Preparation of Hetero-Chitooligosaccharides and Their Antimicrobial Activity on Vibrio parahaemolyticus

  • This study was performed to investigate the antimicrobial effects of hetero-chitosans and their oligosaccharides on the halophilic bacterium, Vibrio parahaemolyticus. Nine classes of hetero-chitosan oligosaccharides were prepared based on their molecular weights, using an ultrafiltration membrane reactor system with chitosanase and celluase, from partially different deacetylated chitosans, 90%, 75%, and 50% deacetylated chitosan, respectively. Thirty-two strains of V. parahaemolyticus were isolated from various marine organisms such as shellfish, shrimps, octopus, and seabirds. Seventy-five percent deacetylated chitosan showed the highest antimicrobial acitivity. The minimal inhibitory concentration (MIC) was 0.5 mg/ml on 14 strains of V. parahaemolyticus, and MIC of the rest strains (18 strains) was 1.0 mg/ml. In addition, MIC of most hetero-chitosan oligosaccharides was 8.0 mg/ml. The results revealed that the antimicrobial effects of hetero-chitosans and their oligosaccharides against V. parahaemolyticus depend on the degree of deacetylation, their molecular weights, and strains tested.

Improvement of the Stability of Lactobacillus casei YIT 9018 by Microencapsulation Using Alginate and Chitosan

  • Lactobacillus casei YIT 9018 was microencapsulated within alginate or alginate/chitosan double membrane using an air atomizer. Microbiological analysis revealed that the viability of encapsulated L. casei in gastric juice, hydrogen peroxide, and pepsin was 2-3 log cycle higher than that of the nonencapsulated cell. However, the encapsulated cells did not show a signifciant increase in survival when subjected to in virto high acid and 0.6% bile salt condition. Alginate-encapsulated, alginate/chitosan-encapsulated, and nonencapsulated cells were stored at different temperatures eencapsulated cells showed similar stability at $4^{\circ}C$. However, at $22^{\circ}C$, the alginate/chitosan-encapsulted cell was the most stable.

Antibacterial Activity of Low Molecular Weight Water-Soluble Chitosan (저분자량 수용성 키토산의 항균 활성에 관한 연구)

  • Chitosan is a natural polymer derived from chitin that has been widely used as a dietary supplement and in a variety of pharmacological and biomedical applications. In addition, water-soluble chitosan has been used to enhance the stability of chitosan in water and reduce cytotoxic activity induced by acetic acid. In this study, the antibiotic activity and mechanism of low molecular weight water-soluble chitosan (LMWSC; MW1, MW3, MW5, and MW10) were examined in pathogenic bacteria cells and vesicles containing bacterial membrane lipids. MW10 displayed potent antibacterial activity against pathogenic bacteria strains and no cytotoxicity against mammalian cells. In addition, the degree of calcein leakage was examined as a function of lipid composition (PE/PG=7/3 w/w). The results of these experiments demonstrated that MW10 promoted leakage in negatively-charged membranes. Furthermore, confocal microscopy revealed that MW10 was located in the plasma membrane.

Preparation of Flame Retardant and Antibacterial Wood with Composite Membrane Coating

  • A novel flame retardant and antibacterial composite membrane coating for wood surfaces was prepared by adding POSS-based phosphorous nitrogen flame retardant (later referred to as NH2-POSS) and silver nanoparticles (Ag NPs) to chitosan (CS). The effects of NH2-POSS content (mass fractions of CS 0%, 0.5%, 1%, 3%, 5%, and 7%) on the structure and properties of the composite membrane coating on wood were investigated. The composite film was prepared by the method of blending and ducting. Contact angle, tensile property and antibacterial effects of the composite film were measured, and infrared spectroscopy was used. The results show that the addition of NH2-POSS can not only improve the toughness of the membrane, but also the flame retardancy of the membrane, which improves the application of the membrane in wood products. However, with the addition of NH2-POSS, the transparency of the composite membrane was weakened. The inhibitory effect of the composite membrane on the growth of Escherichia coli was enhanced with the increase in Ag NPs. This research provides a foundation for the application of functional wood.

