The major aim of this work is the research of secondary metabolites isolated from the aerial parts of Petasites japonicus. The plant material is extracted with a polar solvent, which is 95% by volume methanol at room temperature. The concentrated extract was partitioned as EtOAc, n-BuOH, and $H_2O$ fractions. From the EtOAC and n-BuOH fraction, two bakkenolides and two caffoylquinic acid were isolated using the Diaion HP-20, silica gel, ODS-A, and Sephadex LH-20 column chromatographies. According to the results of the results of physico-chemical and spectroscopic data including NMR, MS and UV. The chemical structures of the compounds were respectively determined as bakkenolide B (1), bakkenolide D (2), 1,5-dicaffeoylquinic acid (3), and 5-O-caffeoylquinic acid (4). These results suggest that the compounds isolated from the aerial parts of this plant were almost identical with known components of Petasites japonicus. However, it is necessary to investigate more about the difference of amounts of constituents according to harvest area and time.

In the course of study on the roles of phytotoxins in the pathogenicity of Botrytis cinerea, we isolated two novel phytotoxins. They were identified as 3-O-acetyl botcinol and 3-O-acetyl botcinolide. In this study, we investigated correlation between the two phytotoxins and the pathogenicity of B. cinerea. In liquid cultures, the two phytotoxins were not produced by three low pathogenic isolates out of 25 B. cinerea isolates. Among strong or moderate pathogenic isolates, some produced the two phytotoxins, but the others did not. On the other hand, the ethyl acetate extracts of fermentation broths of 10 out of 25 isolates showed phytotoxic activity against various plants tested in a whole plant assay. The phytotoxins were detected in all of the 10 phytotoxic ethyl acetate extracts. In planta, the two phytotoxins were detected in all of the plant tissues infected with strong pathogenic isolates. However, there was no correlation between the ability of B. cinerea isolates to produce the two phytotoxins and their pathogenicities. The two phytotoxins began to detect in tomato plant tissues infected with B. cinerea 2-16 at 3 days after inoculation, increased gradually till 4 days after inoculation, and then decreased. The above results suggest that 3-O-acetyl botcinol and 3-O-acetyl botcinolide are one of pathogenicity factors for B. cinerea, but not a primary determinant of its pathogenicity.
This study was carried out to develop the improved useful mutants for yield or composition of stevia plants using the gamma ray or chemical mutagens treatments. The seeds of stevia 'Suwon No. 11' were irradiated up to 400 Gy of gamma ray. Chemical mutagens were treated on the seeds of the 'Suwon No. 11' using 0.07% colchicine, 10 mM sodium azide, or 10 mM NMU for various durations. The germination rate, and shoot and root growth of seedling were estimated at 30 days after gamma ray irradiation or chemical mutagen treatment, and the plant height, the number of branches, and leaf length and width were examined at 3 months after mutagenesis treatments. In the case of gamma ray treatments, the germination rate and early-stage growth were decreased as the increase of radiation dose, and the 50% lethal dose was found to be 200 Gy. the plant height was decreased as the increase of radiation dose, while the number of branches per plant and leaf length were increased. Leaf shape was modified to the relatively longer one compared to the control, which was identified more apparently at the treatments of higher than 150 Gy. In the treatment of chemical mutagens, the rate of germination and survival were decreased as the increase of incubation time. The 50% lethal dose for germination rate were identified as the conditions of the 15 hours incubation in 0.07% colchicine, the 4 hrs in 10 mM sodium azide, and the 2 hrs in 10 mM NMU, in the three chemical mutagens treatments. Chemical mutagens had no influence on shoot growth, while root growth was increased, especially as the incubation time was extended. The highest root growth occurred in the NMU treatment at 6 hrs incubation time. The plant height was decreased as the increase of incubation time in the chemical mutagens treatments. Among the chemical mutagens, NMU was the most effective to induce the mutants with long-shaped or the least lobed leaves.

We discovered two novel phytotoxins produced by the pathogenic fungus, Botrytis cinerea. Among the twenty-five B. cinerea isolates, which were obtained from various host plants in 1994 and 1996, twenty-two showed strong or moderate pathogenicity on five plants such as cucumber, tomato, red pepper, tobacco and Chinese cabbage. The culture filtrate of the B. cinerea 2-16 strain showed the most potent phytotoxic activity in a tobacco leaf-wounding assay. Two novel phytotoxins were isolated from the liquid cultures of B. cinerea 2-16 by ethyl acetate extraction, flash silica gel column chromatography, silica gel column chromatography, Sephadex LH-20 column chromatography, preparative TLC and subsequently preparative HPLC. Their chemical structures were determined to be 3-O-acetyl botcinol and 3-O-acetyl botcinolide, respectively, by mass and NMR spectral analyses. These two phytotoxins caused leaf necrosis in a leaf-wounding bioassay, and significant electrolyte leakage from leaf tissues of tobacco. In the two bioassays tested, 3-O-acetyl botcinol exhibited stronger phytotoxic activity than 3-O-acetyl botcinolide. This is the first report on the production of both 3-O-acetyl botcinol and 3-O-acetyl botcinolide from B. cinerea.

