  • Human ChlR1 protein (hChlR1), a member of the cohesion establishment factor family, plays an important role in the segregation of sister chromatids for maintenance of genome integrity. We previously reported that hChlR1 interacts with hFen1 and stimulates its nuclease activity on the flap-structured DNA substrate covered with RPA. To elucidate the relationship between hChlR1 and Okazaki fragment processing, the effect of hChlR1 on in vitro nuclease activities of hFen1 and hDna2 was examined. Independent of ATPase activity, hChlR1 stimulated endonuclease activity of hFen1 but not that of hDna2. Our findings suggest that the acceleration of Okazaki fragment processing near cohesions may aid in reducing the size of the replication machinery, thereby facilitating its entry through the cohesin ring.

  • To estimate the effect of algae to particulate organic matter in the upper regions of brackish Lake Sihwa, temporal and spatial variations of particulate organic carbon (POC) and phytoplankton pigments (chlorophyll a; Chl-a, pheophytin-a; Pheo-a), and their relationships were studied at seven sites of the brackish regions from March to October 2005 and 2006. POC concentration varied from 1.0 to $76.6mgL^{-1}$ (mean $7.4mgL^{-1}$), with maximal concentrations occurring in the middle parts of the study area in spring of 2005 and 2006. Concentrations of Chl-a and Pheo-a varied from 1.3 to $942.9{\mu}gL^{-1}$ (mean $71.0{\mu}gL^{-1}$) and $1.4{\sim}1,545.5{\mu}gL^{-1}$ (mean $59.9{\mu}gL^{-1}$), respectively, and corresponded closely with variation in POC. During the study period Pheo-a concentration was 44.2% of total Chl-a, implying that non-living or inactive phytoplankton is also the important part of phytoplankton-derived POC in brackish regions of Lake Sihwa. From the positive linear relationships between POC and phytoplankton pigments (POC with Chl-a (r=0.93), total Chl-a (r=0.88), and Pheo-a (r=0.81)), it is suggested that phytoplankton was a significant component of POC in the upper regions of brackish Lake Sihwa. On the other hand, the ratios of POC/Chl-a and POC/total Chl-a (Chl-a+Pheo-a) were 82.9 and 35.9, respectively. The ratio of POC/total Chl-a is similar to those reported in previous studies, including 40~60 in estuaries. This study suggests that Pheo-a concentration is considered in estimation of POC concentration from phytoplankton pigments in aquatic systems with high content of Pheo-a, like an upper region of blackish Lake Sihwa.

  • The objective of this study was to evaluate the relationship between nutrients and phytoplankton; and phytoplankton and organic matter. In order to examine the limiting nutrient for phytoplankton, Redfield ration was used and revealed nitrogen limitation. Nitrogen limitation was greatest with a 4.7 DIN/DIP ratio especially during the summer season. Chl.-a increase by 79% and 97% in spring and summer, respectively, compared to winter. COD was lowest with 0.84mg/l in winter and highest with 1.12mg/l in summer. The interrelationship between nutrients and Chl.-a was high. Relationship coefficient$(r^2)$ between DIN and Chl.-a, and DIP and Chl.-a were 0.93 and 0.89, respectively. This suggests Nutrients might be utilized at the increase of phytoplankton. Also, Relationship coefficient$(r^2)$ between Chl.-a and COD was 0.78. COD production rate was calculated with Regression Equation. The COD production rate was 25% in winter and 40% in summer.

  • The objectives of this study were to characterize long-term annual and seasonal trophic state of Topjeong Reservoir using conventional variables of Trophic State Index (TSI) and to determine the empirical relations between the trophic parameters. For the analysis, we used water quality dataset of 1995$\sim$2007, which is obtained from the Ministry of Environment, Korea and the number of parameters was 9. Annual ambient mean values of TN and TP were 1.78 mg $L^{-1}$ and 0.03 mg $L^{-1}$, respectively and TN : TP ratios averaged 76, indicating that this system was nitrogen-rich hypertrophic, and was probably phosphorus-limitation for algal growth. Therefore, nitrogen varied little with seasons and years, and total phosphorus (TP) varied depending on season and year. Monsoon dilutions of TP occurred in August and monthly fluctuations of suspended solid (SS) was similar to those of chlorophyll-$\alpha$ (CHL). Annual mean values of BOD and $COD_{Mn}$ were 1.61 mg $L^{-1}$ and 4.23 mg $L^{-1}$, respectively and the interannual values were directly influenced by the intensity of annual rainfall. There were no significant differences in the trophic variables between the two sampling sites. Mean values of Trophic State Index (TSI, Carlson, 1977), based on TN, TP, CHL, and SD (Secchi depth), turned out as eutrophic state, except for the TN (hypertrophic). Regression analyses of log-transformed seasonal CHL against TP and TN showed that variation of the CHL was explained 37% by the variation of TP ($R^2$=0.37, p<0.001, r=0.616), but not by TN ($R^2$=0.03, p>0.05). Regression coefficient of $Log_{10}$CHL vs $Log_{10}SD$ was 0.330 (p<0.003, r=0.580), indicating that transparency is regulated by the organic matter in the system. Results, data suggest that one of the ways controlling the eutrophication would be a reduction of phosphorus from the watershed.

