The new cationic meso-substituted N-quarternized 4-pyridylporphyrins and their metal derivatives were synthesized as novel chemotherapeutics. The level of DNA damage induced by porphyrins TOBut4PyP, TOBut4PyP, TOEt4PyPMn and TOBut4PyPMn and its dependence on the chemical structure of compounds were analyzed by the Comet-assay. On the base of data obtained, the investigated porphyrins may be arranged by their genotoxic activity in the following order: TOEt4PyP>TOEt4PyPMn>TOBut4PyP>TOBut4PyPMn. Thus, i) the genotoxicity of the Mn-derivatives of TOEt4PyP and TOBut4PyP is higher than the original porphyrins and ii) the genotoxicity of TOEt4PyP and TOEt4PyPMn is increased after substitution of a butyl radical for ethyl one. The applied Comet-assay permits to reveal the dependence of DNA damage induction on the chemical structure of porphyrins.
In vitro antimutagenic activity of methanol extract from brrwn rice and its physico-chemical characteristics were investigated using Salmonella typhimurium reversion assay and SOS chromotest. Methanol extracts of brown rice were not mutagenic compared with direct and indirect, mutagenicities of 4NQO (4-nitroquinoline oxide), 2NF(2-nitrofluorene), Trp-p-1(3-Amino-1,4-dimethyl-5H-pyrido-[4,3-b]indole), and Trp-p-2(3-Amino-1-methy-5H-pyrido-[4,3-b]indole). Antimutagenic activity against the indirect mutagenicties induced by Trp-p-1, Trp-p-2 and AFB1 (aflatoxin B1) was found in methanol extract. Even though antimutagenic activity showed dose-dependent, it remained constant at inhibition rate ranging 60~90% when the concentration was abov 3mg/plate in the S. typhimurium reversion assay and 0.2~0.6 mg/assay in the SOS chromotest. The antimutagenic activity of the methanol extracts was stable at various pH (2, 7 and 10), temperatures (60, 80 and 10$0^{\circ}C$)and heation times (2, 4, 6, 8, 10 min at 10$0^{\circ}C$).

Natural substances have various physiological activities. Substances isolated from natural substances are known to be safer and more potent than pharmaceuticals. Potatoes not only act as energy sources but also contain active ingredients such as vitamins and minerals. In particular, the potato contains a large amount of polar compounds, including the saponin in the polar compounds, and the physiological activity of the saponins, such as immunity enhancement, antioxidant and anti-inflammatory is known. In this study, the antioxidative activity of polar compounds from five potatoes was examined by chemical base anti-oxidation assay and cell based anti-oxidation assay. In the chemical base anti-oxidation assay, DPPH experiment showed activity in the order of Hongyoung, Haryung, Seohong, Sumi, and Jayoung. In the LPA experiment, IC50 was lower in the order of Jayoung, Seohong, Sumi, Hongyoung, and Haryung. In the cell based anti-oxidation assay, the smallest amount of ROS was generated when the compound was derived from Haryung and hongyoung, and strong SOD activity was observed in Sumi and Jayoung. The results of this study reveal the antioxidative effect of polar compounds extracted from various kind of potatoes, which will enable the acquisition of new bioactive candidates and the establishment of new profit generation models for farmers.

