The Effectiveness of Arbuscular Mycorrhizal Fungi (AMF) Inoculation on the Growth of Lettuce (상추에 대한 Arbuscular 균근균(AMF) 접종원 처리 효과)

  • To evaluate the effectiveness of AMF on the growth of horticultural crops, we compared mycorrhizal and non-mycorrhizal plants, lettuce (Lactuca sativa L.), that were inoculated with AMF propagules. As compared to the AMF- seedlings, in AMF+ seedlings at 3 weeks after sowing, the number of leaves increased 9%, leaf fresh weight increased 59%, leaf area increased 58%, and leaf length and width increased 21-22%, and chlorophyll content increased 2%. Furthermore, at 9 weeks after sowing, compared to the AMF- plants, in lettuce plants inoculated with AMF at the sowing and transplanting stages, the number of leaves increased 21% and 18%, leaf fresh weight increased 51% and 41%, root fresh weight increased 56% and 47%, and chlorophyll content increased 18% and 19%, respectively. Further this experiment indicated that the growth responses of lettuce plants inoculated with AMF during transplanting were similar to those inoculated with AMF during sowing. The results imply that the AMF infection timepoint is not important. The P content in the leaves of lettuce plants inoculated with AMF during transplanting was significantly higher (217%) than that of leaves from lettuce plants not inoculated with AMF. In contrast, the P content of the leaves of lettuce plants inoculated with AMF during the sowing stage was similar to that of leaves of control lettuce plants. In this experiment, P and chlorophyll content in AMF+ lettuce plants were higher than in AMF- plants, indicating that the photosynthetic rate was improved with AMF inoculation.

Studies on the Physiological Characteristics and Cambial Electrical Resistance of Street Trees in Cheonan City (천안시 가로수의 생리적 특성과 형성층 전기저항치에 관한 연구1)

  • This study was conducted to diagnose the health of street trees with physiological characters, and to figure out the relation of physiological characters and cambial electrical resistances. Ginkgo biloba, Prunes serrulata and Salix koreensis were chosen in the Cheonan City. Soils under trees were collected to analyze dehydrogenase activities, and chlorophyll content, nitrate reductase and superoxide dismutase activities were analyzed from leaves sampled at the edge of crown in July. Cambial electrical resistances were measured in May, July and September, Soils with low dehydrogenase activity reflected the level of pollution. Chlorophyll content was the lowest in the leaves of P. serrulata at the Dongseo-street. Nitrate reductase activity of Ginkgo biloba was higher than P. serruluta and Satix koreensis. Nitrate reductase activity showed higher activity in the city than control(Independence Hall and Yonam College), but superoxide dismutase activity in the city lower than control. P. serruzatu in the Dongseo-street that cambial electrical resistance increase continuously during the growing season, showed the loss of vitality Cambial electrical resistance was negatively or positively correlated with nitrate reductase($r^2$=-0.566) and superoxide dismutase activity($r^2$=0.579). It was concluded that cambial electrical resistance might be suitable for diagnosing the tree health.

The Distribution of $Cd^2+$ and its Physiological Toxicity in Commelina communis L. (닭의장풀 내 $Cd^2+$의 분포와 생리적 독성)

  • The effect of $Cd^2+$ on chlorophyll content, water potentials, ion transport, photosynthesis, stomatal apertures and $Cd^2+$ accumulation of organs in Commelina communis was investigated. 3-weeks old Commelina. communis was transferred to and grown in Hoagland solution in the presence or absence of 5 mM $Cd^2+$ for 4 days. $Cd^2+$ was accumulated in all parts of the organs including leaves, roots and stem. The proximity from the root and the age of leaf were significant factors responsible for the distribution of cadmium. Most of $Cd^2+$ was accumulated in the first leaf which was the nearest from the root. $Cd^2+$) accumulation in the leaves led to significant reductions in a series of physiological metabolism. $Cd^2+$ reduced total chlorophyll content up to 70%, and changed chlorophyll a/b ratio to 2. $Cd^2+$ also reduced about 20% of water potential. The treatment of $Cd^2+$ showed about 60% inhibition of photosynthetic activity when measured at various light intensity (100-1,000 $\mu$mol $Em^-2s^-1$). Similar effect was found in terms of stomatal conductance. Therefore, it could be concluded that the treatment of $Cd^2+$ decrease or block various physiological activities. Electron Transport Activities of Chloroplasts Isolated from the Detached Rice Leaves Stored under Low Temperature with Illumination (광 및 저온처리한 벼잎 절편에서 분이한 엽록체의 전자전달 활성)

