%, extremely dry stress), and the 16 CF parameters were measured. In addition, re-irrigation was performed on the drought stressed seedlings(D5 - D9) to determine the growth and photosynthesis recovery level, which was not confirmed by visual judgment. The kinetic curve patterns of CF in three different drought stressed seedling groups were found to be different for the early detection of drought stress. All the 16 CF parameters decreased continuously with exposure to drought stress and drastically decreased from D5 (32.1%) where the visual judgment was possible. The fluorescence decline ratio (Rfd_Lss) started to decrease from the initial drought stress level (D5 - D6), and the Maximum PSII quantum yield (Fv/Fm) was significantly decreased in the later extreme drought stress range (D7 - D9) by re-irrigation recovery test. Thus, Rfd_Lss and Fv/Fm parameters were finally selected as potent indicators of growth and photosynthesis recovery in the initial and later stages of drought stress. Also, to the differences in the numerical values of the individual chlorophyll fluorescence parameters, the drought stress level was intuitively confirmed through the image. These results indicate that Rfd and Fv/Fm can be considered as potential CF parameters for the detection of low and extremely high drought stress, respectively. Furthermore, Fv/Fm can be considered as the best CF parameters for recovery at re-irrigation.

Characterizations of Water Quality, and Potential Relationships of Nitrogen Components and Microbes in the Mulgol Pond on Dokdo, Korea (독도 물골의 수질 특성 및 질소화합물과 미생물간의 잠재적 관계)

  • Water in the Mulgol pond on Dokdo (island), Korea, was historically used for drinking water, but now it has been no longer used for this purpose due to regionally low water quality. Since 2007, this pond has been covered with a metal lid to protect from pollutants of seabirds, indicating limited light penetration into the Mulgol pond. Here, we investigated water quality in the pond and potential relationships of nitrogen components and microbes in May, June, August, and November 2020. The source salinity ranged from 1.39 to 1.57 psu. Suspended solids (0.8~5.1 mg L-1) and chlorophyll-a (<0.01~0.49 ㎍ L-1) remained low. The concentration of dissolved inorganic nitrogen (DIN) was between 35.9 and 47.2 mg L-1. Thus, water in the Mulgol pond proves to be brackish water with low chlorophyll-a and high nutrients. This unique environment may be established by limited light intensity, sea fog (or seawater), and fecal pellets from many seabirds. Although the light source (800~8000 lux) was exposed to the four subsamples, chlorophyll-a concentrations were below <0.5 ㎍ L-1 during the incubation periods. This result suggests that the biomass of phytoplankton does not increase along with an increase in light intensity. Furthermore, the content of nitrate constituted more than 90% of DIN, and a significant negative correlation between nitrate concentration and bacterial abundance was shown in May and June 2020 during the light exposure experiments (R=-0.762, p<0.05). Thus, it is possible that bacteria may be a significant agent to reduce nitrate concentration in the Mulgol pond, the relationship between nitrate concentration and bacterial abundance may vary seasonally.

Evaluation of the Growth and Yield of Sweetpotato (Ipomoea batatas L.) at Different Growth Stages under Low Light Intensity (생육시기별 차광 처리에 의한 고구마 생육 및 수량성 평가)

  • This study was conducted to determine the degree of reduction in the yield of sweetpotato subjected to different shading treatments according to the growing season of the typical viscous sweetpotato 'Hogammi' and the powdery sweetpotato 'Jinyulmi'. Shading was provided using commercially available shading nets (55% and 75% shading level), and the treatments were applied at the following stages of storage root growth: SFS: the storage root formation stage (planting-50th day), SSS: the storage root swelling stage (50-90th day), and SAS: the storage root actively swelling Stage (90-120th day). The growth characteristics according to shading treatments during each growth period, the number of tubers obtained at harvest, and sugar contents were investigated. For both assessed cultivars, there was no significant difference between the control group and the 55% shading treated group with respect to the maximum quantum yield (Fv/Fm) of photosystem II under different shading treatments, whereas the 75% shading group showed slightly higher values than the control group. In both cultivars, the contents of chlorophyll a and b tended to increase in plants subjected to shading treatments compared with the control plants, particularly that of chlorophyll b. Compared with the control group, the chlorophyll b content of 'Hogammi' subjected to 55% and 75% shading increased by 47% and 41%, respectively, whereas that of 'Jinyulmi' increased by 39% and 34%, respectively. We also detected reductions in the dry weights of the above- and belowground parts of the two varieties in response to shading compared with the control, with the reduction in the dry weight of belowground parts being significant. Furthermore, in both varieties, the T/R rate tended to increase in response to shading treatment. Owing to the lack of sunlight, both cultivars tended to suppress the formation and enlargement of tuber roots. Consequently, post-harvest yield analysis revealed that under shading treatments, both cultivars were characterized by poor tuber root growth according to growing season, with the yield of 'Hogammi' showing a greater reduction compared with that of 'Jinyulmi'. In addition, we found that the higher shading level also significantly reduced yields. Compared with the storage root formation and storage root actively swelling stages, shading treatments during the storage root swelling stage significantly affected yield reduction in both varieties.
