  • 원자력발전소의 이종용접부에서 일차응력부식균열이 발생하고 있으며 용접부의 잔류응력이 균열발생 및 성장에 기여할 수 있다. 용접부의 잔류응력은 기본적으로 용접에 의해 형성되지만 기계가공에 의해 표면잔류 응력상태가 변화할 수 있다. 본 논문에서는 기계가공이 원전재료인 SA508과 오스테나이트 스테인리스강에 표면잔류응력에 미치는 영향을 평가하였다. 이를 위해 SA508, TP304, F316L 재료를 연마, 연삭, 방전가공으로 가공한 후 표면에 형성되는 잔류응력을 측정하였다. 측정방법은 구멍뚫기법과 엑스선회절법을 사용하였다. 기계가공방법에 따라 각 재료에 미치는 잔류응력의 크기 및 방향, 잔류응력이 형성되는 깊이 등의 특성을 확인하였다.

  • Tumor of the trachea are uncommon. Trachea tumor are usually diagnosed late in their natural history, because the tracheal lumen may be extensively involved before symptoms occur, and because symptoms may be confused with those of asthma. Some 19% to 29% of the trachea tumor have laryngeal or lung malignancy. Squamous carcinoma is by far the most frequent among tracheal tumor, although adenocarcinoma and oat cell carcinoma have been described so. Oat cell carcinoma in the trachea are aggressive tumors as they are in the lung and elsewhere. Oat cell carcinoma as well as squamous cell carcinoma are related with tobacco exposure. We presented a case of small cell carcinoma nearly obstructing the trachea and right main bronchus which was partially removed after bronchoscopic ethanol injection, along with a review of the literature.

Curcumin Derivatives Inhibit the Formation of Jun-Fos-DNA Complex Independently of their Conserved Cysteine Residues

  • Curcumin, a major active component of turmeric, has been identified as an inhibitor of the transcriptional activity of activator protein-1 (AP-1). Recently, it was also found that curcumin and synthetic curcumin derivatives can inhibit the binding of Jun-Fos, which are the members of the AP-1 family, to DNA. However, the mechanism of this inhibition by curcumin and its derivatives was not disclosed. Since the binding of Jun-Fos dimer to DNA can be modulated by redox control involving conserved cysteine residues, we studied whether curcumin and its derivatives inhibit Jun-Fos DNA binding activity via these residues. However, the inhibitory mechanism of curcumin and its derivatives, unlike that of other Jun-Fos inhibitors, was found to be independent of these conserved cysteine residues. In addition, we investigated whether curcumin derivatives can inhibit AP-1 transcriptional activity in vivo using a luciferase assay. We found that, among the curcumin derivatives examined, only inhibitors shown to inhibit the binding of Jun-Fos to DNA by Electrophoretic Mobility Shift Assay (EMSA) inhibited AP-1 transcriptional activity in vivo. Moreover, RT-PCR revealed that curcumin derivatives, like curcumin, downregulated c-jun mRNA in JB6 cells. These results suggest that the suppression of the formation of DNA-Jun-Fos complex is the main cause of reduced AP-1 transcriptional activity by curcuminoids, and that EMSA is a suitable tool for identifying inhibitors of transcriptional activation.

이종금속 평판 맞대기용접의 용접잔류응력 분포특성 (Distribution Characteristics of Weld Residual Stress on Butt Welded Dissimilar Metal Plate)

  • 본 연구에서는 최근 원자력 발전소에서 이종금속용접 접합 소재로 많이 사용되고 있는 SA 508과 Type 304 스테인리스강의 이종금속 용접잔류응력 분포특성을 살펴보기 위하여 이종금속 용접시편을 제작하여 직접 용접을 실시하고 X-선 회절법을 이용하여 용접시험편의 표면에서 용접 잔류응력을 측정하였다. 또한 3차원 유한요소해석을 통하여 이종금속 맞대기용접을 전산모사하고 시편의 용접잔류응력 분포를 구하였다. 측정한 잔류응력과 유한요소해석 결과 계산된 잔류응력을 고찰함으로써 3차원 평판 이종금속 맞대기 용접부의 잔류응력분포 특성을 살펴보았다.