  • The electron transport activities of choloroplasts isolated from hte detached rise (Oryza sativa L. cv. Chucheong) leave stored under low temperature(4$^{\circ}C$) with light illumination were investigated to understand the role of light in the low temperature inhibition of photosynthesis in the chilling-sensitive plants. Chlorophyll content of the detached leaves upon incubation at 28$^{\circ}C$ and 4$^{\circ}C$ in the dark was also measured. The rice seedlings were grown with Hoagland medium in the growth chamber of 28$^{\circ}C$ temperature and 400 ft.c fluorescent light with the photoperiod of 16 h. Although chlorophyll content of the detached leaves stored in the dark declined by 61.7% after 28$^{\circ}C$ treatement, there occurred only 5.2% decrease of chlorophyll with 4$^{\circ}C$ treatment. Low temperature treatment(4$^{\circ}C$) for 6 days brought about decreases in total photosystem(PS II+PS I) activities by 35.2% and 73.6% in te presence and absence of light, respectively, while after 28$^{\circ}C$ treatment of the detached leaves for 6 days in the dark there was only 27.6% decrease in PS II+PS I activity. PS II activities were also decreased by 35.6% and 72.2% in the light and dark, respectively. PS I activities were decreased slightly, however, by 7.6% and 16.2% in the light and dark, respectively. Investigations into DPClongrightarrowDCPIP and NH2OHlongrightarrowDCPIP activities revealed that low temperature inhibition of PS II activities was not due to the inactivation of the water oxidation capacity at low temperature. It was concluded that light protects the electron transport activities of isolated rice chloroplasts from the inhibitory effect of low temperature in the detached leaves.

  • To determine the optimum light intensity for growth of ginseng plants, change of temperature, moisture content in son, occurrence alternaria blight, defoliation rate, chlorophyll contents, and growth of shoots and roots were investigated under different light intensity such as 5%, 10%, 20% and 30% light transmittance rare(L.T.R.). The results obtained were as follows. 1. Maximum temperature under the shading was increased as the increase of light intensity, whereas soil moisture content decreased 2. As the increase of light intensity, stem and Peduncle length, leaf area, and chlorophyll contents decreased significantly but length and width of the leaf was not significant, while stem diameter, special leaf weight and chlorophyll a/chl. b ratio increased 3. Stem color was shown dark purp!e as the increase of light intensity. 4. Photosynthesis during the day was highest at 9 A.M. and decreased as time passed in all plots. The means of photouynthesis during the day showed in the order of 20%, 10%, 30%, 5% L.T.R., and optimum light intensity for highest photosxthesis was 18.4% L.T.R. by theoritical equation. 5. It was showed a tendency that alternaria leaf blight of ginseng plants was increased as the increase of light intensity. 6. Defoliation rate of ginseng plants was increased as the increase of light intensity, especially all plants were defoliated by late June without shading. 7. Yield percentage of the rear line was increased as the increase of light intensity. Root weight per plant showed in the order of 20%, 10%, 30%, 5% L.T.R., and optimum light intensity for the best yield was 18.5% L.T.R. by theeritical equation.

Growth, Protein and Pigment Content of Rice Seedlings under Phosphorus Deprivation Condition

  • Phosphorus (P) is a macronutrient playing important roles in many plant processes. Significant interest has been devoted to search and utilize genotypic variations in P use efficiency in rice but with little effort to understand its physiological and biochemical bases. In this study, we examined responses to P deprivation of some primary and secondary traits in 3-week-old seedlings of the three genotypes, Sobi-byeo (japonica), Dasan-byeo (japonica $\times$ indica) and Palawan (indica). In general, percent weight due to root was increased up to 26%, but amounts of root protein and proteins secreted from roots were decreased by 11 to 19% and 31 to 51 %, respectively, by 3 to 21 days of P deprivation in the three genotypes. Interestingly, however, responses of Palawan to short-term P deprivation were contrasting to those of Dasan-byeo and Sobi-byeo in seedling weight and contents of shoot protein, chlorophyll and anthocyanin. Seedling weight was not decreased, but shoot protein content was decreased in P-deprived seedlings of Palawan. Contents of chlorophyll in leaves and anthocynin in roots were increased in Dasan-byeo and Sobi-byeo, but decreased in Palawan. The results suggest that responses of protein and pigment synthesis to P deficiency are different in modem and traditional varieties and the difference may at least in part be due to the selection for high yield under highly fertilized conditions.

