The fluorescence yields of chl-a and-b complexes bound to silicon dimer, tetramer and hexamer containing pyridine group in diethyl ether solvent, were shown the transition of excited energy through silicon polymer chain, and the maximum energy quenching were appeared at 1 : 1 ratio of chl-a and pyridine group in silicon polymer but the chl-b complexes were shown the maximum energy of fluorescene emission at the same ratio.
BACKGROUND: Purpose of the study was to establish the extinction coefficients of chlorophyll a and b in N,N-dimethylformamide(DMF) and Dimethylsulfoxide(DMSO) solvents and to find out the conditions of optimal extraction temperature and time in citrus leaves. METHODS AND RESULTS: Chlorophyll a and b standards were dissolved in DMF and DMSO. Extinction coefficients of chlorophyll pigments were determined and their contents were quantified using spectrophotometer. Chlorophyllous pigments of citrus(Citrus unshiu Marc. cv. Okitsu wase) leaves were extracted at 25, 40, 60 and $80^{\circ}C$ for 4, 6, 8, 24 and 48 hours to determine the optimal extraction condition. CONCLUSION: The extinction coefficients of chlorophyll a(Chl a) and chlorophyll b(Chl b) of DMF extracts for high extinction wavelength were 663.8 and 647.2 nm. Similarly, the high extinction wavelength of DMSO extracts were 665.8 and 649.0 nm for chl a and b respectively. Chl a, Chl b and total chlorophyll content of DMF extracts were Chl a = $12.10A_{663.8}-2.74A_{647.2}$, Chl b = $21.94A_{647.2}-5.06A_{663.8}$ and total $Chl=19.193A_{647.2}+7.04A_{663.8}$. Similarly, Chl a, Chl b and total Chl of DMSO extracts were Chl a = $14.53A_{665.8}-5.40A_{649.0}$, Chl b = $26.98A_{649.0}-7.11A_{665.8}$ and total $Chl=21.58A_{649.0}+7.43A_{665.8}$. The chlorophyll extracts of DMF and DMSO were very stable in dark. High chlorophyll contents of citrus leaves were found at $80^{\circ}C$ extraction for 6 hours in DMF and at $80^{\circ}C$ extraction for 24 hours in DMSO. However, the chlorophyll content was decreased significantly after 8 hours in DMF extraction while it was remained up to 30 hours in DMSO extraction.

The wavelength of chlorophyll-b on the absorbance and fluorescence emission were shifted to the longer depending on the increasing of solvent polarities but fluorescence excitation spectra were not. The presence of chl-b oligomers and monomers were identified by the specra of fluorescence emission. Fluorescence excitation, absorbance and the measurement of its intensities vs. the concentration of n-prOH added to chl-b solution. The calibration curve of chl-b solution were not obeyed to Beer's law in the range of concentrated soln. because of the presence as the oligomers.
Lincomycin, an inhibitor of plastid protein synthesis, was found to block the synthesis of apoprotein P700 with a molecular mass of 72 kDa and the assembly of the Chl a-protein of PS I. Synthesis of the polypeptides of 48, 43.5, and 32 kDa of the PS II complex is also suppressed. This process is accompanied by the disappearance of the PS Two reaction center Chl a at 683 nm, and of the PS One reaction center Chl a at 690, 696, and 705 nm on the fourth derivative of the absorption spectra at 77K. Lincomycin does not affect the synthesis of LHC subunits. It increases the content of the two main Chl forms of LHC at 648 nm (Chl b) and 676 nm (Chl a). The low-temperature fluorescence ratio F736/F685 is also increased. However, the effect of cycloheximide (an inhibitor of cytoplasmic protein synthesis) leads to the reduction of polypeptides of the light-harvesting Chl a/b-protein complex in the range of 29.5-22 kDa. Under these conditions, the relative amount of Chl b and the F736/ F685 fluorescence ratio decrease significantly. This is obviously the result of blocking the LHC I and LHC II synthesis. At the same time rifampicin and actinomycin D (inhibitors which block transcription in chloroplast and nuclear genome, respectively) inessentially affect the characteristics of these complexes.

