  • The purpose of this study was to investigate effects of inner skin of chestnut on the activation of a living body's function (regional cerebral blood flow and mean arterial blood pressure in Sprague-Dawley rats, proliferation of thymocytes in normal mice and L1210 cells transplanted mice) . We used inner skin of chestnut extract(Sample A : inner skin of chestnut-panbroiled after driedextract (100$\^{C}$ ), Sample B , inner skin of chestnut-panbroiled-extract(100$\^{C}$ ) , Sample C : inner skin of chestnut -panbroiled after dried-extract(80$\^{C}$ ), Sample D : inner skin of chestnut-panbroiled-extract(80$\^{C}$)} Regional cerebral blood flow(rCBF) and Mean arterial blood pressure(MABP) were tested using Leser -Doppler Flowmetry(LDF), and the proliferation of thymcytes was tested using a colorimetric tetrazoliun assay ( MTT assay) The experimental results as follows 1. rCBF was significantly increased by Sample C in a dose-dependent manner. 2. MABP was not changed by Sample C in a 0.1mg/kg∼10.0mg/kg treated group. 3. Proliferation of thymocytes was not changed by Sample C in normal mice. 4. Proliferation of thymocytes was significantly accelerated by Sample C in L1210 cells transplanted mice.

우피의 울금 염색시 사용된 율피의 매염 효과 (Mordanting effect of Chestnut's inner skin on the cow leather dyed with Turmeric powder)

  • This research was carried out to find the mordant effect of Chestnut's inner skin on the cow leather dyed by Turmeric powder. The best proper mordanting conditions were examined by changing mordant method, concentration, temperature, bath ratio, time, and repetition. Also dyeability and surface color changes were evaluated by various mordanting methods. The optimum mordanting conditions of chestnut's inner skin extract on the cow leather were pre mordant, 80%, $50^{\circ}C$, 50:1, 40minutes, 4 repetition. The K/S values as a mordant were higher in pre mordant than post mordanting condition. Although the ${\Delta}E$ was slightly higher in post mordant than pre mordant, it was too small to find any means. The surface colors of all dyed cow leathers were yellow. Among light, dry cleaning, and abrasion fastnesses, only light fastness was increased one degree, others were same degrees.

Effects of the Chestnut Inner Shell Extract on the Expression of Adhesion Molecules, Fibronectin and Vitronectin, of Skin Fibroblasts in Culture

  • The inner shell of the chestnut (Castanea crenata S. et Z., Fagaceae) has been used as an anti-wrinkle/skin firming agent in East Asia, and preliminary experiments have found that a 70% ethanol extract from this plant material can prevent cell detachment of skin fibroblasts from culture plates. In order to examine the molecular mechanisms underlying this phenomenon, its effects on the expression of adhesion molecules, such as fibronectin and vitronectin, were investigated using the mouse skin fibroblast cell line, NIH/3T3. Using fixed-cell ELISA, Western blotting and immunofluorescence cell staining, it was clearly demonstrated that the chestnut inner shell extract enhanced the expression of the cell-associated fibronectin and vitronectin. Scoparone (6,7-dimethoxycoumarin), isolated from the extract, also possessed similar properties. These findings suggest that the enhanced expression of the adhesion molecules may be one of the molecular mechanisms for how the chestnut inner shell extract preventing cell detachment and may be also responsible for its anti-wrinkle/skin firming effect.

국내산 식물성 식품중 페놀성 물질의 함량 분석 (Analysis of Phenolic Substances Content in Korean Plant Foods)