Effect of different water levels on the photosynthetic pigments of crops

  • An excess soil water condition is one of the major problems for the field crops growing in paddy fields because of their poor drainage and less availability for oxygen uptake which leads to adversely affect the photosynthesis. Therefore, the current study was undertaken with aim to investigate the effects groundwater level on the photosynthetic response of soy bean (Urum), red bean (Arari), sesame (Geonbaek), perilla (Dayu) after the transplanting to the lysimeter to investigate the plant-water relation and their effect on photosynthesis. The chlorophyll content of the crops according to the humid conditions of the soy bean, sesame and the perilla was found to be 5%, 6.89 % and 13.7% higher than that of the groundwater treated at 40cm, respectively. On the other hand, the chlorophyll content of adzuki bean decreased 6.6% from the groundwater level of 40cm, and the sorghum decreased by 5.7%. As a result of investigating the Fv / Fm value of groundwater, the adzuki bean at 20cm above groundwater was lower than that of groundwater by 40cm immediately before flowering. The Fv / Fm value of soy bean and sesame at 40cm above groundwater were lowered by flowering under groundwater 20 cm and Fv / Fm value of sorghum is increased at 40 cm treatment immediately before flowering while the Fv / Fm values of the perilla had no significant difference in comparison to those at 20 cm and 40 cm of groundwater. In the case of chlorophyll fluorescence reaction, it is known that the when the absolute value is closer to 0.82, the stress is considered less. As a result of comparing the numerical values of the crops, it was found that the sorghum was the most stressed followed by adzuki bean and sesame, while the soy beans and perilla was found on the average, as they received less stress.

Mitochondrial activity in illuminated leaves of chlorophyll-deficient mutant rice (OsCHLH) seedlings

  • The rice CHLH gene encodes the $Mg^{2+}$-chelatase H subunit, which is involved in chlorophyll biosynthesis. Growth of the chlorophyll-deficient oschlh mutant is supported by mitochondrial activity. In this study, we investigated the activity of mitochondrial respiration in the illuminated leaves during oschlh seedling development. Growth of mutant plants was enhanced in the presence of 3% sucrose, which may be used by mitochondria to meet cellular energy requirements. ATP content in these mutants was, however, significantly lowered in light conditions. Low cytosolic levels of NADH in illuminated oschlh mutant leaves further indicated the inhibition of mitochondrial metabolism. This down-regulation was particularly evident for oxidative stressresponsive genes in the mutant under light conditions. Hydrogen peroxide levels were higher in oschlh mutant leaves than in wild-type leaves; this increase was largely caused by the impairment of the expression of the antioxidant genes, such as OsAPXl, OsRACl, and OsAOXc in knockout plants. Moreover, treatment of mesophyll protoplasts with ascorbic acid or catalase recovered ATP content in the mutants. Taken together, these results suggest that the light-mediated inhibition of mitochondrial activity leads to stunted growth of CHLH rice seedlings.

Photosynthetic Responses of four Oak Species to Changes in Light Environment (광환경 변화에 대한 네 참나무 수종의 광합성 반응)

  • This study was conducted to investigate the photosynthetic responses of four Oak species (Quercus mongolica, Q. serrata, Q. acutissima and Q. variabilis) by shading treatment. We investigated light response curve, photosynthesis (A)-intercellular $CO_2$ concentration (Ci) curve, leaf growth and chlorophyll content at the level of 35, 55 and 75% shading treatments and under the full sunlight. In our results, Q. variabilis and Q. acutissima showed increased leaf growth, chlorophyll content and net apparent quantum yield but reduced chlorophyll a/b and carboxylation efficiency under the low light intensity. Therefore, light absorption and light utilization efficiency were improved under the low light intensity. Q. mongolica showed the similar responses that Q. variabilis and Q. acutissima showed, but net apparent quantum yield was reduced. The effects of shading treatment on Q. serrata were lower than those of other three species.

Phosphate Fertilizer Influences Growth and Photosynthesis of Pepper Seedlings (인산시비가 고추 플러그묘의 생장과 광합성에 미치는 영향)

  • Seedlings of ‘Nokgwang’ and ‘Kwari’ green pepper were cultured in 32-cell plug trays in TK$_2$ medium to find out optimal concentrations of phosphate. Seedling growth of both ‘Nokgwang’ and ‘Kwari’ cultivars was affected by phosphate strength. Applying P fertilizer to the plug system caused a marked increase in plant height and leaf area at 1.0 phosphate strength. On the other hand, total dry weight increased with increasing phosphate strength. Higher chlorophyll content was observed in ‘Nokgwang’ than ‘Kwari’ in all treatments. Phosphate strength greater than 0.5 gave similar chlorophyll content. Photosynthetic rate was higher for plants fertilized with 1.0 strength for ‘Nokgwang’ and 0.5 strength for ‘Kwari’than the other treatments. Higher concentrations of phosphate reduced photosynthesis in both cultivars. With ‘Nokgwang’increasing concentration of phosphate up to 1.0 strength resulted in increase in stomatal conductance and transpiration rate.