  • In this study, organic matters production was calculated with long term data in Wando costal area where was selected for non influent of fresh water. The objective of this study was to evaluate relationship between nutrients and phytoplankton and, between phytoplankton and organic matter. The result of DIN was the highest with 0.138 mg/L in winter season and the lowest with 0.052 mg/L in summer season. Similarly, DIP was shown to be 0. 017 mg/L in winter and 0.011 mg/L in summer. Limiting nutrient was revealed with nitrogen in Wando costal area. Specially in summer season, nitrogen limitation was the greatest with 10.5 of N/P ratio. Chl. -a was increased 80%, 108% in spring and summer compare with winter. COD was the lowest with 0.84 mg/L in winter and the highest with 1.10 mg/L in summer. The interrelation between nutrients and Chl. -a was high. Relationship $coefficient(r^2)$ were 0.93(P<0.05), 0.89(P <0.05) between DIN and Chi. -a, DIP and Chl. -a. This results suggest dissolved nutrients might be utilized at the production of phytoplankton. Also Relationship $coefficient(r^2)$ was 0.77(P<0.05) between Chl. -a and COD. COD production rate was calculated with regression equation. The COD production rate was 17% in winter and 36% in summer. It was revealed nutrients were decreased according to temperature increasing and then Phytoplankton and organic matter were increased. The Relationship of Nutrients, Chl. -a and COD was very high.

  • The purpose of this study was to determine trophic state, based on nutrients (TN, TP), transparency (SD), and chlorophyll-${\alpha}$ (Chl) and identify their empirical relations of TN-Chl, TP-Chl and Chl-SD depending on the dataset used along with dynamics of conductivity and suspended solids. Analysis of trophic states showed that more than half of 36 reservoirs were judged as eutrophic-hypertrophic conditions depending on the trophic variables. Seasonal values of TP varied by nearly 500% and showed greater in August than any other months. In contrast, TN varied within less than 90% and all monthly mean values of TN were never fall less than 1.2 mg L$^{-1}$ indicating low seasonal variations and high ambient concentrations (eutrophic-hypertrophic state). Analysis of empirical relations in the trophic variables showed that transparency had greater functional relations with Chl (R$^2$=0.31, p<0.001) than TP (R$^2$=0.15, p<0.001) and TN (R$^2$=0.20, p<0.001). Ratios of TN : TP in the ambient water indicated that most reservoirs showed a potential phosphorous limitation on the algal growth. Thus, algal biomass, based on Chl values, was more regulated by phosphorous than nitrogen. Analysis of linear regression model, based on log-transformed annual mean values, showed that only 30% in the variation of Chl was explained by TP (R$^2$=0.295, p=0.001, n=36) and 15% by TN (R$^2$=0.151, p=0.019, n=36). However, linear regression model, based on individual system, showed that Chl-TP model had strong positive relations (R$^2$=0.62, p=0.002, n=12), whereas the model had no any relations (p=0.892, n=12). Overall, our data suggested that averaging effect in the empirical model developments may influence the significance in the statistical analysis.

  • This study analyzed the water quality characteristics and developed empirical models prior to and after the construction of Baekje Weir, in the Geum River watershed between 2004-2017. The comparative evaluation of the surface water chemistry before and after the four major river projects on the weirs indicated that total phosphorus (TP), based on annual data, rapidly decrease after the construction of the weir while the total nitrogen(TN) decreased. Conversely, chlorophyll-a (CHL) concentration, which is a good indicator of primary productivity, increased after the construction of the weir together with an increase in specific conductivity. Simply put, the construction of the weir led to the decrease in concentrations of N and P due to the increased water residence time (WRT), whereas the CHL :TP ratio greatly increased in magnitude. The regression analysis of the empirical model indicated that CHL had no significant relation (r=0.068, p=0.6102, n=58) with TP before the weir construction, but had a relation with TP after the weir construction (r=0.286, p<0.05, n=56). Therefore, such conditions resulted in an increase in primary productivity on a given unit of phosphorus, resulting in frequent algal blooms. In contrast, seasonal suspended solids (SS) and TP increased during the monsoon period, compared to the pre-monsoon, thereby showing positive correlations (r>0.40, p<0.01, n=163) with precipitation. If the government consistently discharges water from the weir, the phosphorus concentration will be increased due to its reversion to a lotic waterbody from a lentic waterbody hereby reducing algal blooms in the future.