  • 한국인의 식생활에서 널리 이용되고 있는 식물성 식품 및 민간요법에서 이용되는 비상용식품 45가지를 선택하여 총페놀함량은 Folin-Denis 법으로, 축합형 탄닌의 함량은 vanillin 법과 modified vanillin 법으로, 단백질 침전성 페놀물질의 함량은 단백질 침전법으로 측정하였다. 총페놀 함량(건물기준)은 $0.1{\sim}5.8%$로 감잎, 밤속껍질, 모과, 호두, 해바라기씨, 칡뿌리에서는 2% 이상의 높은 함량을 보였다. 축합형 탄닌함량(건물기준)은 $0{\sim}48%$로 모과 밤속껍질에서 매우 높이 나타났다. 단백질 침전성 페놀성분의 함량(건물 기준)은 $0.4{\sim}2.2%$로 밤속껍질, 호두, 모과에서 매우 높았고 그들의 단백질 침전능은 트립신보다 펩신이나 알부민에서 더 잘 나타났다. 식품중 페놀성물질 함량간의 상관관계를 보면 총페놀 함량과 단백질 침전성 페놀물질 함량(r=0.65)과 축합형 탄닌함량(r=0.56)간에 높은 상관관계를 보였으나, 축합형 탄닌함량과 단백질 침전성 페놀물질 함량간에는 비교적 낮은 상관관계(r=0.39)를 보였다. 축합형 탄닌의 중합도를 예측하는 vanillin/Folin-Denis 비를 살펴본 결과 모과, 밤속껍질, 수수에서 높은 값을 보여 결국 낮은 중합도를 보였다. 메밀, 도토리, 쑥, 칡 등을 이용한 가공식품에서의 페놀물질 함량은 원료상태에서 보다 낮은 함량을 보여 식품단백질의 이용율에는 큰 장애를 주지 않을 것으로 판단되었다.

Antioxidative Activity of the Extract from the Inner Shell of Chestnut

  • The ethanolic extract of chestnut (Castanea crenata S. et Z., Fagaceae) inner shell (CISE) and one of its components, ellagic acid (EA), were evaluated for their protective effects against 1, 1-diphenyl-2-picryl hydrazine (DPPH) free radical generation and hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line. CISE and EA were shown to possess the free radical scavenging effect against DPPH radical generation, significantly. They were also found to strongly inhibit hydrogen peroxide-induced DNA damage from Chinese hamster lung (CHL) cell, assessed by single cell gel electrophoresis assay and 8-hydroxy -2'-deoxy guanosine (8-OH-2'dG) assay. Furthermore, topical application of CISE [$12.5\%$(w/w) cream] and ellagic acid [$1.0\%$(w/w) cream] for 14 days potently inhibited malondialdehyde (MDA) formation of mouse dorsal skin (a marker of lipid peroxidation) induced by ultraviolet B exposure. Therefore, CISE and its component, ellagic acid, may be the useful natural antioxidants by scavenging free radicals, inhibition of lipid peroxidation and protecting oxidative DNA damage when topically applied.

국내산 밤 일부 품종의 기능성 성분분석과 항산화 및 대식세포 활성 (Antioxidative and Macrophage Phagocytic Activities and Functional Component Analyses of Selected Korean Chestnut (Castanea crenata S. et Z.) Cultivars)

  • 국내산 밤 일부 품종(단택, 대보, 석추, 옥광, 병고)의 기능성 성분 분석과 이들 물질들의 항산화력 및 밤 추출물이 대식세포 활성에 미치는 영향력을 알아보았다. 국내산 밤 일부 품종의 내피와 과육에서 coumarin, gallic acid 그리고 catechin이 HPLC에 의해 검출되었다. 밤 내피에 가장 풍부한 기능성 성분은 catechin이고 그 다음 gallic acid와 coumarin 순이었으나, 과육에서는 gallic acid만 검출되었다. 따라서 밤의 기능성 성분인 gallic acid와 catechin의 항산화력을 DPPH radical 소거작용과 SOD 유사활성을 통해 알아보았다. Gallic acid과 catechin을 6.0 mg/100 g 처리하였을 때 DPPH radical 소거작용이 각각 69.4%와 38.3%였으며, gallic acid의 DPPH radical 소거작용은 농도 의존적이었다. Gallic acid를 동일한 농도의 catechin과 비교하면 모든 농도 구간에서 catechin보다 gallic acid가 높은 DPPH radical 소거활성을 나타내었다. 그러나 SOD 유사활성은 gallic acid보다 catechin이 높았다. 더하여 밤의 과육, 노란과육 및 흰과육 추출물들이 대식세포 활성에 미치는 영향력을 알아보았다. 병고 흰과육 추출물은 현저하게 대식세포 활성을 증가시켰고, 병고, 단택, 대보 및 옥광의 노란과육 추출물 역시 아무 것도 처리하지 않은 세포와 비교하여 현저히 대식세포를 활성화시켰다. 노란과육 추출물이 흰과육 추출물보다 더욱 더 대식세포 활성을 증가시켰고, 5품종 중 병고가 대식세포 활성을 가장 증가시켰다.