수용모델을 이용한 서울지역 미세입자 (PM2.5)에 영향을 미치는 배출원 특성에 관한 연구 (A Study on the PM2.5 Source Characteristics Affecting the Seoul Area Using a Chemical Mass Balance Receptor Model)

  • The purpose of this study is to study the $PM_{2.5}$ source characteristics affecting the Seoul area using a chemical mass balance (CMB) receptor model. This study was also to evaluate the $PM_{2.5}$ source profiles, which were directly measured and developed. Asian Dust Storm usually occurred in the spring, and very high $PM_{2.5}$ concentrations were observed in the fall among the sampling periods. So the ambient data collected in the spring and fall were evaluated. The CMB model results as well as the $PM_{2.5}$ source profiles were validated using the diagnostic categories, such as: source contribution estimate, t-statistic, R-square, Chi-square, and percent of total mass explained. In the spring months, the magnitude of $PM_{2.5}$ mass contributors was in the following order: Chinese aerosol $(31.7\%)>$ secondary aerosols ($22.3\%$: ammonium sulfate $13.4\%$ and ammonium nitrate $8.9\%)>$ vehicles ($16.1\%$: gasoline vehicle $1.4\%$ and diesel vehicles $14.7\%)>$biomass burning $(15.5\%)>$ geological material $(10.5\%)$. In the fall months, the general trend of the $PM_{2.5}$ mass contributors was the following: biomass burning $(31.1\%)>$ vehicles ($26.9\%$: gasoline vehicle $5.1\%$ and diesel vehicles $21.8\%)>$ secondary aerosols ($23.0\%$: ammonium sulfate $9.1\%$ and ammonium nitrate $13.9\%)>$ Chinese aerosol $(10.7\%)$. The results show that the $PM_{2.5}$ mass in the Seoul area was mainly affected by the Chinese area.

당뇨병성 족부 질환에 의한 하지 절단 후 임상적 분석과 결과 (Clinical Analysis and Results after the Amputations of Lower Extremities due to Diabetic Foot)

  • Purpose: The authors evaluated the clinical results and prognosis after amputating the lower extremity due to diabetic foot. Materials and Methods: From 1991 to 2003, the patients who had suffered amputation of his lower extremity due to diabetic foot ulcer were evaluated retrospectively. 79 patients were male and 6 patients were female. The author evaluated the patient who had the ipsilateral additional surgery, contralateral amputation, level of blood sugar, combined disease and mortality rate within 5 years from medical record. Statistical analysis was done by Chi-square test and Kaplan-Meier survival test. Results: Mean age of patients who had first experienced amputation was 63.4 years old. The mean duration of diabetes until amputation was $14.5{\pm}7.5$ years. Major amputations were 50 cases and minor amputations 35 cases. 20 patients (23.5%) were suffered ipsilateral secondary surgery including revised stump. Overall 5-year mortality rate was 18.8% (16 cases). Death rate within 1 year was 8.2% (7 cases), mortality rate within 3 years was 14.1% (12 cases). 5-year mortality rate after major amputation was 20% (10 cases) and after minor amputation was 17.1% (6 cases). It was statistically significant (p<0.05). Patient who underwent more than 2 combined vascular related disease had higher mortality rate than diabetic amputee without combined disease (p<0.05). Conclusion: Mortality rate after major amputation was significant higher than amputation after minor amputation in diabetic patients from our data.