  • The purpose of this study was to evaluate trophic conditions of various Korean reservoirs using Trophic State Index (TSI) and predict the reservoir conditions by empirical models. The water quality dataset (2000, 2001) used here were obtained from the Ministry of Environment, Korea. The water quality, based on multi-parameters of dissolved oxygen (DO), biological oxygen demand (BOD), chemical oxygen demand (COD), total phosphorus (TP), total nitrogen (TN), suspended solid (SS), Secchi depth (SD), chlorophyll-${\alpha}$ (CHL), and conductivity largely varied depending on the sampling watersheds and seasons. In general, trophic conditions declined along the longitudinal axis of headwater-to-the dam and the largest seasonal variations occurred during the summer monsoon of July-August. Major inputs of TP occurred during the monsoon (r=0.656, p=0.002) and this pattern was similar to solid dynamics of SS (r=0.678, p<0.001). Trophic parameters including CHL, TP, SD, and TN were employed to evaluate how the water systems varies with season. Trophic State Index (TSI, Carlson, 1977), based on TSI (CHL), TSI (TP), and TSI (SD), ranged from mesotrophic to eutrophic. However, the trophic state, based on TSI (TN), indicated eutrophic-hypereutrophic conditions in the entire reservoirs, regardless of the seasons, indicating a N-rich system. Overall, nutrient data showed that phosphorus was a primary factor regulating the trophic state. The relationships between CHL (eutrophication index) vs. trophic parameters (TN, TP, and SD) were analysed to develop empirical models which can predict the trophic status. Regression analyses of log-transformed seasonal CHL against TP showed that the value of $R^2$ was 0.31 (p=0.017) in the premonsoon but was 0.69 (p<0.001) during the postmonsoon, indicating a greater algal response to the phosphorus during the postmonsoon. In contrast, SD had reverse relation with TP, CHL during all season. TN had weak relations with CHL during all seasons. Overall, data suggest that TP seems to be a good predictor for algal biomass, estimated by CHL, as shown in the empirical models.

  • To understand the growth of attached microalgae to the immersed artificial surfaces in seawater with exposure time, chlorophyll a (chl a) concentration and abundance of attached microalgae to glass slides, and primary productivity and chl a concentration on coverglasses were investigated in Incheon Harbour in May, June 1996 and January-February 1997. Chl a concentrations of microalgae and abundances of diatoms attached to glass slides reached 62.5 mg chl a $m^{-2}$ and $144{\times}10^3$ cells $cm^{-2}$, respectively, during the study period. Chl a concentrations increased with exposure time, and they were significantly correlated with the abundances of attached diatoms ($r^2=0.79$, p<0.001). The chl a concentrations of attached micro algae on coverglass reached the maximum values of 31.1 mg chl a $m^{-2}$ and 65.4 mg chl a $m^{-2}$, and then decreased in May, June 1996. But in January-February 1997, the chl a concentration increased continuously up to 98.9 mg chl a $m^{-2}$. The primary productivity reached the maximum values of 63.1 mgC $m^{-2}\;h^{-1}$, 347.0 mgC $m^{-2}\;h^{-1}$ and 78.3 mgC $m^{-2}\;h^{-1}$, respectively, in May, June and January-February. The primary productivity in May and June varied in accordance with chl a concentrations. But in January-February, the primary productivity decreased from 26 days of exposure while chl a concentration continued to increase. Two cases that primary productivity decreased abruptly seemed to be caused by decrement of chl a and light specific $P^B$ (chl a specific primary productivity) (May and June) and by decrement of light specific $P^B$ due to photoinhibition (January-February). The results of present study indicated that chl a concentrations and the primary productivity of microalgae attached to artifical surfaces immersed in seawater would expedite analysis of dynamics of biomass and physiological status of attached microalgae during biofilm formations.

  • The objectives of this study were to evaluate seasonal and interannual variations of water quality and nutrient input (N, P) in Junam Reservoir, a nesting waterbody of migratory birds, over 10 years during 1998$\sim$2007 along with dynamic relations of trophic parameters using empirical models. Concentrations of COD averaged 7.8 mg $L^{-1}$ during the study, while TN and TP were $1.4\;mg\;L^{-1}$ and $83{\mu}g\;L^{-1}$, respectively, indicating an eutrophic-hypereutrophic state. Values of monthly COD had strong positive relations (r=0.669, p<0.001) with conductivity, indicating that summer rainfall resulted in an ionic dilution of the reservoir water by rainwater and contributed better water quality. One-way ANOVA tests showed significant differences (F=$5.2{\sim}12.9$, p<0.05) in TN and TP between the before and after the bird migration. In other words, nutrient levels were greater in the absence of migratory birds than in the presence of the migratory birds, suggesting a no-effect on nutrient inputs by the birds. Also, one-way ANOVA indicated no significant differences (F=$0.37{\sim}0.48$, p>0.05) in $NO_{3^-}N$ and $NH_{3^-}N$ between the before and after the birds migration. Linear empirical models using trophic parameters showed that algal biomass as CHL, had significant low correlations with TN ($R^2$=0.143, p<0.001, n=119) and TP ($R^2$=0.192, p<0.001, n=119). These results suggest that influences of nutrients on the CHL were evident, but the effect was weak. This fact was supported by analysis of Trophic State Index Deviation (TSID). Over 70% in the observed values of "TSI (CHL)-TSI (SD)" and "TSI (CHL)-TSI (TP)" were less than zero, suggesting a light limitation on the CHL by inorganic suspended solids.