An antagonistic bacterium of Bacillus subtilis BD0310 against Colletotrichum theae-sinensis was isolated from the phylloplane of tea trees at a tea plantation in Korea. SC (suspension concentrate)-type biofungicide was formulated with the antagonist. Cell viability and antifungal activity of B. subtilis were maintained in the formulation more than 12 months at room temperature. The antagonist was sensitive only to copper sulfate among the chemical pesticides currently registered for tea trees in Korea. Greenhouse application demonstrated that the biofungicide controlled more effectively the disease in a protective mode than in a curative mode. Field trial showed that alternate applications of the biofungicide and chemical fungicide were more effective in controlling tea anthracnose than single application of the biofungicide or chemical fungicide with less use of chemicals. This study suggests that the biofungicide of B. subtilis 8D0310 is an effective method for biological control of anthracnose in tea plantations.
Characteristics of sophorolipid and rhamnolipid were evaluated as antifungal agents against plant pathogenic fungi. Eight percent of mycelial growth of plant pathogen (Phytophthora sp. and Pythium sp.) was inhibited by 200 mg/l of rhamnolipid or 500 mg/l of sophorolipid, and zoospore motility of Phytophthora sp. decreased by $90\%$ at 50 mg/l of rhamnolipid and $80\%$ at 100 mg/l of sophorolipid. The effective concentrations for zoospore lysis were two times higher than those of zoospore motility inhibition. The highest zoospore lysis was observed with Phytophthora capsici; $80\%$ lysis at 100 mg/I of di-rhamnolipid or lactonic sophorolipid, showing the dependency of structure on the lysis. In the pot test, the damping-off disease incidence ratio decreased to $42\%\;and\;33\%$ of control value at 2,000 mg/l sophorolipid and rhamnolipid, respectively. These results showed the potential of microbial glycolipid biosurfactants as an effective antifungal agent against damping-off plant pathogens.
Spherical nanosized cobalt powder with an average size of 150-400 nm was successfully prepared at room temperature from cobalt sulfate heptahydrate ($CoSO_4{\cdot}7H_2O$). Wet chemical reduction method was adopted to synthesize nano cobalt powder and hypophosphorous acid ($H_3PO_2$) was used as reduction agent. Both the HCP and the FCC Co phase were developed while $CoSO_4{\cdot}7H_2O$ concentration ranged from 0.7 M to 1.1 M. Secondary phase such as $Co(OH)_2$ and $CO_3O_4$ were also observed. Peaks for the crystalline Co phase having HCP and FCC structure crystallized as increasing the concentration of $H_3PO_2$, indicating that the amount of reduction agent was enough to reduce $Co(OH)_2$. Consequently, a homogeneous Co phase could be developed without second phase when the $H_3PO_2/CoSO_4{\cdot}7H_2O$ ratio exceeded 7.

An experiment of alum sludge application was carried out on a mountainous site, to know its effect on the growth of forage sorghum hybrid, Pioneer 93 1 (Sorghum bicolor x S. bicolor). When alum sludge with 80% water content was applied at the level of 133 ton per ha, the effect of the application on the growth of Pioneer 931 was much smaller than that of chemical fertilizer. But Eresh yield of the forage, with the sludge application, was greater than that without fertilizer when the soil is not fertile. But when the soil was comparatively fertile, the sludge application gave an adverse affect on the growth of the forage plant. The content of exchangeable K in the soil has a very close positive relation with the Eresh yield and plant height of the plant, while that of exchangeable Ca has a close negative relation with those growing factors. There was a tendency of yield decrease of the forage plant, when alum or compost was applied with NPK to the comparatively infertile soil especially for alum sludge. As a result, when soil is not fertile more attention will be necessary for applying alum sludge with chemical fertilizers. But to soil with better fertility, it is recommended to use alum sludge with chemical fertilizer to obtain more yield of the forage plant.
The objectives of this study are to investigate the suitability of various coal fly ashes and incineration ashes for zeolite synthesis. Zeolite P and hydroxysodalite are produced from coal fly ash and paper sludge incineration ash. When soluble and acid-soluble materials in incineration fly ash are removed by the water washing or acid washing before hydrothermal synthesis, hydroxysodalite can be produced. The factors to make solid-liquid separation difficult are the calcium component and the unburned carbon in ash.