It was reported that the digestion ratio of chlorella was low because it had a low metabolic rate in body. Generally, the thickness of a cell membrane of it is $200-250\;{\AA}$, the weight of it is approximatly 13% of the total weight of a dry cell. And it is composed of protein, lipid, hemicellulose and ash etc. So, in order to elevate the digestion ratio of chlorella in body, we experimented the crude treatment methods of chlorella. The results obtained in this experiment are summarized as follows : 1. The digestion ratios calculated from ordinary N- balance method were 83.05% for 10% chl. (b) plus diastase group ; 81.25% for 10% chl. (b) plus amylase group, and 79.23% for 10% chl. (b), 58.55% for 10% chl. (a). 2. Biological values from this method were 80.25% for 10% chl. (b) plus diastase group, and 60% for 10% for chlorella(a).
The Beer-Lambert law states that absorbance is proportional to the concentration of a solute in a solution at a given wavelength. This linearity works for an ideal or a 'sufficiently diluted' solution, so this linearity is often used as a criterion for the fidelity of the absorbance value measured. In this study, we used a chlorophyll (Chl) solution, isolated from rice leaves with 80% acetone to test the use of the Chl a/b ratio as an additional criterion for checking the fidelity of measured values using four different absorption spectrophotometers: Cary4E, UV-1650PC, Versamax (a microplate reader), and NanoDrop 1,000(which can handle a $4{\mu}l$ aliquot). We used Chl solutions of varying concentrations from $0.2{\mu}g/ml$ to $200{\mu}g/ml$ to measure absorbance values at 645 nm and 663 nm and checked the linearity first. The results indicated that the range of Chl concentrations that we can rely on based on the linearity was similar to the range in which the calculated Chl concentrations based on the measured absorbance values agreed with the known concentrations. However, some border cases or cases with very low Chl concentrations inside the fidelity range of Chl concentrations did not agree with the criterion that the Chl a/b ratio should not change after dilution of the Chl in the solution. These results suggest that the Chl a/b ratio is a better criterion for the reliability of the absorbance values measured for the determination of chlorophyll concentration than the criterion based on the linearity suggested by the Beer-Lambert law.

The effect of ABA on the chloroplast disassembly of Zea mays was investigated by measuring the changes in the relative distribution of chlorophyll(Chl) between the Chl-protein complexes in ABA treated and untreated sensecting leaves. The reaction center(RC)-light harvesting complex(LHC) regions were rapidly disassembled in the late stage of dark-induced senescence. Plus, during dark-induced senescence, the disassembly of a reaction center of P700 apoproteins containing mainly Chl a was faster than that of a reaction center of LHCI apoproteins containing both Chl a and Chl b. The increase in the relative distribution of Chl-protein complexes in the RC-Core2 in the late stage of senescence was due to the accumulation of core complexes such as CP47/43 and reaction centers including D1/D2 apoproteins disassembled from the RC-Corel containing the dimer of D1/D2 apoproteins. The LHCII region was more stable than the other Chl-protein complexes throughout leaf senscence. Accordingly, it is suggested that the preferential breakdown of Chl a gives rise to the disassembly of Chl a-binding proteins, particularly reaction centers and core complexes during dark-induced senescence, plus the primary target of the photosynthetic apparatus in sensecing leaves would seem to be Chl a along with the proteins associated with Chl a. The application of ABA promoted the disassembly of the P700 apoproteins in the PSI reaction center and the dimer of D1/D2 apoproteins, and the conversion of the trimeric LHCII apoprotein to the monometirc LHCII apoprotein during the middle stage of leaf senescence, thereby suggesting that ABA accelerates the disassembly of both Chl a-binding and Chl a+b-binding proteins, particularly Chl a-binding proteins during the middle stage of leaf senescence.