A number of uracil derivatives have been developed as anti-AIDS drugs having a mechanism of inhibiting cellular reverse transcriptase. A simple and rapid assay technique for recently synthesized KR-V analogues was developed using a high-performance liquid chromatography, and oral pharmacokinetics was examined for assessing their oral bioavailabilites. Plasma samples were analyzed by reversed-phase HPLC using an ODS column with an ultraviolet detection system. All the analogues were eluted within 12 min and the LOQ was 15-30 ng/$m\ell$. The extraction recoveries were higher than 85%, except KR-V1039, 1068 and 1720 having ester group. This chromatic method was well applied to the kinetic studies for KR-V analogues. Among 16 analogues tested in the present work, the 6 compounds including KR-V1123, 1122, 1784, 1783, 1736 and 1700 were found to be bioavailable for oral administration to rats.
In this study, sixty chemicals including 47 pesticides were screened for estrogenic activity using E-screen assay. MCF-7 cell, used in E-screen assay, is known to be proliferated by addition of estrogenic chemicals. Eight of the measured pesticides showed estrogenic activity at the concentration range of 100-10,000nM. Their relative proliferative effect (RPE) and the relative proliferative potency (RPP) were 20-65% and 0.01-1.0%, respectively, when compared with 1.0nM of $\beta-Estradiol-17-acetate(E_{2}).$ DDVP and diazinone showed most strong estrogenic potency(RPP; 1.0%) and effect(RPE; 65%) of the eight pesticides. These results are in agreement with estrogenic activity of bisphenol A is known as a positive endocrine disrupter. Also, in this study, paraquat, DDVP, 4-chloro-3-methylphenol, 2,4,6-trichlorophenol and diazinon of the measured pesticides are estimated to estrogenic chemical.
Safflower is called as the 'beneficial flower' because 'it helps human health', and it was introduced as red flower in Tonguibogam due to the red color of floral leaf. From old times, it has been used for the material of cloth and rouge. Recently, polyphenol compound, the main ingredient of safflower, known as anti-aging and anti-oxidizing material in the healthy food industry becomes the emerging hot topic. This study aims to confirm by DDT (Disk Diffusion Test) assay, MTT assay, and NF-${\kappa}$B Luciferase activity inhibition assay in vitro that polyphenol compound, which is the main ingredient of safflower, has the anti-microbial efficacy to inhibit the growth of acne germs that make troubles for the teenagers or middle aged. Also it aims to evaluate its clinical efficacy on the acne skin, utilizing the facial cleansing cosmetic form of soap sample. This study can contribute to take a major step forward to the development of cosmetic soap for acne in the cosmeceutical industry.
A liquid-based colorimetric assay using a pH indicator was introduced for high-throughput monitoring of lysine decarboxylase activity. The assay is based on the color change of bromocresol purple, measured at 595 nm in liquid reaction mixture, due to an increase of pH by the production of cadaverine. Bromocresol purple was selected as the indicator because it has higher sensitivity than bromothymol blue and pheonol red within a broad range and shows good linearity within the applied pH. We applied this for simple determination of lysine decarboxylase reusability using 96-well plates, and optimization of conditions for enzyme overexpression with different concentrations of IPTG on lysine decarboxylase. This assay is expected to be applied for monitoring and quantifying the liquid-based enzyme reaction in biotransformation of decarboxylase in a high-throughput way.

Fucoidan is a kind of polysaccharides in brown seaweeds. For the past years have been extensively studied due to their numerous biological activities : anticancer, anticoagulant, antithrombotic, anti-inflammatory and antiviral. In this study, we h ave extracted fucoidan from the Korean brown seaweeds and examined it's anticancer activities for employed SV40 DNA replication assay, RPA-ssDNA binding assay of replication protein A(RPA: known as human single-stranded DNA-binding protein essential for DNA rep-lication) and MCF7 cell growth inhibition assay. In addition to, we found that chemically sulfated fucoidan'santicancer activity is more higher than natural and desulfated fucoidan. It seem that fucoidan's sulfate group affect on DNA replication, cause of decrease RPA's DNA binding activity. These results suggests that sulfated fucoidan from Korean brown seaweeds have anticancer activity.

A voltammetric assay of phenol ions was investigated using three electrode systems of graphite pencil working, reference and counter electrodes. Under optimum analytical parameters, square wave stripping working ranges were attained at a mili range of 10~80 mg/L and a micro range of 20~90 ug/L using seawater electrolyte. The developed sensor was applied to tap water and the human body system of a smoker. It was found that the methods can be applied to in vivo fluid or medicinal diagnosis.
12 samples from an assay process chain were submitted to qualitative and quantitative neutron activation analysis for the determination of gold. Gold was detected and quantitatively determined in three samples after a chemical separation consisting of solvent extraction and precipitation steps. Recoveries ranged between 81.0 and 93.6% and results of duplicated determinations were reproducible. Quantitative data were obtained from gamma-spectrometric photopeak-area counting. Interference from fast neutron reactions was negligible.