Antimicrobial Activity of Hetero-Chitosans and Their Oligosaccharides with Different Molecular Weights

  • This study was performed to investigate the antimicrobial effects of hetero-chitosans and their oligosaccharides against three Gram-negative bacteria and five Gram-positive bacteria. Nine classes of hetero-chitosan oligosaccharides consisted of partially deacetylated chitosans; 90%, 75%, and 50% deacetylated chitosans. Based on molecular weight, they were prepared using an ultrafiltration membrane reactor system. Seventy-five percent deacetylated chitosan showed the highest antimicrobial acitivity as compared with the 90% and 50% deacetylated chitosan, and the activity was dependent on their molecular weights. It was apparent that the growth of Gram-negative bacteria is less inhibited in the presence of the heterochitosans and their oligosaccharides than Gram-positive bacteria. These results revealed that the antimicrobial effects of hetero-chitosans and their oligosaccharides depend on the degree of deacetylation, and their molecular weights.

Colon Delivery of Prednisolone Based on Chitosan Coated Polysaccharide Tablets

  • Colon drug delivery is advantageous in the treatment of colonic disease and oral delivery of drugs unstable or suceptible to enzymatic degradation in upper GI tract. In this study, multilayer coated system that is resistant to gastric and small intestinal conditions but can be easily degraded by colonic bacterial enzymes was designed to achieve effective colon delivery of prednisolone. Variously coated tablets containing prednisolone were fabricated using chitosan and cellulose acetate phthalate (CAP) as coating materials. Release aspects of prednisolone in simulated gastrointestinal fluid and rat colonic extracts (CERM) were investigated. Also, colonic bacterial degradation study of chitosan was performed in CERM. From these results, a three layer (CAP/Chitosan/CAP) coated system exhibited gastric and small intestinal resistance to the release of prednisolone in vitro most effectively. The rapid increase of prednisolone in CERM was revealed as due to the degradation of the chitosan membrane by bacterial enzymes. The designed system could be used potentially used as a carrier for colon delivery of prednisolone by regulating drug release in stomach and the small intestine.

Antifungal Activity of Chitosans on Candida albicans and Trichophyton rubrum and its Induction of Apoptosis (키토산의 Candida albicans와 Trichophyton rubrum에 대한 항진균 작용과 Apoptosis 유도작용)

  • The antifungal activity of chitosan ($M.W.\;400,000{\sim}500,000$) and chitooligosaccharide ($M.W.\;3,500{\sim}5,000$) was investigated against Candida albicans and Trichophyton rubrum. Chitosan showed antifungal activity against C. albicans and T. rubrum at 50 and 100 ng/ml, respectively while chitooligosaccharide did not suppress the growth of fungus. The mode of antifungal activity of chitosan was found to be fungicidal activity. In order to investigate the induction of apoptosis by chitosan, exposure of phosphatidylserine on the surface of the cytoplasmic membrane was observed by the FITC-annexin V reaction. The results showed that chitosan induced apoptosis on C. albicans.

Effects of Chitosan on the Mouse Hepatotoxicity Toxicated by Mercury (수은으로 중독된 흰쥐의 간독성에 대한 키토산의 효과)

  • This study aims demonstrate the effect of chitosan, one of the natural chelator, on the ultrastructural changes in the mouse liver caused by $HgCl_2$. The experimental group was divided in two groups; group A and group B. The group A administrated $HgCl_2$ (5.0 mg/kg) to the oral. The group B treated with $HgCl_2$ (5.0 mg/kg) and chitosan (3%) solution, 2 times/day). Each group was observed 24, 48, 72 and 96 hours after treated $HgCl_2$ and chitosan. Histological changes of the livers were investigated by electron microscope. 1. Croup A Nuclear membrane was shrinked. The inner and outer membrane of the mitochondria were dilated. Destruction of lamellae of rough endoplasmic reticulum showed. Smooth endoplasmic reticulum showed over cytoplasm. 2. Group B Nuclear membrane was more rounded, The cristae of the mitochondria were almost normal shape and electron-density showed compacted. Dilation of inner cavity of rough endoplasmic reticulum showed at the pre-time but formed typical lamellae at the 48Hrs. Smooth endoplasmic reticulum showed over cytoplasm. Therefore, we concluded that chitosan has significantly protective effects in liver to harmful $HgCl_2$.