Abundance and distribution of phytoplankton in seawater at southwestern East/Japan Sea near Gampo were investigated by HPLC analysis of photosynthetic pigments during summer of 1999. Detected photosynthetic pigments were chlorophyll a, b, $c_{1+2}$ (Chl a, Chl b, Chl $c_{1+2}$), fucoxanthin (Fuco), prasinoxanthin (Pras), zeaxanthin (Zea), 19'-butanoyloxyfucoxanthin (But-fuco) and beta-carotene (B-Car). Major carotenoid was fucoxanthin (bacillariophyte) and minor carotenoids were Pras (prasinophyte), Zea (cyanophyte) and But-fuco (chrysophyte). Chl a concentrations were in the range of $0.16-8.3\;{\mu}g/land$ subsurface chlorophyll maxima were observed at 0-10m at inshore and 30-50 m at offshore. Thermocline and nutricline tilted to the offshore direction showed a mild upwelling condition. Results from size-fraction showed that contribution from nano+picoplankton at Chl a maximum layer was increased from 18% at inshore to 69% at offshore on average. The maximum contribution from nano+picoplankton was found as 87% at St. E4. It was noteworthy that contribution from nano+picoplanktonic crysophytes and green algae to total biomass of phytoplankton was significant at offshore. Satellite images of sea surface temperature indicated that an extensive area of the East/Japan Sea showed lower temperature ($<18\;^{\circ}C$) but the enhanced Chi a patch was confined to a narrow coastal region in summer, 1999. Exceptionally high flux of low saline water from the Korea/Tsushima Strait seemed to make upwelling weak in summer of 1999 in the study area. Results of comparisons among Chi a from SeaWiFS, HPLC and fluorometric analysis showed that presence of Chi b cause underestimation of Chi a about 30% by fluorometric analysis but overestimation by satellite data about 30-75% compared to HPLC data.
To investigate the effects of strong acidic electrolytic water on the chloroplast, barley leaves were treated with strong acidic electrolytic water(pH 2.5). And to investigate the effects of weak acidic electrolytic water on the chloroplast development, etiolated barley leaves were treated with weak acidic electrolytic water(pH 6.5) during greening period. Chl contents, Fo, Fv, and Chl fluorescence quenching coefficient in barley leaves were measured during and after treatment of acidic electrolytic water. The following results were obtained. Chl a, b, and carotenoid were decreased with treatment of strong acidic electrolytic water. Chl contents were significantly decreased than that of the control after 5 min. These results provide evidence that the strong acidic electrolytic water dissimilate the Chl and so that the value of Fo was slightly increased. The strong acidic electrolytic water damaged PS II because Fo was increased and Fv, Fm, and Fv/Fm ratio were decreased. qP, qNP and qE were decreased. On the other hand qI was increased than that of the control. But Chl content and Chl fluorescence patterns were a little changed as the pH increase over 4.0 Chl a, b, and carotenoid were increased with treatment of weak acidic electrolytic water during greening period. Chl contents were significantly increased than that of control after 12 hours greening. These results provide evidence that the weak acidic electrolytic water accelerated the chlorophyll synthesis. And the weak acidic electrolytic water accelerated PS II development because Fv, Fm, qP and Fv/Fm ratio were increased than that of the control.
The formation of chlorophyll-protein complexes (CP-complexes) during the greening of rape cotyledons (Brassica napus cv. Yongdang) was investigated by the SDS-polyacrylamide gel electrophoresis. The total chlorophyll content and Chl a/b ratio were also determined. In addition, the effects of dark treatment on the CP-complex patterns during greening have been examined with respect to their photosynthetic electron transport activity. Greening has brought about the increasein total chlorophyll content and the decrease in Chl a/b ratio, but there have been no changes in Chl a/b ratio after 24 hrs of greening. The light-harvesting chlorophyll a/b-protein complex (LHCP-complex0 was predominant during the initial greening period. Thereafter, the amout of chlorophyll a-protein complex (CP I-complex) was gradually increased. Twenty-four-hr dark treatment immediately after illumination for 6 hrs and 12 hrs resulted in the increase of the Chl a/b ration and the CP I complex, otherwise the decrease of the LHCP-complex. The LHCP/CP I ratio was gradually decreased with further greening, and appeared no change after 48 hrs illumination. The investigation of the photosynthetic electron transport activity indicated that photosystem (PS) II activity (H2Olongrightarrowp-PD*+FeCy**) did not change, but the activity of PS I was increased suddenly due to the dark treatment. The data suggests that the increase of CP I-complex may result in that of P-700, that is, the increase of PS I activity.