  • Since the birth of Dolly using fully differentiated somatic cells as a nuclear donor, viable clones were generated successfully in many mammalian species. These achievements in animal cloning demonstrate developmental potential of terminally differentiated somatic cells. At the same time, the somatic cell nuclear transfer (SCNT) technique provides the opportunities to study basic and applied biosciences. However, the efficiency generating viable offsprings by SCNT remains extremely low. There are several explanations why cloned embryos cannot fully develop into viable animals and what factors affect developmental potency of reconstructed embryos by the SCNT technique. The most critical and persuasive explanation for inefficiency in SCNT cloning is incomplete genomic reprogramming, such as DNA methylation and histone modification. Numerous studies on genomic reprogramming demonstrated that incorrect DNA methylation and aberrant epigenetic reprogramming are considerably correlated with abnormal development of SCNT cloned embryos even though its mechanism is not fully understood. The SCNT technique is useful in cloning farm animals because pluripotent stem cells are not established in farm animal species. Therapeutic cloning combined with genetic manipulation will help to control various human diseases. Also, the SCNT technique provides a chance to overcome excessive demand for the organs by production of transgenic animals as xenotransplantation resources. Here, we describe the factors affecting the efficiency of generating cloned farm animals by the SCNT technique and discuss future directions of animal cloning by SCNT to improve the cloning efficiency.

Proteomic analysis of rice mutants susceptible to Magnaporthe oryzae

  • To identify genes involved in rice Pi5-mediated disease resistance to Magnaporthe oryzae, we compared the proteomes of the RIL260 rice strain carrying the Pi5 resistance gene with its susceptible mutants M5465 and M7023. Proteins were extracted from the leaf tissues of both RIL260 and the mutant lines at 0, 24, and 48 h after M. oryzae inoculation and separated by two-dimensional polyacrylamide gel electrophoresis (2-DE). Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) analysis identified eight proteins that were differently expressed between the resistant and susceptible plants (three down- and five up-regulated proteins in the mutants). The down-regulated proteins included a triosephosphate isomerase (spot no. 2210), a 2,3-bisphosphoglycerate-independent phosphoglycerate mutase (no. 3611), and an unknown protein (no. 4505). In addition, the five up-regulated proteins in the mutants were predicted to be a fructokinase I (no. 313), a glutathione S-transferase (no. 2310), an atpB of chloroplast ATP synthase (no. 3616), an aminopeptidase N (no. 3724), and an unknown protein (no. 308). These results suggest that proteomic analysis of rice susceptible mutants is a useful method for identifying novel proteins involved in resistance to the M. oryzae pathogen.

수열합성법으로 제조된 지르코니아의 나노분말 특성 (Characteristics of Zirconia Nanoparticles with Hydrothermal Synthesis Process)

  • 지르코니아 나노분말을 수열합성법으로 제조하였으며, 반응온도, 반응시간, 침전제의 종류 및 농도, 전구체의 종류를 실험 변수로 하였고, 각각의 제조된 분말을 X-선 회절(XRD)과 주사전자현미경(FE-SEM)을 통해 입자크기 및 결정상을 측정하였다. 침전제의 농도 증가에 따라 지르코니아 입자크기가 증가하였으며, 입자의 결정화도도 높아졌고, KOH 보다 NaOH 침전제를 사용하였을 경우, 입자 성장 속도가 증가하였다. 이는 입자의 크기 컨트롤에 있어 NaOH를 사용하는 경우보다 KOH를 사용하는 것이 더 효과적이었다. 4 h의 수열반응시간에서는 4 h의 수열반응시간에서는 비정질의 지르코니아 입자가 발견되었지만, 8 h 이상 합성 시, 단사정상의 지르코니아 입자가 생성됨을 확인하였고, 반응시간이 길어짐에 따라 지르코니아 입자의 폭이 소폭으로 줄어들고, 길이는 소폭 늘어남을 확인하였다. 동일한 합성 조건에서 전구체로서 zirconium (IV) acetate, zirconium nitrate, zirconium chloride 중에서 zirconium chloride를 사용하여 합성하였을 경우, 입자의 크기가 가장 작게 형성되었다.
